Stevia rebaudiana Bertoni, a semi-perennial herb from the Asteraceae family, is native to the Paraguay–Brazil border region. The growing industrial interest in this species is due to its natural sweetening properties, such as steviol and its derivatives, which offer sweetness without adding calories. Morphological traits are crucial for assessing genetic variability and ensuring distinctness, homogeneity, and stability (DHS) for cultivar protection. This study characterized 19 elite Stevia genotypes from Embrapa Cerrados’ Active Germplasm Bank (BAG) using 21 morphological descriptors from Brazil’s Ministry of Agriculture, Livestock, and Supply (MAPA). Genetic distances were calculated using the simple coincidence index complement method, and clustering was performed via the Unweighted Pair-Group Method with Arithmetic Mean (UPGMA). The results showed that 17 of the 21 descriptors (>80%) effectively differentiated the genotypes, revealing significant genetic variability. Dendrogram analysis identified at least four major similarity groups, highlighting the potential of these genotypes for Stevia breeding programs. These findings underscore the suitability of these elite genotypes for developing superior varieties adapted to Cerrado conditions, supporting future cultivation and genetic improvement efforts. Water and nitrogen (N) availability are among the primary limiting factors for the productivity of tomato (Solanum licopersicum L.). This study evaluated the interaction between these factors by assessing the effects of different N concentrations (85.5, 128.3, 171.0, 213.8, and 256.1 ppm N) on the water consumption, growth, and photosynthetic efficiency of hydroponically-grown tomato plants. The variables that were analyzed included the leaf N content, leaf chlorophyll index (LCI), maximum quantum efficiency of photosystem II (the ratio of variable to maximum chlorophyll fluorescence; Fv/Fm), non-photochemical quenching (NPQ), fresh mass (FM), dry mass (DM), cumulative water consumption, and water use efficiency (WUE). Increasing N concentrations led to higher water consumption and FM accumulation. Dry biomass was quadratically related to the N concentration, which peaked between doses of 213.8 and 256.1 ppm N. The LCI and Fv/Fm increased with the N supply, reaching a peak at N concentrations above 171 ppm, and then remained relatively constant. Conversely, the NPQ was reduced at the highest N level (256.1 ppm), which indicated diminished excess energy dissipation capacity. The highest WUE was observed at 213.8 ppm N, which was associated with greater DM and reduced water consumption compared to the highest N treatment. These findings suggest that the N concentration significantly affects the biomass production and water use in hydroponically-grown tomato plants, with 213.8 ppm N being the most efficient for vegetative growth under the studied conditions. Background/Objectives: Familial hypercholesterolemia (FH) is an increasingly common inherited disorder that increases cardiovascular risk. Despite the importance of lifestyle interventions, adherence to a healthy diet among individuals with FH remains suboptimal. This pilot, multicenter, double-blind, placebo-controlled randomized trial aimed to evaluate the feasibility and preliminary effects of a culturally adapted cardioprotective diet (DICA-FH), alone or in combination with phytosterol and/or krill oil supplementation, on lipid parameters in Brazilian adults with probable or definitive FH. Methods: Between May and August 2023, 58 participants were enrolled across nine Brazilian centers and randomized (1:1:1:1) into four groups: DICA-FH + phytosterol placebo + krill oil placebo; DICA-FH + phytosterol 2 g/day + krill oil placebo; DICA-FH + phytosterol placebo + krill oil 2 g/day; and DICA-FH + phytosterol 2 g/day + krill oil 2 g/day. Interventions lasted 120 days. The primary outcomes were mean low-density lipoprotein cholesterol (LDL-c) and lipoprotein(a) (Lp[a]) levels, as well as adherence to treatment at follow-up. Secondary outcomes included mean levels of other lipids, frequency of adverse events, and assessment of protocol implementation components. All data were presented separately for the allocation groups: phytosterol vs. placebo and krill oil vs. placebo. Results: Mean age was 54.5 ± 13.7 years, and 58.6% were women. Both adherence to protocol (91.8% attendance; 79.1% investigational product intake) and retention (86.2%) were high. No significant differences between groups were found for LDL-c or Lp(a). However, regardless of allocation to active supplementation or placebo, a significant reduction in Lp(a) concentrations was observed following the DICA-FH intervention (median difference: −3.8 mg/dL [interquartile range: −7.5 to −1.2]; p < 0.01). Significant reductions in oxidized LDL (LDL-ox) and LDL-ox/LDL-c ratio were also observed in the overall sample (p < 0.01). Although not statistically significant, all groups showed improvements in diet quality after 120 days. No serious adverse events related to the interventions were reported. Additionally, most protocol implementation components were successfully achieved. Conclusions: The DICA-FH strategy, with or without supplementation, was safe and well-tolerated. Although not powered to detect clinical efficacy (which is acceptable in exploratory pilot trials), the study supports the feasibility of a larger trial and highlights the potential of dietary interventions in the management of HF. Background/Objective: Yellow fever (YF) is an acute infectious disease caused by the yellow fever virus which is transmitted by mosquitoes. Neotropical primates are susceptible to infection, which is often presented as epizootic outbreaks. The aim was to evaluate and characterize the immune response against YF in different species of neotropical primates from the Belo Horizonte Zoo. Methods: Vaccine 17DD was administered to 24 neotropical primates, with a single subcutaneous dose. Clinical exams, RNAemia, and detection of IgG and neutralizing antibodies against YFV were performed. In addition, an ethogram was performed to assess clinical changes and animal welfare. Results: At 4 days post-vaccination, RNAemia was detected in nine animals. There was seroconversion and persistence of immune response in Alouatta guariba clamitans, Sapajus xanthosternos, Saguinus imperator and Aotus infulatus. However, the vaccine was not immunogenic for Lagothrix cana. In Pithecia irrorata seroconversion did not persist long term, while the Ateles sp. had a transient immune response. No significant clinical manifestations were observed in any of the vaccinated animals. Conclusions: This study demonstrated a safe, immunogenic and persistent immune response induced by the attenuated 17DD vaccine strain in A. guariba clamitans, S. xanthosternos, S. imperator, and A. infulatus. Pseudoplacentational endometrial hyperplasia (PEH) and cystic endometrial hyperplasia (CEH) are both hyperplastic conditions that may occur in the canine uterus during diestrus. CEH can impair fertility, and, although the consequences of PEH are poorly known, this condition is significantly associated with pyometra. The aim of this study was to investigate frequencies of both PEH and CEH in female dogs according to age, size, breed, and breed group. Uteri from 300 female dogs were sampled and processed for histopathologic analyses. Lesions were identified, and frequencies were statistically compared. PEH was significantly more frequent in dogs between 4 and 12 years of age and significantly less frequent in Yorkshires than Shih-tzus, as well as in the breed group that includes Brazilian Terriers and Yorkshires. CEH was significantly more frequent in dogs older than 12 years, but no breed predisposition was observed. The frequency of PEH was significantly higher than that of CEH. The frequency of pyometra in PEH cases was significantly higher than endometritis or uterus without inflammation. This study identified age, breed group, and size as predisposing factors for PEH and further demonstrated the association between this condition and pyometra in dogs. Full article

Background/Objectives: The optimal dietary approach for managing glycemic and metabolic control in type 2 diabetes (T2D) is still uncertain, though it should be tailored for clinical settings. Therefore, we sought to assess the impact of a multicomponent nutritional strategy on glycemic control in T2D patients within a public health system. Methods: NUGLIC was an open-label, parallel-group, superiority, multicenter randomized controlled trial. Participants aged 30 and older with poorly controlled T2D were randomly assigned to either (1) a personalized dietary prescription (control group, n = 185) or (2) a strategy involving targeted nutritional advising, mindfulness techniques, and short message services (NUGLIC [intervention] group, n = 186). The primary outcomes were glycated hemoglobin (HbA1c, %) measured after 24 weeks and glycemic control, defined as having an HbA1c > 7% at baseline and achieving ≤7% after follow-up, or having HbA1c ≤ 7% at baseline and reducing the use of glucose-lowering medications post-follow-up. The secondary outcomes included cardiometabolic features, self-care practices, diet quality, and quality of life. Results: A total of 371 participants were included in an intention-to-treat analysis for the primary outcomes. At six months, both groups exhibited a reduction in HbA1c levels compared to the baseline (NUGLIC group: −0.6% [95% confidence interval (CI) −0.9; −0.3], p < 0.001; control group: −0.5% [95% CI −0.7; −0.3], p < 0.001). However, no significant differences were observed between the groups in terms of HbA1c after follow-up (intervention group: 8.1%; control group: 8.3%; difference: −0.2% [95% CI −0.5; 0.1], p = 0.30) or glycemic control (NUGLIC group: 19.9%; control group: 18.9%; odds ratio 0.96 [95% CI 0.56; 1.67], p = 0.89). While the control group showed an improvement in overall diet quality, no significant differences emerged between the groups by the end of this study (p = 0.13). There were also no significant differences in other secondary outcomes nor in the use of glucose-lowering medications and adverse events after follow-up. Conclusions: The multicomponent nutritional strategy did not demonstrate superiority over personalized dietary prescriptions in achieving glycemic control for participants with poorly managed T2D. In this sense, both nutritional interventions could be used in clinical practice to improve HbA1c levels, considering the profile and preferences of individuals. Full article

Multidrug-resistant bacteria have complicated the treatment of gastrointestinal diseases; their microbial resistance stems from the indiscriminate use of medications and the transfer of resistance genes. Varronia curassavica Jacq., a plant traditionally used to treat rheumatic and gastrointestinal diseases in underserved populations, has sparked interest as a potential source of antimicrobial compounds. This study aimed to investigate the chemical composition and antibacterial effects of V. curassavica essential oil and to evaluate its toxicity in Drosophila melanogaster. The essential oil was extracted through hydrodistillation and its chemical composition was determined using GC-MS. Antibacterial tests were performed with microdilution. The results showed the presence of major compounds including α-pinene and β-caryophyllene. The essential oil did not show relevant MIC, but it enhanced the effects of the antibiotics, gentamicin, norfloxacin, and oxacillin. It exhibited no toxicity and did not affect geotaxis, even at high concentrations. The in silico analysis of α-pinene revealed low toxicity; however, its permeability to the BBB shows that caution is needed in its application. These results indicate that the essential oil of V. curassavica shows promising potential in enhancing pharmaceuticals to prevent increased bacterial resistance. In addition, it demonstrated safe aspects when tested on D. melanogaster. Full article

Immunoglobulins Y (IgY) purified from egg yolks of hens represents an attractive, cost-effective alternative for the development of new diagnostic and therapeutic platforms. In this study, we evaluated the therapeutic efficacy of rotavirus-specific IgY in a cynomolgus monkey (Macaca fascicularis) model. Animals were experimentally infected with human rotavirus Group A (RVA), the most common cause of severe acute diarrhoea among young children worldwide. Animals were administered human RVA (3.1 × 107 FFU/mL) by oral gavage, challenged with 2.5 mg of anti-RVA IgY orally, and monitored for five days according to clinical, haematological and biochemical parameters; serum electrolyte levels; viral shedding; and histopathological changes. Immunotherapy with anti-RVA IgY had a protective effect against severe rotavirus-induced enteritis in four of the ten treated monkeys, as evidenced by histopathological findings. Although only one animal had diarrhoea, all but one exhibited virus shedding regardless of the treatment. Full article

Chlorpyrifos, an organophosphate insecticide widely used to control agricultural pests, poses a significant environmental threat due to its toxicity and persistence in soil and water. Our work aimed to evaluate the acute (survival) and chronic (regeneration, locomotion, and reproduction) toxicity of chlorpyrifos to the non-target freshwater planarian Girardia tigrina. The 48 h lethal concentration (LC50) of the commercial formulation, containing 480 g L−1 of chlorpyrifos, the active ingredient, was determined to be 622.8 µg a.i. L−1 for planarians. Sublethal effects were translated into a significant reduction in locomotion and delayed head regeneration (lowest observed effect concentration—LOEC = 3.88 µg a.i. L−1). Additionally, chlorpyrifos exposure did not affect planarian fecundity or fertility. Overall, this study demonstrates the potential of chlorpyrifos-based insecticides to harm natural populations of freshwater planarians at environmentally relevant concentrations. The observed toxicity emphasizes the need for stricter regulations and careful management of chlorpyrifos usage to mitigate its deleterious effects on aquatic ecosystems. By understanding the specific impacts on non-target organisms like G. tigrina, we can make more informed suggestions regarding the usage and regulation of organophosphate insecticides, ultimately promoting sustainable agricultural practices and environmental conservation. Morinda citrifolia Essential Oil: A Plant Resistance Biostimulant and a Sustainable Alternative for Controlling Phytopathogens and Insect Pests

With the growing demand for sustainable and safe agricultural practices, plant compounds emerge as a solution for biological activities. Here, we evaluated the potential of using Morinda citrifolia essential oil to induce plant resistance and to control phytopathogens (Curvularia lunata) and insect pests (Daubulus maidis). We conducted a chromatographic analysis to unveil the essential oil components. We also quantified the activity levels of antioxidant enzymes and chitinase for resistance induction. The antifungal action was evaluated through disease progression and the inhibition of mycelial growth in addition to in silico studies that made it possible to predict the interaction site between the fungal protein and the compounds. We assessed the toxicity and repellent actions towards the D. maidis. Octanoic acid (58.43%) was identified as the essential oil major compound. Preventive treatment with essential oil and octanoic acid (25.0 µL mL−1) increased not only the plant defense activities (i.e., the activity of the enzymes superoxide dismutase, catalase, phenol peroxidase, ascorbate peroxidase, and chitinase) but also controlled Curvularia leaf spot. The stable interactions between octanoic acid and tyrosine-tRNA ligase from C. lunata suggested protein synthesis inactivation. The essential oil inhibited 51.6% of mycelial growth, and this effect was increased to 75.9% with the addition of adjuvants (i.e., angico gum). The essential oil reduced 76% of the population of D. maidis adults and repelled 50% of the number of D. maidis after 48 h under field conditions. The repellency effect in the field reduced the population of D. maidis adults, transmitters of the stunting complex, by 50%. The results highlight the potential of M. citrifolia as a resistance activator, fungicide, insecticide, and an effective biorational alternative. Full article

Natural fiber reinforcements have the potential to enhance mechanical properties, thereby improving performance and durability in various applications. In this study, we comprehensively evaluated the impact of environmental degradation over 120 days on reprocessed polypropylene (PP) reinforced with corn husk fiber (CHF) composites. The manufactured systems underwent rigorous analysis using various techniques, including Fourier transform infrared spectroscopy, thermogravimetric analysis, optical microscopy, scanning electron microscopy, and tensile testing. These analyses revealed that climatic conditions significantly influenced (p < 0.05) the mechanical properties of all systems. Photodegradation led to surface morphological changes and chemical structures. Regardless, adding CHF filler proved a key factor, as it allowed for less susceptibility to environmental degradation than the reprocessed matrix. These findings, therefore, provide robust evidence supporting the feasibility of using CHF composites for manufacturing agricultural containers. Full article

The citrus blackfly (CBF), Aleurocanthus woglumi Ashby, is an exotic pest native to Southeast Asia that has spread rapidly to the world’s main centers of citrus production, having been recently introduced to Brazil. In this study, a maximum entropy niche model (MaxEnt) was used to predict the potential worldwide distribution of CBF under current and future climate change scenarios for 2030 and 2050. These future scenarios came from the Coupled Model Intercomparison Project Phase 6 (CMIP6), SSP1-2.6, and SSP5-8.5. The MaxEnt model predicted the potential distribution of CBF with area under receiver operator curve (AUC) values of 0.953 and 0.930 in the initial and final models, respectively. The average temperature of the coldest quarter months, precipitation of the rainiest month, isothermality, and precipitation of the driest month were the strongest predictors of CBF distribution, with contributions of 36.7%, 14.7%, 13.2%, and 10.2%, respectively. The model based on the current time conditions predicted that suitable areas for the potential occurrence of CBF, including countries such as Brazil, China, the European Union, the USA, Egypt, Turkey, and Morocco, are located in tropical and subtropical regions. Models from SSP1-2.6 (2030 and 2050) and SSP5-8.5 (2030) predicted that suitable habitats for CBF are increasing dramatically worldwide under future climate change scenarios, particularly in areas located in the southern US, southern Europe, North Africa, South China, and part of Australia. On the other hand, the SSP5-8.5 model of 2050 indicated a great retraction of the areas suitable for CBF located in the tropical region, with an emphasis on countries such as Brazil, Colombia, Venezuela, and India. In general, the CMIP6 models predicted greater risks of invasion and dissemination of CBF until 2030 and 2050 in the southern regions of the USA, European Union, and China, which are some of the world’s largest orange producers. Knowledge of the current situation and future propagation paths of the pest serve as tools to improve the strategic government policies employed in CBF’s regulation, commercialization, inspection, combat, and phytosanitary management. Infectious and Parasitic Diseases (IPD) remain a challenge for medicine due to several interconnected reasons, such as antimicrobial resistance (AMR). American tegumentary leishmaniasis (ATL) is an overlooked IPD causing persistent skin ulcers that are challenging to heal, resulting in disfiguring scars. Moreover, it has the potential to extend from the skin to the mucous membranes of the nose, mouth, and throat in both humans and various animals. Given the limited effectiveness and AMR of current drugs, the exploration of new substances has emerged as a promising alternative for ATL treatment. Arrabidaea brachypoda (DC). Bureau is a native Brazilian plant rich in dimeric flavonoids, including Brachydin (BRA), which displays antimicrobial activity, but still little has been explored regarding the development of therapeutic formulations. In this work, we present the design of a low-cost liquid formulation based on the use of Pluronic F127 for encapsulation of high BRA concentration (LF-B500). The characterization techniques revealed that BRA-loaded F127 micelles are well-stabilized in an unusual worm-like form. The in vitro cytotoxicity assay demonstrated that LF-B500 was non-toxic to macrophages but efficient in the inactivation of forms of Leishmania amazonensis promastigotes with IC50 of 16.06 µg/mL. The results demonstrated that LF-B500 opened a new perspective on the use of liquid formulation-based natural products for ATL treatment. Full article

Toxoplasmosis is an important zoonotic disease caused by the parasite Toxoplasma gondii and is especially fatal for neotropical primates. In Brazil, the Ministry of Health is responsible for national epizootic surveillance, but some diseases are still neglected. Here, we present an integrated investigation of an outbreak that occurred during the first year of the COVID-19 pandemic among eleven neotropical primates housed at a primatology center in Brazil. After presenting non-specific clinical signs, all animals died within four days. A wide range of pathogens were evaluated, and we successfully identified T. gondii as the causative agent within four days after necropsies. The liver was the most affected organ, presenting hemorrhage and hepatocellular necrosis. Tachyzoites and bradyzoite cysts were observed in histological examinations and immunohistochemistry in different organs; in addition, parasitic DNA was detected through PCR in blood samples from all specimens evaluated. A high prevalence of Escherichia coli was also observed, indicating sepsis. This case highlights some of the obstacles faced by the current Brazilian surveillance system. A diagnosis was obtained through the integrated action of researchers since investigation for toxoplasmosis is currently absent in national guidelines. An interdisciplinary investigation could be a possible model for future epizootic investigations in animals. Full article

The COVID-19 pandemic, caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), challenged public health systems worldwide. Individuals in low-income countries/regions are still at individual and community risk concerning inequality, sanitation, and economic conditions. Besides, during the pandemic, the transmission in municipalities and communities in the countryside and less developed regions kept viral spread and required structured and strengthened clinical and laboratory surveillance. Here, we present an observational, analytic, cross-sectional study conducted using secondary data from the Laboratório de Farmacogenômica e Epidemiologia Molecular (LAFEM)-Universidade Estadual de Santa Cruz (UESC), to evaluate individual and community factors associated to SARS-CoV-2 infection in outpatients from different cities from Southern Region of Bahia State, in Brazil. The data were collected between June 2021 and May 2022. The SARS-CoV-2 positivity by RT-qPCR was correlated with low socio-economic indicators, including the Human development index (HDIc) and Average worker salary (AWSc). Besides, in general, females were less likely to test positive for SARS-CoV-2 (OR = 0.752; CI 95% 0.663–0.853; p < 0.0001), while brown individuals had more positivity for infection (p < 0.0001). In addition, those who had clinical symptoms were more likely to test positive for SARS-CoV-2 (OR = 6.000; CI 95% 4.932–7.299; p < 0.0001). Although dry cough, headache, and fever were the most frequent, loss of taste (OR = 5.574; CI 95% 4.334–7.186) and loss of smell (OR = 6.327; CI 95% 4.899–8.144) presented higher odds ratio to be positive to SARS-CoV-2 by RT-qPCR. Nonetheless, the distribution of these characteristics was not homogenous among the different cities, especially for age and gender. The dynamic of SARS-CoV-2 positivity differed between cities and the total population and reinforces the hypothesis that control strategies for prevention needed to be developed based on both individual and community risk levels to mitigate harm to individuals and the health system.
