The potato (Solanum tuberosum L.) ranks among the most consumed agricultural products globally and is nutrient-rich. Exposure of potato tubers’ epidermal and subcutaneous tissues to light results in greening and the production of neurotoxic steroidal glycoalkaloids, which significantly reduces tuber quality, increases food safety risks, and leads to rejection by consumers and the processing industry. This review conducts an in-depth analysis of three key aspects: (1) the differences in various light conditions, namely, the disparities in light sources, wavelengths, intensities, and durations; (2) the characteristics of potato cultivars resistant to greening; and (3) the molecular mechanisms of light-induced biosynthesis of chlorophyll and steroidal glycoalkaloids (SGAs). This review is expected to provide technical support for potato food safety measures and a theoretical foundation for the molecular breeding of green-resistant potato varieties. Full article

Potato peels (PPs) represent a significant agro-industrial by-product with notable potential for valorization due to their rich composition of bioactive compounds, including phenolics, glycoalkaloids, dietary fiber, and essential minerals. This review explores the functional applications of PPs in the food industry by examining their chemical profile, extraction methods, and biological activities. Phenolic compounds, mainly chlorogenic acid and its derivatives, are the most abundant bioactives and contribute to antioxidant and anti-inflammatory properties. Glycoalkaloids, such as α-solanine and α-chaconine, exhibit antimicrobial activity but require careful monitoring due to their potential toxicity, although recent evidence suggests that controlled doses may provide health benefits. The choice of extraction technique influences the recovery of these compounds, with ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE) proving to be efficient and environmentally friendly alternatives to conventional methods. The incorporation of PP-derived ingredients into food formulations, including cereal, dairy, meat, and fish products, as well as vegetable oils, has shown promising results in the improvement of nutritional quality, oxidative stability and functional properties. However, challenges remain with regard to the standardization of PP composition, bioavailability of bioactive compounds and their stability within food matrices. Advancing research on PPs will not only contribute to circular economy goals but also provide innovative solutions for the food industry, reinforcing the link between sustainability and human health. Wireworms (Agriotes spp., Coleoptera, Elateridae) are a major threat to potatoes, as are the current commercial standards for assessing potato damage. To reduce wireworm impacts on potato crops and comply with IPM legislation, we started research to assess the potential for new Italian 4x-breeding clones to reduce wireworm feeding on daughter tubers. Two sets of trials were carried out over a six-year period (2018–2023): in-field and in semi-natural conditions, with pots used to introduce a set number of reared wireworms. In the field trials, the varieties were planted in 4.8 × 9–12 m plots in a randomized-block layout with at least three replications. The same wireworm damage assessment was used for both sets of trials. The assessment involved counting all the erosions/scars caused by wireworm feeding activity. The prevalent wireworm species studied was Agriotes sordidus. Both sets of trials showed that some 4x-breeding clones were tolerant to wireworm attacks. The percentages of tubers damaged (any symptom) or with at least one ordinary/large hole was up to five times lower than in the commercial varieties. Glycoalkaloids and the caffeic acid content in tubers are considered to be the main cause of lower appetibility to wireworms. Full article

Deciphering the genetic mechanisms underlying key agronomic and quality traits in potato (Solanum tuberosum L.) is essential for advancing varietal improvement. Phenotypic instability in early clonal generations and inbreeding depression, coupled with the complexity of tetrasomic inheritance, pose significant challenges in constructing mapping populations for the genetic dissection of complex traits. Genome-wide association studies (GWASs) offer an efficient method to establish trait–genome associations by analyzing genetic recombination and mutation events in natural populations. This review systematically examines the application of GWASs in identifying agronomic traits in potato, such as plant architecture, yield components, tuber shape, and resistance to early and late blight and nematodes, as well as quality traits including dry matter, starch, and glycoalkaloid content. Some key chromosomal hotspots identified through GWASs include chromosome 5 associated with tuber yield, starch content, and late blight resistance; chromosome 4 and 10 associations with tuber shape and starch content; chromosomes 1, 9, and 11 associated with plant height, tuber number, glycoalkaloid content, and pest resistance. It elucidates the advantages and limitations of GWASs for genetic loci identification in this autotetraploid crop, providing theoretical insights and a reference framework for the precise localization of key genetic loci and the discovery of underlying genes using GWASs. Glycoalkaloids (TGAs, total glycoalkaloids), toxic secondary metabolites, are found in potatoes (110–335 mg·kg−1 DW), mainly in the peel. Colorful, unpeeled potatoes are an innovative raw material for the production of snacks which are poorly tested in terms of their glycoalkaloid content. Third-generation snacks and French fries made from red-fleshed Mulberry Beauty (MB) and purple-fleshed Double Fun (DF) potatoes were produced with the use of 1% solutions of ascorbic, citric, lactic, malic, and tartaric acids to stabilize the structure of anthocyanins in the raw material and maintain their color in obtained products. The influence of the type of acid and thermal processes, like frying, microwaving, and baking, on the content of glycoalkaloids in ready-made products was examined. Only 0.45–1.26 mg·100 g−1 of TGA was found in pellet snacks and 1.32–1.71 mg·100 g−1 in French fries. Soaking blanched potatoes in organic acid solution reduced the α-chaconine content by 91–97% in snacks and by 57–93% in French fries in relation to the raw material to the greatest extent after the use of malic acid and the DF variety. The effect of lactic and citric acid was also beneficial, especially in the production of baked French fries from MB potatoes. Full article

Solanum is the largest genus within the Solanaceae family and has garnered considerable attention in chemical and biological investigations over the past 30 years. In this context, lobeira or “fruta-do-lobo” (Solanum lycocarpum St. Hill), a species predominantly found in the Brazilian Cerrado, stands out. Beyond the interesting nutritional composition of the fruits, various parts of the lobeira plant have been used in folk medicine as hypoglycemic, sedative, diuretic, antiepileptic, and antispasmodic agents. These health-beneficial effects have been correlated with various bioactive compounds found in the plant, particularly alkaloids. In this review, we summarize the alkaloid composition of the lobeira plant and its biological activities that have been reported in the scientific literature in the last decades. The compiled data showed that lobeira plants and fruits contain a wide range of alkaloids, with steroidal glycoalkaloid solamargine and solasonine being the major ones. These alkaloids, but not limited to them, contribute to different biological activities verified in alkaloid-rich extracts/fractions from the lobeira, including antioxidant, anti-inflammatory, anticancer, antigenotoxic, antidiabetic, antinociceptive, and antiparasitic effects. Despite the encouraging results, additional research, especially toxicological, pre-clinical, and clinical trials, is essential to validate these human health benefits and ensure consumers’ safety and well-being. While numerous Fabaceae seeds are a good nutritional source of high-quality protein, the use of some species is hampered by toxic effects caused by exposure to metabolites that accumulate in the seeds. One such species is the faba or broad bean (Vicia faba L.), which accumulates vicine and convicine. These two glycoalkaloids cause favism, the breakdown of red blood cells in persons with a glucose-6-phosphate dehydrogenase deficiency. Because this is the most common enzyme deficiency worldwide, faba bean breeding efforts have focused on developing cultivars with low levels of these alkaloids. Consequently, quantification methods have been developed; however, they quantify vicine and convicine only and not the derivatives of these compounds that potentially generate the same bio-active molecules. Based on the recognition of previously unknown (con)vicine-containing compounds, we screened the fragmentation spectra of LC-MS/MS data from five faba bean cultivars using the characteristic fragments generated by (con)vicine. This resulted in the recognition of more than a hundred derivatives, of which 89 were tentatively identified. (Con)vicine was mainly derivatized through the addition of sugars, hydroxycinnamic acids, and dicarboxylic acids, with a group of compounds composed of two (con)vicine residues linked by dicarboxyl fatty acids. In general, the abundance profiles of the different derivatives in the five cultivars mimicked that of vicine and convicine, but some showed a derivative-specific profile. The description of the (con)vicine diversity will impact the interpretation of future studies on the biosynthesis of (con)vicine, and the content in potentially bio-active alkaloids in faba beans may be higher than that represented by the quantification of vicine and convicine alone. α-Solanine and α-chaconine are the two most predominant glycoalkaloids (GAs) present in potato. Potato peel contains a high concentration of GAs, which are especially interesting for application in the pharmaceutical industry due to their different beneficial properties (such as anticarcinogenic, anti-inflammatory, antiallergic, antipyretic, antiviral, fungicide, and antibiotic activities, among others); so, potato peel waste can be valorized by extracting these biologically active compounds. For this, a green, quick, and efficient miniaturized analytical approach based on ultrasound-assisted extraction (UAE) combined with HPLC-DAD was developed to quantify α-solanine and α-chaconine in potato peel. Some parameters of the extraction were optimized, including the extraction method, the type of solvent, and the sample/solvent ratio, by a three-factor, three-level (33) full factorial experimental design. The optimal extraction conditions were obtained with UAE using methanol as a solvent and a sample/solvent ratio of 1:10 (w/v, g/mL). The analytical greenness metric for sample preparation (AGREEprep) tool was used to assess the greenness of the methods used. The tool revealed an acceptable green analysis, with 0.61 points. The method was validated and applied to the evaluation of GAs in the peel of 15 commercial varieties of potato. The amount of glycoalkaloids found in the samples evaluated ranged from 143 to 1273 mg/kg and from 117 to 1742 mg/kg dry weight for α-solanine and α-chaconine, respectively. These results reveal the important variability that exists between potato varieties; so, their analysis is of great importance to select the most suitable ones for biovalorization (e.g., the Amandine and Rudolph varieties, with around 3000 mg/kg, in total, of both GAs). To provide higher stability to the peel during storage, freeze-drying or a medium-temperature drying process resulted preferable to avoid GA degradation. Overall, this study will contribute to the expansion of the future biovalorization of potato peel waste as well as provide a powerful analytical tool for GA analysis. Full article

Unripe tomatoes represent an agri-food waste resulting from industrial by-processing products of tomatoes, yielding products with a high content of bioactive compounds with potential nutraceutical properties. The food-matrix biological properties are attributed to the high steroidal glycoalkaloid (SGA) content. Among them, α-tomatine is the main SGA reported in unripe green tomatoes. This review provides an overview of the main chemical and pharmacological features of α-tomatine and green tomato extracts. The extraction processes and methods employed in SGA identification and the quantification are discussed. Special attention was given to the methods used in α-tomatine qualitative and quantitative analyses, including the extraction procedures and the clean-up methods applied in the analysis of Solanum lycopersicum L. extracts. Finally, the health-beneficial properties and the pharmacokinetics and toxicological aspects of SGAs and α-tomatine-containing extracts are considered in depth. In particular, the relevant results of the main in vivo and in vitro studies reporting the therapeutic properties and the mechanisms of action were described in detail. Full article

Potato is susceptible to light exposure and wounding during harvesting and transportation. However, the metabolite profile changes in these potato tubers are unclear. The potato cultivars in this study included Hezuo88 (HZ88), Shida6 (SD6), and Jianchuanhong (JCH); the effects of light exposure (L), wounding (W), and the cooking process on potato metabolites were determined. In total, 973 metabolites were identified, with differential metabolites (mainly alkaloids, flavonoids, and phenolic acids) accumulated on days 0 and 2 (0 d and 2 d) in the 0dHZ88 vs. 0dJCH (189), 0dHZ88 vs. 0dSD6 (147), 0dSD6 vs. 0dJCH (91), 0dJCH vs. 2dIJCH (151), 0dJCH vs. 2dWDJCH (250), 0dJCH vs. 2dWLJCH (255), 2dIJCH vs. 2dWDJCH (234), and 2dIJCH vs. 2dWLJCH (292) groups. The flavonoid content in the light exposure group was higher than that in the dark group. The alkaloid content in the wounded group was higher than that in the uninjured potato tuber group, but the lipid content in the wounded group was lower. Importantly, only 5.54% of the metabolites changed after potato tuber steaming. These results provide valuable information for the breeding and consumption of potato tubers. Full article

The rising global demand for animal-based food has an increasingly detrimental ecological impact, exacerbated by significant food waste (approximately one-third of all food). This research aimed to analyze the possibility of changing the usually balanced feed with sustainable alternatives that remain as a by-product of the production of farms, grain processing, and breweries, thus promoting the sustainability of agriculture. The mealworm larvae were reared on different substrates: (1) agar-agar gels, wheat bran, and brewer’s yeast, (2) carrots, wheat bran, and brewer’s yeast, (3) sprouted potatoes, wheat bran, and brewer’s yeast, and (4) carrots, brewers’ spent grain and brewer’s yeast. For analysis, the frozen larvae were lyophilized and tested for chemical safety in three accredited laboratories. The results have shown that all tested samples had lower levels of pesticides than the detection limit. In scientific literature, we didn’t find studies on polycyclic aromatic hydrocarbons (PAH). In our study, we found PAH in the substrate and these toxins, as our study shows, can also enter the larvae, but no significant accumulation was observed (sum of benzo(a)pyrene, benz(a)anthracene, benzo(b)fluoranthene and chrysene was 0.0007 mg/kg). Furthermore, the total content of PAH, benzo(a)pyrene and histamine did not exceed recommended levels. We have noticed that the highest concentration of heavy metals (e.g., chromium—1.45 ± 0.02) was found in the sample with the brewer’s by-products. While numerous studies utilize plant-derived by-products, the accumulation of glycoalkaloids has not been explored. Among the all glycoalkaloids (tomatidin, tomatine, α-solanine, α-chaconine and solanidin), amounts of α-solanine and α-chaconine were the highest, detected in the sample with sprouted potatoes (175.12 ± 0.21 and 139.32 ± 0.32 mg/kg, respectively). The amount of total putrescine, tyramine, spermine, and spermidine in mealworm larvae was statistically higher compared to the amount detected in the substrate, and histamine level-on the contrary, was statistically significantly lower compared to the amount detected in the substrate. Considering the amount of toxic substances found in the substrate from the by-products, we can assume that mealworms did not accumulate high levels of toxins, which would violate regulations. Glycoalkaloids (GAs) are produced naturally by plants and affect insect survivability and fertility. These compounds can be considered potential bioinsecticides; however, the mechanisms and effects of their action remain undiscovered. As lipids are essential molecules for the proper functioning of an insect organism, this research aimed to determine the effects of GAs on the lipid metabolism of the Tenebrio molitor beetle. Solanine, chaconine, tomatine, and tomato leaf extract were applied to larvae by injection at two concentrations, 10−8 and 10−5 M. Then, the tissue was isolated after 2 and 24 h to determine the levels of free fatty acids, sterols and esters using the GC–MS technique. Moreover, the triacylglyceride level and the activity of the key β-oxidation enzyme, 3-hydroxyacyl-CoA dehydrogenase (HADH), were measured. The results indicate that GAs affect the content and composition of lipid compounds in the beetles’ haemolymph and fat body. The effects depend on the GA concentrations, incubation time, and kind of tissue. Moreover, the tested compounds decrease HADH activity, especially in the fat body, which may affect energy production. To our knowledge, this is the first study concerning lipid metabolism in T. molitor after GA application. Our results provide some insights into that topic. Full article

The UN 2030 Agenda for Sustainable Development established the goal of cutting the use of pesticides in the EU by 50% by 2030. However, a ban on pesticides could seriously affect the productivity of agriculture, resulting in severe issues due to global hunger and food deficiency. Controlled release (CR) of bioactive chemicals could play a valid alternative in this context. To this aim, two biodegradable polymers, namely sodium alginate (AL) and sodium carboxymethylcellulose (CMC), were employed to obtain crosslinked hydrogel beads for the encapsulation and CR of glycoalkaloids extracted from tomato and potato leaves to be used as biocompatible disinfectants for agricultural soils. The physico-chemical characterization of the controlled-release systems was carried out by means of Attenuated Total Reflectance–Fourier Transform Infrared (ATR-FTIR) spectroscopy, Scanning Electron Microscopy (SEM), thermogravimetry (TGA), differential scanning calorimetry (DSC) (FWI > 80%) and drying kinetics. The plant extracts and the encapsulation efficiency (~84%) were, respectively, characterized and evaluated by High-performance Liquid Chromatography–Mass Spectrometry (HPLC-MS). Finally, preliminary microbiological tests were conducted to test the efficacy of the most promising systems as biocidal formulations both in the lab and on a model soil, and interesting results were obtained in the reduction of bacterial and fungal load, which could lead to sustainable perspectives in the field. Tomato by-products represent a good source of phytochemical compounds with health properties, such as the steroidal glycoalkaloid α-tomatine (α-TM) and its aglycone tomatidine (TD). Both molecules have numerous beneficial properties, such as potential anticancer activity. Unfortunately, their therapeutic application is limited due to stability and bioavailability issues. Therefore, a valid strategy seems to be their encapsulation into Solid Lipid Nanoparticles (SLN). The nanoformulations containing α-TM (α-TM-SLN) and TD (TD-SLN) were prepared by solvent-diffusion technique and subsequently characterized in terms of technological parameters (particle size, polydispersity index, zeta potential, microscopy, and calorimetric studies). To assess the effect of α-TM and TD on the percentage of cellular viability in Olfactory Ensheathing Cells (OECs), a peculiar glial cell type of the olfactory system used as normal cells, and in SH-SY5Y, a neuroblastoma cancer cell line, an MTT test was performed. In addition, the effects of empty, α-TM-SLN, and TD-SLN were tested. Our results show that the treatment of OECs with blank-SLN, free α-TM (0.25 µg/mL), and TD (0.50 µg/mL) did not induce any significant change in the percentage of cell viability when compared with the control. In contrast, in SH-SY5Y-treated cells, a significant decrease in the percentage of cell viability when compared with the control was found. In particular, the effect appeared more evident when SH-SY5Y cells were exposed to α-TM-SLN and TD-SLN. No significant effect in blank-SLN-treated SH-SY5T cells was observed. Therefore, SLN is a promising approach for the delivery of α-TM and TD. Full article
