Over the years, macromycete fungi have been used as a source of food, part of religious rites and rituals, and as a medicinal remedy. Species with strong health-promoting potential include Hericium erinaceus, Cordyceps militaris, Ganoderma lucidum, Pleurotus ostreatus, Flammulina velutipes, and Inonotus obliquus. These species contain many bioactive compounds, including β-glucans, endo- and exogenous amino acids, polyphenols, terpenoids, sterols, B vitamins, minerals, and lovastatin. The level of some biologically active substances is species-specific, e.g., hericenones and erinacines, which have neuroprotective properties, and supporting the production of nerve growth factor in the brain for Hericium erinaceus. Due to their high health-promoting potential, mushrooms and substances isolated from them have found applications in livestock nutrition, improving their welfare and productivity. This phenomenon may be of particular importance in the nutrition of laying hens and broiler chickens, where an increase in pathogen resistance to antibiotics has been observed in recent years. Gallus gallus domesticus is a key farm animal for meat and egg production, so the search for new compounds to support bird health is important for food safety. Studies conducted to date indicate that feed supplementation with mushrooms has a beneficial effect on, among other things, bird weight gain; bone mineralisation; and meat and egg quality, including the lipid profile and protein content and shell thickness, and promotes the development of beneficial microbiota, thereby increasing immunity. This research examined the impact of dietary supplementation with Phlogacanthus pulcherrimus extract (PPE) on the growth, disease resistance, and expression of immune-, growth-, and antioxidant-related genes in Labeo chrysophekadion. Over 150 days, 90 fish from each group were fed diets with 0 (control), 0.25, 0.50, or 0.75 g/kg of PPE. Phytochemical analysis revealed phenolics (96.00 mg GAE/g), flavonoids (17.55 mg QE/g), anthraquinones, and triterpenoids, along with moderate antioxidant activity (IC50 = 1314.08 μg/mL). One-way ANOVA of growth indices, including weight gain, specific growth rate, feed conversion ratio, and survival rate, revealed no significant differences (p > 0.05); however, PPE supplementation significantly enhanced immune and antioxidant gene expression. IL-1β was significantly (p < 0.05) upregulated at all doses, with the highest expression observed at 0.50 g/kg, showing a fivefold increase compared to the control. In addition, the highest relative expressions of IGF-1 and CAT were found at 0.75 g/kg, with 4.5-fold and 3.5-fold increases compared to the control, respectively. PPE at 0.75 g/kg decreased the cumulative mortality rate (CMR) by 20% compared to the control group, which had a CMR of 50% following exposure to Aeromonas hydrophila. PPE acted as an effective immunostimulant and antioxidant, supporting reduced antibiotic reliance in aquaculture. Full article

Cinnamaldehyde (CA) is a potential substitute for antibiotic growth promoters in animal breeding. In this study, we investigated its effects as a dietary supplement on growth performance, serum antioxidant capacity, intestinal digestive enzyme activities, intestinal morphology, and caecal microbiota in meat rabbits. Weaned meat rabbits (n = 450) were randomly assigned to five groups, Groups A, B, C, D, and E, and fed 0, 50, 100, 150, and 200 mg/kg CA diets, respectively, for 47 days. Biological samples including serum (antioxidants), duodenal/caecal content (enzymes), intestinal tissue (morphology), and caecal digesta (microbiota) were collected at day 47 postweaning for analysis. Groups C and D showed significantly higher final body weights than Group A, with Group D (150 mg/kg CA) demonstrating superior growth performance including 11.73% longer duodenal villi (p < 0.05), 28.6% higher microbial diversity (p < 0.01), and 62% lower diarrhoea rate versus controls. Digestive enzyme activity as well as serum antioxidant capacity increased with increasing CA dose, Microbiota analysis revealed CA increased fibre-fermenting Oscillospiraceae (+38%, p < 0.01) while reducing Ruminococcaceae (−27%, p < 0.05). Thus, dietary CA supplementation at 150 mg/kg was identified as the optimal CA dose for improving meat rabbit production. These findings highlight CA as a functional feed additive for promoting sustainable rabbit production. Full article

This article summarizes research on the application of L. acidophilus in aquaculture, focusing on growth and nutrient utilization, intestinal structure and microbial communities, disease prevention and control in aquatic organisms, and the regulation of water quality. This review holds significant implications for the development of compound feed additives and environmental regulators involving L. acidophilus, as well as for future aquatic food safety. Background/Objectives: In many parts of the world, pharmacists hold the primary responsibility for providing safe and effective pharmacotherapy. A key aspect is the availability of appropriate medicines for each individual patient. When industrially manufactured medicines are unsuitable or unavailable, pharmacists can prepare tailor-made medicines. While this principle applies globally, practices vary between countries. In the Netherlands, the preparation of medicines in pharmacies is well-established and integrated into routine healthcare. This narrative review explores the role and significance of extemporaneous compounding, pharmacy preparations and related product care in the Netherlands. Methods: Pharmacists involved in pharmacy preparations across various professional sectors, including community and hospital pharmacies, central compounding facilities, academia, and the professional pharmacists’ organisation, provided detailed and expert insights based on the literature and policy documents while also sharing their critical perspectives. Results: We present arguments supporting the need for pharmacy preparations and examine their position and role in community and hospital pharmacies in the Netherlands. Additional topics are discussed, including the regulatory and legal framework, outsourcing, quality assurance, standardisation, education, and international context. Specific pharmacy preparation topics, often with a research component and a strong focus on product care, are highlighted, including paediatric dosage forms, swallowing difficulties and feeding tubes, hospital-at-home care, reconstitution of oncolytic drugs and biologicals, total parenteral nutrition (TPN), advanced therapy medicinal products (ATMPs), radiopharmaceuticals and optical tracers, clinical trial medication, robotisation in reconstitution, and patient-centric solid oral dosage forms. Conclusions: The widespread acceptance of pharmacy preparations in the Netherlands is the result of a unique combination of strict adherence to tailored regulations that ensure quality and safety, and patient-oriented flexibility in design, formulation, and production. This approach is further reinforced by the standardisation of a broad range of formulations and procedures across primary, secondary and tertiary care, as well as by continuous research-driven innovation to develop new medicines, formulations, and production methods. The article presents a new AM (Additive Manufacturing) process development, necessary to repair parts made from Aluminum 6061 material, with T6 treatment. The laser Directed Energy Deposition (DED) and Extreme High-Speed Directed Energy Deposition (EHLA) capabilities are evaluated for repairing Al large components. To optimize the process parameters, single-track depositions were analyzed for both laser-powder DED (feed rate of 2 m/min) and EHLA (feed rate 20 m/min) for AlSi10Mg and Al6061 powders. The cross-sections of single tracks revealed the bonding characteristics and provided laser-powder DED, a suitable parameter selection for the repair. Three damage types were identified on the Al component to define the specification of the repair process and to highlight the capabilities of laser-powder DED and EHLA in repairing intricate surface scratches and dents. Our research is based on variation of the powder mass flow and beam power, studying the influence of these parameters on the weld bead geometry and bonding quality. The evaluation criteria include bonding defects, crack formation, porosity, and dilution zone depth. The bidirectional path planning strategy was applied with a fly-in and fly-out path for the hatching adjustment and acceleration distance. Samples were etched for a qualitative microstructure analysis, and the HV hardness was tested. The novelty of the paper is the new process parameters for laser-powder DED and EHLA deposition strategies to repair large Al components (6061 T6), using AlSi10Mg and Al6061 powder. Our experimental research tested the defect-free deposition and the compatibility of AlSi10Mg on the Al6061 substrate. The readers could replicate the method presented in this article to repair by laser-powder DED/EHLA large Al parts and avoid the replacement of Al components with new ones. Full article

Biomarker-Driven Optimization of Saponin Therapy in MASLD: From Mouse Models to Human Liver Organoids

(1) Background: Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by liver damage similar to alcoholic fatty liver disease, including triglyceride infiltration of hepatocytes, regardless of alcohol consumption. It leads to progressive liver damage, such as loss of liver function, cirrhosis, and liver cancer, and the response rate of drugs under clinical research is less than 50%. There is an urgent need for biomarkers to evaluate the efficacy of these drugs. (2) Methods: MASLD was induced in mice using a High-Fat diet (HF), Western diet (WD), and Methionine/Choline-Deficient diet (MCD) for 20 weeks (4 weeks for MCD). Liver tissue biopsies were performed, and the treatment effects of saponin and non-saponin feeds were evaluated. Fat accumulation and hepatic inflammation were measured, and mRNA sequencing analysis was conducted. The therapeutic effects were validated using patient-derived liver organoids. (3) Results: The NAFLD Activity Score (NAS) significantly increased in all MASLD models compared with controls. Saponin treatment decreased NAS in the HF and WD groups but not in the MCD group. RNA sequencing and PCA analysis showed that the HF saponin response samples were similar to normal controls. DAVID analysis revealed significant changes in lipid, triglyceride, and fatty acid metabolic processes. qRT-PCR confirmed decreased fibrosis markers in the HF saponin response group, and GSEA analysis showed reduced HAMP1 gene expression. (4) Conclusions: Among the diets, red ginseng was most effective in the HF diet, with significant effects in the saponin-treated group. The therapeutic efficacy was better when HAMP1 expression was increased. Therefore, we propose HAMP1 as a potential exploratory biomarker to assess the saponin response in a preclinical setting. In addition, the reduction of inflammation and hepatic iron accumulation suggests that saponins may exert antioxidant effects through modulation of oxidative stress. Full article

Teleoperation emerged as a promising fallback for situations beyond the capabilities of automated vehicles. Nevertheless, teleoperation still faces challenges, such as reduced situational awareness. Since situational awareness is primarily built through the remote operator’s visual perception, the graphical user interface (GUI) design is critical. In addition to video feed, supplemental informational elements are crucial—not only for the predominantly studied remote driving, but also for emerging desk-based remote assistance concepts. This work develops a GUI for different teleoperation concepts by identifying key informational elements during the teleoperation process through expert interviews (N = 9). Following this, a static and dynamic GUI prototype was developed and evaluated in a click dummy study (N = 36). Thereby, the dynamic GUI adapts the number of displayed elements according to the teleoperation phase. Results show that both GUIs achieve good system usability scale (SUS) ratings, with the dynamic GUI significantly outperforming the static version in both usability and task completion time. However, the results might be attributable to a learning effect due to the lack of randomization. The user experience questionnaire (UEQ) score shows potential for improvement. To enhance the user experience, the GUI should be evaluated in a follow-up study that includes interaction with a real vehicle. Full article

Understanding how habitat conditions drive morphological adaptations in animals is critical in ecology, yet amphibian studies remain limited. This study investigated intraspecific variation in ecomorphological traits of three montane stream-dwelling frogs (Quasipaa boulengeri, Amolops sinensis, and Odorrana margaratae) across elevation gradients in Tianping Mountain, China. Using morphological measurements and environmental variables collected from ten transects, we analyzed functional traits related to feeding and locomotion and assessed their associations with microhabitat variables. Significant trait differences between low- and high-elevation groups were detected only in Q. boulengeri, with high-elevation individuals exhibiting greater body mass and shorter hindlimbs. Redundancy analysis demonstrated that microhabitat variables, particularly air humidity, flow rate, and rock coverage, were linked to trait variations. For example, air humidity and flow rate significantly influenced Q. boulengeri’s body and limb proportions, while flow rate affected A. sinensis’s snout and limb morphology. In addition, sex and seasonal effects were also associated with trait variations. These results underscore amphibians’ phenotypic plasticity in response to the environment and highlight the role of microhabitat complexity in shaping traits. By linking habitat heterogeneity to eco-morphology, this study advocates for conservation strategies that preserve varied stream environments to support amphibian resilience amid environmental changes. Dietary supplementation under high ambient temperatures can be considered crucial for supporting immune responses in livestock. In the present paper, Rosmarinus officinalis essential oil (REO) alone and in combination with dried sugar beet pulp (REO + B) was included in the diet of lambs during the fattening period under heat stress conditions. Environmental conditions and physiological and growth parameters of lambs were monitored throughout the trial. Plasma samples were collected to evaluate cytokine secretion (IL-1β, IL-6, and IL-10). Notably, REO inclusion reduced rectal temperature and respiration rate while increasing pro-inflammatory cytokines (IL-1β and IL-6) at the same time, suggesting an enhanced immune response without compromising growth performance. Dietary tannin supplementation represents a potential strategy to modulate rumen fermentation and enhance lactation performance in dairy cows, though responses remain inconsistent. A 21-day feeding trial was conducted to evaluate the effect of chestnut tannin (CNT) extract on the enteric methane emissions (EME), blood metabolites, and milk production traits in mid-lactation dairy cows. Thirty-six Holstein cows were allocated to three homogeneous treatment groups: control (CNT0, 0 g/d CNT), CNT40 (40 g/d CNT), and CNT80 (80 g/d CNT). Measurements of EME, dry matter intake (DMI), milk yield (MY), and blood and milk parameters were carried out pre- and post-21-day supplementation period. Compared with the no-additive group, the CNT extract reduced methane production, methane yield, and methane intensity in CNT40 and CNT80 (p < 0.001). CNT40 and CNT80 cows exhibited lower blood urea nitrogen (p = 0.019 and p = 0.002) and elevated serum insulin (p = 0.003 and p < 0.001) and growth hormone concentrations (p = 0.046 and p = 0.034), coinciding with reduced aspartate aminotransferase (p = 0.016 and p = 0.045), and lactate dehydrogenase (p = 0.011 and p = 0.008) activities compared to control. However, CNT80 had higher circulating NEFA and BHBA than CNT0 (p = 0.003 and p = 0.004) and CNT40 (p = 0.035 and p = 0.019). The blood glucose, albumin, and total bilirubin concentrations were not affected. MY and fat- and protein-corrected milk (FPCM), MY/DMI, and FPCM/DMI were higher in both CNT40 (p = 0.004, p = 0.003, p = 0.014, p = 0.010) and CNT80 (p = 0.002, p = 0.003, p = 0.008, p = 0.013) cows compared with controls. Feeding CNT80 resulted in higher protein content (p = 0.015) but lower fat percentage in milk (p = 0.004) compared to CNT0. Milk urea nitrogen and somatic cell counts were significantly lower in both CNT40 (p < 0.001, p = 0.009) and CNT80 (p < 0.001 for both) compared to CNT0, while milk lactose did not differ between treatments. Changes in Ruminal Dynamics and Microbial Populations Derived from Supplementation with a Protein Concentrate for Cattle with the Inclusion of Non-Conventional Feeding Sources

Feed supplementation strategies are essential for optimizing cattle productivity, and the incorporation of non-conventional feed resources may reduce both production costs and environmental impact. This study evaluated the effects of pelletized protein concentrates (including Acacia farnesiana, A. schaffneri, and Agave duranguensis bagasse) on rumen fermentation parameters, microbial communities, and gas emissions. Fistulated bullocks received the concentrate daily, and ruminal contents were collected and filtered before and after supplementation to assess in vitro gas and methane production, pH, and microbial composition using high-throughput sequencing of 16S rRNA and mcrA amplicons. In addition, in situ degradability was evaluated during and after the supplementation period. Supplementation led to a significant (p < 0.05) reduction in degradability parameters and methane production, along with a marked decrease in the abundance of Methanobrevibacter and an increase in succinate-producing taxa. These effects were attributed to the enhanced levels of non-fiber carbohydrates, hemicellulose, crude protein, and the presence of bioactive secondary metabolites and methanol. Rumen microbiota composition was consistent with previously described core communities, and mcrA-based sequencing proved to be a valuable tool for targeted methanogen detection. Overall, the inclusion of non-conventional ingredients in protein concentrates may improve ruminal fermentation efficiency and contribute to methane mitigation in ruminants, although further in vivo trials on a larger scale are recommended. In this study, the seeking behaviour and food acceptance of larvae of Plodia interpunctella Hübner (Lepidoptera: Pyralidae) were analysed under laboratory conditions. Larval orientation and feeding preferences were assessed using a selection arena for neonate larvae and a four-way olfactometer for third-instar larvae. Stimulants included amaranth bars with additives (honey and chocolate) and natural amaranth (toasted grain only). The results showed that amaranth volatiles influence the orientation and feeding behaviour of this polyphagous insect. A marked preference for sugar-rich foods was observed, with amaranth with honey and amaranth with chocolate being the food sources most frequently chosen by the neonate larvae. These individuals exhibited a gregarious feeding behaviour and did not engage in cannibalism. The third-instar larvae also showed a preference for sweet food but were more attracted to the amaranth–additive combination. In the four-way olfactometer bioassays, chocolate was the most frequently chosen stimulus, while cellophane did not differ significantly from air. An analysis of volatile compounds by gas chromatography mass spectrometry (GC-MS) revealed that amaranth with chocolate releases more volatile compounds (16) compared with honey (12) and natural amaranth (6), suggesting that these volatiles could possibly influence the larvae’s choice of food source. Leaves, the main by-product of lingonberry harvesting, can be effectively used as a functional feed additive due to their health-promoting properties. This study evaluated the effects of lingonberry leaf (LL) supplementation on rumen fermentation, protozoal populations, and gastrointestinal morphology in sheep. Eight one-year-old Polish Mountain Sheep ewes (mean body weight: 33 kg) were allocated to a control (basal diet; forage-to-concentrate ratio 60:40) or an experimental group (basal diet + 9.30 g/kg DM dried LLs) in a completely randomised design (n = 4 per group) over 34 days. Both diets were formulated to be isoenergetic and isonitrogenous. LL additive significantly reduced Isotrichidae protozoal counts (p < 0.001) and ruminal pectinolytic activity (p = 0.043), without altering short-chain fatty acid (SCFA) or methane concentrations (p > 0.1). Histological analyses showed increased ruminal papilla width and surface area (p < 0.001) and decreased duodenal villus height and muscular layer thickness (p < 0.01). Inflammatory lesions (reddish foci) were identified in the liver in both groups. These findings demonstrate that LL supplementation affected specific protozoal population, fibrolytic activity, and gastrointestinal morphology. Further study on a larger number of animals is recommended to validate the effects and assess the safety and efficacy of LLs as a dietary additive in ruminant nutrition. Full article

Background/Objectives: Green tripe (GRET) is rich in essential fatty acids, vitamins, calcium, phosphorus, and other nutrients and contains various beneficial microorganisms, including lactobacillus, along with feed components consumed by ruminants. Methods: In this study, Latilactobacillus sakei and L. curvatus were isolated from GRET and evaluated for their potential as probiotics, focusing on their anti-inflammatory properties and ability to modulate inflammatory responses. Results: When heat-killed L. sakei or L. curvatus (108 CFU/mL) and their metabolites (0.5 mg/mL) were applied to RAW 264.7 macrophages stimulated with LPS, nitric oxide (NO) production was reduced by approximately 10–35% and 2–11%, respectively. Furthermore, the expression levels of key anti-inflammatory cytokines, TNF-α and IL-6, were suppressed by more than 5%. These effects were not due to cytotoxicity but instead due to genuine anti-inflammatory activity. In addition, both strains exhibited antioxidant activity, as demonstrated by their performance in ABTS and FRAP assays. Conclusions: These findings suggest that L. sakei and L. curvatus have significant antioxidant and anti-inflammatory properties, highlighting their potential as probiotics and prebiotics. Moreover, these newly isolated strains from GRET are expected to serve as valuable functional ingredients for developing health-promoting foods and dietary supplements. Full article
