Morphological descriptions of 13 wild fighting fish species were revised by examining specimens collected as near as possible to the type specimen localities. The description has been revised for certainty and consistency. Principal Component Analysis (PCA) and Kruskal–Wallis with Dunn’s post hoc tests were analyzed and revealed previously described morphological characteristics were evaluated for distinguishing features. The PCA showed that the high loading factor of the principal component 1 (PC1) indicated that head length (0.221) was beneficial for the fish species identification. The Kruskal–Wallis test indicated the significant differences in morphometric characteristics among the fish species (p < 0.05). Then the significant differences in Dunn’s post hoc test (p < 0.001) in head length distinguished the mouth-brooders (i.e., Betta apollon, B. ferox, B. kuehnei, B. pallida, B. pi, B. prima, B. pugnax, and B. simplex) from bubble-nesters (i.e., B. imbellis, B. mahachaiensis, B. siamorientalis, B. smaragdina, and B. splendens) with a head length range of 29.27–36.42 vs. 24.14–28.31% of standard length, including the symmetrical caudal fin. Some characteristics distinguish the fish into subgroups and some species, such as a greater number of anal soft fin rays and lateral line scales of B. pi among the mouth-brooding species (28–29 vs. 22–26 fin rays and 33–35 vs. 26–32 scales) or of B. smaragdina among the bubble-nesting species (26–28 vs. 22–25 fin rays and 31–33 vs. 27–30 scales). Additionally, descriptive characteristics such as anal and caudal fin shape and melanophore pigmentation on the head, body, and fins, including the coloration of live specimens, are useful for distinguish among species and generating a dichotomous identification key. Glutathione S-transferases (GSTs) are an important family of enzymes involved in plant detoxification, maintenance of redox homeostasis, and responses to abiotic stresses. However, the evolutionary characteristics and functional roles of the potato GST pan-gene family have not yet been systematically investigated at the pan-genome level. In this study, based on high-quality potato genomes constructed from 45 diploid accessions, GST gene family members were systematically identified, and their evolutionary features and expression patterns were analyzed. Phylogenetic analysis classified the GST family into six subgroups, among which the soft-core gene StGST7 and the near-core genes StGST8 and StGST16 were assigned to the Phi and Tau subgroups, respectively. Selection pressure analysis indicated that five StGST genes may have undergone positive selection, whereas most of the remaining genes were mainly subjected to purifying selection. Structural variation significantly affected the expression of StGST42 and the conserved domains of its encoded protein. Expression profiling revealed that GST family members exhibited clear tissue-specific expression patterns and responded differentially to drought, salt, high temperature, ABA, and IAA treatments. Co-expression network analysis revealed significant positive and negative correlations between multiple transcription factors and StGST gene expression, suggesting their potential involvement in the coordinated regulation of StGST genes. Further analyses demonstrated that StGST7 was significantly differentially expressed under multiple stress conditions, and its heterologous expression enhanced yeast tolerance to salt and drought stress. This study revealed the evolutionary characteristics and potential functions of the potato GST gene family and provides a theoretical basis for elucidating the molecular mechanisms underlying its regulation of environmental adaptation. Background: Profound musculoskeletal alterations encompassing bones and soft tissues are common in people with anorexia nervosa (AN). This study aims to examine spinal posture and mobility in adolescents with AN, and to compare these outcomes with those obtained from normal-weight female controls. Methods: Spinal posture and movements were measured in 37 adolescents with AN and 31 normal-weight controls using the Idiag M360 scan tool, and between-group differences were analyzed using analysis of covariance. Results: Spinal postures and the lumbar-to-hip ratio were not different between subgroups. By contrast, AN had reduced thoracic (−21.8 degrees, p < 0.0001) and lumbar (−18.2 degrees, p < 0.0001) mobility in the frontal plane, as well as decreased hip flexion (−14 degrees, p = 0.001) and extension (−18.6 degrees, p < 0.0001) compared to the CG. Conclusions: Thoracic and lumbar spinal mobility, mainly in the frontal plane, and also hip mobility in the sagittal plane, are decreased in AN. These findings provide clinically relevant insights into spinal characteristics in adolescents with AN. Full article

In this paper, a new type of soft group called the soft symmetric difference group (SSD-group) is introduced and systematically developed. This structure is constructed by integrating soft set theory with group theory through the symmetric difference operation and set inclusion. Fundamental concepts such as characteristic soft symmetric difference groups, soft symmetric difference subgroups, normal soft symmetric difference subgroups, soft normalizers, and soft cosets are defined, and their essential algebraic properties are investigated. Several characterizations of soft normality are also established through these concepts. Various axiomatic results are obtained, providing necessary and sufficient conditions for a soft set to form an SSD-group. Furthermore, soft quotient (factor) groups of SSD-groups are introduced and their structural properties are examined in detail. The relationship between SSD-group theory and classical group theory is also established through several corresponding concepts. Illustrative examples are provided to demonstrate the applicability and internal consistency of the proposed framework. Overall, the results obtained in this study extend existing soft group structures and contribute to the development of algebraic theory within the context of soft sets, while also providing a foundation for further generalizations to other algebraic frameworks such as semigroups, rings, and fields. Background: Yoga has gained popularity worldwide and is generally considered a safe physical activity. However, injuries associated with yoga practice are increasingly reported, while data on cases requiring emergency care remain limited. Methods: A retrospective single-center study was conducted, analyzing cases of yoga-related injuries treated at a Swiss emergency department between 2013 and 2023. Medical records of 67 adult patients (aged ≥16 years) were reviewed for demographics, injury characteristics, management, and clinical outcomes. The study population consisted predominantly of females (76.1%), with a median age of 35 years. Results: Most injuries were musculoskeletal in nature and predominantly affected a single body region (95.5%). The most frequently involved areas were the head (29.9%), lower extremities (25.4%), and spine (19.4%). Soft tissue injuries, particularly muscle and tendon strains as well as contusions, were most common. Injury patterns differed across subgroups: older patients were more likely to sustain head injuries, whereas younger individuals more frequently presented with extremity injuries, including the rare cases of fractures and dislocations. Conservative treatment was sufficient in 94% of cases, although 20.9% of patients required hospitalization. Conclusion: Yoga-related injuries presenting to emergency care are generally minor and mainly involve soft tissues; however, injury patterns vary across demographic subgroups. Older adults appear more susceptible to balance-related and head injuries, while younger practitioners are more prone to acute extremity trauma. Recognizing these population-specific differences may support targeted prevention strategies and safer yoga practice. Full article

Background and Objectives: This study aimed to describe the clinical characteristics and survival outcomes of three representative complex karyotype soft tissue sarcoma (STS) subtypes—undifferentiated sarcoma (US, primarily undifferentiated pleomorphic sarcoma (UPS)), myxofibrosarcoma (MFS), and leiomyosarcoma of soft tissue (LMS-ST)—using data from a single-institution cohort. Materials and Methods: A retrospective review of 124 patients treated at a single tertiary referral center between 2002 and 2024 was conducted. Clinicopathologic characteristics and survival outcomes were analyzed. Kaplan–Meier methods were used to estimate overall survival (OS). Cox proportional hazards regression models were applied to identify independent prognostic factors for survival, incorporating variables such as age, sex, tumor stage, and treatment modality. Results: The cohort comprised 36 cases of US, 64 of MFS, and 24 of LMS-ST. OS and survival after cohort enrollment (S-NCC) were evaluated both by subtype and across the entire cohort to assess potential differences across tumor subgroups. In both univariable and multivariable analyses, US subtypes showed poorer survival than MFS and LMS-ST. FNCLCC grade 3 emerged as a significant adverse prognostic factor for survival across all three subtypes. For FNCLCC grade 3 patients, the presence of benign prostatic hyperplasia (BPH) was significantly associated with an increased risk of death. Conclusions: Among the three subtypes, US demonstrated the most aggressive clinical course, MFS was notable for frequent local recurrence but relatively favorable survival, and LMS-ST showed intermediate outcomes. These findings highlight the clinical heterogeneity of complex karyotype STS and provide a foundation for future studies integrating molecular and multi-omics data to refine risk stratification and therapeutic strategies. Carbohydrates are a primary nutrient for plant growth, and sugar transporter proteins play a crucial role in sugar allocation. In this study, hexose transporter genes encoding in the genome of colored calla lily ‘Jingcai Yangguang’ (Zantedeschia elliottiana cv. Jingcai Yangguang) were identified, and their expression patterns following infection by Pectobacterium carotovora subsp. Carotovora were investigated. Additionally, the transport characteristics of three hexose transporters, ZeSTP7, ZeSTP15, and ZeSTP17, were determined. The results showed that the sugar transporter protein family in Z. elliottiana comprises 18 members, most of which possess 12 transmembrane domains. Phylogenetic analysis revealed that the ZeSTP gene family was divided into five subgroups. Tandem gene duplication events were identified on the 16 chromosomes of Z. elliottiana, with multiple tandemly duplicated genes detected. Comparative analysis of synteny between species identified ZeSTP8 and OsSTP22 as homologous gene pairs, while OsSTP6 (OsMST6) was identified as a homologous gene pair with both ZeSTP14 and ZeSTP17. Following infection by P. carotovora subsp. carotovora, the transcript levels of ZeSTP7, ZeSTP15, and ZeST17 were all significantly elevated. Yeast mutant hexose complementation tests indicated that ZeSTP7 could transport glucose and galactose, whereas ZeSTP15 and ZeSTP17 exhibited limited transport capacity in this respect. This study provides a systematic identification and analysis of hexose transporter genes at the genome-wide level, highlighting the role of ZeSTP genes in the response of colored calla lily to soft rot and laying a theoretical foundation for further understanding the functions of sugar transporter genes. Peritoneal sarcomatosis is a rare malignant disease with a poor prognosis, secondary to peritoneal dissemination of abdominopelvic soft tissue sarcomas. Its rarity, together with the characteristic histological heterogeneity and the historically poor response to systemic treatments, has prevented the establishment of widely accepted treatment criteria with curative intent. In this sense, radical cytoreductive surgery (CRS) with peritonectomy procedures and hyperthermic intraperitoneal chemotherapy (HIPEC), widely used in peritoneal carcinomatosis with excellent results, have not had the same evolutionary development in patients with peritoneal sarcomatosis. A multidisciplinary working group of experts in sarcomas and peritoneal oncological surgery established a series of recommendations based on current scientific evidence for the management of peritoneal sarcomatosis, taking into account the different histological subgroups of abdominopelvic sarcomas that can cause it depending on their origin: retroperitoneal sarcomas, uterine sarcomas, and visceral/peritoneal sarcomas of GIST (gastrointestinal stromal tumor) and non-GIST origin. This article shows the results of sarcoma experts’ voting on the recommendations presented during the I Ibero-American Consensus on the Management of Peritoneal Sarcomatosis, which took place during the recent celebration of the III Hispanic-Portuguese Meeting for Updates on the Treatment of Sarcomas. Background: Whether re-excision (RE) of a soft tissue sarcoma (STS) of limb or trunk should be systematized as adjuvant care and if it would improve metastatic free survival (MFS) are still debated. The impact of resection margins after unplanned macroscopically complete excision (UE) performed out of a NETSARC reference center or after second resection was further investigated. Methods: This large nationwide series used data from patients having experienced UE outside of a reference center from 2010 to 2019, collected in a French nationwide exhaustive prospective cohort NETSARC. Patient characteristics and survival distributions in patients reexcised (RE) or not (No-RE) are reported. Multivariate Cox proportional hazard model was conducted to adjust for classical prognosis factors. Subgroup analysis were performed to identify which patients may benefit from RE. Results: Out of 2371 patients with UE for STS performed outside NETSARC reference centers, 1692 patients were not reviewed by multidisciplinary board before treatment decision and had a second operation documented. Among them, 913 patients experienced re-excision, and 779 were not re-excised. Characteristics were significantly different regarding patient age, tumor site, size, depth, grade and histotype in patients re-excised (RE) or not (No-RE). In univariate analysis, final R0 margins are associated with a better MFS, patients with R1 margins documented at first surgery had a better MFS as compared to patients with first R0 resection. The study identified RE as an independent favorable factor for MFS (HR 0.7, 95% CI 0.53–0.93; p = 0.013). All subgroups except older patients (>70 years) and patients with large tumors (>10 cm) had superior MFS with RE. Conclusions: RE might be considered in patients with STS of limb or trunk, with UE with macroscopic complete resection performed out of a reference center, and also in originally defined R0 margin resections, to improve LRFS and MFS. Systematic RE should not be advocated for patients older than 70 years, or with tumors greater than 10 cm. Full article

Group theory is the part of mathematics which addresses the study of symmetry. This paper extends the investigation of the soft group theory which Aktaş and Çağman have defined. We study new concepts in the soft group theory such as the center of the soft group, the kernel of soft homomorphism and soft automorphisms with their basic properties. Furthermore, the concept of soft point groups is introduced and the properties of these soft groups are studied. We state the concept of characteristic soft subgroups of a given soft group. Also, some theorems related to this concept are investigated. We study the characteristic soft subgroups of a given soft point group. The characteristic soft subgroups play a significant part in the study of the soft group theory and are useful for understanding the structure of a soft group and its soft automorphisms. As an application of characteristic soft subgroups, they allow us to identify and study important soft subgroups that are preserved under soft automorphisms. Also, practical applications for our theory can be conducted in future work such as the relation with other disciplines in sciences. Full article

Background: Bone and soft-tissue tumor patients experience long-lasting physical and psychological challenges. It is unknown to what extent Health-Related Quality of Life (HRQoL) is already affected during the diagnostic process. This study assesses the HRQoL of bone and soft-tissue tumor patients around time of diagnosis and explores which patient or tumor characteristics are associated with a reduced HRQoL. Methods: All patients with a suspected benign/malignant bone tumor (BT), benign soft-tissue tumor (STT), or malignant soft-tissue sarcoma (STS) visiting the Leiden University Medical Center between 2016 and 2020 were invited to complete the Patient-Reported Outcomes Measurement Information System (PROMIS) 29-item profile questionnaire. Mean scores of all included patients and per diagnosis group were compared to mean scores of the general population using one-sample t-tests. Results: Overall, patients (n = 637) reported statistically significantly worse HRQoL-scores on anxiety (51.3 ± 9.6), pain (55.3 ± 10.1), physical functioning (46.0 ± 9.7), and social functioning (48.1 ± 10.8) with the difference in pain and physical functioning being clinically relevant (based on a 3-point difference on t-metric). HRQoL-scores differed between diagnosis subgroups, i.e., patients with malignant tumors had higher anxiety levels and experienced more pain, where patients with bone tumors had worse physical functioning. Conclusion: The HRQoL of patients with suspected bone and soft-tissue tumors is already affected during the diagnostic process. Full article

Integrated Analysis of TME and Hypoxia Identifies a Classifier to Predict Prognosis and Therapeutic Biomarkers in Soft Tissue Sarcomas

Soft tissue sarcoma (STS) is one of the rarest but most aggressive cancer. It is important to note that intratumoral hypoxia and tumor microenvironment (TME) infiltration play a significant role in the growth and therapeutic resistance of STS. The goal of this study was therefore to determine whether linking hypoxia-related parameters to TME cells could provide a more accurate prediction of prognosis and therapeutic response. An analysis of 109 hypoxia-related genes and 64 TME cells was conducted in STS. Hypoxia-TME classifier was constructed based on 6 hypoxia prognostic genes and 8 TME cells. As a result, we evaluated the prognosis, tumor, and immune characteristics, as well as the effectiveness of therapies in Hypoxia-TME-defined subgroups. The Lowplus group showed a better prognosis and therapeutic response than any other subgroup. It is possible to unravel these differences based on immune-related molecules and somatic mutations in tumors. Further validation of Hypoxia-TME was done in an additional cohort of 225 STS patients. Additionally, we identified five key genes through differential analysis and RT-qPCR, namely, ACSM5, WNT7B, CA9, MMP13, and RAC3, which could be targeted for therapy. As a whole, the Hypoxia-TME classifier demonstrated a pretreatment predictive value for prognosis and therapeutic outcome, providing new approaches to therapy strategizing for patients. Full article

Soft tissue sarcomas (STSs) are tumors that are challenging to treat due to their pathologic and molecular heterogeneity and their tumor biology that is not yet fully understood. Recent research indicates that dysregulation of cyclin-dependent kinase (CDK) signaling pathways can be a strong driver of sarcogenesis. CDKs are enzyme forms that play a crucial role in cell-cycle control and transcription. They belong to the protein kinases group and to the serine/threonine kinases subgroup. Recently identified CDK/cyclin complexes and established CDK/cyclin complexes that regulate the cell cycle are involved in the regulation of gene expression through phosphorylation of critical components of transcription and pre-mRNA processing mechanisms. The current and continually growing body of data shows that CDKs play a decisive role in tumor development and are involved in the proliferation and growth of sarcoma cells. Since the abnormal expression or activation of large numbers of CDKs is considered to be characteristic of cancer development and progression, dysregulation of the CDK signaling pathways occurs in many subtypes of STSs. This review discusses how reversal and regulation can be achieved with new therapeutics and summarizes the current evidence from studies regarding CDK modulation for STS treatment. Full article

Desmoid-type fibromatosis (DTF) is a rare, soft-tissue tumour. These tumours do not metastasize, but their local aggressive tumour growth and unpredictable behaviour can have a significant impact on health-related quality of life (HRQoL). Little is known about which DTF patients are particularly affected by an impaired HRQoL. The objectives of this study were to assess HRQoL among different groups of DTF patients and to investigate which socio-demographic and clinical characteristics were associated with DTF-specific HRQoL. A cross-sectional study was conducted among DTF patients from the United Kingdom and the Netherlands. HRQoL was assessed using the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire Core 30 (EORTC QLQ-C30), accompanied by the DTF-QoL to assess DTF-specific HRQoL. The scores were compared amongst subgroups, based on the socio-demographic and clinical characteristics of DTF patients. Multiple linear regression analyses with a backward elimination were conducted to identify the factors associated with DTF-specific HRQoL. A total of 235 DTF patients completed the questionnaires. Female patients, patients with more than two comorbidities, or patients who received treatment other than only active surveillance (AS) or surgery scored significantly worse on the subscales of both the EORTC QLQ-C30 and DTF-QoL. Patients that were ≥ 40 years scored significantly worse on the physical functioning scale of the EORTC QLQ-C30, while younger patients (18–39 years) scored significantly worse on several DTF-QoL subscales. Differences in the DTF-QoL subscales were found for tumour location, time since diagnosis and the presence of recurrent disease. Furthermore, treatments other than AS or surgery only, female sex, younger age and the presence of comorbidities were most frequently associated with worse scores on the DTF-QoL subscales. This study showed that (DTF-specific) HRQoL differs between groups of DTF patients. Awareness of these HRQoL differences could help to provide better, personalised care that is tailored to the needs of a specific subgroup. Full article

Machine learning approaches, such as soft independent modeling of class analogy (SIMCA) and pathway analysis, were introduced in depression research in the 1990s (Maes et al.) to construct neuroimmune endophenotype classes. The goal of this paper is to examine the promise of precision psychiatry to use information about a depressed person’s own pan-omics, environmental, and lifestyle data, or to tailor preventative measures and medical treatments to endophenotype subgroups of depressed patients in order to achieve the best clinical outcome for each individual. Three steps are emerging in precision medicine: (1) the optimization and refining of classical models and constructing digital twins; (2) the use of precision medicine to construct endophenotype classes and pathway phenotypes, and (3) constructing a digital self of each patient. The root cause of why precision psychiatry cannot develop into true sciences is that there is no correct (cross-validated and reliable) model of clinical depression as a serious medical disorder discriminating it from a normal emotional distress response including sadness, grief and demoralization. Here, we explain how we used (un)supervised machine learning such as partial least squares path analysis, SIMCA and factor analysis to construct (a) a new precision depression model; (b) a new endophenotype class, namely major dysmood disorder (MDMD), which is a nosological class defined by severe symptoms and neuro-oxidative toxicity; and a new pathway phenotype, namely the reoccurrence of illness (ROI) index, which is a latent vector extracted from staging characteristics (number of depression and manic episodes and suicide attempts), and (c) an ideocratic profile with personalized scores based on all MDMD features.
