Ancient Lineages of the Western and Central Palearctic: Mapping Indicates High Endemism in Mediterranean and Arid Regions

The Palearctic region is characterised by high endemism in the west and east, and a low endemism centre. The endemic lineages occurring at the two ends are largely distinct, and eastern endemics are typically associated with humid climates and forests, representing the start of a continuum from temperate to tropical forest groups and leading to Indo-Malay endemics. In contrast, western Palearctic endemics are typically associated with arid or seasonally dry (Mediterranean) climates and vegetation. Those lineages occurring in the central Palearctic are typically of western origin. Here, we use phylogenetic age (older than 34 million years (My)) to define a list of tetrapod and vascular plant lineages endemic to the western and central Palearctic, map their distributions at the ecoregion scale, and combine these maps to illustrate and understand lineage richness and endemism patterns. Sixty-three ancient lineages were recovered, approximately half of them reptiles, with several herbaceous and shrubby angiosperms, amphibians, and rodents, and single lineages of woody conifers, insectivores, and birds. Overall, we show high lineage richness in the western Mediterranean, eastern Mediterranean, and Iran, with the highest endemism values recorded in the western Mediterranean (southern Iberian Peninsula, southern France). This paints a picture of ancient lineage survival in areas of consistently dry climate since the Eocene, but also in association with persistent water availability (amphibians in the western Mediterranean). The almost complete absence of ancient endemic bird lineages is unusual and perhaps unique among the world’s biogeographic regions. The factors accounting for these patterns include climate since the end of the Eocene, micro-habitats and micro-climates (of mountain terrain), refugia, and patchiness and isolation (of forests). Despite their aridity adaptations, some of the lineages listed here may be tested under anthropogenic climatic change, although some may extend into the eastern Palearctic. We recommend using these lineages as flagships for conservation in the study region, where their uniqueness and antiquity deserve greater recognition. Full article

A new species of Solenopsis (Campanulaceae), S. gutermannii is described and illustrated. It is endemic to Kefalonia island, Greece, where it is very rare and localized in flat clay surfaces that are periodically submerged. This species is a very small annual hygrophyte, characterized by rosulate leaves or with slightly caulescent stems, long-pedunculated flowers, bilabiate corolla, and papillate near the throat. It exhibits close affinities primarily with S. minuta, with which it was previously identified, and additionally with S. antiphonitis, due to similarities in habit and certain flower traits. Nevertheless, several relevant morphological features serve to distinguish it from these species. Its morphology, seed coat and pollen SEM micro-sculpturing, ecology, phenology, distribution, conservation status, and taxonomic relationships are also examined. Full article

Background: Osteoporosis is characterized by the microstructural depletion of bone tissue and decreased bone density, leading to an increased risk of fractures. Cotoneaster wilsonii Nakai, an endemic species of the Korean Peninsula, grows wild in Ulleungdo. In this study, we aimed to investigate the effects of C. wilsonii and its components on osteoporosis. Methods and Results: The alkaline phosphatase (ALP) activity of C. wilsonii extracts and fractions was evaluated in MC3T3-E1 pre-osteoblasts, and the n-hexane fraction (CWH) showed the best properties for ALP activity. The effects of the CWH on bone formation were assessed in MC3T3-E1 cells and ovariectomized mice. Biochemical assays and histological analyses focused on the signaling activation of osteoblast differentiation and osteogenic markers, such as ALP, collagen, and osterix. The CWH significantly activated TGF-β and Wnt signaling, enhancing osteoblast differentiation and bone matrix formation. Notably, CWH treatment improved micro-CT indices, such as femoral bone density, and restored serum osteocalcin levels compared to OVX controls. Conclusions: These results highlight the potential of the C. wilsonii Nakai n-hexane fraction as a promising therapeutic agent for managing osteoporosis. Full article

Over the last two decades, the field of microRNA (miRNA) research has grown significantly. MiRNAs are a class of short, single-stranded, non-coding RNAs that regulate gene expression post-transcriptionally. Thereby, miRNAs regulate various essential biological processes including immunity. Dysregulated miRNAs are associated with various infectious and non-infectious diseases. Recently co-infection with soil-transmitted helminths (STHs) and herpes simplex virus type 2 (HSV-2) has become a focus of study. Both pathogens can profoundly influence host immunity, particularly in under-resourced and co-endemic regions. It is well known that STHs induce immunomodulatory responses that have bystander effects on unrelated conditions. Typically, STHs induce T-helper 2 (Th2) and immunomodulatory responses, which may dampen the proinflammatory T-helper 1 (Th1) immune responses triggered by HSV-2. However, the extent to which STH co-infection influences the host immune response to HSV-2 is not well understood. Moreover, little is known about how miRNAs shape the immune response to STH/HSV-2 co-infection. In this article, we explore the potential influence that STH co-infection may have on host immunity to HSV-2. Because STH and HSV-2 infections are widespread and disproportionately affect vulnerable and impoverished countries, it is important to consider how STHs may impact HSV-2 immunity. Specifically, we explore how miRNAs contribute to both helminth and HSV-2 infections and discuss how miRNAs may mediate STH/HSV-2 co-infections. Insight into miRNA-mediated immune responses may further improve our understanding of the potential impact of STH/HSV-2 co-infections. Full article

Background: Leprosy control remains challenging in Brazil and has been aggravated by the COVID-19 pandemic. Objective: To analyze the impact of the COVID-19 pandemic on the epidemiological scenario of leprosy through the detection rate of new cases, the risk of illness, and the hidden prevalence of leprosy according to high-risk micro-region in Minas Gerais, Brazil. Methods: An ecological study conducted in the health micro-regions of Minas Gerais, using data on new leprosy cases diagnosed between 2015 and 2023. The annual detection rate of new cases, the risk of illness index and the hidden prevalence of leprosy were evaluated. The time trend was evaluated by calculating the annual percentage change (APC) of the detection rate, based on segmented linear regression, considered significant when it showed p < 0.05. Results: The state of Minas Gerais showed a significant negative annual increase between 2015 and 2020 (APC = −7.91; 95%CI −21.76–−1.72), and in 2020 it showed an inflection point, with an annual increase of 9.91 in the period from 2020 to 2023. When evaluating the hidden prevalence of leprosy in Minas Gerais, we observed a reduction in the estimates’ average from 2015–2019 (2.78) to 2020–2023 (2.00). The state as a whole showed an upward trend in the risk of illness, with the average index varying from 0.28 (medium risk) to 0.55 (high risk). Conclusions: The pandemic has had a considerable and heterogeneous impact on the detection of new cases, as well as on the risk of becoming ill and the hidden prevalence of leprosy, with repercussions for the control of the endemic in populations. There is a need to implement public health policies that prioritize the early identification of cases and ensure that vulnerable populations are monitored. Full article

Incarvillea semiretschenskia (B. Fedtsch.) Grierson is listed in the Red Data Book of Kazakhstan as a rare relic, narrowly endemic species of the Shu-Ile low mountains (Kazakhstan). The aim of this research was to advance our knowledge of the ecological conditions of its habitats, the floristic composition of plant communities, and molecular phylogeny, as well as to identify threats to the species’ existence. The ecological conditions of I. semiretschenskia habitats are rocky slopes and intermountain valleys of the low mountains in the altitude range from 812 to 1075 m asl with light chestnut mountain soils of little development, having a light granulometric composition and containing insignificant amounts of organic matter. We revealed 164 species of vascular plants in the communities of I. semiretschenskia, including five endemics and five species from the Red Data Book of Kazakhstan. Anthropogenic factors associated with grazing, fires, and limestone mining were identified as leading to a decrease in the number and density of populations. New insights into the relationship and time of diversification in the genus Incarvillea are gained through the use of several accessions of I. semiretschenskia and I. potaninii Batalin in the phylogenetic study. Preliminary fingerprint analysis shows relatively high genetic variability within populations of I. semiretschenskia. This unique relic species has survived since the Miocene epoch and exists to this day only in the Shu-Ile low mountains. To preserve this rare species, measures are proposed to create plant micro-reserves to provide ex situ collections and ensure future in situ restoration efforts. Full article

Background/Objectives: Leptospirosis is a zoonotic disease that is widely distributed around the world and presents symptoms similar to other febrile illnesses in tropical regions, which complicates clinical diagnosis. This study aimed to evaluate the performance and agreement between serological diagnostic tests for detecting both acute and convalescent human leptospirosis, using the micro agglutination test (MAT) as a reference in an endemic region of the Colombian Caribbean. Methods: A prospective descriptive study was conducted on 275 participants with suspected leptospirosis. Paired serum samples were obtained, and an epidemiological survey was conducted. Using the MAT as the gold standard, we calculated positive and negative predictive values, sensitivity, specificity, and kappa index. A Bayesian latent class model was also used to compare the diagnostic tests. Results: In 223 paired serum samples, the sensitivity values for various stages of the disease ranged between 10.8% to 54.1% in the acute and 6.1% to 66.7% during the convalescent phase compared to the MAT. According to the Bayesian model, sensitivity was 9.5% to 75.3% in the acute phase and 5.7% to 85.3% in the convalescent phase. The Kappa value, an indicator of agreement, was moderate for the IgM ELISA in the acute phase (0.553) and substantial in the convalescent phase (0.692). Conclusions: The MAT was the best confirmatory test in both acute and convalescent phases of leptospirosis. Despite the high specificity of ELISA, 21.62% of participants identified as negative by IgM-ELISA in both phases were subsequently confirmed as positive by the MAT. It is necessary to re-evaluate diagnostic guidelines that do not employ the MAT for confirmation and to enhance the diagnostic and clinical identification of leptospirosis within healthcare institutions and public health laboratories while providing a rapid and reliable test for its implementation. This study aimed to describe the experiences of healthcare personnel and patients in the organization of free surgical campaigns and care for trachomatous trichiasis in the health district of Siguiri in Guinea, including challenges experienced in providing surgical care. This was an explanatory qualitative study conducted in 2022 in the health district of Siguiri. A total of 20 participants were interviewed including patients (n = 7; 35%), community health workers (n = 4; 20%), health services managers, and healthcare providers (n = 8; 40%). Two main data collection technics were used: documentary review and in-depth individual interviews. All interviews were transcribed and manually coded using an Excel extraction spreadsheet. Data were analysed using inductive and deductive approaches. The results showed several organizational, structural, and community challenges that underlined the low surgical coverage of trachomatous trichiasis in the health district of Siguiri. Organizational challenges included the low involvement of local actors in planning activities, the limited timeframe of the campaigns, and the lack of logistics for activities supervision and patients’ transportation to surgery sites. Structural challenges included the inadequacy of health centres to provide surgical services, poor health infrastructures, and sanitation conditions in some areas. Individual challenges included remoteness of surgical sites and costs associated with services provision including medicines. Community challenges included fear of surgery and the coincidence of the campaigns with agricultural and mining activities. The results of the study call on the national neglected tropical disease program and its partners to adopt and promote micro-planning of trachomatous trichiasis surgical activities, with the effective participation of local stakeholders in endemic health districts. They should also envision integrating the management of surgical treatment including costs associated with care (transportation, food, rehabilitation support for patients who have undergone surgery) and complications of surgical procedures for an expansion of the trachomatous trichiasis free surgical care coverage in endemic health districts in Guinea. The yak (Bos grunniens) is a valuable livestock animal endemic to the Qinghai–Tibet Plateau in China with low reproductive rates. Cryptorchidism is one of the primary causes of infertility in male yaks. Compared with normal testes, the tight junctions (TJs) of Sertoli cells (SCs) and the integrity of the blood–testis barrier (BTB) in cryptorchidism are both disrupted. MicroRNAs are hairpin-derived RNAs of about 19–25 nucleotides in length and are involved in a variety of biological processes. Numerous studies have shown the involvement of microRNAs in the reproductive physiology of yak. In this study, we executed RNA sequencing (RNA-seq) to describe the expression profiles of mRNAs and microRNAs in yaks with normal testes and cryptorchidism to identify differentially expressed genes. GO and KEGG analyses were used to identify the biological processes and signaling pathways which the target genes of the differentially expressed microRNAs primarily engaged. It was found that novel-m0230-3p is an important miRNA that significantly differentiates between cryptorchidism and normal testes, and it is down-regulated in cryptorchidism with p < 0.05. Novel-m0230-3p and its target gene CSF1 both significantly contribute to the regulation of cell adhesion and tight junctions. The binding sites of novel-m0230-3p with CSF1 were validated by a dual luciferase reporter system. Then, mimics and inhibitors of novel-m0230-3p were transfected in vitro into SCs, respectively. A further analysis using qRT-PCR, immunofluorescence (IF), and Western blotting confirmed that the expression of cell adhesion and tight-junction-related proteins Occludin and ZO-1 both showed changes. Specifically, both the mRNA and protein expression levels of Occludin and ZO-1 in SCs decreased after transfection with the novel-m0230-3p mimics, while they increased after transfection with the inhibitors, with p < 0.05. These were achieved via the CSF1/CSF1R/Ras signaling pathway. In summary, our findings indicate a negative miRNA-mRNA regulatory network involving the CSF1/CSF1R/Ras signaling pathway in yak SCs. These results provide new insights into the molecular mechanisms of CSF1 and suggest that novel-m0230-3p and its target protein CSF1 could be used as potential therapeutic targets for yak cryptorchidism. Madagascar, as one of the global biodiversity hotspots, hosts numerous unique terrestrial mammal species that need urgent protection. To identify priority species for conservation, an updated list of terrestrial Malagasy mammal species was compiled, including their threat status, distribution, endemism level, Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) listing, and Evolutionarily Distinct and Globally Endangered (EDGE) score. An overview of these species kept in zoos worldwide was created using the Zoological Information Management System and Zootierliste to assess ex situ conservation efforts. Nearly 60% of the 212 native terrestrial mammal species are threatened with extinction, with 18% being regional endemics, 39% microendemics, and 42% endemics. The majority of these species (92%) occur within protected areas. About half of Madagascar’s mammals are listed under CITES, and less than half have an EDGE score. Only 34 species are kept in zoos globally, with 26 Red-Listed as threatened. Nine out of seventeen families are not represented in zoos. A total of 1545 institutions, primarily in Europe and North America, keep Malagasy mammal species, with successful reproduction reported for 28 species in the last 12 months, 23 of them listed as threatened on the IUCN Red List. To maximize conservation, we recommend reallocating resources towards priority species and implementing concerted ex situ and in situ actions as proposed by the IUCN’s One Plan Approach. Full article

Millions of individuals throughout the world suffer from lymphatic filariasis (LF), which is a morbid disease caused by Wuchereria bancrofti, Brugia malayi, and Brugia timori. These infections belong to tissue-invading nematodes and are one of the major neglected tropical diseases that often result in permanent and enduring disability among individuals in endemic regions. Due to combination therapy, LF eradication has drastically decreased infections globally. The development of blood micro-sampling techniques allowing precise quantitation of drugs in blood would facilitate pharmacokinetic (PK) studies in remote populations. Therefore, an LC-MS/MS bioanalytical method was utilized to analyze albendazole (ABZ), albendazole sulfone (ABZ-ON), albendazole sulfoxide (ABZ-OX), and probenecid (PR) in plasma and dried blood spots. Solid-phase extraction was utilized to extract the analyte from both plasma and blood-spiked DBS. Analytes of interest were eluted with a gradient mobile system using 0.05% formic acid in water (A) and 0.05% formic acid in methanol (B) and separated using a reversed-phase Acquity ®BEH C18 UPLC column (100 × 2.1 mm, 1.7 µm). Precision and accuracy at each QC level were within the acceptable limit, i.e., ±15% for all analytes in both the matrices. Tests for stability under laboratory and storage conditions indicated that no notable changes were observed for plasma and DBS. The LC-MS/MS method demonstrated its capability to consistently identify all target analytes (ABZ, ABZ-ON, ABZ-OX, and PR) at low concentrations, even at the small specimen volumes obtained from DBS cards. This confirms the efficacy and durability of DBS cards as a micro-sampling technique. Moreover, it enhances collection efforts for therapeutic drug monitoring in remote locations for patients infected with lymphatic filariasis. Full article

The extraction of museum DNA from a unique collection of samples of the “Crocidura pergrisea” species complex, which comprises local endemics of Central and West Asia, allowed us to determine their inter- and intragroup relationships. The first step of this study was the re-evaluation of heavily damaged type specimens of C. armenica via a microcomputed-tomography-based cybertaxonomic approach (CTtax), which enabled a precise description of the species’ morphology; three-dimensional models of the cybertypes were made available through the MorphoBank Repository. We developed the “AProMaDesU” pipeline on the basis of five requirements for micro-CT-based cyber-datasets in relation to mammalian collections. Our second step was a combination of several meticulous approaches to morphological investigation against a background of a cytb-based phylogeny, which helped us to make a taxonomic decision about the status of species of the “pergrisea” group, e.g., C. arispa, C. armenica, and C. serezkyensis, when the morphological results were partly incongruent with the molecular phylogeny. Nevertheless, under two assumptions, our findings preserved a separate species-level status of C. serezkyensis and C. arispa. In addition, we restored the species-level status of C. armenica. This taxonomic decision is based on our morphospace analysis, which revealed unique craniomandibular shape transformations within the rocky shrews that helped them with the transition to a new area of morphospace/trophic niches and consequently separated them from the other analyzed Crocidura groups. A novel species, Gymnadenia winkeliana, has been identified in the Bucegi Natural Park ROSCI0013, located in the Southern Carpathians of Central Romania. Two moderately sized populations of Gymnadenia winkeliana, totalling 120–140 individuals, were discovered inhabiting the alpine grasslands of the park, situated 2.000 m above sea level. To describe this newly found population as comprehensively as possible, 44 vegetative and floral organs/organ parts were directly studied and measured from living plants. Special attention was focused on the characteristics that proved to have taxonomic significance, particularly those involving distinctive details in the morphology of the leaves, perianth, labellum and gynostemium. A total of 223 characteristics were analysed encompassing the morphology of every organ of the plant, cytology and breeding system. Furthermore, comprehensive taxonomic treatment and description, accompanied by colour photographs illustrating the holotype, are provided. Voucher specimens were deposited at the Herbarium of the University of Agriculture and Veterinary Medicine, Bucharest (USAMVB Herbarium barcode: 40102, NEA); Gymnadenia winkeliana, a (micro)endemic species, is characterized as a putative allogamous, facultatively apomict that significantly differs from other Gymnadenia R.Br. species found in Romania. Notably, it distinguishes itself through its smaller habitus (reaching heights of up to 8–10 cm), its two-coloured, rounded/hemispherical inflorescence displaying a gradient of pink hues in an acropetal fashion (ranging from whitish-pink at the base to vivid-pink at the topmost flowers), and its limited distribution in high-altitude areas, encompassing approximately 8–10 km2 in the central area of the Bucegi Natural Park. This species has been under observation since 2005, with observed population numbers showing a significant increase over time, from ca. 50–55 (counted at the time of its discovery) to 120–140 individuals (counted in June 2023). Additionally, comprehensive information regarding the habitat, ecology, phenology and IUCN conservation assessments of Gymnadenia winkeliana are provided, including maps illustrating its distribution. The mountaintops of eastern Brazil harbor the highest rates of plant endemism in South America. However, local biodiversity faces constant threats due to habitat loss and mining activities. About 89 rare and endangered species are exclusive to this region, including the threatened species Arthrocereus glaziovii. This study aims to evaluate the potential distribution of A. glaziovii based on abiotic variables and soil elements and to characterize the distribution of mineral titles that may restrict the species’ occurrence areas. We used the Bioclim, Domain, MaxEnt, GLM, and Random Forest algorithms to model this ecological niche under future climatic scenarios, in addition to modeling the layers of mineral titles corresponding to areas already mined and those slated for future mining projects. Our predictions indicate an expansion in the future distribution of A. glaziovii. Nevertheless, the future predicted occurrence areas of the species are already compromised due to mining. According to our findings, we emphasize the looming threat of the predicted extinction of this species. Therefore, implementing conservation strategies to ensure the survival of A. glaziovii is imperative. Full article

Medicinal and Aromatic Plants (MAPs) play a critical role in providing ecosystem services through their provision of herbal remedies, food and natural skin care products, their integration into local economies, and maintaining pollinators’ diversity and populations and ecosystem functioning. Mountainous regions, such as Chelmos-Vouraikos National Park (CVNP), represent unique reservoirs of endemic MAP diversity that require conservation prioritisation. This study aims to provide insights into the sustainable management of MAPs, contributing to efforts to protect Mediterranean biodiversity amid the dual challenges of climate and land-use change, using a suite of macroecological modelling techniques. Following a Species Distribution Modelling framework, we investigated the vulnerability of endemic and non-endemic MAPs to climate and land-use changes. We examined the potential shifts in MAP diversity, distribution, and conservation hotspots within the CVNP. Our results revealed species-specific responses, with endemic taxa facing severe range contractions and non-endemic taxa initially expanding but eventually declining, particularly under land-use change scenarios. Local biodiversity hotspots are projected to shift altitudinally, with considerable area losses in the coming decades and elevated species turnover predicted throughout the CVNP, leading to biotic homogenization. Climate and land-use changes jointly threaten MAP diversity, calling for adaptive conservation strategies, thus highlighting the importance of proactive measures, such as awareness raising, establishing plant micro-reserves, assisted translocation, and promoting sustainable harvesting to protect these species within the CVNP. Our study offers vital insights for managing biodiversity hotspots amid global change pressures, stressing the need to integrate ecological and socioeconomic factors. Full article
