Slaughterhouse detection of lesions compatible with bovine tuberculosis represents a key passive surveillance component in Santa Catarina, Brazil, yet subsequent trace-back investigations often fail to identify infected farms. This study developed a quantitative framework to prioritize epidemiological investigations by estimating the probability of infection associated with each farm connected to PCR-confirmed cases. Using official movement records and historical diagnostic data, we reconstructed the lifetime contact networks of slaughtered cattle presenting confirmed Mycobacterium bovis lesions (n = 502). For each sentinel animal–farm interaction (n = 1452), infection probability was estimated through a non-homogeneous Poisson process incorporating exposure duration and the time-weighted average herd size as determinants of infectious pressure. After evaluating stochastic variability through Monte Carlo simulation, a deterministic model using the mean infectious-pressure parameter was applied to classify farms into high-, medium-, and low-risk categories. Model performance was assessed using validated field diagnostic outcomes within a three-year temporal window. High-risk farms represented most validated contacts (58%) and demonstrated a relative risk of 3.48 compared with lower-risk category. These findings indicate that a standardized risk-based classification can substantially improve the prioritization of trace-back investigations, offering a practical decision-support tool to enhance bovine tuberculosis surveillance and contribute to eradication strategies in Santa Catarina. The rapid global spread of antimicrobial resistance among pathogenic microorganisms poses a serious challenge to both human and animal health, underscoring the urgent need for new strategies to combat resistance. Antimicrobial peptides (AMPs), key components of the innate immune system, are promising candidates because they disrupt the membranes of bacteria, fungi, and viruses, thereby reducing the risk of resistance development. Lactoferrin (LF), a multifunctional iron-binding glycoprotein abundant in mammalian milk, is a rich source of AMPs. Cationic peptide fragments such as lactoferricin and lactoferrampin exhibit more potent direct antimicrobial activity than the intact protein. Our previous studies have shown that peptides derived from Equine milk lactoferrin exhibit antihypertensive, anti-inflammatory, and anti-oncogenic activity in silico, highlighting their multifunctional bioactive potential. Building on these results, the present study aims to investigate the antimicrobial properties of these peptides. We used an integrated approach combining computer modeling and in vitro studies to identify and validate novel antimicrobial peptides from equine milk lactoferrin. Bioinformatics tools, including AMPScanner and CAMP, were used to predict antimicrobial domains, followed by experimental testing against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The results showed that equine milk lactoferrin peptides possess potent and selective antimicrobial activity, with efficacy varying across bacterial species. These data expand the functional profile of lactoferrin-derived peptides, demonstrating their multifunctionality, and suggest that equine milk lactoferrin represents a promising natural source of antimicrobial agents, supporting alternative strategies to reduce antibiotic use in human and veterinary medicine. The transition toward sustainable food systems requires innovative approaches to managing perishable products, where inefficient inventory practices contribute significantly to global food loss and environmental degradation. This study develops a circular-economy-oriented inventory optimisation framework for dairy supply chains that integrates environmental externalities and waste valorisation pathways into operational decision-making. Departing from traditional linear “produce–consume–dispose” models, this study embeds three core sustainability mechanisms into a stochastic dynamic-programming framework: (1) progressive environmental cost internalisation aligned with EU Emissions-Trading System carbon pricing, capturing both waste-related emissions and cold-chain energy footprints; (2) circular-economy value-recovery channels that redirect near-expiry products to secondary applications (animal feed, biogas production, industrial processing) rather than disposal; and (3) deterioration-aware demand management that minimises resource throughput while maintaining service levels. Empirical calibration using Ukrainian dairy industry data demonstrates that sustainability-integrated inventory policies reduce waste generation by 4.8–10% relative to conventional approaches, with high-deterioration products showing the greatest potential for improvement. The authors identify a critical threshold in the circular economy: when salvage recovery rates exceed 35%, waste becomes an economic and ecological asset, fundamentally altering the sustainability calculus of inventory decisions. Environmental costs account for 4.6% of total operating expenses at current carbon prices, a share projected to increase substantially as climate regulations tighten. The findings provide actionable guidance for dairy supply chain stakeholders pursuing the Sustainable Development Goals (SDGs 2, 12, 13): processors should establish circular-economy partnerships that achieve salvage rates above 35%, implement product-specific policies for high-deterioration items, and proactively integrate carbon pricing into inventory optimisation. The framework bridges sustainable operations theory and circular economy practice, offering a replicable model for transitioning perishable food supply chains toward closed-loop, low-waste configurations that simultaneously reduce environmental impact and enhance economic performance. Zearalenone (ZEA) is an estrogenic Fusarium mycotoxin widely contaminating feed and feedstuffs, and posing significant risks to animal health. This preliminary study aimed to evaluate the toxicological effects of dietary exposure to purified ZEA at doses ranging from below to above the Chinese regulatory limit (0.15 mg/kg) in weaned female piglets. Twenty piglets were randomly assigned to five groups (four piglets per group) receiving 0, 0.075, 0.15, 0.3, or 0.6 mg/kg ZEA for 42 days. Results suggested that ZEA promoted systemic oxidative stress, evidenced by decreased serum total antioxidant capacity (T-AOC) and increased malondialdehyde (MDA) content in liver across all doses, and in jejunal mucosa at ≥ 0.15 mg/kg (p < 0.01). Growth performance declined only at 0.6 mg/kg during days 29–42 (p < 0.01), while hemoglobin (HGB) levels (p < 0.01) and ileal villus height (p < 0.05) were reduced at all doses. ZEA also caused inflammatory dysregulation, as evidenced by decreased interleukin-4 (IL-4) levels in serum, liver, and intestinal tissues across all doses (p < 0.01), and disrupted reproductive hormones even at 0.075 mg/kg, as indicated by suppressed serum luteinizing hormone (LH) levels (p < 0.01), which progressed to histopathological damage in uterine and ovarian tissues at higher doses. These preliminary findings, together with significant correlations between oxidative stress markers and multi-organ parameters, suggest that low doses of purified ZEA may induce systemic oxidative stress and subclinical multi-organ toxicity in weaned female piglets, highlighting the need to incorporate redox status into risk assessment and to explore potential antioxidant-based mitigation strategies. However, given the small sample size, these results should be interpreted with caution and warrant validation in larger samples. Full article

Milk production in dairy cattle is increasingly challenged by thermal variability. This underscores the need for reliable assessment of microclimatic conditions and their interaction with animal- and management-related factors to ensure sustainable dairy production. The aim of this study was to evaluate the effects of thermal variability and selected environmental and biological factors on key milk production traits in dairy cattle. The influence of fixed factors related to production conditions and microclimatic variability, including the Temperature–Humidity Index (THI) as an indicator of thermal variability, on daily milk yield (MY), milk fat content (MF), and milk protein content (MP) was assessed. This study used a dataset covering two observation periods of daily milk production traits in cows of different breeds (Simmental, Holstein-Friesian, Red Holstein, and Brown Swiss) reared in three regions of the Republic of Serbia (Mačva, Podunavlje, and Šumadija), enabling an assessment of thermal variability under diverse production and microclimatic conditions. The expression and variability of the investigated traits were determined using the PROC FREQ and PROC MEANS procedures, while the effects of individual factors were analysed using general linear and regression models, with results expressed as least squares means. All examined factors showed a highly significant effect on MY, MF, and MP (p < 0.0001). Milk production was highest within the THI range of 51–60, corresponding to thermoneutral conditions, whereas higher THI values (>60) reflect increasing thermal load and were associated with measurable reductions in milk yield, as confirmed by statistical analysis. These findings demonstrate that thermal variability and heat stress significantly influence milk production and composition and highlight the importance of integrating microclimatic indicators into sustainability-oriented dairy management and breeding strategies. The urban trade in non-timber forest products (NTFPs) plays a key role in sustaining livelihoods in the Global South, while also suggesting potential pressure on resource supply systems. This study provides an integrated analysis of NTFP diversity, market structure, economic importance, and perceived drivers of resource decline in Kolwezi, a rapidly expanding mining city where such dynamics remain poorly documented. Data were collected through surveys conducted with 35 sellers across two major urban markets and 384 consumers from different neighbourhoods and analysed using descriptive and inferential statistics to examine patterns, associations, and socio-demographic influences. A total of 65 NTFP species were recorded, including 49 plant, 14 animal, and 2 fungal species, reflecting strong dependence on Miombo ecosystems. Medicinal (59.3%) and food uses dominate, with multifunctional species such as Bobgunnia madagascariensis (Desv.) J.H.Kirkbr. & Wiersama, Canarium schweinfurthii Engl., Terminalia mollis M.A.Lawson, Gardenia ternifolia subsp. jovis-tonantis (Welw.) Verdc., and Albizia antunesiana Harms, playing a central role in both household use and market supply. The trade is largely female-dominated (79.1%) and constitutes a major component of the informal urban economy, with monthly incomes ranging from USD 9 to 429.3, primarily driven by sales volume rather than unit price. However, the sector is constrained by structural and logistical limitations, including remoteness of supply areas, seasonality, and limited value addition. The perceived declining availability of high-use-value species, attributed by respondents to deforestation, mining expansion, and overexploitation, highlights perceived sustainability concerns. These pressures are perceived differently across socio-demographic groups, indicating heterogeneous understandings of environmental change. Overall, the results indicate a perceived mismatch between rising urban demand and declining resource availability, which may reflect an emerging socio-ecological imbalance between urban demand and perceived resource availability. Addressing these challenges requires integrated strategies that combine the domestication of priority species, the development of processing chains, improved infrastructure, and strengthened governance mechanisms. Such approaches are essential to reconcile livelihood support with the sustainable management of NTFPs in rapidly transforming urban landscapes. Full article

Extensive animal production systems based on dryland pastures in Mediterranean regions have low profit margins. Improvements in soil fertility or grazing management and stocking rates are recognized strategies for reversing this situation and to ensure long-term agricultural sustainability. This article aims to assess whether this strategy of possible intensification of sheep production has a significant impact on soil compaction, which is a manifestation of soil functionality degradation resulting from trampling. An experimental design with four treatments was implemented (with and without dolomitic limestone application; continuous grazing with low stocking rates, CG-LSR, and deferred grazing with high stocking rates, DG-HSR). The study involved cone index (CI, in kPa) measurements (48 sampling areas, 12 in each treatment) on eight dates during two annual pasture/grazing cycles (2023/2024 and 2024/2025). Other soil parameters, the presence of trees and grazing preferences were also monitored and correlated with CI. The main results showed: (i) significantly higher soil compaction under CG-LSR than under DG-HSR; (ii) a negative and significant effect of soil moisture content (SMC) on CI (r = −0.381; p < 0.05); (iii) a significant CI increase in preferential grazing areas, but only in the topsoil layer (0–10 cm) and with a very weak correlation coefficient (r = 0.172; p < 0.05); and (iv) no significant differences in CI under and outside tree canopy areas (UTC and OTC, respectively) for the depth range of 0–30 cm. These results are good indicators of the desired and sustainable intensification of extensive livestock grazing systems. Full article

Hepatitis E virus (HEV) is a zoonotic pathogen of global concern that circulates in both domestic and wild swine populations. Understanding its presence and dynamics in wildlife reservoirs is crucial for assessing spillover risks and designing One Health surveillance strategies. This study investigated the occurrence, genetic diversity, and evolutionary relationships of HEV in wild boars from mainland Portugal. A total of 120 animals from seven districts were tested, with HEV RNA detected in four cases (3.3%), all from the Évora district near the Spanish border. One positive sample was successfully sequenced, and phylogenetic analysis based on the complete genome classified it within the HEV-3m subtype, clustering with predominantly human-derived sequences from Spain and France, which highlights its zoonotic potential. A second phylogenetic analysis based on a partial genomic fragment, including sequences from domestic pigs in Portugal, revealed the co-circulation of subtypes 3e, 3f, and 3m without clear spatial or temporal patterns. Phylogeographic analysis suggested that the identified strain was most likely introduced from Spain, supporting the hypothesis of cross-border transmission through wild boar movement. No recombination events were detected in the sequence obtained in this study. These findings provide the first molecular evidence of HEV-3m circulation in wild boars in Portugal, offering valuable insight into the HEV strain circulation in European wildlife populations. The zoonotic potential of HEV and the likelihood of interspecies transmission highlight the need for coordinated cross-border surveillance and integrated One Health strategies. Full article

Pasteurella multocida (P. multocida) is not only the core pathogen of bovine respiratory disease (BRDC) but also a significant zoonotic agent, posing a dual threat to global animal husbandry and public health. This study utilized untargeted metabolomics to systematically dissect the metabolic regulatory network of P. multocida in response to tylosin within a One Health framework. The results revealed significant “defense–growth” metabolic reprogramming: activation of amino sugar and nucleotide sugar pathways (e.g., CDP-glucose) indicated cell wall remodeling, while directional shifts in the phenylalanine–tyrosine network directed flux toward defensive secondary metabolites. Concurrently, amino acid disorders and the overactivation of the ABC transporter system exacerbated an internal energy crisis, characterized by a shift from respiration to glycolysis, ATP depletion, and ROS accumulation. SEM observations confirmed membrane integrity disruption and cytoplasmic leakage. Crucially, this metabolic stress and the transition into a “persister-like” dormant state are closely linked to the adaptive expression of antimicrobial resistance (AMR) genes. Under the selective pressure of tylosin, these metabolic perturbations may facilitate the emergence and horizontal transfer of resistance determinants, which can circulate through the animal–human–environment interface. By revealing the metabolic physiological basis of tylosin’s action and its role in inducing bacterial tolerance, this study provides critical theoretical insights for antimicrobial stewardship, aiming to mitigate the risk of AMR transmission and preserve the efficacy of macrolides for both veterinary and human medicine. Environmental pollutants, lifestyle factors, and intrinsic metabolism can amplify reactive oxygen and nitrogen species (ROS/RNS) generation beyond antioxidant capacity. The resulting oxidative stress damages macromolecules, perturbs redox signaling, and may accelerate biological aging. This review synthesizes evidence published mainly in 2020–2025 on how major pollutant classes (air pollutants, metals, pesticides, nanoparticles, and micro-/nanoplastics) induce ROS through shared nodes mitochondrial electron transport disruption, NADPH oxidase activation, and redox cycling/Fenton chemistry and how these signals propagate to epigenetic remodeling (DNA methylation, histone modifications, and non-coding RNAs). To move beyond descriptive cataloging, we grade the strength of evidence by study context (cell culture, animal models, human observational studies, and clinically oriented biomarker research), highlight convergent findings and unresolved controversies, and specify key methodological limits. We then compare oxidative-stress biomarker platforms by analytical specificity, pre-analytical susceptibility, and translational readiness, distinguishing validated markers from exploratory redox-epigenetic and multi-omics signatures. Finally, we discuss how exposomics and AI-assisted multi-omics integration may support biomarker discovery while emphasizing current constraints (confounding, batch effects, and limited prospective validation) that must be addressed for clinical translation. Highly pathogenic avian influenza (HPAI) H5N1 clade 2.3.4.4b continues to circulate globally across wild birds, poultry, and an expanding range of mammalian hosts, highlighting the need for antiviral strategies that address the animal–environment–human interface. The influenza A polymerase acidic (PA) endonuclease, a key enzyme in viral transcription, represents a conserved antiviral target across host species. In this study, we present a computational prioritization framework integrating homology modeling, molecular docking, molecular dynamics simulations, and physicochemical filtering to identify candidate PA endonuclease inhibitors relevant to a One Health context. Homology models of contemporary H5N1 clade 2.3.4.4b PA sequences were constructed based on the crystallographic template 6FS8 and used for cross-host docking against a targeted ligand library. Docking analysis identified baloxavir, a reference inhibitor, and entecavir, a nucleoside analog, as compounds of interest, with entecavir demonstrating favorable binding behavior, particularly in the poultry-associated model. Molecular dynamics simulations of the poultry PA–entecavir complex indicated stable interaction over 170 ns, supported by low structural deviation and favorable binding free energy (ΔG ≈ −85 kJ/mol). Physicochemical profiling suggested that entecavir possesses properties such as high polarity and predicted aqueous solubility, which were considered within the translational filtering step of this computational workflow. However, these properties do not establish antiviral efficacy or practical suitability for field use. The study provides a structured framework for integrating cross-host structural analysis with basic translational considerations, supporting the identification of candidate compounds for further biochemical and virological evaluation within the context of H5N1 control. Characterization and Genetic Evolution of H6N2 Subtype AIV Isolates from Aquatic Birds

Birds serve as the primary natural reservoirs for avian influenza viruses (AIVs), harboring nearly all known AIV subtypes. The seasonal migratory movements of wild birds play a significant role in the transmission and dissemination of AIVs. Jianhu Lake in Dali, Yunnan Province, serves as a vital congregation point along avian migratory routes, providing an ideal habitat for birds. In this study, a total of 619 avian samples were collected from the Jianhu area, from which four H6N2 subtype AIV strains were successfully isolated. Among these, A/grey heron/Jianhu/JH-89/2024 (hereafter referred to as JH-89) and A/grey heron/Jianhu/JH-91/2024 (JH-91) were isolated from grey herons (Ardea cinerea); A/mareca penelope/Jianhu/JH-2-11/2025 (JH-2-11) from a Eurasian wigeon (Mareca penelope); and A/duck/Jianhu/JH-1-1/2025 (JH-1-1) from a domestic duck (Anas platyrhynchos domesticus). Genomic analyses revealed that these four H6N2 isolates belong to the Eurasian lineage, with all eight gene segments originating from complex reassortment events among diverse Asian isolates. In vitro assays demonstrated that the representative strain JH-2-11 replicated efficiently in various human- and animal-derived cell lines. In vivo infection models revealed that, without prior adaptation, the JH-2-11 strain successfully infected BALB/c mice, resulting in suppressed body weight gain and severe pathological lesions in the respiratory tract (nasal turbinates, trachea, and lungs), without causing mortality or extrapulmonary dissemination. Collectively, although these H6N2 viruses evolve primarily within avian hosts, they exhibit potential for mammalian adaptation and require continuous epidemiological monitoring. Full article

A Retrospective Analysis of Wildlife Rehabilitation Trends in Lithuania over Two Decades

The primary aim of wildlife rehabilitation centres is the release of treated animals back into the wild after treatment. Data collected during rehabilitation can provide valuable insights into current trends and can help with conservation strategies aiming to reduce human-related negative impact. This review analyses records from past and currently operating wildlife rescue and rehabilitation centres in Lithuania over a 23-year period. Data were compiled for 7847 individual animals representing 216 species of birds, mammals, and reptiles. The study evaluates patterns of seasonal admission, taxonomic composition, known causes of injury or admission, and rehabilitation outcomes. The results showed that 83% of cases involved birds, mammals comprised 16%, and reptiles were recorded only infrequently (0.52%). Admissions peaked in summer, comprising 42% of all cases. Injuries of unknown origin were the most common (55%). Among cases with identified causes, a substantial proportion were associated with human activities, including road accidents (5% of all cases), collisions with anthropogenic structures (4%), and attacks by domestic cats or dogs (3%). The presence of a specialized rehabilitation centre, together with active public involvement in the rescue of injured wildlife, contributes to release rates reaching approximately 30% of admitted animals. Datasets such as those analyzed in this study may contribute to improved preparedness for managing unavoidable human–wildlife interactions in the future. Full article

Cerebral palsy (CP) is a non-progressive neurological condition associated with neuroinflammation, motor impairments, and gastrointestinal dysfunction mediated by the gut–brain axis. Preserving the intestinal epithelial barrier integrity may represent a therapeutic strategy, and quercetin is a bioactive compound with potential intestinal protective effects. This study investigated the effects of neonatal quercetin treatment on morphometric parameters, oxidative stress markers, and epithelial barrier gene expression in an experimental CP model. Wistar rats were distributed into four groups according to health status and treatment with a vehicle (V) or quercetin (Q, 10 mg/kg, intraperitoneally): healthy control (CV and CQ) and CP (CPV and CPQ) (n = 10/group). Intestinal morphology, oxidative stress markers, and gene expression (occludin, zonulin, and mucin 2) were evaluated. CP animals showed segment-specific alterations, with structural impairment predominantly in the ileum and increased oxidative damage in the jejunum. Quercetin attenuated oxidative stress markers and modulated antioxidant enzyme activity in CP, increased jejunal tight-junction gene expression in both healthy and CP groups, and enhanced MUC2 expression only in healthy animals, without fully reversing CP-induced morphological changes. In conclusion, neonatal quercetin modulates oxidative stress and epithelial barrier-related gene expression, supporting its potential as an adjuvant strategy for intestinal barrier protection in experimental CP. Alpacas have become increasingly popular as farm and companion animals in many countries and were introduced in Hungary approximately 15 years ago. However, reliable data on their population size and management practices have remained limited. Therefore, this study aimed to provide the first nationwide overview of alpaca keeping in Hungary. An anonymous Google Forms-based questionnaire comprising 55 questions was used to collect data on farm characteristics, herd size, husbandry practices, and animal health management. A total of 53 valid responses were analyzed. The results indicated that approximately 260 alpacas are currently kept in Hungary, with most herds being small and typically consisting of 1–5 animals. Most owners planned to expand their herds, primarily through domestic breeding or purchases; however, only a minority considered alpaca keeping economically profitable. Responses related to quarantine, veterinary access, and disease management suggested limited knowledge of biosecurity and animal health. While most owners reported high levels of trust in veterinarians, many had difficulty accessing specialists with expertise in alpacas. In conclusion, alpaca keeping in Hungary is a growing but still small-scale sector. Improving owner education and strengthening professional support may enhance animal welfare and reduce health risks, thereby supporting the One Health approach. Full article
