Insect pests cause severe damage and yield losses to many agricultural crops globally. The use of chemical pesticides on agricultural crops is not recommended because of their toxic effects on the environment and consumers. In addition, pesticide toxicity reduces soil fertility, poisons ground waters, and is hazardous to soil biota. Therefore, applications of entomopathogenic nematodes (EPNs) and plant growth-promoting rhizobacteria (PGPR) are an alternative, eco-friendly solution to chemical pesticides and mineral-based fertilizers to enhance plant health and promote sustainable food security. This review focuses on the biological and ecological aspects of these organisms while also highlighting the practical application of molecular communication approaches in developing a novel plant health product. This insight will support this innovative approach that combines PGPR and EPNs for sustainable crop production. Several studies have reported positive interactions between nematodes and bacteria. Although the combined presence of both organisms has been shown to promote plant growth, the molecular interactions between them are still under investigation. Integrating molecular communication studies in the development of a new product could help in understanding their relationships and, in turn, support the combination of these organisms into a single plant health product. A thermosensitive collagen-based gel (TSV gel), containing type I and III collagen, has been developed to improve the handling and stability of bone graft materials. However, its direct effect on osteoblasts is not well understood. This in vitro study evaluated the biological response of human oral osteoblasts to four bone substitutes: OsteoBiol® GTO® (larger granules with 20% TSV gel), Gen-OS® (smaller granules), Gen-OS® combined with 50% TSV gel (Gen-OS®+TSV), and TSV gel alone. Cell proliferation, adhesion, morphology, collagen and calcium deposition, alkaline phosphatase (ALP) activity, gene expression of osteogenic markers and integrins, and changes in pH and extracellular calcium and phosphate levels were investigated. All materials supported osteoblast activity, but Gen-OS+TSV and GTO showed the most pronounced effects. These two groups promoted better cell adhesion and proliferation, higher ALP activity, and greater matrix mineralization. GTO improved cell adhesion, while the addition of TSV gel to Gen-OS enhanced biological responses compared with Gen-OS alone. Integrins α2, α5, β1, and β3, important for cell attachment to collagen, were notably upregulated in Gen-OS+TSV and GTO. Both groups also showed increased expression of osteogenic markers such as BMP-2, ALP, and osteocalcin (OCN). Higher extracellular ion concentrations and a more alkaline pH were observed, particularly in conditions without cells, suggesting active ion uptake by osteoblasts. In conclusion, combining TSV gel with collagen-based granules improves the cellular environment for osteoblast activity and may support bone regeneration more effectively than using either component alone. Influence of Mineral Fertilizers and Application Methods on Raspberry Composition Cultivated in an Acid Soil

Acid soils are often a limiting factor in the production of most cultivated plants. In practice, the application of inadequate, physiologically acidic fertilizers, urea and NPK, is often encountered, which further worsens the already poor physicochemical properties of such soils. In this study, the influence of different amounts of NPK and urea fertilizers and methods of their application on the chemical properties of a very acidic soil and the accumulation of essential biogenic elements (N, P, K, Ca, Mg, and Al) in raspberry plants (leaves and fruits) was evaluated. The field trial with the raspberry plants was set up on a very acidic soil (pH in KCl 3.6), type Dystric Cambisol, and was monitored for 2 years. The application of NPK and urea mainly increased soil acidity in the second year in all treatments (for 0.10–0.18 pH unit) (except for urea applied in rows). The application of higher amounts of NPK increased the content of available forms of P (for 9.3–30.8 mg/kg) and K (for 57–95 mg/kg) in soil in both years, as well as exchangeable Ca (for 200–510 mg/kg) and Mg in the first year (15–165 mg/kg). The introduction of fertilizers in rows, compared to fertilization of the entire surface, influenced the reduction in mobile Al (especially when applying NPK, from 5.89 to 7.13 mg/100 g), the increase in mineral N and K content in the soil, and the increase in Ca and Mg only when applying urea, i.e., P when applying NPK in rows. In the leaves, the application of fertilizers in rows increased the content of Ca and Mg in the first year and P and K in the second year. In the fruits, the content of all estimated elements was not in correlation with their content in leaves and the fertilizer application, which indicates the influence of other ecological and biological factors on plant nutrition. The purpose of this work was to obtain specialized enrichment cultures from an original extreme acidophilic consortium of extremely acidophilic microorganisms and to study their microbial community composition and biotechnological potential. At temperatures of 25, 35, 40 and 50 °C, distinct enrichments of extremely acidophilic microorganisms used in the processes of bioleaching sulfide ores were obtained using nutrient media containing ferrous sulfate, elemental sulfur and a copper sulfide concentrate as nutrient inorganic substrates, with and without the addition of 0.02% yeast extract. The microbial community composition was studied using the sequencing of the V3–V4 hypervariable region of the 16S rRNA genes. The different growth conditions led to changes in the microbial composition and relative abundance of mesophilic and moderately thermophilic, strict autotrophic and mixotrophic microorganisms in members of the genera Acidithiobacillus, Sulfobacillus, Leptospirillum, Acidibacillus, Ferroplasma and Cuniculiplasma. The dynamics of the oxidation of ferrous iron, sulfur, and sulfide minerals (pyrite and chalcopyrite) by the enrichments was also studied in the temperature range of 25 to 50 °C. The study of enrichment cultures using the molecular biological method using the metabarcoding method of variable V3–24 V4 fragments of 16S rRNA genes showed that enrichment cultures obtained under different conditions differed in composition, which can be explained by differences in the physiological properties of the identified microorganisms. Regarding the dynamics of the oxidation of ferrous ions, sulfur, and sulfide minerals (pyrite and chalcopyrite), each enrichment culture was studied at a temperature range of 25 to 50 °C and indicated that all obtained enrichments were capable of oxidizing ferrous iron, sulfur and minerals at different rates. The obtained enrichment cultures may be used in further work to increase bioleaching by using the suitable inoculum for the temperature and process conditions. Full article

Over the years, macromycete fungi have been used as a source of food, part of religious rites and rituals, and as a medicinal remedy. Species with strong health-promoting potential include Hericium erinaceus, Cordyceps militaris, Ganoderma lucidum, Pleurotus ostreatus, Flammulina velutipes, and Inonotus obliquus. These species contain many bioactive compounds, including β-glucans, endo- and exogenous amino acids, polyphenols, terpenoids, sterols, B vitamins, minerals, and lovastatin. The level of some biologically active substances is species-specific, e.g., hericenones and erinacines, which have neuroprotective properties, and supporting the production of nerve growth factor in the brain for Hericium erinaceus. Due to their high health-promoting potential, mushrooms and substances isolated from them have found applications in livestock nutrition, improving their welfare and productivity. This phenomenon may be of particular importance in the nutrition of laying hens and broiler chickens, where an increase in pathogen resistance to antibiotics has been observed in recent years. Gallus gallus domesticus is a key farm animal for meat and egg production, so the search for new compounds to support bird health is important for food safety. Studies conducted to date indicate that feed supplementation with mushrooms has a beneficial effect on, among other things, bird weight gain; bone mineralisation; and meat and egg quality, including the lipid profile and protein content and shell thickness, and promotes the development of beneficial microbiota, thereby increasing immunity. Wildfires can significantly alter soil physicochemical conditions and microbial communities in forest ecosystems. This study aimed to characterize the culturable soil fungal community and evaluate biological activity in Banco Totumo Bijibana, a protected dry tropical forest in Atlántico, Colombia, affected by a wildfire in 2014. Twenty soil samples were collected for microbiological (10 cm depth) and physicochemical (30 cm) analysis. Basal respiration was measured using Stotzky’s method, nitrogen mineralization via Rawls’ method, and fungal diversity through culture-based identification and colony-forming unit (CFU) counts. Diversity was assessed using Simpson, Shannon–Weaver, and ACE indices. The soils presented low organic matter (0.70%) and nitrogen content (0.035%), with reduced biological activity as indicated by basal respiration (0.12 kg C ha−1 d−1) and mineralized nitrogen (5.61 kg ha−1). Four fungal morphotypes, likely from the genus Aspergillus, were identified. Simpson index indicated moderate dominance, while Shannon–Weaver values reflected low diversity. Correlation analysis showed Aspergillus-3 was positively associated with moisture, whereas Aspergillus-4 correlated negatively with pH and sand content. The species accumulation curve reached an asymptote, suggesting an adequate sampling effort. Although no control site was included, the findings provide a baseline characterization of post-fire soil microbial structure and function in a dry tropical ecosystem. Full article

Climbing vines have recently induced increasing threats to forest growth under favourable environmental changes. In mangrove forests, the native vine Derris trifoliata became invasive and is now one of the main threats. Yet current management relies on manual removal with low efficiency. Exploring an alternative, cost-effective method is required. To assess the potential of a proposed biological control method, this study performed a pot-plant experiment using Cuscuta japonica to infect D. trifoliata and three common mangrove species in Beihai, China. Results showed that D. trifoliata had a higher infection rate and high host mortality (90%) than mangrove (0%). It also had significantly decreased moisture by 4%, nitrogen by 14%, phosphorus by 27%, potassium by 49% and increased soluble sugar by 49% and protein by 20%, whereas only moisture (2% reduction) and one or two minerals of Excoecaria agallocha and Aegiceras corniculatum were influenced. Only Kandelia obovata had neither effective haustoria nor any nutrients impact from the infection. This study indicated that C. japonica can cause more damage to D. trifoliata than to mangrove species and has the potential to be used as a biological control agent for the threatened mangrove forests of A. corniculatum and K. obovata with monitoring and control. This study explored the extraction, characterization, and biological properties of polysaccharides derived from Spirulina (Limnospira platensis), a microalga known for its rich nutritional benefits. Polysaccharides were successfully isolated and characterized using optimized biorefinery water extraction techniques to detail their structural and functional characteristics. Results revealed notable antioxidant activity and effective α-glucosidase inhibition, indicating potential health benefits. X-ray fluorescence (XRF) analysis was conducted to assess the elemental composition, offering insights into the mineral contents of the polysaccharides. Our findings underscore the promising applications of polysaccharides from Limnospira platensis as functional ingredients in health-related fields, advocating the need for further research into their mechanisms of action and therapeutic applications. Here, we report the discovery of silica- and sulfur-enriched deposits forming on the seafloor off Zannone Island (western Mediterranean Sea), where hydrothermal activity is ongoing. Our multidisciplinary investigation reveals that these deposits form through the interplay between hydrothermal processes and microbial activity. The deposits result from a dynamic equilibrium involving microbial mediation, sedimentation, and episodic lithification, driven primarily by two mineralization pathways: silica and sulfur precipitation. This study provides new insights into the bio-sedimentary processes shaping authigenic crusts in shallow submarine hydrothermal settings, contributing to a broader understanding of mineralization in marine environments influenced by both geological and biological factors. The food industry consumes large amounts of water across various processes, and generates wastewater characterized by parameters like biochemical oxygen demand, chemical oxygen demand, pH, suspended solids, and nutrients. To meet environmental standards and enable reuse or valorization, treatment methods such as physicochemical, biological, and membrane-based processes are applied. This review focuses on the valorization of food industry wastewater in the biotechnological production of high-value products, with an emphasis on starch-rich wastewater, wineries and confectionery industry wastewater, and with a focus on new technologies for reduces environmental burden but also supports circular economy principles. Starch-rich wastewaters, particularly those generated by the potato processing industry, offer considerable potential for biotechnological valorization due to their high content of soluble starch, proteins, organic acids, minerals, and lipids. These effluents can be efficiently converted by various fungi (e.g., Aspergillus, Trichoderma) and yeasts (e.g., Rhodotorula, Candida) into value-added products such as lipids for biodiesel, organic acids, microbial proteins, carotenoids, and biofungicides. Similarly, winery wastewaters, characterized by elevated concentrations of sugars and polyphenols, have been successfully utilized as medium for microbial cultivation and product synthesis. Microorganisms belonging to the genera Aspergillus, Trichoderma, Chlorella, Klebsiella, and Xanthomonas have demonstrated the ability to transform these effluents into biofuels, microbial biomass, biopolymers, and proteins, contributing to sustainable bioprocess development. Additionally, wastewater from the confectionery industry, rich in sugars, proteins, and lipids, serves as a favorable fermentation medium for the production of xanthan gum, bioethanol, biopesticides, and bioplastics (e.g., PHA and PHB). Microorganisms of the genera Xanthomonas, Bacillus, Zymomonas, and Cupriavidus are commonly employed in these processes. Although there are still certain regulatory issues, research gaps, and the need for more detailed economic analysis and kinetics of such production, we can conclude that this type of biotechnological production on waste streams has great potential, contributing to environmental sustainability and advancing the principles of the circular economy. Osteoporosis is a multifactorial, polygenic disease characterized by reduced bone mineral density (BMD) and increased fracture risk. Genome-wide association studies (GWASs) have identified numerous loci associated with BMD and/or bone fractures, but functional characterization of these target genes is essential to understand the biological mechanisms underlying osteoporosis. This review focuses on current methodologies and key examples of successful functional studies aimed at evaluating gene function in osteoporosis research. Functional evaluation typically follows a multi-step approach. In silico analyses using omics datasets expression quantitative trait loci (eQTLs), protein quantitative trait loci (pQTLs), and DNA methylation quantitative trait loci (mQTLs) help prioritize candidate genes and predict relevant biological pathways. In vitro models, including immortalized bone-derived cell lines and primary mesenchymal stem cells (MSCs), are used to explore gene function in osteogenesis. Advanced three-dimensional culture systems provide additional physiological relevance for studying bone-related cellular processes. In situ analyses of patient-derived bone and muscle tissues offer validation in a disease-relevant context, while in vivo studies using mouse and zebrafish models enable comprehensive assessment of gene function in skeletal development and maintenance. Integration of these complementary methodologies helps translate GWAS findings into biological insights and supports the identification of novel therapeutic targets for osteoporosis. Full article

This review paper presents a comprehensive overview of DNA preservation in hard tissues (bones and teeth) for applications in forensic and archaeogenetic analyses. It presents bone structure, DNA location in bones and teeth, and extensive information about postmortem DNA location and preservation. Aged bones are a challenging biological material for DNA isolation due to their low DNA content, degraded DNA, and the potential presence of PCR inhibitors. In addition, the binding of DNA to the mineral matrix necessitates the inclusion of a demineralization process in extraction, and its contribution to the resulting increase in both DNA quality and quantity is explained. Guidelines and recommendations on bone sample selection to obtain higher DNA yields are discussed in terms of past, recent, and possible future recommendations. Interskeletal and intraskeletal differences in DNA yield are also explained. Recent studies have shown that current recommendations for the genetic identification of skeletal remains, including femurs, tibias, and teeth, may not be the most effective sampling approach. Moreover, when mass disasters and mass graves with commingled skeletal remains are considered, there is a greater possibility that the recommended set of skeletal elements will not be available for sampling and subsequent genetic testing. This review highlights interskeletal and intraskeletal variability in DNA yield, with a focus on studies conducted on poorly preserved skeletal remains, including both postwar (1945) victims from Slovenia and ancient human skeletons. Special emphasis is placed on anatomical differences and potential mechanisms influencing DNA preservation, as demonstrated in research on both modern and historical skeletons. Finally, the petrous part of the temporal bone and tooth cementum were reviewed in greater detail because they have been recognized as an optimal sampling type in both ancient DNA studies and routine forensic case analyses. Our experiences with the Second World War and archaeological petrous bones are discussed and compared to those of other bone types. Full article

Azospirillum is a well-studied genus of plant growth-promoting rhizobacteria (PGPR) and one of the most extensively researched diazotrophs. This genus can colonize rhizosphere soil and enhance plant growth and productivity by supplying essential nutrients to the host. Azospirillum–plant interactions involve multiple mechanisms, including nitrogen fixation, the production of phytohormones (auxins, cytokinins, indole acetic acid (IAA), and gibberellins), plant growth regulators, siderophore production, phosphate solubilization, and the synthesis of various bioactive molecules, such as flavonoids, hydrogen cyanide (HCN), and catalase. Thus, Azospirillum is involved in plant growth and development. The genus Azospirillum also enhances membrane activity by modifying the composition of membrane phospholipids and fatty acids, thereby ensuring membrane fluidity under water deficiency. It promotes the development of adventitious root systems, increases mineral and water uptake, mitigates environmental stressors (both biotic and abiotic), and exhibits antipathogenic activity. Biological nitrogen fixation (BNF) is the primary mechanism of Azospirillum, which is governed by structural nif genes present in all diazotrophic species. Globally, Azospirillum spp. are widely used as inoculants for commercial crop production. It is considered a non-pathogenic bacterium that can be utilized as a biofertilizer for a variety of crops, particularly cereals and grasses such as rice and wheat, which are economically significant for agriculture. Furthermore, Azospirillum spp. influence gene expression pathways in plants, enhancing their resistance to biotic and abiotic stressors. Advances in genomics and transcriptomics have provided new insights into plant-microbe interactions. This review explored the molecular mechanisms underlying the role of Azospirillum spp. in plant growth. Additionally, BNF phytohormone synthesis, root architecture modification for nutrient uptake and stress tolerance, and immobilization for enhanced crop production are also important. A deeper understanding of the molecular basis of Azospirillum in biofertilizer and biostimulant development, as well as genetically engineered and immobilized strains for improved phosphate solubilization and nitrogen fixation, will contribute to sustainable agricultural practices and help to meet global food security demands. Buckwheat, a dicotyledonous pseudocereal from the Polygonaceae family, has emerged as a crop of scientific and industrial interest due to its exceptional phytochemical profile, adaptability to different environments, and minimal agronomic input requirements. This paper aims to highlight the proximate composition (carbohydrates, protein, dietary fiber, lipids, starch, vitamins, and minerals) of the buckwheat principal species, Fagopyrum esculentum Moench (common buckwheat) and Fagopyrum tataricum (L.) Gaertn (Tartary buckwheat). Other bioactive compounds, including flavonoids (e.g., rutin, quercetin), phenolic acids, and anthocyanins, were emphasized, together with their influence on human health. These constituents confer a broad range of biological activities such as anti-inflammatory, antimicrobial, antidiabetic, antihypertensive, and hypoglycemic effects. Moreover, buckwheat is inherently gluten-free, making it a valuable alternative in formulations targeting gluten-sensitive populations. Finally, the review addresses the possibility of using starch buckwheat as a raw material in starch-based films. Further research is needed to elucidate the potential of buckwheat starch as a viable material for the development of biodegradable food packaging films. Full article

Effect of Farming System and Irrigation on Physicochemical and Biological Properties of Soil Under Spring Wheat Crops

A field experiment in growing spring wheat (Triticum aestivum L.—cv. ‘Monsun’) under organic, integrated and conventional farming systems was conducted over the period of 2020–2022 at the Czesławice Experimental Farm (Lubelskie Voivodeship, Poland). The first experimental factor analyzed was the farming system: A. organic system (control)—without the use of chemical plant protection products and NPK mineral fertilization; B. conventional system—the use of plant protection products and NPK fertilization in the range and doses recommended for spring wheat; C. integrated system—use of plant protection products and NPK fertilization in an “economical” way—doses reduced by 50%. The second experimental factor was irrigation strategy: 1. no irrigation—control; 2. double irrigation; 3. multiple irrigation The aim of the research was to determine the physical, chemical, and enzymatic properties of loess soil under spring wheat crops as influenced by the factors listed above. The highest organic C content of the soil (1.11%) was determined in the integrated system with multiple irrigation of spring wheat, whereas the lowest one (0.77%)—in the conventional system without irrigation. In the conventional system, the highest contents of total N (0.15%), P (131.4 mg kg−1), and K (269.6 mg kg−1) in the soil were determined under conditions of multiple irrigation. In turn, the organic system facilitated the highest contents of Mg, B, Cu, Mn, and Zn in the soil, especially upon multiple irrigation of crops. It also had the most beneficial effect on the evaluated physical parameters of the soil. In each farming system, the multiple irrigation of spring wheat significantly increased moisture content, density, and compaction of the soil and also improved its total sorption capacity (particularly in the integrated system). The highest count of beneficial fungi, the lowest population number of pathogenic fungi, and the highest count of actinobacteria were recorded in the soil from the organic system. Activity of soil enzymes was the highest in the integrated system, followed by the organic system—particularly upon multiple irrigation of crops. Summing up, the present study results demonstrate varied effects of the farming systems on the quality and health of loess soil. From a scientific point of view, the integrated farming system ensures the most stable and balanced physicochemical and biological parameters of the soil due to the sufficient amount of nutrients supplied to the soil and the minimized impact of chemical plant protection products on the soil. The multiple irrigation of crops resulting from indications of soil moisture sensors mounted on plots (indicating the real need for irrigation) contributed to the improvement of almost all analyzed soil quality indices. Multiple irrigation generated high costs, but in combination with fertilization and chemical crop protection (conventional and integrated system), it influenced the high productivity of spring wheat and compensated for the incurred costs (the greatest profit).
