Current electrolytic refining processes for crude solder commonly employ fluosilicic acid (H2SiF6) as the electrolyte with bone glue and β-naphthol additives yet suffer from poor electrolyte stability, coarse cathode crystallization, low current efficiency, and high energy consumption, adversely affecting product quality and economic viability. In order to solve these limitations, electrochemical techniques, XRD, SEM, and ICP-OES were used to study the effects of gelatin and sodium lignosulfonate on the deposition overpotential and cathode morphology, as well as the effects of process parameters on current efficiency and energy consumption. A novel approach was developed using an H2SiF6 system enhanced by gelatin and sodium lignosulfonate for crude solder refining. After optimization, 120 h electrolysis achieved a current efficiency >97.8%, smooth/dense cathode surface, average cell voltage of 0.24 V, and energy consumption of 98.15 kWh/t. Efficient deposition of 81.2% Sn and 75.2% Pb on the cathode was realized, while >93.3% of Sb, Bi, Ag, Cu, and As were enriched in anode slime to facilitate valuable metal recovery, and >90.6% of In/Al concentrated in the electrolyte enabled effective Sn-Pb impurity separation. This study provides theoretical and technical foundations for advancing sustainable and economical electrolytic refining of crude solder. Full article

β-galactosidase (β-Gal) has emerged as a pivotal biomarker for the comprehensive investigation of diseases associated with cellular senescence. The development of a fluorescent sensor is of considerable importance for precisely detecting the activity and spatial distribution of β-Gal. In this study, we developed two excited-state-altering responsive fluorescent sensors (TF1 and TF2) for ratiometric detection of β-Gal. Two TCF dyes, composed of tricyanofuran (TCF) and naphthol units, feature electron “pull–push” systems and are quenched fluorescence by β-Gal. Upon β-Gal hydrolysis, a significant ratiometric shift in absorption from ca. 475 nm to 630 nm is observed, accompanied by the emergence of a fluorescence signal at ca. 660 nm. The enzyme-responsive optical red-shifts are attributed to the excited-state transition from intramolecular charge transfer (ICT) state to local excited (LE) state, which was confirmed by density functional theory (DFT) calculations. Both fluorescent sensors display exceptional sensitivity and selectivity for the response of β-Gal in PBS solution and are capable of tracking β-Gal within senescent A549 cells. This study introduces a framework for developing multimodal optical probes by systematically modulating excited-state properties, demonstrating their utility in senescence studies, diagnostic assay design, and therapeutic assessment. Here, we report for the first time on the mechanisms of action of the essential oil of Ruta graveolens (REO) against the plant pathogen Colletotrichum gloeosporioides. In particular, the presence of REO drastically affected the morphology of hyphae by inducing changes in the cytoplasmic membrane, such as depolarization and changes in the fatty acid profile where straight-chain fatty acids (SCFAs) increased by up to 92.1%. In addition, REO induced changes in fungal metabolism and triggered apoptosis-like responses to cell death, such as DNA fragmentation and the accumulation of reactive oxygen species (ROS). The production of essential enzymes involved in fungal metabolism, such as acid phosphatase, β-galactosidase, β-glucosidase, and N-acetyl-β-glucosaminidase, was significantly reduced in the presence of REO. In addition, C. gloeosporioides activated naphthol-As-BI phosphohydrolase as a mechanism of response to REO stress. The data obtained here have shown that the essential oil of Ruta graveolens has a strong antifungal effect on C. gloeosporioides. Therefore, it has the potential to be used as a surface disinfectant and as a viable replacement for fungicides commonly used to treat anthracnose in the postharvest testing phase. Full article

Plant extracts and essential oils can be alternative environmentally friendly agents to combat pathogenic microbes and malaria vectors. Myrrh is an aromatic oligum resin that is extracted from the stem of Commiphora spp. It is used in medicine as an insecticide, cytotoxic, and aromatic. The current study assessed the effect of Commiphora myrrha resin extracts on the biological potency of the third larval stage of Aedes aegypti, as well as its antioxidant and cytotoxic properties against two types of tumor cells (HepG-2 and Hela cell lines). It also used GC–MS to determine the chemical composition of the C. myrrha resin extracts. Fifty components from the extracted plant were tentatively identified using the GC–MS method, with curzerene (33.57%) typically listed as the primary ingredient, but other compounds also make up a significant portion of the mixture, including 1-Methoxy-3,4,5,7-tetramethylnaphthalene (15.50%), β-Elemene (5.80%), 2-Methoxyfuranodiene (5.42%), 2-Isopropyl-4,7-Dimethyl-1-Naphthol (4.71%), and germacrene B (4.35%). The resin extracts obtained from C. myrrha exhibited significant efficacy in DPPH antioxidant activity, as evidenced by an IC50 value of 26.86 mg/L and a radical scavenging activity percentage of 75.06%. The 50% methanol extract derived from C. myrrha resins exhibited heightened potential for anticancer activity. It demonstrated substantial cytotoxicity against HepG-2 and Hela cells, with IC50 values of 39.73 and 29.41 µg mL−1, respectively. Notably, the extract showed non-cytotoxic activity against WI-38 normal cells, with an IC50 value exceeding 100 µg mL−1. Moreover, the selectivity index for HepG-2 cancer cells (2.52) was lower compared to Hela cancer cells (3.40). Additionally, MeOH resin extracts were more efficient against the different growth stages of the mosquito A. aegypti, with lower LC50, LC90, and LC95 values of 251.83, 923.76, and 1293.35 mg/L, respectively. In comparison to untreated groups (1454 eggs/10 females), the average daily number of eggs deposited (424 eggs/L) decreases at higher doses (1000 mg/L). Finally, we advise continued study into the possible use of C. myrrha resins against additional pests that have medical and veterinary value, and novel chemicals from this extract should be isolated and purified for use in medicines. Leishmaniasis remains a significant global health concern, with current treatments relying on outdated drugs associated with high toxicity, lengthy administration, elevated costs, and drug resistance. Consequently, the urgent need for safer and more effective therapeutic options in leishmaniasis treatment persists. Previous research has highlighted selenium compounds as promising candidates for innovative leishmaniasis therapy. In light of this, a library of 10 selenium-containing diverse compounds was designed and evaluated in this study. These compounds included selenium-substituted indole, coumarin, chromone, oxadiazole, imidazo[1,2-a]pyridine, Imidazo[2,1-b]thiazole, and oxazole, among others. These compounds were screened against Leishmania amazonensis promastigotes and intracellular amastigotes, and their cytotoxicity was assessed in peritoneal macrophages, NIH/3T3, and J774A.1 cells. Among the tested compounds, MRK-106 and MRK-108 displayed the highest potency against L. amazonensis promastigotes with reduced cytotoxicity. Notably, MRK-106 and MRK-108 exhibited IC50 values of 3.97 µM and 4.23 µM, respectively, and most of the tested compounds showed low cytotoxicity in host cells (CC50 > 200 µM). Also, compounds MRK-107 and MRK-113 showed activity against intracellular amastigotes (IC50 18.31 and 15.93 µM and SI 12.55 and 10.92, respectively). In conclusion, the identified selenium-containing compounds hold potential structures as antileishmanial drug candidates to be further explored in subsequent studies. These findings represent a significant step toward the development of safer and more effective therapies for leishmaniasis, addressing the pressing need for novel and improved treatments. The electrolytic refining of crude tin is generally carried out in the fluorosilicic acid (H2SiF6) system with the assistance of bone glue and β-naphthol. However, the high saturated vapor pressure and low stability of H2SiF6 present environmental concerns and do not align with sustainable development goals. In this paper, the electrochemical behavior of tin on a glassy carbon (GC) electrode was studied in a relatively green and environmentally friendly methanesulfonic acid (MSA) system. Bone glue, sodium lignosulfonate, and β-naphthol were used as additives to make the deposit morphology smooth and to ensure grain refinement. The electrochemical reduction process of Sn2+ in an MSA system is a quasi-reversible process controlled by diffusion. The apparent activation energy Ea = 14.189 kJ/mol for the ion diffusion of Sn2+ was further calculated. The results of chronoamperometry showed that the electrocrystallization of Sn2+ on the GC electrode gradually tended to three-dimensional instantaneous nucleation with the increase in applied potential. The morphology and phase of the deposits were analyzed using scanning electron microscopy (SEM) and X-ray diffraction (XRD). The results showed that the deposits were uniform and dense pure tin. This work elucidates the electrochemical behavior of tin in a methanesulfonic acid system. Full article

The probiotic features of Lactiplantibacillus (L.) pentosus and L. paraplantarum strains, endogenous in Cobrançosa table olives from northeast Portugal, were assessed in terms of functional properties and health benefits. Fourteen lactic acid bacteria strains were compared with Lacticaseibacillus casei from a commercial brand of probiotic yoghurt and L. pentosus B281 from Greek probiotic table olives, in attempts to select strains with higher probiotic performances than those references. For functional properties, the i53 and i106 strains, respectively, exhibited: 22.2 ± 2.2% and 23.0 ± 2.2% for Caco-2 cell adhesion capacity; 21.6 ± 7.8% and 21.5 ± 1.4% for hydrophobicity; 93.0 ± 3.0% and 88.5 ± 4.5% for autoaggregation ability by 24 h of incubation; and ability to co-aggregate with selected pathogens—from 29 to 40% to Gram+ (e.g., Staphylococcus aureus ATCC 25923 and Enterococcus faecalis ATCC 29212); and from 16 to 44% for Gram− (e.g., Escherichia coli ATCC 25922 and Salmonella enteritidis ATCC 25928). The strains proved to be resistant (i.e., halo zone ≤14 mm) to some antibiotics (e.g., vancomycin, ofloxacin, and streptomycin), but susceptible (i.e., halo zone ≥ 20 mm) to others (e.g., ampicillin and cephalothin). The strains exhibited health-beneficial enzymatic activity (such as acid phosphatase and naphthol-AS-BI-phosphohydrolase), but not health-harmful enzymatic activity (such as β-glucuronidase and N-acetyl-β-glucosaminidase). Additionally, the antioxidant activity and cholesterol assimilation features, respectively, of the strains were 19.6 ± 2.8% and 77.5 ± 0.5% for i53, and 19.6 ± 1.8% and 72.2 ± 0.9% for i106. This study indicated that the addition of L. pentosus strains i53 and/or i106 to Cobrançosa table olives is likely to enhance the added value of the final product, in view of the associated potential benefits upon human health. 2-naphthol is a low-molecular-weight (LMW) polycyclic aromatic hydrocarbon (PAH) and air pollutant associated with childhood obesity. There has been a recent emergence of studies on the consequences of PAHs on human health. Current epidemiological reports suggest LMW-PAHs may contribute to obesity incidences in children, yet most studies focus on high-molecular-weight PAHs. This study explores 2-naphthol’s impact on obesity and obesity-associated metabolic disorders. To investigate 2-naphthol’s effect on lipid metabolism and inflammation, we employed 3T3-L1 and BAT1 cell lines to model white and brown adipocytes, respectively, alongside a murine macrophage cell line (RAW264.7). We found that 2-naphthol increased the expression of key adipogenic and lipogenic genes while decreasing lipolytic gene expression in chronically treated 3T3-L1 and BAT1 adipocytes. In addition, chronic 2-naphthol treatment also suppressed adrenergic-stimulated thermogenic gene expression in BAT1 brown adipocytes. In consistence, an increase in lipid accumulation was demonstrated in BODIPY and Oil Red O-stained adipocytes. Additionally, 3T3-L1 adipocytes and RAW264.7 macrophages chronically exposed to 2-naphthol showed upregulated mRNA expression of major inflammatory cytokines (e.g., tumor necrosis factor α (Tnfα), interleukin-1β (Il-1β), and Il-6). In summary, chronic exposure to 2-naphthol stimulates lipid accumulation in adipocytes and inflammation in adipocytes and macrophages. These findings support previous research that demonstrates 2-naphthol has obesogenic potential. Full article

The present study aimed to evaluate the safety of Bacillus subtilis (BS) IDCC1101, newly isolated from Cheonggukjang in Korea. Genome sequencing of BS IDCC1101 was performed to investigate the presence of secondary metabolites, virulence, antibiotic resistance, and mobile elements. Its phenotypic safety analyses included antibiotic susceptibility, enzyme activity, carbohydrate utilization, production of biogenic amines (BAs) and D-/L-lactate, hemolytic activity, and toxicities in HaCaT cells and rats. The genome of BS IDCC1101 consisted of 4,118,950 bp with 3077 functional genes. Among them, antimicrobial and antifungal secondary metabolites were found, such as fengycin, bacillibactin, and bacilysin. Antibiotic resistance and virulence genes did not exhibit transferability since they did not overlap with mobile elements in the genome. BS IDCC1101 was susceptible to almost all antibiotics suggested for assessment of BS’s antibiotic susceptibility by EFSA guidelines, except for streptomycin. BS IDCC1101 showed the utilization of a wide range of 27 carbohydrates, as well as enzyme activities such as alkaline phosphatase, esterase, esterase lipase, naphthol-AS-BI-phosphohydrolase, α-galactosidase, β-galactosidase, α-glucosidase, and β-glucosidase activities. Additionally, BS IDCC1101 did not exhibit the production of D-/L-lactate and hemolytic activities. Its toxicity in HaCaT cells and rats was also not detected. Thus, these genotypic and phenotypic findings indicate that BS IDCC1101 can be safely used for industrial applications. Full article

Gummy stem blight (GSB) caused by Didymella bryoniae (D. bryoniae) is a worldwide fungal soil-borne disease that can cause severe yield reduction of watermelon. To shorten the monitoring time of carbendazim-resistant strains of D. bryoniae in the field, in this study, we developed a loop-mediated isothermal amplification (LAMP) assay for rapid detection of carbendazim-resistant strains of D. bryoniae. The β-tubulin gene of carbendazim-resistant strains was selected as the target for primer design. Based on the color change of hydroxy naphthol blue (HNB) and gel electrophoresis, the optimal reaction conditions for LAMP were determined at 65 °C for 50 min. In specificity tests, the LAMP assay was able to distinguish between carbendazim-resistant and sensitive strains of D. bryoniae. Moreover, in sensitivity tests, the detection limit was 1 ng/μL D. bryoniae DNA of the carbendazim-resistant strain. In addition, the LAMP method was successfully applied to detect carbendazim-resistant strains in D. bryoniae-infested samples. Therefore, the developed LAMP assay provides a new method for the rapid detection of carbendazim-resistant strains of D. bryoniae. Full article

The high expression of 17β-hydroxysteroid dehydrogenase type 1 (17β-HSD1) mRNA has been found in breast cancer tissues and endometriosis. The current research focuses on preparing a range of organic molecules as 17β-HSD1 inhibitors. Among them, the derivatives of hydroxyphenyl naphthol steroidomimetics are reported as one of the potential groups of inhibitors for treating estrogen-dependent disorders. Looking at the recent trends in drug design, many halogen-based drugs have been approved by the FDA in the last few years. Here, we propose sixteen potential hydroxyphenyl naphthol steroidomimetics-based inhibitors through halogen substitution. Our Frontier Molecular Orbitals (FMO) analysis reveals that the halogen atom significantly lowers the Lowest Unoccupied Molecular Orbital (LUMO) level, and iodine shows an excellent capability to reduce the LUMO in particular. Tri-halogen substitution shows more chemical reactivity via a reduced HOMO–LUMO gap. Furthermore, the computed DFT descriptors highlight the structure–property relationship towards their binding ability to the 17β-HSD1 protein. We analyze the nature of different noncovalent interactions between these molecules and the 17β-HSD1 using molecular docking analysis. The halogen-derived molecules showed binding energy ranging from −10.26 to −11.94 kcal/mol. Furthermore, the molecular dynamics (MD) simulations show that the newly proposed compounds provide good stability with 17β-HSD1. The information obtained from this investigation will advance our knowledge of the 17β-HSD1 inhibitors and offer clues to developing new 17β-HSD1 inhibitors for future applications. Full article

Research into the biodeteriorative potential of fungi can serve as an indicator of the condition of heritage items. Biodeterioration of canvas paintings as a result of fungal metabolic activity is understudied with respect to both the species diversity and mechanisms involved. This study brings new evidence for the physiology of fungi biodeteriorative capacity of canvas paintings. Twenty-one fungal isolates were recovered from four oil paintings (The Art Museum, Cluj-Napoca) and one gouache painting (private collection), dating from the 18th to 20th centuries. The species, identified based on the molecular markers Internal Transcribed Spacer (ITS), beta-tubulin (tub2), or translation elongation factor 1 (TEF-1), are common colonisers of canvas paintings or indoor environments (e.g., Penicillium spp., Aspergillus spp., Alternaria spp.). Fungi enzymatic profiles were investigated by means of hydrolysable substrates, included in culture media or in test strips, containing components commonly used in canvas paintings. The pigment solubilisation capacity was assessed in culture media for the primary pigments and studied in relation to the organic acid secretion. Caseinases, amylases, gelatinases, acid phosphatase, N-acetyl-β-glucosaminidase, naphthol-AS-BI-phosphohydrolase, and β-glucosidase were found to be the enzymes most likely involved in the processes of substrate colonisation and breakdown of its components. Aureobasidium genus was found to hold the strongest biodeteriorative potential, followed by Cladosporium, Penicillium, Trichoderma, and Aspergillus. Blue pigment solubilisation was detected, occurring as a result of organic acids secretion. Distinct clusters were delineated considering the metabolic activities detected, indicating that fungi specialise in utilisation of certain types of substrates. It was found that both aged and modern artworks are at risk of fungal biodeterioration, due to the enzymatic activities’ diversity and intensity, pigment solubilisation capacity or pigment secretion. The three-dimensional electro-Fenton (3D-EF) system using a nano zero-valent iron-supported biochar (NZVIs-BC) particle electrode and a Cu-supported stainless steel electrode (Cu-SSE) was proposed to avoid the disadvantages of the homogeneous electro-Fenton. In this work, the 3D-EF system was developed, which consisted of a Cu-SSE (cathode), a graphite rod (anode) and a NZVIs-BC particle electrode. The effect of the ratio of ferrous sulfate heptahydrate (FS) to rice straw (RS), CuSO4•5H2O amount, initial pH of 2-naphthol wastewater and current intensity (the output current of the power supply) on the removal rate of 2-naphthol were investigated. It is noteworthy that more than 98.36% of the 2-naphthol in aqueous solution was removed by the 3D-EF system, and only about 60.09% of 2-naphthol was removed by the homogeneous electro-Fenton system. Furthermore, naphthalene, benzoic acid, β-naphthoquinone, 1, 2-naphthalenedione, phenol and aromatic hydrocarbon were the main degradation products of 2-naphthol by the 3D-EF system; the toxicity of 2-naphthol wastewater was also greatly reduced. The synthesis of 14-aryl 14H-dibenzo[a,j]xanthenes is achieved by a simple condensation reaction between β-naphthol with aryl or alkyl aldehydes in an effective synergetic catalytic system created by combining basic bleaching earth clay and PEG-600. The advantages of the present method include catalyst recyclability, superior product yield, a shorter reaction time and the avoidance of hazardous reagents. Synthesized xanthene derivatives were also screened for their antibacterial activity against Staphylococcus aureus (MTCC 96) and Pseudomonas aeruginosa (Wild). Full article
