Evaluation of the Genotoxicity and Cytotoxicity of Bioceramic Endodontic Sealers in HepG2 and V79 Cell Lines: An In Vitro Study Using the Comet and Micronucleus Assays

Background: The primary objective of this study was to evaluate the cytotoxic and genotoxic effects of calcium silicate-based sealers (BioRoot RCS and MTA Fillapex) compared to an epoxy-based sealer (AH Plus). Materials and methods: The study was conducted in vitro with the cell lines HepG2 and V79 to evaluate cytotoxicity and genotoxicity using the comet and micronucleus assays. Eluates of the materials were tested at two different concentrations (3 cm2/mL and 0.5 cm2/mL) after an exposure time of 72 h. Data were analyzed using the Mann–Whitney and Kruskal–Wallis tests (p < 0.05). Results: At lower concentrations in both cell lines, MTA Fillapex showed no significant difference in the measured comet assay parameters compared to the negative control (p > 0.05). In addition, it showed significantly lower genotoxic effects compared to AH Plus for all comet assay parameters, concentrations, and cell lines (p ≤ 0.001). BioRoot RCS showed lower primary DNA damage (p ≤ 0.001) than AH Plus, only at higher concentrations and in the HepG2 cell line. Concerning the two tested bioceramic sealers, BioRoot RCS showed higher tail intensity values compared to MTA Fillapex (p < 0.05). In contrast to the results of the comet assay, BioRoot RCS significantly reduced the number of nuclear buds and nucleoplasmic bridges in the HepG2 cell line compared to MTA Fillapex, whereas reduction in the V79 cell line was only observed for nuclear buds (p < 0.05). Both materials increased the number of apoptotic cells compared to the negative control (p < 0.05). In comparison to AH Plus, BioRoot RCS and MTA Fillapex significantly reduced the number of cells with micronuclei and increased the number of cells with undamaged chromatin (p < 0.05). Conclusions: The findings suggest that MTA Fillapex and BioRoot RCS exhibit superior biocompatibility over AH Plus, as evidenced by their lower cytotoxic and genotoxic effects in vitro. These results support the use of calcium silicate-based sealers in clinical practice, highlighting the need for further studies to evaluate their performance in vivo and their implications for patient safety. A future radioactive waste management centre is under development in central Croatia. One of the activities in the centre’s development was to monitor environmental radioactivity before the disposal of radioactive materials. Part of the monitoring programme focused on soil characterisation in the municipality (total area 1308 km2) surrounding the centre, where about 40% of the soil is today used in organic farming. The study included a physico-chemical and radionuclide characterisation of the soil as well as ambient dose rate measurements. The aim of this study was to investigate how the physical and chemical composition of soil affects the concentration of radionuclides 238U, 40K, and 137Cs in soil, based on the measured radionuclide concentrations and values of selected soil parameters. Additionally, the ambient equivalent dose rate H*(10)/t was measured and the annual effective dose was calculated for the average person living in the area of interest. The observed ranges of radionuclide concentrations in the soil samples were: 9–72 Bq/kg for 238U, 65–823 Bq/kg for 40K, and 4–80 Bq/kg for 137Cs. Ambient dose equivalent rate measurements were in the range of 52–130 nSv/h. The highest measured values were in correlation with higher 238U activity concentrations in these parts of the investigated area. The results of this study showed that 238U had a significant correlation with pH; plant available P; sand, silt, and clay content; hydrolytic acidity; CaCO3; total carbon, organic matter, and total inorganic and organic carbon; and concentrations of Al, Si, Fe, Ca, Ti, K, Rb, Zr, Nb, Y, Sr, Th, and W. 40K showed a significant correlation with pH, sand content, hydrolytic acidity, total hydrogen, total nitrogen, CaCO3, total carbon, total inorganic carbon, and concentrations of Al, Si, Fe, Ca, Ti, Rb, Zr, Nb, P, Y, Zn, and Th. 137Cs showed a significant correlation with silt content, total nitrogen, and Si concentration. Effective antimicrobial stewardship (AMS) is crucial for combating the rise of antimicrobial resistance (AMR), particularly in veterinary medicine. Educational programs targeting veterinary students can play a significant role in shaping their knowledge and attitudes toward antimicrobial use and stewardship. This study aimed to evaluate the impact of educational programs on the knowledge and attitudes regarding AMS among veterinary students in Serbia. A structured educational program on AMS was implemented for veterinary students at the Faculty of Agriculture, University of Novi Sad. Pre- and post-symposium assessments were conducted to measure changes in students’ knowledge and attitudes. The study employed a mixed-methods approach, including surveys and focus groups, to gather quantitative and qualitative data. The study resulted in significant short-term improvements in students’ knowledge of AMS principles and their attitudes toward responsible antimicrobial use. Participants demonstrated a better understanding of the mechanisms of resistance and the importance of adhering to stewardship guidelines. Qualitative feedback indicated increased awareness of the consequences of inappropriate antimicrobial use and a stronger commitment to applying stewardship practices in their future careers. The educational programs effectively enhanced immediate veterinary students’ knowledge and attitudes regarding AMS. These findings underscore the importance of integrating targeted educational programs into veterinary curricula to promote responsible antimicrobial use and combat resistance in veterinary practice. Patient satisfaction is a crucial measure of the quality of healthcare, especially with regard to nursing services in hospital settings. Understanding and increasing patient satisfaction with nursing care is critical to improving overall healthcare and ensuring positive patient experiences during their hospital stay. The aim of this research was to evaluate the psychometric properties of the Croatian version of the Satisfaction with Nursing Care Quality Questionnaire (PSNCQQ), test the reliability and validity of the tool after translation, and investigate differences in patient satisfaction based on demographic variables, as well as their contribution to satisfaction with the quality of nursing care. After translation and adaptation, the Croatian version of the PSNCQQ was applied to 350 hospitalized patients (average age 51.19 years (range: 18–87)), of whom 194 (55.4%) were men and 156 (44.6%) were women. The results showed high internal consistency (Cronbach’s α = 0.977) and confirmed the one-factor structure of the questionnaire, explaining 70.64% of the total variance. Confirmatory factor analysis supported the unidimensional model, showing strong fit indices (χ2 = 583.047, df = 149, p < 0.001, GFI = 0.861, AGFI = 0.818, NFI = 0.936, TLI = 0.946, CFI = 0.955, RMSEA = 0.080, PCLOSE < 0.001). In conclusion, the validation of the PSNCQQ in the Croatian language increases resources for evaluating and improving the quality of nursing care. This research lays the foundation for future studies and practical applications aimed at improving patient satisfaction and nursing care outcomes in Croatia, but there are also limitations to this study, including its one-institution scope, the possible influence of factors outside the current treatment on satisfaction, and the lack of comparison with objective clinical indicators. The purpose of this research was to examine whether demographic variables, personality traits, and workplace variables (working in shifts, job tenure, and perceived organizational justice) contribute the most to the prediction of job satisfaction in nurses. The survey included 161 nurses. The instruments used in this research were as follows: the Demographic Data Questionnaire, the Perceived Organizational Justice Scale, the Job Satisfaction Scale, and the NEO five-factor inventory. The study findings indicated that age, health status, distributive justice, and procedural justice positively contribute to job satisfaction among nurses, while neuroticism contributes negatively. Older nurses, those in better health, those who are satisfied with the organization’s decision-making process, and those who feel adequately rewarded for their contributions tend to be more satisfied with their jobs. Conversely, nurses with a higher level of the neuroticism personality trait tend to be less satisfied with their job. The strongest predictors of job satisfaction among nurses were found to be health status, the personality trait of neuroticism, and distributive and procedural justice, with the age of nurses being slightly less powerful but still significant. Maize is considered a promising candidate for biofortification through breeding, given its widespread cultivation and significance as a food crop. This cost-effective and sustainable approach could be used to increase the content of different tocol compounds, i.e., vitamin E, in maize grain due to the well-documented genetic variability. In the present study, an evaluation of the content of nine different tocol traits was performed in a genebank collection of 88 inbred lines of temperate maize grown at two locations in Croatia in 2019. A large genotypic variability within the studied material was observed for α-tocopherol, γ-tocopherol, δ-tocopherol, total tocopherols, α-/γ-tocopherol ratio, α-tocotrienol, γ-tocotrienol, total tocotrienols and total tocols with corresponding coefficients of variation of 52, 61, 51, 45, 106, 24, 54, 33 and 38%, respectively. Heritability estimates ranged from 0.66 for α-tocotrienol to 0.95 for γ-tocopherol. The content of α-tocopherol, which has the highest vitamin E activity and is therefore most interesting for selection, was not significantly correlated with either grain color or grain type. Comparison of the effects of simulated selection with an intensity of 20% on increased α-tocopherol content using the two selection criteria, absolute α-tocopherol content and α-/γ-tocopherol ratio, showed that the highest absolute α-tocopherol content was a better selection criterion. Indeed, simulated selection based on the absolute α-tocopherol content resulted in a 64% increase in this compound without significantly affecting the total tocopherols, the total tocotrienols, and the α-/γ-tocopherol ratio. Low-income regions do not have the economic strength to use conventional isolators; therefore, low-cost alternatives are needed. The recent series of earthquakes in Turkey has once again demonstrated the destructive power of this natural disaster and highlighted the need for high-quality earthquake-resistant structures. In this context, a comprehensive experimental parametric study on the use of natural stone pebbles (ASL-1) and stone pebbles in combination with a geogrid layer (ASL-2) as suitable materials for a GSI system is conducted and the main results are presented. The seismic behavior of five different models was tested using four different acceleration diagrams with different peak ground accelerations (PGAs). Characteristic displacements, accelerations, and strains were measured. The results and conclusions presented are based on the integration and synthesis of several previously conducted studies. Increasing pressure on animal and human health as well as food security, due to antimicrobial resistance, is leading to increased demand for natural-product-derived antibiotics worldwide. Considering the importance of bovine mastitis as well as the huge challenge posed by mastitis therapy in cattle production, the main goal of this research was to evaluate the effectiveness of the essential oil-based pharmaceutical (Phyto-Bomat), as an alternative to the existing treatment with antibiotics. The therapeutic response of the cows was monitored using clinical and bacteriological cure of the proposed formulation. Among 550 dairy cows, 75 were diagnosed with a clinical or subclinical form of mastitis and divided into three experimental groups: treated with antibiotic; Phyto-Bomat; antibiotic and Phyto-Bomat. Indicators of bacteriological cure were somatic cell count (SCC), total bacterial count, and causative agent isolation, while the clinical cure was accessed by observing physical udder conditions. All mentioned indicators were assessed before and after the treatment (1st and 7th day post-treatment), with all of the three therapeutic protocols. After Phyto-Bomat treatment, no visible signs of irritation were noticed, while no significant effect in reducing SCC and total bacterial count was observed. Phyto-Bomat has shown promising results in the treatment of mastitis, without the risk of udder irritation, alone or in combination with antibiotics as part of a mastitis control program during lactation. This paper aims to propose a statistical model to assess pubertal growth spurt using the ratio of the anterior height projection to the posterior (Vba) of the fourth cervical vertebra body (C4) on cephalograms and to calculate the residual proportion of skeletal maturation and the time for the pubertal growth spurt to end for a given Vba. A sample of 538 cephalograms from healthy-living children aged between 5 and 15 years was analyzed. A segmented regression model was used to explain the different Vba stages relative to the pubertal growth spurt. In addition, the time to achieve skeletal maturation was evaluated for a given Vba between the beginning (Vba1) and the end (Vba2) of the pubertal growth spurt. A longitudinal sample of 25 males and 25 females was analyzed to validate the proposed method. The values of Vba corresponding to higher pubertal development rate ranged from Vba1 = 0.677 (95%CI, 0.644–0.711) to Vba2 = 0.966 (95%CI, 0.905–1.028) and from Vba1 = 0.669 (95%CI, 0.645–0.693) to Vba2 = 1.073 (95%CI, 1.044–1.101) in males and females, respectively. The validation process results showed that our model did not produce any incorrect forecasts. The proposed method estimates the beginning and the end of the pubertal growth spurt together with the residual proportion of skeletal maturation for a given Vba. Full article

Due to the emergence of antibiotic-resistant bacteria, the risk it represents to public health, and the possible consequences for animal health and welfare, there is an increasing focus on reducing antimicrobial usage (AMU) in animal husbandry. Therefore, a great interest in developing alternatives to AMU in livestock production is present worldwide. Recently, essential oils (EOs) have gained great attention as promising possibilities for the replacement of antibiotics. The current study aimed to test the potential of using a novel EO-based pharmaceutical formulation (Phyto-Bomat) in bovine mastitis treatment. The antibacterial activity was performed using the microdilution technique. Lactating dairy cows were treated with 15 mL of Phyto-Bomat in the inflamed quarter for 5 consecutive days in order to analyze blood and milk samples for thymol and carvacrol residues using gas chromatography and mass spectrometry (GC–MS). Antimicrobial activity expressed as the minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) indicates that this formulation has the highest activity against Gram-positive strains. The dominant compounds in Phyto-Bomat were thymol and carvacrol, at 12.58 ± 1.23 mg/mL and 23.11 ± 2.31 mg/mL, respectively. The quantification of these two compounds in evaluated biological samples showed that 24 h after administration the concentration of thymol and carvacrol in milk samples was at the same level as before application. On the other hand, thymol and carvacrol were detectable in plasma samples even after 24 h post-treatment, with values ranging from 0.15–0.38 and 0.21–0.66 µg/mL, respectively. The tested formulation showed encouraging results of antibacterial activity against bovine mastitis pathogens, as well as the withdrawal period of dominant compounds, which implies that further testing regarding the bacteriological and clinical cure rates in clinical settings is needed. The main subject of the research is the assessment of the knowledge, attitudes and behaviors of veterinarians regarding the use of antibiotics (AMU) and antimicrobial resistance (AMR) through a questionnaire conducted among veterinarians in the northern region of Serbia. A total of 62 respondents completed the questionnaire, which represents a response rate of 44.3%. Male veterinarians are less likely to be in the group of veterinarians with insufficient knowledge (p < 0.05). Veterinarians engaged in mixed practice (small and large animals) (p < 0.001) and veterinarians who have over 100 patients per month (p < 0.005) are also less likely to be in the group with insufficient knowledge of antimicrobial resistance. The proportion of those with insufficient knowledge is growing among veterinarians whose source is the Internet (p < 0.01), while the proportion of those with insufficient knowledge about antimicrobial resistance is declining among veterinarians whose source of information is continuous education (p < 0.05). The majority of the respondents (n = 59, 95.2%) completely agreed that AMR is a very big issue in the global health sector right now. Unfortunately, there are crucial gaps in the knowledge and attitudes of the surveyed participants. They do not appear to be aware of the importance of AMU in veterinary medicine and its influence on overall AMR, or the crucial part that non-prescribed antibiotics have in all of it. Positively, many veterinarians use good practice AMU guidelines in their everyday practice and in line with the global trend of AMU reduction, respondents have also decreased their AMU compared to the previous year. The study was conducted on a commercial pig farm located in Serbia. Thirty Duroc or Landrace breed boars were randomly selected for this study. The experimental group was fed a compound feed with added organic selenium and Oxynat 3D. The antioxidant status parameters of boar seminal plasma were evaluated using a biochemical analyzer and commercial Randox kits. The sperm chromatin structure assay (SCSA) using flow cytometry (FC) provided information about spermatozoa’s DNA status. Additionally, the total number of motile spermatozoa and spermatozoa kinematic parameters were measured using the computer-assisted sperm analysis (CASA) system. The aim of this study was to improve the parameters of semen by combining two preparations that have a potential antioxidant effect, but also to establish the level of various antioxidant enzymes in native sperm. There was no statistically significant difference in total antioxidant capacity and glutathione peroxidase activity in the seminal plasma obtained from the experimental and control groups of boars. Regarding the superoxide dismutase activity, the research results showed a difference in the control group compared to the experimental one. Moreover, spermatozoa DNA fragmentation and the total number of motile spermatozoa showed statistically significant lower and higher values, respectively, in experimental compared to the control groups. The combination of these two preparations shows significantly enhanced vital parameters of semen. To the best of our knowledge, this study is the first in which the ejaculate parameters were examined after the application of a combination of these two antioxidant supplements. Full article

Development of new cultivars and agronomic improvements are key factors of increasing in future grain yield in maize grown in environments affected by climate change. Assessment of value for cultivation and use (VCU) reflects the results of latest breeding efforts showing yield trends, whereby external environmental covariates were rarely used. This study aimed to analyze several environmental effects including stress degree days (SDD) on grain yields in Croatian VCU trials in three maturity groups using linear mixed model for the estimation of fixed and random effects. Best linear unbiased predictions (BLUPs) of location-year interaction showed no pattern among maturity groups. SDD showed mostly non-significant coefficients of regression on location BLUPs for yield. Analyzing location BLUPs, it was shown that the effect became consistently stronger with later maturity, either positive or negative. The effects of management might play more critical role in maize phenology and yield formation compared with climate change, at least in suboptimum growing conditions often found in Southeast Europe. To facilitate more robust predictions of the crop improvement, the traditional forked approach dealing with G × E by breeders and E × M by agronomists should be integrated to G × E × M framework, to assess the full gradient of combinations forming the adaptation landscape. The successful exploration of dimension stone mainly depends on the quality, size, and shape of extractable blocks of dimension stone. The investigated area is in the Pelješac Peninsula (Croatia), in the External Dinarides orogeny, built from thick carbonate succession, characterized by relatively small deposits of high-quality dimension stone. These conditions demand challenging geological investigations in the “pre-quarry” phase to find optimal quarry location. The size and shape of dimension stone blocks are mainly controlled by fracture pattern systems. In the rugged, covered terrains, it is very hard to obtain a satisfactory amount of fracture data from the surface, so it is necessary to collect them from the underground. Borehole camera technology can visualize the inner part of the rock mass and measure the fracture characteristics. The main conclusions are as follows: (1) the digital borehole camera technology provides a quick, effective, and low-cost geological survey of fractured rock mass; (2) statistical fracture distribution parameters, P10, fracture spacing, Volumetric Joint Count (Jv) based on borehole wall survey can reflect the integrity of rock mass, providing a solid decision-making base for further investment plans and dimension stone excavation method.
