Investigation of Effects of Low Ruminal pH Values on Serum Concentrations of Macrominerals, Trace Elements, and Vitamins and Oxidative Status of Dairy Cows

Due to the feeding system (high-concentrate diet) during the early lactation stage, ruminal pH in dairy cows follows a diurnal pattern and can remain below the critical level of 5.5 for extended periods of the day. This study aimed to evaluate the effect of low ruminal pH on blood concentrations of certain macrominerals, trace minerals, and fat-soluble vitamins and on the oxidative status of dairy cows during the first half of lactation. Fifty-three randomly selected lactating Holstein cows were used; blood and ruminal fluid samples were collected from all cows on days 30, 90 and 150 of lactation. Blood samples were obtained via coccygeal venipuncture, while the ruminal fluid was obtained by rumenocentesis and the pH was measured immediately after collection. Using a threshold pH of 5.5, samples were classified as normal (pH > 5.5) or low pH (pH ≤ 5.5). Serum concentrations of Ca, Mg, K, Cr, Mn, Zn, Se, and vitamins A, D3, E, and K were not significantly affected by ruminal pH, either by days in milk or by their interaction (p > 0.05). Plasma malondialdehyde and reduced glutathione followed the same trend (p > 0.05). Copper concentration was significantly higher (p < 0.05), and Fe concentration tended to be higher in cows with low pH compared to those with normal pH (p = 0.052). On day 150 of lactation, Cu, Fe, and Co concentrations were significantly higher in low-pH cows compared to normal-pH cows (p < 0.05). Low ruminal pH is associated with significant changes in serum concentrations of copper, iron, and cobalt but has no significant effect on the oxidative status of the animals or on the serum concentrations of the macro elements and fat-soluble vitamins studied. Silvopastoral systems, though ecologically beneficial, remain underrepresented in the European Union’s Common Agricultural Policy and are seldom studied in Mediterranean contexts. The current study assesses both the environmental and economic aspects of five typical silvopastoral systems in central Greece, encompassing cattle, sheep, and goat farming. A Life Cycle Assessment approach was implemented to quantify greenhouse gas emissions using economic allocation, distributing impacts between milk and meat outputs. Enteric fermentation was the major emission source, accounting for up to 65.14% of total emissions in beef-based systems, while feeding and soil emissions were more prominent in mixed and small ruminant systems. Total farm-level emissions ranged from 60,609 to 273,579 kg CO2eq per year. Economically, only beef-integrated systems achieved an average annual profitability above EUR 20,000 per farm, based on financial data averaged over the last five years (2020–2024) from selected case studies in central Greece, while the remaining systems fell below the national poverty threshold for an average household, underscoring concerns about their economic viability. The findings underline the dual challenges of economic viability and policy neglect, stressing the need for targeted support if these multifunctional systems are to add value to EU climate goals and rural sustainability. Kefiran, the extracellular polysaccharide produced from the Generally Recognized as Safe (GRAS) bacteria in kefir grains, with its well-documented functional and health-promoting properties, constitutes a promising biopolymer with a variety of possible uses. Its compatibility with other biopolymers, such as milk proteins, and its ability to form standalone cryogels allow it to be utilized for the fabrication of films with improved properties for applications in the food and biomedical–pharmaceutical industries. In the present work, the properties of kefiran films were investigated in the presence of milk proteins (sodium caseinate, native and heat-treated whey proteins, and their mixtures), alongside glycerol (as a plasticizer) and cryo-treatment of the film-forming solution prior to drying. A total of 24 kefiran films were fabricated and studied for their physical (thickness, moisture content, water solubility, color parameters and vapor adsorption), mechanical (tensile strength and elongation at break), and optical properties. Milk proteins increased film thickness, solubility and tensile strength and reduced water vapor adsorption. The hygroscopic effect of glycerol was mitigated in the presence of milk proteins and/or the application of cryo-treatment. Glycerol was the most effective at reducing the films’ opacity. Heat treatment of whey proteins proved to be the most effective in increasing film tensile strength, reducing, at the same time, the elongation at break, while sodium caseinates in combination with cryo-treatment resulted in films with high tensile strength and the highest elongation at break. Cryo-treatment, carried out in the present study through freezing followed by gradual thawing of the film-forming solution, proved to be the most effective factor in decreasing film roughness. Based on our results, proper selection of the film-forming solution composition and its treatment prior to drying can result in kefiran–glycerol films with favorable properties for particular applications. A unique line of Holstein cattle has been maintained without selection in Minnesota since 1964. After many generations, unselected cattle produce less milk, but have better reproductive performance and health traits when compared with contemporary cows. Comparisons between this line of unselected Holstein and those under selection provide useful insights that connect selection and complex traits in cattle. Utilizing these unique resources and sequence data, we sought to identify genome changes due to selection. We sequenced 30 unselected and 54 selected Holstein cattle and compared their sequence variants to identify selection signatures. After many years, the two populations showed completely different patterns in their genome-level population structures and linkage disequilibrium. By integrating signals from five different detection methods, we detected consensus selection signatures from at least four methods covering 14,533 SNPs and 155 protein-coding genes. An integrated analysis of selection signatures with gene annotation, pathways, and the cattle QTL database demonstrated that the genomic regions under selection are related to milk productivity, health, and reproductive efficiency. The polygenic nature of these complex traits is evident from hundreds of selection signatures and candidate genes, suggesting that long-term artificial selection has acted on the whole genome rather than a few major genes. In summary, our study identified candidate selection signatures underlying phenotypic differences between unselected and selected Holstein cows and revealed insights into the genetic basis of complex traits in cattle. Açaí (Euterpe oleracea) seeds account for up to 95% of the fruit’s weight and are commonly discarded during pulp processing. Roasted açaí seed extract (RASE) has recently emerged as a caffeine-free coffee substitute, although its composition and functionality remain underexplored. This study characterized commercial açaí seed powder and evaluated the effect of temperature on the recovery of total phenolic content (TPC) in the aqueous extract using a Central Composite Rotatable Design (CCRD). An intermediate extraction condition (6.0 ± 0.5 g 100 mL−1 at 100 °C) was selected, resulting in 21.78 mg GAE/g TPC, 36.23 mg QE/g total flavonoids, and notable antioxidant capacity (FRAP: 183.33 µmol TE/g; DPPH: 23.06 mg TE/g; ABTS: 51.63 mg TE/g; ORAC: 31.46 µmol TE/g). Proton Nuclear Magnetic Resonance (1H NMR) analysis suggested the presence of amino acids, carbohydrates, and organic acids. During in vitro digestion, TPC decreased from 54.31 to 17.48 mg GAE 100 mL−1 when RASE was combined with goat milk. However, higher bioaccessibility was observed with skimmed (33%) and semi-skimmed (35%) cow milk. These findings highlight RASE as a phenolic-rich, antioxidant beverage with functional stability when prepared with boiling water. This is the first study to report the phytochemical profile of RASE and its interactions with different milk types, supporting its potential as a coffee alternative. Full article

Inflammatory processes within the body have been hypothesised to be causative agents of various health complications, including reproductive issues. This study investigates the effects of mastitis on ovarian morphometry and fertility outcomes while also comparing the effectiveness of different synchronisation protocols in affected cows. Healthy multiparous Holstein dairy cows, in their second to fifth lactations and with an average milk yield of approximately 12,000 kg in the preceding lactation, were selected for this study. The average milk yield per lactation was approximately 12,000 kg, with an average daily milk yield of 30 kg. The aim of this study was to determine the impact of mastitis on the reproductive cycle, ovarian morphometry, and function in dairy cows, as well as its effect on synchronisation efficiency. This study focused on both clinically healthy cows and those diagnosed with clinical mastitis postpartum. Three different synchronisation protocols—OvSynch, G7G, and Presynch—were evaluated at 60 days in milk (DIM). A total of 110 cows were included in this study, with 53 being clinically healthy and 57 affected by mastitis. The results indicated that inflammation affects ovarian morphometric parameters such as the area and the number of functional structures. In addition, the G7G protocol was observed to result in higher conception rates in cows suffering from mastitis following the second insemination. This study demonstrated that cows that developed mastitis within the first 30 DIM exhibited higher conception rates after the first insemination compared to those in cows that developed mastitis later in lactation. The role of Enterococcus spp. in food is debated since this group of lactic acid bacteria contains opportunistic pathogenic strains, some of which exhibit a multidrug-resistant profile. In livestock farms, the use of antibiotics is the most common practice to deal with mastitis-causing bacteria. However, the heavy usage and/or misuse of antibiotics has led to the emergence of antibiotic resistance. This study aimed to genetically and phenotypically characterize Enterococcus strains isolated from raw sheep milk. Samples were collected over one year from the bulk tank of a dairy sheep farm and cultured on selective media. Isolates were purified and analyzed by whole-genome sequencing and antimicrobial susceptibility testing. The isolates were divided into clusters and the corresponding species were identified along with their genes related to virulence and antibiotic resistance. The pan-, core- and accessory-genomes of the strains were determined. Finally, the antibiotic-resistant profile of selected strains was examined and associated with their genomic characterization. These findings contribute to a better understanding of Enterococci epidemiology, providing comprehensive profiles of their virulence and resistance genes. The presence of antibiotic-resistant bacteria in raw sheep milk destined for the production of cheese should raise awareness. Full article

Background: Cow’s milk allergy (CMA) is the most common food allergy in children, typically resolving by adolescence. In contrast, the clinical spectrum of allergies to non-cow mammalian milk and their patterns of IgE cross-reactivity are less well documented. Nutritional differences between various mammalian milks may also impact dietary management in milk-allergic patients. Objectives: To characterize clinical features, onset age, and IgE cross-reactivity patterns of non-cow mammalian milk allergies in adult patients seen at a tertiary allergy center, and to compare these findings with published cases. Methods: A retrospective analysis of patients included in the “Extended Laboratory Investigation for Rare Causes of Anaphylaxis study” with mammalian milk allergy was performed using clinical history, skin testing, and serum-specific IgE measurements. Cross-reactivity patterns were assessed in selected cases using immunoblotting, specific IgE inhibition, and basophil activation testing, and compared with published reports of non-cow mammalian milk allergy. Results: In our case series of 22 patients with mammalian milk allergy and 10 healthy control subjects, 3 patients were identified with isolated adult-onset non-cow mammalian milk allergy (n = 1 buffalo milk; n = 2 mare milk), confirmed via immunoblotting and basophil activation testing. Streptavidin-based specific IgE measurement for buffalo cheese was positive in the buffalo milk allergic patient. The literature review identified 82 cases of non-cow mammalian milk allergy. These cases typically showed late onset (mean age 8.6 years; range 1–70 years), severe reactions (CoFAR (Consortium for Food Allergy Research) grade 3 or 4 in 66%, and one fatality), and selective sensitization (affecting sheep and/or goat, camel, mare, buffalo, donkey, or combinations thereof in 56, 10, 5, 5, 4, and 2 cases, respectively). Conclusions: Non-cow mammalian milk allergies are rare but generally present later in life with selective IgE cross-reactivity, differing from the broader cross-reactivity observed in CMA. This selectivity may allow for safe dietary alternatives. These findings underscore the need for improved diagnostics and personalized dietary management in this patient population. Full article

Staphylococcus aureus frequently causes intramammary infections in dairy cows (bovine mastitis), which impair animal welfare, milk yield, and food safety. This study determined the prevalence and genetic diversity of S. aureus in bulk tank milk (BTM) samples from community-based Alpine dairy pastures in Tyrol, a major milk-producing region in Austria. Throughout the 2023 Alpine season (May–September), 60.3% (94/156) of BTM samples tested positive for S. aureus at least once over the course of up to four samplings. A total of 140 isolates collected from the 94 S. aureus-positive community-based Alpine dairy pastures revealed 33 distinct spa types, with t2953 (n = 33), t529 (n = 12), t267 (n = 11), and t024 (n = 10) being the most common. Selected isolates representing the different spa types were characterised by DNA microarray-based genotyping, multi-locus sequence typing (MLST), and antimicrobial susceptibility testing. Isolates with spa types associated with bovine-adapted CC8 (CC8bov/GTB) were identified as the most common subtype, being detected in BTM samples from 35.3% (55/156) of the pastures. This emphasises the high prevalence of this subtype in dairy herds across European Alpine countries. Other common bovine-associated subtypes were also detected, including CC97, CC151, and CC479. While antimicrobial resistance was rare, enterotoxin-producing genes were detected in all CC8bov-associated spa types. Overall, these findings underscore the importance of rigorous hygiene practices in dairy farming, particularly in community-based Alpine dairy pastures, where the risk of transmission is particularly high. It also emphasises the need for continued surveillance and subtyping to improve animal health, ensure food safety, and promote sustainable milk production. Full article

This study analyzes the use of a lightweight image-based deep learning model to classify dairy cows into low-, medium-, and high-milk-yield categories by automatically detecting the udder region of the cow. The implemented model was based on the YOLOv11 architecture, which enables efficient object detection and classification with real-time performance. The model is trained on a public dataset of cow images labeled with 305-day milk yield records. Thresholds were established to define the three yield classes, and a balanced subset of labeled images was selected for training, validation, and testing purposes. To assess the robustness and consistency of the proposed approach, the model was trained 30 times following the same experimental protocol. The system achieves precision, recall, and mean Average Precision (mAP@50) of 0.408 ± 0.044, 0.739 ± 0.095, and 0.492 ± 0.031, respectively, across all classes. The highest precision (0.445 ± 0.055), recall (0.766 ± 0.107), and mAP@50 (0.558 ± 0.036) were observed in the low-yield class. Qualitative analysis revealed that misclassifications mainly occurred near class boundaries, emphasizing the importance of consistent image acquisition conditions. The resulting model was deployed in a mobile application designed to support field-level assessment by non-specialist users. These findings demonstrate the practical feasibility of applying vision-based models to support decision-making in dairy production systems, particularly in settings where traditional data collection methods are unavailable or impractical. The objective of this study was to assess the impacts of milk thistle seed (MTS) and Achyranthes japonica extract (AJE), both individually and in combination, on growth performance, nutrient digestibility, fecal score, fecal gas emissions, and cytokine responses in n = 120 crossbred [(Landrace Yorkshire) × Duroc] weaning piglets with an initial body weight of 6.53 ± 1.24 kg. Pigs were selected based on sex and randomly assigned to one of four dietary treatments for 6 weeks. The experimental diets were as follows: (1) CON—control/basal diet; (2) AJE (CON + 0.10% AJE); (3) MTS (CON + 0.10% MTS); and (4) CMB—combo feed (CON + 0.05% of AJE + 0.05% of MTS). Each treatment consisted of six replicates with five pigs (three ♀ and two barrows ♂) per pen. The incorporation of MTS has the potential to enhance (p < 0.05) the average daily gain in weaning pigs, particularly when used alone or in combination with AJE. But there were no significant effects or adverse effects observed on other growth parameters such as body weight, average daily feed intake, and gain-to-feed ratio. Also, there were no notable changes found in nutrient digestibility, fecal score, fecal gas emissions, or cytokine production. Increasing the shelf-life of fruits and vegetables using edible natural substances after harvest is economically important and can be useful for human health. Postharvest techniques help maintain the quality of edible tissues resulting in extended marketing periods and reduced food waste. The edible coating on perishable commodities is a common technique used by the food industry during the postharvest supply chain. The objective of this research was to study the effect of edible oil to minimize the loss of postharvest physio-chemical and nutritional attributes of bananas. The study selected two banana cultivars (Musa, ‘Cavendish’ and ‘Milk’) to conduct this experiment, and two edible oils (olive oil (Olea europaea) and moringa oil (Moringa peregrina)) were applied as an edible coating under two different storage conditions (15 and 25 °C). The fruit’s physio-chemical properties including weight loss, firmness, color, total soluble solids (TSS), pH, titratable acidity (TA), TSS: TA ratio, and mineral content were assessed. The experiment lasted for 12 days. The physicochemical properties of the banana coated with olive and moringa oils were more controlled than the non-coated (control) banana under both storage temperatures (15 °C and 25 °C). Coated bananas with olive and moringa oils stored at 15 °C resulted in further inhibition in the ripening process. There was a decrease in weight loss, retained color, and firmness, and the changes in chemical parameters were slower in banana fruits during storage in the olive and moringa oil-coated bananas. Minerals were highly retained in coated Cavendish bananas. Overall, the coated samples visually maintained acceptable quality until the final day of storage. Our results indicated that olive and moringa oils in this study have the potential to extend the shelf-life and improve the physico-chemical quality of banana fruits. Full article

In this paper, screen-printed ion-selective electrodes combined with single-walled carbon nanotubes (SWCNTs) and gold nanoparticles (AuNPs) were used to rapidly and accurately measure serum Ca2+ concentration. Due to the susceptibility of cows to hypocalcemia after delivery, this disease can affect the health of cows and reduce milk production. Therefore, the development of an economical and swift detection method holds paramount importance for facilitating early diagnosis and subsequent treatment. In this study, by combining the high electrical conductivity and large surface area of SWCNTs with the strong catalytic activity of AuNPs, a SWCNTs/AuNPs composite with high sensitivity and good stability was prepared, achieving efficient selective recognition and signal conversion of Ca2+. The experimental results indicate that the screen-printed electrode modified with SWCNTs/AuNPs exhibited excellent performance in the determination of Ca2+ concentration. Its linear response range is 10−5.5–10−1 M, covering the normal and pathological concentration range of Ca2+ in cow blood, and the detection limit is far below the clinical detection requirements. In addition, the electrode also has good anti-interference ability and fast response time (about 15 s), showing good performance in the range of 5–45 °C. In practical applications, the combination of the electrode and portable detection equipment can realize the field rapid determination of cow blood Ca2+ concentration. This method is easy to operate, cost-effective, and easy to promote, providing strong technical support for the health management of dairy farms. Full article

Milk is frequently susceptible to contamination by potential pathogens, posing risks to both food safety and public health. Cheesemaking often relies on raw milk, where microbial communities—including Staphylococcus xylosus—can play a dual role: (i) contributing to fermentation and (ii) acting as opportunistic pathogens that can be often present in subclinical mastitis and be subjected to carry over in dairy products. In this study, Staphylococcus xylosus was isolated from raw bovine milk (preclinical mastitis) and identified via matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (Biotyper scores: 1.87–2.19). Its susceptibility to erythromycin and to an essential oil blend composed of Myrtus communis, Salvia officinalis, and Cistus ladanifer was evaluated. The essential oil blend produced inhibition zones ranging from 9 mm to 13.3 mm, indicating moderate antibacterial activity. Further Minimum Inhibitory Concentration analysis revealed that Myrtus communis, Salvia officinalis, and the essential oil blend inhibited Staphylococcus xylosus growth at concentrations between 0.5 and 0.25 percent, while Cistus ladanifer required higher levels (1 to 0.5 percent). These findings suggest that selected essential oils—especially in combination—hold promise as complementary antimicrobial agents in food safety and antimicrobial resistance mitigation efforts. Prediction of Mature Body Weight of Indigenous Camel (Camelus dromedarius) Breeds of Pakistan Using Data Mining Methods

The determination of the live body weight of camels (required for their successful breeding) is a rather difficult task due to the problems with handling and restraining these animals. Therefore, the main aim of this study was to predict the ABW of eight indigenous camel (Camelus dromedarius) breeds of Pakistan (Bravhi, Kachi, Kharani, Kohi, Lassi, Makrani, Pishin, and Rodbari). Selected productive (hair production, milk yield per lactation, and lactation length) and reproductive (age of puberty, age at first breeding, gestation period, dry period, and calving interval) traits served as the predictors. Six data mining methods [classification and regression trees (CARTs), chi-square automatic interaction detector (CHAID), exhaustive CHAID (EXCHAID), multivariate adaptive regression splines (MARSs), MLP, and RBF] were applied for ABW prediction. Additionally, hierarchical cluster analysis with Euclidean distance was performed for the phenotypic characterization of the camel breeds. The highest Pearson correlation coefficient between the observed and predicted values (0.84, p < 0.05) was obtained for MLP, which was also characterized by the lowest root-mean-square error (RMSE) (20.86 kg), standard deviation ratio (SDratio) (0.54), mean absolute percentage error (MAPE) (2.44%), and mean absolute deviation (MAD) (16.45 kg). The most influential predictor for all the models was the camel breed. The applied methods allowed for the moderately accurate prediction of ABW (average R2 equal to 65.0%) and the identification of the most important productive and reproductive traits affecting its value. However, one important limitation of the present study is its relatively small dataset, especially for training the ANN (MLP and RBF). Hence, the obtained preliminary results should be validated on larger datasets in the future. Full article
