Comprehensive Genomic Analysis of Pseudomonas aeruginosa PSU9449 Isolated from a Clinical Case in Thailand

Background/Objectives: Pseudomonas aeruginosa is one of the most significant multidrug-resistant bacteria. It poses considerable challenges in terms of treatment and causes hospital-acquired infections that lead to high morbidity and mortality. Colonization by P. aeruginosa in a patient without clinical signs of infection is a concern in hospital settings, as it is an opportunistic pathogen and can potentially be a multidrug-resistant strain. The objective of this study was to characterize and provide a detailed genomic analysis of this strain of the P. aeruginosa PSU9449 genome, an isolate obtained from a patient at Songklanagarind Hospital, Thailand. Methods: Whole-genome sequencing (WGS) and bioinformatics analysis were employed to examine the genomic features of P. aeruginosa PSU9449. We performed sequence type (ST) determination through multilocus sequence typing (MLST), identified antimicrobial resistance genes (ARGs), virulence factor genes (VFGs), and analyzed the presence of mobile genetic elements (MGEs). Additionally, we compared the PSU9449 genome with strains from neighboring countries to understand its phylogenetic relationship. Results: The P. aeruginosa PSU9449 genome contained five insertion sequences and several ARGs, including fosA, aph (3’)-IIb, blaOXA-50, and catB7. It also harbored VFGs related to flagella (fli, fle, and flg), the type 6 secretion system (hcpA, tssA, and las), and the type 3 secretion system (exoS, exoU, and exoT). MLST identified PSU9449 as ST3777, which was reported in Thailand for the first time. Phylogenetic analysis based on core gene SNPs revealed that PSU9449 was closely related to P. aeruginosa HW001G from Malaysia and P. aeruginosa MyJU45 from Myanmar, forming a distinct clade. Conclusions: This study presents a comprehensive genomic analysis of P. aeruginosa PSU9449, shedding light on its genetic characteristics, antimicrobial resistance profile, and virulence potential. Interestingly, ST3777, the novel STs from the published genomes of P. aeruginosa in Thailand, were assigned in this study. The findings enhance valuable insights into the expanding knowledge of P. aeruginosa PSU9449 and highlight the importance of ongoing surveillance of its genetic diversity. Full article

Background/Objectives: Pseudomonas aeruginosa causes chronic infections in humans and animals, especially cats and dogs. This bacterium’s ability to adapt and acquire antibiotic resistance traits may complicate and exacerbate antibacterial therapy. This study aimed to evaluate the antibiotic resistance patterns, virulence factors and ability to form biofilms of P. aeruginosa strains isolated from Algerian dogs and cats. Methods: Nineteen samples were collected from healthy and diseased dogs and cats. Isolates were studied for their antibiotic-resistance patterns (disc diffusion method) and biofilm formation (Microtiter assay) and were whole-genome sequenced (MinION). Results: Nineteen P. aeruginosa strains (15 from dogs and 4 from cats) were isolated. Antibiotic-resistance phenotypes were observed against amoxicillin–clavulanic acid (100%); meanwhile, resistance towards ticarcillin was 40% (dogs) and 25% (cats), ticarcillin–clavulanic acid was 13.33% and 25% for dogs and cats, respectively, and imipenem was 75% (cats) and 20% (dogs). Moreover, 95% of strains were biofilm-producers. Different antimicrobial resistance genes (ARGs) were found: beta-lactamase genes, mainly PAO, OXA-494, OXA-50 and OXA-396 and an aminoglycoside gene (aph(3′)-IIb). The main high-risk sequence types (STs) were ST244, 2788, 388 and 1247. A large panel of virulence genes was detected: exoS, exoT, exoY, lasA, toxA, prpL, algD, rhIA and others. Conclusions: The genetic variety in antibiotic-resistance genes of resistant and virulent P. aeruginosa strains in dogs makes public health protection difficult. Continuous monitoring and research in compliance with the One Health policy are needed to solve this problem. Full article

Background: Pseudomonas aeruginosa is a pathogen that causes infections in animals and humans, with veterinary implications including ear infections in dogs, respiratory diseases in cats, and mastitis in ruminants. In humans, it causes severe hospital-acquired infections, particularly in immunosuppressed patients. This study aimed to identify and assess the prevalence of specific virulence factors in Pseudomonas aeruginosa isolates. Methods: We analyzed 98 Pseudomonas aeruginosa isolates from various animal samples (dogs, cats, ruminants, fowl) from northeastern Poland in 2019–2022 for virulence-related genes (toxA, exoU, exoT, exoS, lasB, plcN, plcH, pldA, aprA, gacA, algD, pelA, endA, and oprF) by PCR and assessed biofilm formation at 48 and 72 h. Genomic diversity was assessed by ERIC-PCR. Results: The obtained results showed that all strains harbored the pelA gene (100%), while the lowest prevalence was found for pldA (24%) and exoU (36%). Regardless of the animal species, strong biofilm forming ability was prevalent among the strains after both 48 h (75%) and 72 h (74%). We obtained as many as 87 different genotyping profiles, where the dominant one was profile ERIC-48, observed in four strains. Conclusions: No correlation was found between presence or absence of determined genes and the nature of infection. Similarly, no correlation was found between biofilm-forming genes and biofilm strength. The high genetic diversity indicates challenges for effective prevention, emphasizing the need for ongoing monitoring and research. Full article

The Chlamydiaceae family consists of Gram-negative, obligate intracellular bacteria that replicate within the cells of a diverse range of hosts. These hosts include domesticated animals such as cats, dogs, and livestock, as well as wildlife like koalas and birds, exotic species such as reptiles and amphibians, and humans. Chlamydial infection can result in various clinical signs, including respiratory diseases, reproductive failures, ocular pathologies, and enteritis, though the infected organism may remain asymptomatic. In recent years, chlamydial nomenclature has undergone several revisions due to the wide range of hosts, the frequent discovery of novel strains, and the reclassification of existing ones. Given this and the clinical significance of these infections, ranging from asymptomatic to fatal, an updated review is essential. This article outlines key characteristics of Chlamydia species and provides an updated overview of their nomenclature, offering a concise reference for future research on chlamydial diseases. Dermatophytic pseudomycetoma (DPM) is a rarely reported invasive fungal infection of humans and animals, especially cats. This study aimed to identify dermatophytes, breed associations, and the frequency of extracutaneous (EC) involvement in feline DPM. Electronic records and formalin-fixed paraffin-embedded tissue (FFPET) from 32 suspected DPM cases in 30 cats were retrieved from a diagnostic laboratory between 2018 and 2024. To confirm DPM and molecular identity, DNA was extracted from FFPET for ITS2 sequencing, and immunohistochemistry was performed on PCR-negative cases. All cases were confirmed as DPM. Microsporum canis was the only dermatophyte identified. The sensitivity and specificity of ITS2 sequencing for M. canis identification in FFPET were 22/32 (68.8%) and 21/22 (95.5%), respectively. Exotic (36.7%) and Persian (23.3%) but not British breeds (26.3%) were over-represented compared to feline admissions at an affiliated veterinary hospital (8.5%, p < 0.001; 3%, p < 0.001; 21.6%, p = 0.817, respectively). Five cases (16.7%) had EC lesions; two had intra-abdominal masses; two had oral cavity masses, including one which extended into the cranial vault; and one had superficial cervical lymph node invasion. Exotic and Persian breeds are over-represented for DPM and M. canis is the primary cause. EC lesions of DPM may occur more commonly than previously thought. Full article

Since late 2019, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has emerged as a significant global threat to public health. Responsible for the COVID-19 pandemic, this new coronavirus has prompted extensive scientific research to comprehend its transmission dynamics, especially among humans. However, as our understanding deepens, it becomes increasingly clear that SARS-CoV-2’s impact goes beyond human populations. Recent investigations have illuminated the transmission of the virus between humans and various animal species, raising important questions about zoonotic spillover events and their potential implications for both human and animal health. Our study set out to investigate the prevalence of SARS-CoV-2 in domestic animals (dogs and cats) and zoo animals in the south of France in 2021 and 2022, covering pre-Omicron and Omicron waves. We identified evidence of SARS-CoV-2 antibodies not only in domestic dogs and cats but also in several mammals in zoos. This study shows the importance of implementing surveillance measures, including serological studies, to identify and monitor cases of SARS-CoV-2 infection in animals. Full article

While people are familiar with the practice of declawing domestic cats, “onychectomy”, as it is also known, is also performed on non-domesticated species, including pantherines, to prolong their use for entertainment purposes. Although the surgery (the partial or complete removal of the distal phalanx) has clear osteological implications, its myological effects have never been studied. As the mass of an animal increases cubically as a product of its volume, while the areas of its paws only increase as a square, larger felids have higher foot pressures and, therefore, the surgery may have particularly substantial functional effects on larger cats. In this study, we evaluate the forearms of clawed and declawed non-domestic felid specimens that spanned the body size range of the whole family to evaluate the effects of onychectomy on muscle fiber architecture. We found that the deep digital flexors (the muscles most directly affected by onychectomy) of declawed felids are significantly lighter (~73%) and less powerful (46–66%) than those of non-declawed felids, while other muscles do not make up for these reductions. Thus, onychectomy has a substantial effect on the myological capabilities of cats, and because these deficiencies are not compensated for in biomechanically disadvantaged larger felids, it probably has even more functionally devastating consequences for these species. Pseudomonas aeruginosa has assumed an increasingly prominent role as the aetiological agent in serious hard-to-treat infections in animals and humans. In this study, 271 P. aeruginosa strains collected from dogs and cats were investigated. The aim of the research was to screen these P. aeruginosa strains for antibiotic resistance and the presence of selected virulence factor genes. Antibiotic resistance was determined using the Kirby–Bauer method, while virulence genes were detected by polymerase chain reaction (PCR). The most frequently detected resistance was to fluoroquinolones, ranging in prevalence from 17.3% for ciprofloxacin up to 83% for enrofloxacin. The resistance to carbapenems was 14% and 4.8% for imipenem and meropenem, respectively. Almost all P. aeruginosa strains harboured the exoT (97.8%) and lasB (93.4%) genes, while the lowest prevalence was found for exoU (17.3%) and plcH (17.3%). P. aeruginosa strains isolated from dogs that harboured the toxA gene were more frequently resistant to ceftazidime (p = 0.012), while the presence of the exoU gene was found to be connected with resistance to marbofloxacin (p = 0.025) and amikacin (p = 0.056). In strains originating from cats, only the connection between the presence of the exoU gene and resistance to enrofloxacin (p = 0.054) was observed. The confirmation of associations between virulence-factor-encoding genes and antibiotic resistance indicates that problems of antibiotic resistance may not only cause complications at the level of antibiotic dosage but also lead to changes in the virulence of the bacteria; thus, further studies in this area are required. Persian and exotic shorthair cats are the most-popular brachycephalic breeds worldwide. This study aimed to explore differences in human-cat bonding, behavioural traits, perception of clinical breed-related problems, and the motivation for purchasing between brachycephalic cat owners (BCOs) and non-brachycephalic cat owners (NBCOs). Using an online questionnaire, human-cat bonding and cats’ behavioural traits were explored using the CORS and Fe-BARQ scales, respectively. Breed-related problems and motivations for purchasing were explored only in BCOs. There were 278 BCOs and 250 NBCOs who completed the survey. Respiratory diseases resulted in being the main health problem of these breeds. Most BCOs were not aware of the incidence of these disorders in brachycephalic cats. Character and appearance were the main motivations for purchasing these breeds. Perceived emotional closeness (EC), cat-owner interaction (COI), and perceived cost (PC) mean scores were significantly higher in BCOs. Playfulness and affection-/attention-seeking scores were significantly higher in NBCOs. BCOs reported a significantly higher score for separation-related behaviours. The higher scores in separation-anxiety-related behaviours and in the EC and COI scales could be related mainly to the high level of care these breeds require. The motivation for acquiring brachycephalic breeds for good companionship seems in contrast with the lower scores obtained from BCOs for playfulness and affection-/attention-seeking. Full article

Corneal sequestra are ophthalmic lesions that are remarkably common in Persian, Himalayan and exotic cats. In this study, the genetic aspects of this disease were investigated in a population of cats originating from a single cattery. Odds ratios were calculated for parents with affected offspring. The heritability of (owner-reported) corneal sequestra was estimated with a Markov chain Monte Carlo procedure. Well-phenotyped cases and controls were used for a genome-wide association study. Data from 692 cats originating from the cattery, of which 61 were affected, were used. Cats from two specific mothers had significantly higher odds of developing corneal sequestra, but no significant effect of the fathers was found (after correction for the mothers). The heritability of corneal sequestra was estimated to be 0.96. A genome-wide association study with 14 cases and 10 controls did not reveal an associated chromosomal region. The large effect that genetic factors had on the development of corneal sequestra in this study suggests that selective breeding could be an effective way to reduce the prevalence of this condition in these cat breeds. The objective of this review was to carry out a comprehensive investigation of the benefits of incorporating insects as a pet food ingredient and the implications this can have in determining a market demand for insect-based pet foods. Black soldier fly larvae (Hermetia illucens), mealworm larvae (Tenebrio molitor) and adult house crickets (Acheta domesticus) are currently used in pet food. These insects are widely fed to exotic pets, mainly in whole, live or dehydrated formats. They are also incorporated as meal or fat and are offered to cats and dogs as dry or wet food and treats. Scientific studies about the use of insects for dog and cat feed are scarce. Most studies are in dogs. Research shows that insect nutrients, mainly amino acids, have high digestibility, are beneficial to health, do not have any detrimental effect on the gut microbiota and are accepted by dogs. In several countries, insects are approved for use in pet food and commercialization has spread throughout the world. Pet owners are willing to try foods made with insect meal for their pets. In conclusion, the use of insects in pet food is a reality that is taking on more and more prominence. Full article

Water scarcity is a major environmental stress that adversatively impacts wheat growth, production, and quality. Furthermore, drought is predicted to be more frequent and severe as a result of climate change, particularly in arid regions. Hence, breeding for drought-tolerant and high-yielding wheat genotypes has become more decisive to sustain its production and ensure global food security with continuing population growth. The present study aimed at evaluating different parental bread wheat genotypes (exotic and local) and their hybrids under normal and drought stress conditions. Gene action controlling physiological, agronomic, and quality traits through half-diallel analysis was applied. The results showed that water-deficit stress substantially decreased chlorophyll content, photosynthetic efficiency (FV/Fm), relative water content, grain yield, and yield attributes. On the other hand, proline content, antioxidant enzyme activities (CAT, POD, and SOD), grain protein content, wet gluten content, and dry gluten content were significantly increased compared to well-watered conditions. The 36 evaluated genotypes were classified based on drought tolerance indices into 5 groups varying from highly drought-tolerant (group A) to highly drought-sensitive genotypes (group E). The parental genotypes P3 and P8 were identified as good combiners to increase chlorophyll b, total chlorophyll content, relative water content, grain yield, and yield components under water deficit conditions. Additionally, the cross combinations P2 × P4, P3 × P5, P3 × P8, and P6 × P7 were the most promising combinations to increase yield traits and multiple physiological parameters under water deficit conditions. Furthermore, P1, P2, and P5 were recognized as promising parents to improve grain protein content and wet and dry gluten contents under drought stress. In addition, the crosses P1 × P4, P2 × P3, P2 × P5, P2 × P6, P4 × P7, P5 × P7, P5 × P8, P6 × P8, and P7 × P8 were the best combinations to improve grain protein content under water-stressed and non-stressed conditions. Certain physiological traits displayed highly positive associations with grain yield and its contributing traits under drought stress such as chlorophyll a, chlorophyll b, total chlorophyll content, photosynthetic efficiency (Fv/Fm), proline content, and relative water content, which suggest their importance for indirect selection under water deficit conditions. Otherwise, grain protein content was negatively correlated with grain yield, indicating that selection for higher grain yield could reduce grain protein content under drought stress conditions. Wanna See My Dog Pic? A Comparative Observational Study of the Presentation of Animals on Online Dating Profiles in Vienna and Tokyo

Online dating applications offer new ways for people to search for social contacts. While previous studies have indicated that the inclusion of animals in profiles can increase users’ dating success rates, the question of how many users display animals, and what kinds of animals are shown on dating profiles, has not yet been empirically investigated. Using a structured observational study of profiles in Vienna and Tokyo on a popular online dating app (n = 2400), we therefore looked at how many profiles show animals and what kinds of animals are shown. We found that 15.5% of the investigated profiles had at least one photo showing an animal. In both cities, dogs were the most frequently shown animal. Taking the cities together, they appeared in 46.4% of the animal pictures, as compared with cats at 25.7%. Other animals such as exotic animals (9.9%), farm animals (6.4%) or horses (4.6%) played a minor role. Users were significantly more likely to show cats in Tokyo (35.8%) than they were in Vienna (18.0%). We found that users in Vienna; women; and older adults were more likely to present animals on their profiles than were users in Tokyo; men; and younger users. Sexual orientation showed no significant differences in the analyses. Full article

Occurrence and Abundance of an Apex Predator and a Sympatric Mesopredator in Rural Areas of the Coastal Range of Southern Chile

The two mammalian carnivores, puma (Puma concolor) and South American grey fox (Lycalopex griseus) were studied, in a remote area located in the humid temperate forest of the coastal range of southern Chile. A total of six locations were selected in three landscapes: pre-mountain range, mountain range, and coast. The chosen study locations are relevant because they correspond to threatened areas with different levels of human intervention., so they offer the ideal setting for studying how different species of carnivores respond to both human presence and activities. A dataset was collected for 24 months during 2016–2018 through photo-trapping (13 camera traps placed along 50 photo-trap stations). Wes estimated the apparent occurrence and relative abundance index (RAI) of the fauna registered, by means of generalized linear models to contrast those of an apex predator, such as the puma and a sympatric mesopredator, the South American grey fox, across the three landscapes. The ecological variables assessed were the RAI of the other carnivore considered, exotic carnivores such as dogs and cats, human intervention, farmland effect, prey availability, and habitat quality. The primary hypothesis was that the apparent occurrence and RAI of puma and fox would be positively associated with the RAI of prey and livestock and negatively with human intervention. On the other hand, the secondary hypothesis dealt with the interactions between puma and fox faced with different degrees of human intervention. The results showed that the apparent occurrence of the puma was statistically explained by location only, and it was highest at the mountain range. The apparent occurrence of foxes was explained by both puma apparent occurrence and relative integrated anthropization index (INRA), being highest in the pre-mountain range. Concerning the RAI of pumas, high values were yielded by location and fox RAI. For the RAI of foxes, they were location, puma RAI, and INRA. It can be suggested that eucalyptus plantations from the pre-mountain range could offer an adequate habitat for the puma and the fox, but not the coastal range, as the mountain range could be acting as a biological barrier. Due to the nature of the data, it was not possible to detect any relevant effect between the two carnivores’ considered, between their respective preys, or the very abundant presence of dogs.
