The role of LAP proteins expressed in skeletal muscles (ERBIN, LANO, and SCRIBBLE) and at neuromuscular junctions (NMJs) remains largely unknown. Our previous data demonstrate that LAP proteins are differentially expressed in muscle cells, nerve endings, and terminal Schwann cells, though they are all expressed in myofibers and accumulate at NMJs. ERBIN and SCRIBBLE align with acetylcholine receptor clusters (CHRNs) at the NMJ. In vivo ablation of Erbin is associated with smaller CHRN and upregulation of Lano and Scribble. However, SCRIBBLE was also shown to influence the fate decision of muscle stem cells. Here, we investigated how the absence of SCRIBBLE in skeletal muscle cells might impair skeletal muscle fibers or NMJs. Although conditional Scribble knockout mice did not exhibit changes in weight or viability, force per weight decreased slightly. This was supported by compromised neuromuscular transmission and increased NMJ fragmentation. Moreover, Scribble knockout muscles transcribe less myosin heavy chain genes. Here, we also showed that RAB5, an effector of endocytic recycling, interacts with all LAP proteins, but in Scribble knockout muscles, reduced interaction was detected with ERBIN and LANO. These data suggest that a delicate signaling network employing LAP proteins is necessary for skeletal muscle fibers and NMJs. Full article

The Inner Mongolia cashmere goat is a local breed valued for both its cashmere and meat production. Early growth traits include birth weight and weaning weight. Generally, cashmere goats with higher birth and weaning weights tend to exhibit a higher carcass weight at slaughter. Consequently, exploring the genetic variation associated with these early growth traits is crucial in enhancing the meat production performance of Inner Mongolia cashmere goats. For this study, we performed genome-wide association analysis and haplotype analysis on the early growth traits of 212 Inner Mongolia cashmere goats (72 rams, 140 ewes) to identify molecular markers and haplotypes significantly associated with birth and weaning weights. Through genome-wide association analysis and gene annotation, we identified 21 SNPs and 117 candidate genes linked to early growth traits. Notably, genes such as RUNX1T1, ERBIN, MYO15B, NT5C, GRB2, ITGB4, and GALK emerged as significant factors influencing the early growth of Inner Mongolia cashmere goats. Additionally, nine haplotype blocks related to early growth traits were constructed, resulting in eight haplotype combinations. In conclusion, the haplotype combinations A1A1, C1C1, and D1D1 were found to be beneficial for the genetic improvement of early growth traits in Inner Mongolia cashmere goats. Full article

Different sulfates (Ca-, Mg, and Fe- sulfates) have been extensively detected on the Martian surface. As one of the Martian sulfates, the presence of ferrous sulfates will provide valuable clues about the redox environment, hydrological processes, and climatic history of ancient Mars. In this study, three hydrated ferrous sulfates were prepared in the laboratory by heating dehydration reactions. These samples were analyzed using X-ray Diffraction (XRD) to confirm their phase and homogeneity. Subsequently, Raman, mid-infrared (MIR) spectra, visible near-infrared (VNIR) spectra, and laser-induced breakdown spectroscopy (LIBS) were measured and analyzed. The results demonstrate that the spectra of three hydrated ferrous sulfates exhibit distinctive features (e.g., the v1 and v3 features of SO42 tetrahedra in their Raman and MIR spectra) that can offer new insights for identifying different ferrous sulfates on Mars and aid in the interpretation of in-situ data collected by instruments such as the Scanning Habitable Environments with Raman & Luminescence for Organics & Chemicals (SHERLOC), SuperCam, and ChemCam, etc. Full article

Magnesium sulfate has been widely detected on the surface of Mars. The occurrence of magnesium sulfate and mixed cationic sulfates preserves clues regarding the sedimentary environment, hydrological processes, and climate history of ancient Mars. In this study, seven magnesium sulfate double salts were synthesized in the laboratory using a high-temperature solid phase reaction or slow evaporation of aqueous solutions. The samples were analyzed using X-ray diffraction to confirm their phase and homogeneity. Subsequently, the Raman, mid-infrared spectra, and visible near-infrared spectra of these samples were collected and analyzed. Our results showed that the spectra of the analyzed magnesium sulfate double salts exhibited distinctive spectral features. These laboratory results may provide new insights for the identification of various magnesium sulfate double salts on Mars during the interpretation of in situ data collected by Scanning Habitable Environments with Raman and Luminescence for Organics and Chemicals (SHERLOC), SuperCam, and the ExoMars Raman Laser Spectrometer (RLS). In addition, the MIR and VNIR spectra features obtained in this study provide an improved reference and spectra library for decipherment of data sourced from the Thermal Emission Spectrometer (TES), Thermal Emission Imaging System (THEMIS), and Mars Mineralogical Spectrometer (MMS). Studies about the pre-stretching effect on the mechanical behavior of cold-rolled 5%Mn medium manganese steel have adopted optical microscopy, scanning electron microscopy, transmission electron microscopy and X-ray diffraction techniques. Results showed that pre-stretching would change the ferrite morphology from massive and lath-like to strip-like. With the pre-stretching increasing from 0% to 14%, the dislocation density and yield strength both grew gradually, which corresponded to growth from 6.49 × 1014 m−2 to 7.98 × 1014 m−2 and growth from 765 MPa to 1109 MPa, respectively. Meanwhile, the austenite volume fraction, elongation and product of strength and elongation were all reduced with the pre-stretch increase. The stabilized retained austenite with pre-stretch delayed the occurrence of the TRIP effect and improved the work hardening rate. As a result, the Lüders band disappeared at 2% pre-stretch and the PLC band vanished from the stress–strain curve at 14% pre-stretch. Full article

DNA methylation is a pivotal epigenetic regulatory mechanism in the development of skeletal muscles. Nonetheless, the regulators responsible for DNA methylation in the development of embryonic duck skeletal muscles remain unknown. In the present study, whole genome bisulfite sequencing (WGBS) and transcriptome sequencing were conducted on the skeletal muscles of embryonic day 21 (E21) and day 28 (E28) ducks. The DNA methylation pattern was found to fall mainly within the cytosine-guanine (CG) context, with high methylation levels in the intron, exon, and promoter regions. Overall, 7902 differentially methylated regions (DMRs) were identified, which corresponded to 3174 differentially methylated genes (DMGs). By using integrative analysis of both WGBS with transcriptomics, we identified 1072 genes that are DMGs that are negatively associated with differentially expressed genes (DEGs). The gene ontology (GO) analysis revealed significant enrichment in phosphorylation, kinase activity, phosphotransferase activity, alcohol-based receptors, and binding to cytoskeletal proteins. The Kyoto Encyclopedia of Genes and Genomes (KEGGs) analysis showed significant enrichment in MAPK signaling, Wnt signaling, apelin signaling, insulin signaling, and FoxO signaling. The screening of enriched genes showed that hyper-methylation inhibited the expression of Idh3a, Got1, Bcl2, Mylk2, Klf2, Erbin, and Klhl38, and hypo-methylation stimulated the expression of Col22a1, Dnmt3b, Fn1, E2f1, Rprm, and Wfikkn1. Further predictions showed that the CpG islands in the promoters of Klhl38, Klf2, Erbin, Mylk2, and Got1 may play a crucial role in regulating the development of skeletal muscles. This study provides new insights into the epigenetic regulation of the development of duck skeletal muscles. Full article

Settlement control of existing stations has consistently been a key issue in tunnel construction. Intermediate soil has a significant influence on the settlement of the existing station as a connector between the new tunnel and the existing station which should be considered when selecting settlement control measures. To clarify the relationship between the state of the intermediate soil and the settlement and control measures of the existing station, this study investigated 49 new tunnel projects under existing stations and elucidated the relationship between the settlement and the settlement control measures of the existing station and the intermediate soil. The relationship between the failure of the intermediate soil and the settlement of the existing station was further analyzed by numerical simulation. Lastly, a simple mechanical model of intermediate soil failure was constructed to determine the stress state of the intermediate soil in constructing a tunnel under the existing station to guide the formulation of settlement control measures for the existing station. Hence, when the intermediate soil is completely destroyed, active control measures, such as jack or grouting lifting, should be implemented; when the intermediate soil is partially damaged, passive control measures should be undertaken. Thus, this study can provide a reference for settlement control of tunnels under existing stations. Full article

Hybrid membranes with three different thicknesses, PMDS_C1, PMDS_C2, and PMDS_C3 (0.21 ± 0.03 mm, 0.31 ± 0.05 mm, and 0.48 ± 0.07 mm), were synthesized by the sol–gel method using polydimethylsiloxane, hydroxy-terminated, and cyanopropyltriethoxysilane. The presence of cyano, methyl, and silicon-methyl groups was confirmed by FTIR analysis. Contact angle analysis revealed the membranes’ hydrophilic nature. Solvent resistance tests conducted under vortex and ultrasonic treatments (45 and 60 min) demonstrated a preference order of acetonitrile > methanol > water. Furthermore, the membranes exhibited stability over 48 h when exposed to different pH conditions (1, 3, 6, and 9), with negligible mass losses below 1%. The thermogravimetric analysis showed that the material was stable until 400 °C. Finally, the sorption analysis showed its capacity to detect furfural, 2-furylmethylketone, 5-methylfurfural, and 2-methyl 2-furoate. The thicker membrane was able to adsorb and slightly desorb a higher concentration of furanic compounds due to its high polarity provided by the addition of the cyano groups. The results indicated that the membranes may be suitable for sorbent materials in extracting and enriching organic compounds. Full article

With the implementation of the “double carbon” policy in various countries in the world, the demand for grain-oriented electrical steel with low iron loss and high magnetic induction is increasing. Reducing the thickness of the steel sheets is an effective method to reduce the iron loss.The sheet thickness reduction means the increasing cold rolling reduction rate, especially the secondary cold rolling reduction rate, will directly affect the texture evolution of the secondary recrystallization process. In this paper, the secondary cold rolling reduction rate of commercial grain-oriented silicon steel was studied by means of X-Ray Diffraction, Electron Backscatter Diffraction. The results showed that Ultra-thin oriented silicon steel cannot be obtained by increasing the secondary cold rolling reduction rate alone; the optimum secondary cold rolling reduction rate was 59.2%. The grain size increased as the secondary cold rolling reduction rate increased and favorable texture content decreased, which was disadvantage to obtain a secondary recrystallization environment. The species living in the Qinghai-Tibet Plateau provide an excellent model system for studying the relationship between molecular convergent evolution and adaptation. Distant species experiencing the same selection pressure (i.e., hypoxia, low temperature and strong ultraviolet radiation) are likely to evolve similar genetic adaptations independently. Here, we performed comparative genomics studies on six independently evolved high-altitude species. The results also showed that the convergent evolution of the six species was mainly reflected at the level of rapidly evolving genes, and the functions of these rapidly evolving genes were mainly related to hypoxia response and DNA damage repair. In addition, we found that high-altitude species had more gene family changes than their low-altitude relatives, except for the order Lagomorpha. The results also show that the convergence of the gene family contraction of high-altitude species is much greater than that of expansion, revealing a possible pattern of species in adapting to high-altitude. Furthermore, we detected a positive selection signature in four genes related to hypoxia response and ultraviolet radiation damage in these six species (FYCO1, ERBIN, SCAMP1 and CXCL10). Our study reveals that hypoxia response might play an important role in the adaptation of independently evolved species to a high-altitude environment, providing a basic perspective for further exploring the high-altitude adaptation mechanism of different related species in the future. Urban coordinated development is an important aspect of regional development. The high-quality development of the Yellow River Basin cannot be separated from the coordinated and sustainable development of its inner cities. However, the network connection and spatial structure of cities in the Yellow River Basin have not received sufficient attention. Therefore, this study considered 11 prefecture-level cities in Shanxi Province, an underdeveloped region in the Yellow River Basin, as case areas and selected data on traffic, migration, and information flow that can better represent the urban spatial network structure and depict the spatial connection between cities. Based on the flow intensity calculation, flow direction judgment, spatial structure index, and social network analysis, the spatial structural characteristics of Shanxi Province were comprehensively analyzed from the perspective of flow space. The results showed the following: (1) Cities in Shanxi Province present a development trend of “one core and multiple centers.” The strong connection concerns mostly Taiyuan and radiates outward and presents a Chinese character “大”—shaped spatial connection pattern. (2) Taiyuan is the first connecting city of most cities in Shanxi Province, and the element flows particularly towards the central city and geographical proximity. (3) The urban spatial pattern of Shanxi Province presents an obvious unipolar development trend, where the network structure is an “absence-type pyramid.” The imbalance of the urban network connection strength is prominent in Shanxi Province, which is strong and numerous in the south but opposite in the north. (4) The overall network element flow density is low, the network connection is weak, Taiyuan agglomeration and radiation are the strongest, and Changzhi centrality ranks second, but the gap between Changzhi and Taiyuan is wide, and the polarization phenomenon is serious. Future research should focus on the rapidly developing provincial capital city of Taiyuan, coordinating the steady development of the central Shanxi city cluster, and driving the common development of neighboring cities. When constructing open excavation and adding stories on the upper part of an existing station structure, the new foundation pit contacts with the existing station at zero distance; subsequently, the construction has a greater impact than that of other close constructions. To explore the cross-impact of new and existing structures on open excavation and story-adding above the existing station, in this study, through numerical simulation and field measurement, we analyze the deformation law of the existing station and foundation pit retaining structure in the excavation and story-adding construction above the existing station. The following conclusions are obtained. The excavation of the upper foundation pit leads to the overall uplift of the existing station, and the sidewall exhibits an internal extrusion trend, which is caused by an increase in the horizontal load on the station caused by the unloading of the vault and expansion of the formation plastic zone. Affected by the existing station structure, some foundation pit retaining structures are end-suspended piles; the bottoms of the end-suspended pile slips are evident. This study offers control measures for the primary support connection between the pile bottom and initial retention and protection of the existing station, deep hole grouting reinforcement of the stratum behind the pile, and reserving earth berm at the pit bottom to reduce the deformation of the end-suspended pile. The research results can provide a reference for constructing the added story of a subway transfer station. Full article

In order to improve the fault diagnosis accuracy of bearings, an intelligent fault diagnosis method based on Variational Mode Decomposition (VMD), Composite Multi-scale Dispersion Entropy (CMDE), and Deep Belief Network (DBN) with Particle Swarm Optimization (PSO) algorithm—namely VMD-CMDE-PSO-DBN—is proposed in this paper. The number of modal components decomposed by VMD is determined by the observation center frequency, reconstructed according to the kurtosis, and the composite multi-scale dispersion entropy of the reconstructed signal is calculated to form the training samples and test samples of pattern recognition. Considering that the artificial setting of DBN node parameters cannot achieve the best recognition rate, PSO is used to optimize the parameters of DBN model, and the optimized DBN model is used to identify faults. Through experimental comparison and analysis, we propose that the VMD-CMDE-PSO-DBN method has certain application value in intelligent fault diagnosis. Full article

Comprehensive Analysis of RNA-Seq Gene Expression Profiling of Brain Transcriptomes Reveals Novel Genes, Regulators, and Pathways in Autism Spectrum Disorder

Background: Autism spectrum disorder (ASD) is a neurodevelopmental disorder with deficits in social communication ability and repetitive behavior. The pathophysiological events involved in the brain of this complex disease are still unclear. Methods: In this study, we aimed to profile the gene expression signatures of brain cortex of ASD patients, by using two publicly available RNA-seq studies, in order to discover new ASD-related genes. Results: We detected 1567 differentially expressed genes (DEGs) by meta-analysis, where 1194 were upregulated and 373 were downregulated genes. Several ASD-related genes previously reported were also identified. Our meta-analysis identified 235 new DEGs that were not detected using the individual RNA-seq studies used. Some of those genes, including seven DEGs (PAK1, DNAH17, DOCK8, DAPP1, PCDHAC2, and ERBIN, SLC7A7), have been confirmed in previous reports to be associated with ASD. Gene Ontology (GO) and pathways analysis showed several molecular pathways enriched by the DEGs, namely, osteoclast differentiation, TNF signaling pathway, complement and coagulation cascade. Topological analysis of protein–protein interaction of the ASD brain cortex revealed proteomics hub gene signatures: MYC, TP53, HDAC1, CDK2, BAG3, CDKN1A, GABARAPL1, EZH2, VIM, and TRAF1. We also identified the transcriptional factors (TFs) regulating DEGs, namely, FOXC1, GATA2, YY1, FOXL1, USF2, NFIC, NFKB1, E2F1, TFAP2A, HINFP. Conclusion: Novel core genes and molecular signatures involved with ASD were identified by our meta-analysis. Full article

In the perspective of information philosophy, this paper discusses Cursive script space-time evolution from a series of all-round information in calligraphy evolution about history tradition which based in keep often and knew change. In this paper, Dudu was thought to change brushwork method and standard cursive script. We can see clearly in the space-time evolution of calligraphy aesthetic realization path, which follow cursive old-information and productive cursive new-information and evolution series relationship holographic, from “often” to “change” to form a new “often”. The complexity of Cursive script had matured in informational-assimilation and dissimilation.
