Background and Clinical Significance: Inherited thrombocytopenia (IT) is a heterogeneous group of disorders caused by mutations in over 45 genes. Among these, ANKRD26-related thrombocytopenia (ANKRD26-RT) accounts for a notable subset and is associated with variable bleeding tendencies and an increased risk of myeloid malignancies. However, the extent of this oncogenic risk appears to vary between specific gene variants. Understanding the genotype–phenotype relationship is essential for patient counseling and management. This report presents a multigenerational family carrying the rare c.−118C > G variant in the 5′ untranslated region of ANKRD26, contributing to the discussion on variant-specific cancer predisposition. Case Presentation: Two sisters aged 57 and 60 presented with lifelong bleeding diathesis and moderate thrombocytopenia. Their symptoms included easy bruising, menorrhagia, and excessive postoperative bleeding. Genetic testing confirmed heterozygosity for the ANKRD26 c.−118C > G variant. Bone marrow analysis revealed abnormal megakaryopoiesis without evidence of dysplasia or somatic mutations. One sister underwent major surgery without complications when managed with prophylactic hemostatic therapy. Their family history included multiple female relatives with similar symptoms, although formal testing was limited. Notably, none of the affected individuals developed hematologic malignancy, and only one developed esophageal cancer, with no current evidence linking this variant to solid tumors. Conclusions: This case underscores the importance of distinguishing between ANKRD26 variants when assessing malignancy risk. While ANKRD26-RT is associated with myeloid neoplasms, the c.−118C > G variant may confer a lower oncogenic potential. Variant-specific risk stratification and genetic counseling are crucial for optimizing surveillance and avoiding unnecessary interventions in low-risk individuals. Full article

Early and accurate detection of mechanical damage in prunes is crucial for preserving postharvest quality and enabling automated sorting. This study proposes a practical and reproducible method for identifying cumulative bruising in prunes using visible–near-infrared (Vis–NIR) reflectance spectroscopy coupled with machine learning techniques. A self-developed impact simulation device was designed to induce progressive damage under controlled energy levels, simulating realistic postharvest handling conditions. Spectral data were collected from the equatorial region of each fruit and processed using a hybrid modeling framework comprising continuous wavelet transform (CWT) for spectral enhancement, uninformative variable elimination (UVE) for optimal wavelength selection, and support vector machine (SVM) for classification. The proposed CWT-UVE-SVM model achieved an overall classification accuracy of 93.22%, successfully distinguishing intact, mildly bruised, and cumulatively damaged samples. Notably, the results revealed nonlinear reflectance variations in the near-infrared region associated with repeated low-energy impacts, highlighting the capacity of spectral response patterns to capture progressive physiological changes. This research not only advances nondestructive detection methods for prune grading but also provides a scalable modeling strategy for cumulative mechanical damage assessment in soft horticultural products. Effectiveness and Safety of Acupuncture for Nausea and Vomiting in Cancer Patients: A Systematic Review and Meta-Analysis

Background and Objectives: Nausea and vomiting (NV) are common and distressing adverse effects among cancer patients undergoing treatment. Despite the widespread use of pharmacological antiemetics, these medications are often insufficient for controlling nausea and may cause medication interactions and side effects. Acupuncture has been proposed as a complementary therapy; however, the comprehensive analysis of its effects on NV across all emetogenic cancer treatments remains limited. This systematic review and meta-analysis aimed to evaluate the effectiveness and safety of acupuncture in managing NV in cancer patients undergoing chemotherapy, radiotherapy, or surgery. Materials and Methods: We conducted a comprehensive search across three electronic databases and two clinical registry platforms from inception to December 2024. Randomized controlled trials (RCTs) evaluating acupuncture for NV in cancer patients were included. Risk ratios (RRs) and 95% confidence intervals (CIs) were calculated using a random-effects model. Safety outcomes were assessed based on the Common Terminology Criteria for Adverse Events (CTCAE). Results: Seventeen RCTs met the inclusion criteria, with twelve studies included in the meta-analysis. Acupuncture did not demonstrate significant effects on acute nausea (RR: 0.98; 95% CI: 0.84–1.15; p = 0.80) or acute vomiting (RR: 0.93; 95% CI: 0.65–1.32; p = 0.67). However, it significantly reduced delayed vomiting (RR: 0.76; 95% CI: 0.61–0.95; p = 0.02). Subgroup analysis demonstrated significant effects when acupuncture was administered for at least five days (RR: 0.56; 95% CI: 0.39–0.81; p = 0.002). The most frequently used acupoints were PC6, ST36, CV12, LI4, LR3, and ST25. No serious adverse events related to acupuncture treatments were reported, with only minor AEs such as localized bleeding and mild bruising observed. Conclusions: Acupuncture represents a safe and effective complementary therapy for managing delayed vomiting in cancer patients receiving emetogenic treatments. Clinicians can anticipate optimal benefits from at least five days of treatment, particularly using acupoints PC6, ST36, CV12, LI4, LR3, and ST25. Further high-quality studies are needed to establish standardized treatment regimens and explore its comprehensive effects on NV. Full article

Dual-energy computed tomography (DECT) is a promising advancement for detecting bone edema-like lesions (BME). However, its application in horses remains limited. The aim of this study was to evaluate DECT virtual-non-calcium (VNCa) imaging in the equine foot and establish which confounding factors could influence its applicability in clinical practice. The DECT VNCa map of 14 standing and 5 cadaveric (recumbent) cases with foot-related lameness was scored in consensus by two readers in comparison to MRI. Overall, 17/19 cases demonstrated BME on MRI, whereas 2 did not. Agreement between DECT VNCa and MRI was found in 15/19 feet (78.9%). Disagreement in 4/19 cases with BME was due to sclerosis (1/19), mild BME extent on MRI (2/19), or scan artifacts (1/19). The extent of BME was significantly underestimated using DECT VNCa compared to MRI (p = 0.016). No significant correlation was found between sclerosis score and the BME extent underestimation on DECT (p = 0.056). Between standing and post-mortem cases, there was no significant difference in the agreement between DECT and MRI (p = 0.53) or DECT VNCa image quality (p = 0.22). In conclusion, DECT VNCa effectively identified moderate and severe BME, and its use was feasible in standing positioning. In case of sclerosis, a case-by-case assessment is recommended. Full article

Hoof disorders in small ruminants pose significant challenges to animal welfare and farm productivity. This study presents the first attempt to determine the prevalence of lameness and hoof disorders and their associated risk factors in goat and sheep farms in Selangor, Malaysia. Locomotion scores were collected from 226 animals (126 sheep and 100 goats) across 10 farms. A hoof examination was conducted, and hoof lesions were identified through detailed photographic evaluation. On-farm assessments and interviews were conducted to gather information on management practices from the farms. Data were analysed using descriptive statistics, bivariate analysis, and logistic regression models. The prevalence of lameness was 42.8% (95% CI 34.2 to 51.9) in sheep and 23.0% (95% CI 16.3–38.4) in goats. Significant variation (p > 0.05) in lameness prevalence was observed across farms, ranging from 26.7% to 61.5% in sheep and 7.7% to 30.8% in goat farms. The majority of lameness and hoof lesions were observed in the hindlimbs of both species. The prevalence of hoof disorders was 91.3% (95% CI 84.6–95.4) in sheep and 43.0% in goats (95% CI 21.4–58.0). The predominant hoof disorders were overgrown wall horn, white line disease, sole bruise, and wall fissures. No hoof affections of infectious origin were observed in the sampled animals. Risk factors for lameness and hoof lesions in sheep included pregnancy, semi-intensive management, and breeds other than Damara. Higher odds of lameness were observed in exotic goat breeds and those with overgrown wall horns. In conclusion, this study revealed a high prevalence of lameness and hoof disorders in goat and sheep farms, highlighting the need to address these important welfare and economic issues. While the identified risk factors could be considered for the management of hoof disorders in small ruminant farms, a larger sample size that is representative of the sheep and goat population is recommended for more generalizable results. Background: Patients diagnosed with coronary artery disease (CAD) have been seen to exhibit increases in health-related quality of life (HRQoL) following percutaneous coronary interventions (PCIs). This study thus aimed to assess the impact of PCI on health outcomes among Jordanian patients three months post-procedure. Methods: This prospective descriptive study evaluated health outcomes three months post-PCI among Jordanian patients who had been originally diagnosed with Chronic Coronary Syndrome (CCS) before being scheduled for PCIs. Quantitative data was collected using the updated version of the Coronary Revascularization Outcome Questionnaire (CROQ v2) across a non-probability sample, based on accessibility, of Jordanian patients who had received the procedure at any of several hospitals in Jordan. Multivariate analysis of covariance (MANCOVA) was employed to examine the mean scores of patient-reported outcomes following revascularization, while partial correlations were employed to investigate associations among patients’ age, weight, gender, and the reported results. Results: A total of 101 patients participated in the study, with a predominance of males (n = 85, 84.2%) relative to females (n = 16, 15.8%). The results indicated a statistically significant improvement across all measures assessed across these patients. Furthermore, the results demonstrated that males showed higher physical function, psychological functioning, and cognitive performance relative to females following coronary revascularization surgery. Nevertheless, the results also revealed varied levels of adverse effects following coronary revascularization, with the most commonly reported being discomfort around the groin or arm wound, followed by pain in the same areas. Conversely, the least significant concerns pertained to the emergence of bruises and similar issues in the groin or arm areas where the catheter was inserted. Conclusions: This study shows that PCI improves CAD patients’ quality of life over the initial three-month period post-procedure. Understanding the positive associations of this and the negative consequences that it entails may help healthcare practitioners better identify those patients likely to benefit or suffer from PCI, enabling more appropriate interventions. To understand how PCI affects HRQoL in CAD patients over time, more research based on rigorous study designs and validated metrics is required, however. Full article

Background/Objectives: Calcium hydroxylapatite (CaHA)-based dermal fillers are widely used in esthetic medicine due to their dual volumizing and biostimulatory properties. Despite their rising popularity, comparative clinical outcome data evaluating different CaHA fillers remain limited. This prospective, single-center, unblinded study aimed to establish the effectiveness of the CaHA filler Rennova® Diamond Intense via comparison to the well-established CaHA filler Radiesse® in terms of clinical performance, safety, and patient satisfaction. Methods: Thirty patients (28 female, 2 male) underwent a single-session bilateral injection of Rennova® Diamond Intense (right side) and Radiesse® (left side) in the lower and medial posterior facial regions. Outcomes evaluated at multiple time points included dermal thickness, skin elasticity, transepidermal water loss, patient-reported outcomes (S-GAIS), physician-assessed outcomes (P-GAIS), and adverse events. Results: Both fillers showed improvements across all parameters. Patient-reported S-GAIS indicated predominantly “improved” outcomes at days 60 and 120, whereas physician assessments (P-GAIS) predominantly indicated “very improved” results at day 120. Ultrasound revealed increases in dermal thickness for both fillers. Similarly, improvements in skin elasticity and decreases in transepidermal water loss were observed bilaterally. Mild, transient adverse events (pain, swelling, redness, bruising) resolved spontaneously within 30 days post injection. Conclusions: Rennova® Diamond Intense effectively increases dermal thickness, improves skin elasticity, and reduces transepidermal water loss, achieving high patient and physician satisfaction. These findings underscore its safety, versatility, and efficacy for esthetic facial rejuvenation, warranting further long-term evaluation. Full article

In forensic pathology, glycophorin A (GPA) presence in tissues has been studied through anti-GPA monoclonal antibodies with immunohistochemical techniques. The use of anti-GPA in distinguishing ante- from postmortem lesions, particularly in advanced stages of decomposition, is explored in this study. This systematic review assesses the current use of glycophorins, focusing on their application in forensic investigations to detect vital hemorrhagic infiltration. A total of 799 studies were identified, and after screening, 16 studies were included, encompassing case reports, experimental studies, and case-control analyses. The data extracted from these studies highlighted the diagnostic accuracy of GPA immunohistochemical staining in forensic cases. Of the 50 specimens analyzed, 48 were stained with anti-GPA serum and 2 with anti-glycophorin C (GPC) serum. The results showed that GPA staining was significantly more reliable in detecting vital hemorrhage compared to routine histology, which had a diagnostic accuracy of only 66%. Moreover, in an experimental study, GPA positivity was observed in 72.2% of bruises and vital tissues, even in highly decomposed specimens. The study further emphasized the robustness of GPA in distinguishing ante- from postmortem injuries, with particular relevance for cases involving significant decomposition. Overall, GPA’s high sensitivity in detecting vital hemorrhage, especially in decomposed bodies, underscores its potential as a reliable tool in forensic pathology. Despite some limitations due to the small number of studies, the evidence supports the inclusion of anti-GPA antibodies in routine forensic examinations, potentially enhancing the accuracy of wound vitality assessments in criminal investigations. Cumulative Ambient Light Exposure Affects Outpatient Transcutaneous Bilirubinometer Readings

Background: We recently reported that the transcutaneous bilirubinometer (TCB) tends to underestimate the severity of neonatal jaundice (NJ). We hypothesize that the cumulative ambient light exposure contributes to the discrepancy. Objectives: This study aimed to identify factors that affect the TCB underestimation. Methods: We analyzed prospectively collected data over a twenty-month period at a level III medical facility. Neonates at risk for NJ who couldn’t secure an appointment with the primary practitioner were followed by the nursery team. Neonates who had phototherapy or forehead bruises were excluded. Concurrently collected total serum bilirubin (TSB) was determined by the diazo method. The primary endpoint was the discrepancy between TCB and the corresponding TSB (TCB-TSB). A mixed-effects model was used to assess the correlation between (TCB-TSB) and potential contributors, including visit age (in hours), gestational age (GA), sex, TSB, season, birth weight, and race. Results: There were 795 visits for 559 neonates, including 341 males, 179 white, 235 black, 103 Hispanic, 41 Asian, and one unrecorded race. The TSB ranged between 1.8 and 33.9 mg/dL. The (TCB-TSB) ranged between −20.0 and 6.4 mg/dL. The median GA and birth weight were 38.7 weeks and 3214.5 g. The visits occurred between 48 and 381 h of age. 133, 148, 132, and 146 visits were in Spring, Summer, Autumn, and Winter, respectively. Fifty-four neonates (9.7%) were admitted for management. 500 sternum TCB readings were also collected from 350 neonates together with the corresponding forehead TCBs. We found that the forehead (TCB-TSB) was significantly less in winter than in spring and summer (p = 0.0014 and 0.0003, respectively). There was a negative correlation between forehead (TCB-TSB) and visit age in hours (p = 0.0006). After adjusting for visit age and season, the (TCB-TSB) is significantly correlated with TSB (p < 0.0001). Similar findings were also seen in the sternum (TCB-TSB) except for the season (p = 0.0808). Conclusions: Cumulative ambient light exposure and the severity of NJ may contribute to (TCB-TSB). Full article

With aging, there come diminishing changes in vascular microcirculation and the accumulation of hemoglobin by-products that gather in the fatty pads beneath the eyes as dark circles, akin to skin bruising. In addition, the extracellular matrix that surrounds the eye is exposed to external threats like UV radiation, weather and pollution, as well as lifestyle choices that create fatigue. This causes the eyes to express wrinkles well before they begin to appear on the rest of the face, particularly in the corners of the eyes called the crow’s feet region. Consumers spend considerable amounts of resources combatting these effects. If consumers could treat some of the sources of these problems, in advance of the inevitable influences of aging, a kind of prejuvenation of the eye infrastructure, then perhaps the inevitable outcomes of aging apparent around the eyes could be slowed. This paper examines the development and in vitro testing of two unique botanical extracts, one based on a traditional medicine mushroom called Phellinus linteus (Huang Sang) and the other based on a traditional medicine root from the plant Angelica polymorpha sinensis (Dong Quai). When combined, these two extracts create a blend called ANGEL-EYE EFX® [INCI: Water (and) Glycerin (and) Phellinus Linteus Extract (and) Angelica polymorpha sinensis Root Extract]. There are several key biomolecules of interest present in this blend, including hispolon, dihydrozingerone, and arginine, as demonstrated using advanced liquid chromatography/mass spectral analyses. The individual extracts were also broadly examined using human genomic microarray assays and then more specifically for their ability to influence several important skin proteins associated with undereye skin aging, including CYGB (Human Cytoglobin), OXSR1 (Oxidative Stress Response Kinase-1), LCE3B (Late Cornified Envelope-3B), EGFR (Epidermal Growth Factor Receptor), VEGFA (Vascular Endothelial Growth Factor-1), and NINJ1 (Ninjurin-1). It was found that the treatment of Normal Human Epidermal Keratinocytes (NHEKs) with increasing concentrations of the active blend between 0.05 and 2.0% showed statistically significant increases in all the proteins noted except VEGFA, which showed a statistically significant decrease in protein expression with the treatment of the Angelica polymorpha sinensis extract at 1.0%. This study aims to identify the prevalence and patterns of sports injuries in recreational sports activities in South Korea. This study utilized data from the “survey of safety accidents” conducted by the Korea Sports Safety Foundation and finally, 3182 recreational sports participants who experienced injuries were selected for the study. For data processing, data related to recreational sports injuries were first collected and organized using Excel 2015, and frequency analysis was conducted using the SPSS 25.0 program. Furthermore, the association rule method was applied via Python 3.13.3 to analyze the patterns of injury sites and types. First, by investigating the prevalence of injuries in recreational sports, it was found that the injury frequency was highest in soccer, followed by cycling, hiking, and badminton. Second, in soccer, it was found that when ankle injuries, which have a high injury frequency, occur, knee, toe, and sprain injuries also occur together (Lift: 1.843). Additionally, in cycling, when knee injuries occur, toe, sprain, and strain (bruise) injuries also occur together (Lift: 2.420). In mountain biking, when ankle injuries, which have a high injury frequency, occur, cuts, sprains, stab wounds (cuts), sprains, and strains (bruises) also occur together (Lift: 1.808). The current survey on recreational sports injuries is expected to be used as basic data to prevent injuries in advance for participants in recreational sports, and it is expected that this will allow them to participate in sports by recognizing common injury sites before participating in sports. Background: Hyaluronic acid (HA)–based bioactive hydrogels have emerged as multifunctional platforms for skin bioregeneration. While traditional mesotherapy using multicomponent substances has been widely practiced for improving skin quality, the time-consuming nature of this approach has led to exploration of alternative delivery methods. Aims: This study evaluated the clinical effectiveness of an HA bioactive hydrogel-based bioregeneration system (containing non-stabilized hyaluronic acid and 14 bioactive ingredients) administered via cannula and its impact on facial skin density as assessed by ultrasound imaging. Methods: We conducted a retrospective review of data from 20 female patients aged 30–42 years who received a single cannula-delivered injection of a bioactive hyaluronic acid hydrogel (TEOSYAL® Redensity [I]) in the midface region. The formulation combines the structural benefits of hyaluronic acid with the biochemical stimulation provided by amino acids, antioxidants, minerals, and vitamins. Skin density was measured using high-frequency ultrasound at baseline, immediately post-procedure, and at 3–4 weeks follow-up. A control group of seven individuals received no treatment. Results: Ultrasound assessments revealed a statistically significant increase in skin density (92.7%, p < 0.001) within the treated area compared to no significant changes in the control group. This substantial improvement in dermal architecture demonstrates the efficacy of bioactive hydrogels in stimulating fibroblast function and extracellular matrix regeneration. Patient satisfaction was high, with 85% of patients reporting being satisfied or very satisfied. Side effects were minimal, with minor bruising (10%) and transient swelling (15%). Conclusions: Cannula-delivered bioactive hyaluronic acid hydrogel effectively enhances facial skin density with high patient satisfaction and minimal downtime, demonstrating the potential of advanced hydrogel formulations as multifunctional therapeutic platforms that extend beyond traditional applications into aesthetic and regenerative dermatology. CRISPR/Cas9-Mediated Development of Potato Varieties with Long-Term Cold Storage and Bruising Resistance

Enzymatic browning and cold-induced sweetening (CIS) affect the post-harvest quality of potato tubers. Browning is caused by polyphenol oxidase 2 (PPO2), which is activated by mechanical damage during harvest and storage. CIS occurs when vacuolar invertase converts sucrose into reducing sugars, which react with amino acids during frying, forming brown pigments and acrylamide. While cold storage prevents sprouting and disease, it also increases vacuolar invertase expression, leading to quality loss. Using CRISPR/Cas9, we developed gene-edited potato lines with improved resistance to browning and CIS. Line 6A (cv. Atlantic) and E03-3 (cv. Spunta) exhibited complete vacuolar invertase (InvVac) knockout, maintaining chip quality for at least 60 days at 4 °C. Line 6A, renamed PIRU INTA, was tested in field trials and preserved frying quality for up to 90 days under cold storage. PIRU INTA is currently undergoing registration as a new variety. Additionally, lines E04-5B and E03-3 (cv. Spunta) showed partial PPO2 gene edits, reducing enzymatic browning by 80% and 40%, respectively. This study demonstrates the potential of CRISPR/Cas9 to develop non-transgenic, gene-edited potatoes with enhanced storage quality, benefiting both growers and the food industry. Full article
