Rhizopogon, a globally distributed genus of edible ectomycorrhizal fungi, is the focus of this study. This study describes a new species, Rhizopogon qujingensis sp. nov., along with two newly recorded species of the genus Rhizopogon from China. The new species is distinguished from other known Rhizopogon species by its distinctly phylogenetic position and unique morphological characteristics, the species is characterized by comparatively smaller basidiomata, a peridium that exhibits yellow tinting after injury and contains red-brown pigment, along with smaller, narrowly oblong to elongate spores. Furthermore, R. rubescens and R. evadens are recorded for the first time in China, with their taxonomic status confirmed through integrated morphological and molecular analysis, thereby extending their known geographical distribution. Phylogenetic analysis was conducted to elucidate the relationships between Chinese Rhizopogon species and other taxa within the genus. This research contributes essential data for advancing the understanding of global Rhizopogon species diversity and biogeographic patterns. Full article

This study presents a comprehensive approach to risk management and planned preventative maintenance (PPM) for cultural heritage, focusing on the hypogeum archaeological site beneath the Chamber of Commerce in Rimini, Italy. Hypogeal environments pose unique conservation challenges due to their microclimates, biological threats, and structural vulnerabilities. Applying the ABC Method—developed by ICCROM and CCI—this research systematically identifies, analyzes, and prioritizes risks associated with agents of risks. The methodology was complemented by the Nara Grid to assess the site’s authenticity and cultural value, aiding in the delineation of risk areas and informing strategic conservation priorities. The study identifies efflorescence formation, flooding risks, and lack of management guidelines as extreme threats, proposing tailored treatments and practical interventions across multiple layers of control. Through environmental monitoring, empirical analysis, and a multidisciplinary framework, the research offers a replicable model for sustainable conservation and preventive heritage management in similar subterranean contexts. Background: Conventional practices for the cultivation of “Cipollotto Nocerino” spring onion are mainly based on growers’ experience, and up to 250 kg/ha for N is commonly furnished among growing cycles. Facing the issue of reduced availability of natural resources for crop production (for example mineral resources), we investigated the optimization of the productivity. Methods: In our research, we tested the use of depletion nutrient supply strategy (CAL-FERT®) to enhance fertilization in accordance with the principle of sustainable agriculture included in the Farm to Fork strategy. In our study, besides the common initial fertilization, three different strategies for cover fertilizations have been elaborated with the support of CAL-FERT® software. The treatments were as follows: (i) commercial standard fertilization as control (named CF); (ii) fertilization equivalent to 50% of the N applied in the control (named F-50); (iii) fertilization corresponding to 25% of the N applied in the control (named F-25); and (iv) strongly reduced fertilization compared to the control (named F-0). The parameters investigated included the following: plant height, yield, SPAD index, nitrogen use efficiency, dry matter, soluble solid content, and pyruvate contents in bulbs and leaves. Nitrogen content was also analyzed for both hypogeous and epigeous apparatuses. Results: Among the most interesting vegetative results, plant height and SPAD readings were reduced only by the extreme treatment F-0 compared with the other treatments at 104 days after planting. Regarding qualitative and productive parameters, the treatments F-50 and F-25 showed the highest yield without prejudging Soluble Solid Content and reducing pungency. Conclusion: In nutritional experiments, onion could be considered as a crop model to investigate quality in vegetables due to its consumption as fresh product and for its particular response, in terms of yield and quality, to fertilization. The use of simulation software can support the identification of strategies to reduce the nutrient supply without any detrimental effect on yield and other vegetative and qualitative parameters in onion crops. Hungary has some of the best-researched hypogeous fungal flora in Europe, with a large number of genera and species already having been discovered in this country. In this study, we performed morphological and molecular analysis of unidentified hypogeous fungi samples collected from Hungary. We confirmed that they belong to the hypogeous earthstar species Geastrum nadalii (Paz et al., 2024), marking the first report of this species in this country and in a continental climate. We also assume that the habitat preference of this mushroom species is similar to the habitat preference of Mattirolomyces terfezioides, with both occurring in planted non-native Robinia pseudoacacia forests, suggesting that these are secondary habitats for these species. We also conclude that this Mediterranean species has appeared only recently in the Hungarian mycota. Full article

For decades, in historical research, archeological vestiges have been linked with geomancy and cults of the mythical ancestors of a group of the population. This is particularly true in Eastern Asia and especially in China, Korea, and Japan. A fundamental problem of Japanese archeology is that few of the remnants were realized in stone. One of the most important parts of archeological sciences is the study of Necropolises or ancient interments. From the 1970s onwards, in the relatively “new” and promising land of Hokkaido, cemeteries were built with the concept of landscape in mind; this is also due to the lavish vegetation features of this northernmost island of Japan. In the case of the Takino cemetery on the plains of Sapporo, Hokkaido, whose construction began in 1982, solemnity and religiousness were incorporated by producing exact stone replicas of famous funerary landmarks from antiquity as such materials were inexistent in the Nipponese Isles. This trend to grant eternity included traditional Buddhist funereal monuments like the Stupa, Seokguram grotto, and Kamakura sites, but at a certain and exuberant point, under the influence of Isamu Noguchi, it reached Stonehenge in England and the Moai from Easter Island in Polynesia (being after all located in a remote isle of the Pacific Ocean). In this article we will outline such process of generation and overall conception, analyzing the inclusion and architectural assembly of the different compounds and the recent and extraordinary additions projected and built by the celebrity architect Tadao Ando. We expect, in this manner, to facilitate the comprehension of the significance of venerable landscape sublimated through archeology for the Nipponese modern civilization. Full article

Ectomycorrhizal fungi of the genus Tuber (Ascomycota) produce hypogeous ascomata commonly known as truffles. Despite their high ecological and economic importance, a considerable gap of knowledge exists concerning the diversity of Tuber species in the eastern Mediterranean region. In the frame of this study, more than 200 Tuber collections, originating from various regions of Greece, were examined. A new species to science, i.e., Tuber leptodermum, is formally described. Tuber leptodermum is grouped in the Maculatum clade, as revealed by the ITS and LSU rDNA concatenated phylogenetic tree, and appears as sister to T. foetidum. In addition, T. leptodermum exhibits distinct morphoanatomic features: it produces medium-sized, dark-brown ascomata with a thin pseudoparenchymatous peridium, composed of globose-to-angular cells and forms one-to-four-spored asci containing reticulate–alveolate, ellipsoid ascospores with broad meshes. Thirty other phylogenetic species are identified: seven of them (i.e., T. anniae, T. buendiae, T. conchae, T. dryophilum, T. monosporum, T. regianum and T. zambonelliae) constitute new records for the Greek mycobiota, while the presence of five other species is molecularly confirmed for the first time. Moreover, the existence of ten undescribed phylogenetic species is revealed, six of which are reported for the first time in Greece. Several taxonomic and phylogenetic issues and discrepancies in the genus Tuber are discussed in relation to the new findings. Full article

This research is focused on the laboratory study of salt crystallization inhibitor products as new materials for conservation treatments which can be applied to mortars and painted plasters; as is well known, salt crystallization is one of the most frequent causes of decay processes on decorated architectural surfaces in a wide range of environments. Specifically, the study targets the field of the preventive conservation of mural paintings within rupestrian heritage sites. For the first time, systematic investigations were performed on mock-ups made of plaster painted with two different pigments: yellow ochre and carbon black. Two types of phosphonate inhibitors, PBTC (2-phosphonobutane-1,2,4-tricarboxylic acid) and ATMP (aminotris (methylene phosphonic acid)), were chosen and applied at two different concentrations. Given the limited literature available, and the presence of pigments potentially sensitive to treatment with salt inhibitors, preliminary tests were required. Their effects on the chromatic features of the pigments were evaluated visually and using colorimetry. The changes in the behaviour of water circulation in the mortar resulting from the treatments were evaluated through water vapour permeability and absorption tests. Accelerated crystallization experiments were carried out to assess how inhibitors could influence the growth of salts and the resulting material damage. The latter was carried out by employing sodium sulphate and calcium sulphate solutions, quantifying the damage to the specimens through material loss in weight and the percentage of painted surface loss. Based on the overall results, the product with the best performance was identified was ATMP 0.1% (by volume) in deionized water. The obtained results show that salt inhibitor treatments are promising for in situ application and could represent an innovative approach to promote the sustainable conservation of mural painting, particularly those located in hypogeal contexts, where the salt supply cannot be removed and slowing the growth of salts and/or changing their crystalline habitus may be effective in limiting their damage. Truffles of the Tuber genus (Pezizales, Ascomycetes) are among the most valuable and expensive foods, but their shelf life is limited to 7–10 days when stored at 4 °C. Alternative preservation methods have been proposed to extend their shelf life, though they may alter certain quality parameters. Recently, a hypogeal display case equipped with an ultrasonic humidity system (HDC) was developed, extending the shelf life to 2–3 weeks, depending on the truffle species. This study assesses the efficacy of HDC in preserving Tuber melanosporum and Tuber borchii ascomata over 16 days, using quantitative magnetic resonance imaging (QMRI) to monitor water content and other parameters. Sixteen T. melanosporum and six T. borchii ascomata were stored at 4 °C in an HDC or a static fridge (SF) as controls. QMRI confirmed that T. borchii has a shorter shelf life than T. melanosporum under all conditions. HDC reduced the rate of shrinkage, water, and mass loss in both species. Additionally, the Apparent Diffusion Coefficient (ADC), longitudinal relaxation time (T1), and transverse relaxation time (T2), which reflect molecular changes, decreased more slowly in HDC than SF. QMRI proves useful for studying water-rich samples and assessing truffle preservation technologies. Further optimization of this method for industrial use is needed. Full article

Imagining life before the advent of modern medical treatments is challenging. Today, congenital dysplasia is typically diagnosed within the first months of a child’s life, allowing for timely intervention. In the past, however, this condition often went unrecognized and untreated, as evidenced by archaeological findings that document the presence of congenital dysplasia persisting into adulthood. We present the case of the individual recovered from the hypogeal cemetery of Santa Maria Maggiore in Vercelli, Italy, a funerary context dated from the 18th to the 19th century. Using macroscopic and radiographic analyses, various morphological irregularities were identified, consistent with the characteristics of developmental hip dysplasia. The skeletal remains identified as FU12 SU151 include a right os coxa and femur, belonging to an adult female. The femur features a 90-degree femoral head angle and a shortened neck with nodules. The acetabulum shows significant morphological changes, including a triangular shape and absence of lunate surfaces, deviating from the normal structure for femoral articulation. CT scans revealed a void within the acetabulum, indicating an absence of material. Despite preservation challenges that restrict the identification of definitive signs, our findings offer valuable insights into possible developmental dysplasia in historic skeletal remains. This research provides insights into the impact of untreated congenital conditions on past populations, underscoring the importance of preserving and studying such remains to enhance our understanding of historical health issues. Seven auxin-producing endophytic bacterial strains (Azospirillum spp., Methylobacterium symbioticum, Bacillus spp.), and two different combinations of these strains were used to verify their influence on tomato during germination and development in hydroponic conditions where, as a novelty for Canestrino di Lucca cultivar, endophytic bacteria were inoculated. To emphasize the presence of bacterial auxins in roots and stems of seedlings, both in situ staining qualitative assessment and quantitative analysis were carried out. Moreover, hypogeal and epigeal growth of the plantlets were measured, and correlation analyses were conducted to examine the relationship between the amount of indolacetic acid (IAA) produced by the bacterial strains and root and stem parameters. Plantlets treated with microbial inoculants showed a significant increase in the survival rate compared to the control treatment. The best results as IAA producers were from Azospirillum baldaniorum Sp245 and A. brasilense Cd, which also induced significant root growth. On the other hand, Bacillus amyloliquefaciens and B. licheniformis induced the best rates in stem growth. These findings highlight the potential for using endophytic bacterial strains in a hydroponic co-cultivation system that enables inoculating plantlets, at an early stage of growth (5 days old). Tuber aestivum, commonly known as the summer truffle, is typically found in various parts of Europe where it grows naturally. However, its presence in Portugal was not confirmed until now. The first fruit bodies were collected in April 2024 at stone pine stands (Alenquer and Arruda dos Vinhos, Lisbon) and in June at holm oak stands (Salir, Faro). These specimens are characterized by hypogeous, subglobose, black ascomata with a peridium surface covered with pyramidal warts. Ascopores are subglobose-to-broadly ellipsoid, distinctively ornamented, usually 1–6 per asci. According to the results of the internal transcribed spacer (ITS) rDNA sequence analysis, these specimens form a well-supported group within the Aestivum clade, with T. aestivum being the closest phylogenetic taxon. This remarkable discovery opens up new opportunities for truffle exploitation in Portugal thanks to the summer truffle’s gastronomical value and high market prices. Full article

Hypogea are natural or artificial spaces located underground often of great interest from an anthropological, archeological, religious, artistic, or historic point of view. Due to their features, these environments usually present conservative problems and biological colonization could be considered as one of the main threats. The present three-year study was carried out by specialists of the Central Institute for Restoration of Rome (ICR) in the hypogeous site preserved in the Roman Archeological Museum of Positano (Positano MAR) and focused on characterizing biological alterations present on the mural paintings; setting up efficient strategies and protocols for biodeterioration control; and monitoring the efficacy of direct and indirect interventions. Patinas with different morphologies were analyzed through microscopic observations, cultural analyses and next-generation sequencing. The results proved that the alterations comprised a great variety of microorganisms forming very distinct communities, differently distributed over space and time. The main taxa represented were bacteria of phyla Pseudomonadota and Actinomycetota, fungi belonging to the genus Fusarium and Gliocladium, and algae of the genus Chlorococcum. Preservation protocols were set up considering the alterations’ composition and included the application of biocides, limiting daily temperature changes, decreasing illuminance values on painted surfaces, and the screening of natural light sources. Fungi of the genus Tuber are famous for their hypogeous ascomata (truffles), many of which possess noteworthy organoleptic properties. T. aestivum shows a wide geographic distribution, has many plant symbionts and is well adapted to various climatic conditions. In this study, five Quercus taxa native to Greece (i.e., Q. coccifera, Q. ilex, Q. ithaburensis subsp. macrolepis, Q. pubescens and Q. trojana subsp. trojana) were inoculated with spore suspensions obtained from a single ascoma of T. aestivum. The fungal colonization of oak roots was evaluated at three, seven and 12 months after inoculation; the respective colonization rates for each time period were as follows: low to medium (17–41%) for Q. pubescens, Q. ithaburensis subsp. macrolepis and Q. trojana subsp. trojana, medium to relatively high (58–80%) for Q. ithaburensis subsp. macrolepis, Q. ilex, Q. pubescens and Q. trojana subsp. trojana, and medium to high (45–87%) for all oak species examined. Positive correlations were assessed between the number of colonized root tips and the total root tips number, but no significant differences were detected between the inoculated plants and the respective control as regards plant growth. The ectomycorrhizae formed by T. aestivum with Q. ithaburensis subsp. macrolepis and Q. trojana subsp. trojana are described for the first time. The outcome of the study evidences the feasibility of generating the seedlings of various indigenous oak species (covering a large range of diverse habitats) successfully inoculated with autochthonous truffles to be readily used for cultivation purposes. Although only a few species of Tuber account for the major truffle sales volume, many species that are not considered delicacies are finding their way to the market, especially in regions where the traditionally appreciated ones do not occur. This is the case for whitish truffles. Specimens of whitish truffles were collected in pecan (Carya illinoinensis) orchards in Uruguay in October 2021. Morphological and molecular methods were used to characterize and assess their identity as Tuber maculatum Vittad. An SPME extraction of volatile compounds and GC–MS analyses were performed to characterize the aromatic profile of these specimens and evaluate their potential applications. Among the 60 VOCs detected, 3-octenone (mushroom odor), 3-octanol (moss, nut, mushroom odor), and 2H-pyran-2-one (no odor), followed by octen-1-ol-acetate (no odor) and 2-undecanone (orange, fresh, green odor) were the major compounds in T. maculatum fruiting bodies. The attributes of exotic edible mushrooms of commercial value in the region are highlighted. In particular, this work emphasizes the characteristics of truffles as a byproduct of pecan cultivation. Seed Propagation of Carapa amorphocarpa W. Palacios Using Various Treatments of Substrates and Mechanical Seed Scarification in a Nursery in the Andean Area of Northwestern Ecuador

Carapa amorphocarpa W. Palacios is a forest species known solely from Cerro Golondrinas in northwestern Ecuador. The species is threatened due to illegal exploitation of its wood and the encroachment of the agricultural frontier. Although information on its ecology and forestry was presented in 2012, there is still very little information available. This study investigated the effects of various pre-germinative treatments on the seed germination and early growth of Carapa amorphocarpa in a nursery. The treatments included combinations of substrates (forest soil alone and forest soil mixed with sand), along with pre-germinative mechanical seed scarification. Through these treatments, the seeds’ germination capacity, growth potential, and survival rates were evaluated in a nursery. Seed germination was found to be cryptocotylar hypogeal, commencing at 41 days and achieving an average of 83.13%. Survival of germinated seedlings was 99.23% at 60 days after germination. Furthermore, a high degree of correlation in growth was observed between basal diameter and plant height (R = 0.94). The mean values for the plants’ basal diameter and total height were 0.91 cm and 50.48 cm, respectively, at 60 days. Plant form was straight with excellent health. These insights offer the potential to enhance species populations, mitigate threat risks, and in the long term, foster sustainable use of Carapa amorphocarpa genetic resources. Moreover, this seed propagation information can be leveraged for other species within the genus exploited for oil extraction from their seeds, thereby broadening knowledge and facilitating sustainable forest resource management.
