In light of the drawbacks of using fossil fuel, this paper investigates the competition between geographically spread charge point operators for future battery electric long-haul trucks along one of the busiest highways in Sweden. This is achieved using an agent-based model where trucks try to charge for a low price and still avoid queues in order to complete their transport mission. The charging need for a typical day at full electrification is derived from data from the Swedish Transport Administration. This typical day is simulated several times and in between these iterations the charge point operators adjust their prices and number of chargers, aiming to increase their profit. After a sufficiently long time of competition, a quasi-equilibrium is reached where, for example, prices and queueing times can be studied. The goal of the study is to estimate conditions for trucks and charge point operators in a future public fast-charging market. Assuming a price for electricity of 0.08 EUR/kWh, the results indicate that a system with low queuing problems is attainable with a mean price of 0.27 EUR/kWh or lower for public fast charging. It is also found that the behaviour of haulage companies, as a collective, can affect the future fast charging market to a great extent. If the hauliers are price-sensitive, they will be offered a low mean price, down to 0.11 EUR/kWh, but with queues, while if they are queue-sensitive, there will be almost no queues, but they will pay more to charge. Flaviviruses are usually transmitted to humans via mosquito or tick bites, whose infections may lead to severe diseases and fatality. During intracellular infection, they remodel the endoplasmic reticulum (ER) membrane to generate compartments scaffolding the replication complex (RC) where replication of the viral genome takes place. In this study, we purified the ER membrane fraction of virus infected cells to identify the proteins that were enriched during flavivirus infection. We found that tripartite motif-containing proteins (TRIMs) including TRIM38, TRIM21, and TRIM14 were significantly enriched during infection with mosquito-borne (West Nile virus strain Kunjin and Zika virus (ZIKV)) and tick-borne (Langat virus (LGTV)) flaviviruses. Further characterizations showed that TRIM21 and TRIM14 act as restriction factors against ZIKV and LGTV, while TRIM38 hinders ZIKV infection. These TRIMs worked as interferon-stimulated genes to mediate IFN-I response against LGTV and ZIKV infections. Restriction of ZIKV by TRIM14 and TRIM38 coincides with their colocalization with ZIKV NS3. TRIM14-mediated LGTV restriction coincides with its colocalization with LGTV NS3 and NS5 proteins. However, TRIM21 did not colocalize with ZIKV and LGTV NS3 or NS5 protein suggesting its antiviral activity is not dependent on direct targeting the viral enzyme. Finally, we demonstrated that overexpression of TRIM21 and TRIM14 restricted LGTV replication. Full article

Staphylococcus aureus is a pathogen known to cause a wide range of infections. To find new targets for identification and to understand host–pathogen interactions, many studies have focused on surface proteins. We performed bacterial-cell surface-shaving, followed by tandem mass tag for quantitative mass spectrometry proteomics, to examine the surfaceome of S. aureus. Two steps were performed, the first step including surface protein-deficient mutants of S. aureus Newman strain lacking important virulence genes (clfA and spa, important for adhesion and immune evasion and srtAsrtB, linking surface-associated virulence factors to the surface) and the second step including isolates of different clinical origin. All strains were compared to the Newman strain. In Step 1, altogether, 7880 peptides were identified, corresponding to 1290 proteins. In Step 2, 4949 peptides were identified, corresponding to 919 proteins and for each strain, approximately 20 proteins showed differential expression compared to the Newman strain. The identified surface proteins were related to host-cell-adherence and immune-system-evasion, biofilm formation, and survival under harsh conditions. The results indicate that surface-shaving of intact S. aureus bacterial strains in combination with quantitative proteomics is a useful tool to distinguish differences in protein abundance of the surfaceome, including the expression of virulence factors. This paper investigates the competition between two charge point operators at the same site for future battery electric long-haul trucks. The charge point operators are located along one of the busiest highways in Sweden. The investigation is carried out using an agent-based model where trucks select charge point operators based on charging prices and the length of any queues, while charge point operators adjust their prices and number of chargers to improve their profitability. The study aims to predict conditions for trucks and charge point operators in a future public fast-charging market. Our findings indicate the potential for a well-functioning future public fast-charging market with small queuing problems, high utilisation, and reasonable prices for public fast charging. Assuming a price for electricity of EUR 0.08/kWh and a minimum profit margin of EUR 0.001/kWh for charge point operators, the findings indicate that the price level outside rush hours will be low, approximately EUR 0.1/kWh. The prices during rush hours will likely be much higher, but it is harder to predict the value due to uncertainties of how charge point operators will act in the future market. Still, from the model result, the price during rush hours is suggested to be just above EUR 0.5/kWh, with an average charging price of around EUR 0.15/kWh. It also seems likely that it is profitable for charge point operators to build enough chargers so that charging queues are short. Full article

Scarce data exist on double maintenance in transplant-eligible high-risk (HR) newly diagnosed multiple myeloma (NDMM) patients. This prospective phase 2 study enrolled 120 transplant-eligible NDMM patients. The treatment consisted of four cycles of ixazomib–lenalidomide–dexamethasone (IRD) induction plus autologous stem cell transplantation followed by IRD consolidation and cytogenetic risk-based maintenance therapy with lenalidomide + ixazomib (IR) for HR patients and lenalidomide (R) alone for NHR patients. The main endpoint of the study was undetectable minimal residual disease (MRD) with sensitivity of <10−5 by flow cytometry at any time, and other endpoints were progression-free survival (PFS) and overall survival (OS). We present the preplanned analysis after the last patient has been two years on maintenance. At any time during protocol treatment, 28% (34/120) had MRD < 10−5 at least once. At two years on maintenance, 66% of the patients in the HR group and 76% in the NHR group were progression-free (p = 0.395) and 36% (43/120) were CR or better, of which 42% (18/43) had undetectable flow MRD <10−5. Altogether 95% of the patients with sustained MRD <10−5, 82% of the patients who turned MRD-positive, and 61% of those with positive MRD had no disease progression at two years on maintenance (p < 0.001). To conclude, prolonged maintenance with all-oral ixazomib plus lenalidomide might improve PFS in HR patients. Full article

This paper investigates the charging needs and charger utilisation of a system of battery electric trucks and their public fast chargers, along the highway between the Swedish cities of Helsingborg and Stockholm, in a possible future scenario in which all trucks are battery electric. The system is investigated via an agent-based model which simulates a typical day with current levels of truck traffic on the road. The traffic flow is based on hourly truck flow data during a day and the annual flow of trucks. The findings indicate major potential for a well-functioning system of public chargers for trucks, with high utilisation, few queuing problems at charging stations, cheap public fast charging and a robust response to queues caused by peaks or increases in the traffic flow. The number of 900 kW chargers along the road needed to achieve this has been estimated at 140, and the utilisation factor for the chargers is predicted to be 30%. Full article

The aim of this study was to evaluate quality and sensory variation in wild boar meat in comparison to pork. Meat quality in wild boar is expected to vary more compared to pork due to different feeding environment, age and gender. In order to be able to promote wild boar meat as a sustainable high-quality product, there is a need to evaluate the variation in meat quality attributes, including technological, compositional and sensory/texture aspects. We evaluated carcass characteristics, pH, colour, lipid profile and sensory aspects of wild boar meat of different age and gender and compared them with pork. Wild boars had lower carcass weight (p = <0.0001) and higher ultimate pH (p = 0.0063) compared to domestic pigs. Intramuscular fat content had a tendency to be higher in wild boar meat (p = 0.1010), as well as the proportion of nutritional valuable n-3 FA (p = 0.0029). The colour of pork was more pink (p = 0.0276) and pale (p = <0.0001) compared to meat from wild boar. Meat from wild boar gilts received the highest sensory scores. Based on these findings, we suggest that meat from younger animals could be sold in different cuts directly while meat from older animals might be more suitable for the production of sausage. This paper investigates the economic consequences of a haulage company replacing its line-haul diesel trucks with battery-electric ones. It also examines how large truck batteries should be, whether the haulage companies should use public fast chargers to complement their own, and whether public fast chargers have the potential to be profitable. The potential extra cost of losing payload capacity is estimated and there is an investigation of whether a charge-point operator should meet the peak demand for charging. The case under analysis is designed to represent a typical line-haul service between terminals in a major logistics system, with the finding that, in this case, a transition to battery-electric trucks seems cost effective for the company. Moreover, it is advisable for the company to use public fast chargers and these will likely become profitable given that the utilisation factor of the investigated public fast chargers may realistically exceed 20%. One possible step for reducing humans’ use of fossil fuel due to transport tasks is to replace diesel trucks with battery electric ones. This paper introduces the energy distribution diagram, which makes it easy to visualise the trucks’ daily energy consumption over their full service life. The energy distribution is used to investigate which driving patterns are suitable for cost-effective battery electric trucks when compared to commercial diesel trucks. It is shown that the battery capacity that results in the lowest cost per kilowatt-hour propulsion energy depends on the driving pattern, and an algorithm for selecting the most cost-effective capacity is presented. In many instances, it was found that battery electric trucks competed favourably with diesel trucks, especially when the trucks had low variations in daily energy consumption. It is beneficial to determine the circumstances under which they may be cheaper, as this will facilitate the transition to battery electric trucks in segments with a reduced overall cost of ownership. The effects of elastic cavity walls on noise generation at transonic speed are investigated for the generic M219 cavity. The flow is simulated with the Spalart–Allmaras (SA) improved delayed detached-eddy simulation (IDDES) turbulence model in combination with a wall function. The structural analysis software uses a modal formulation. The first 50 structural normal mode shapes are included in the simulation, spanning frequencies of 468–2280 Hz. Results are compared with those from a reference simulation with rigid cavity walls. A spectral analysis of pressure fluctuations from a microphone array above the cavity evinces a distinct tone at 816 Hz, which is absent in the reference simulation. Furthermore, the power of the 4th Rossiter mode at 852 Hz is depleted, implying a significant energy transfer from the fluid to the structure. Spectral proper orthogonal decomposition (SPOD) is employed for analyses of cavity wall pressure fluctuations and wall displacements. The SPOD mode energy spectra show results consistent with the spectra of the microphone array with respect to the tone at 816 Hz and the depletion of the energy at the 4th Rossiter mode. Furthermore, the SPOD mode energy spectra show energy spikes at additional frequencies, which coincide with structural eigenfrequencies. Full article

Prostate cancer (PC) bone metastases can be divided into transcriptomic subtypes, by us termed MetA-C. The MetB subtype, constituting about 20% of the cases, is characterized by high cell cycle activity, low androgen receptor (AR) activity, and a limited response to standard androgen deprivation therapy (ADT). Complementary treatments should preferably be introduced early on if the risk of developing metastases of the MetB subtype is predicted to behigh. In this study, we therefore examined if the bone metastatic subtype and patient outcome after ADT could be predicted by immunohistochemical analysis of epithelial and stromal cell markers in primary tumor biopsies obtained at diagnosis (n = 98). In this advanced patient group, primary tumor International Society of Urological Pathology (ISUP) grade was not associated with outcome or metastasis subtype. In contrast, high tumor cell Ki67 labeling (proliferation) in combination with low tumor cell immunoreactivity for PSA, and a low fraction of AR positive stroma cells in the primary tumors were prognostic for poor survival after ADT. Accordingly, the same tissue markers were associated with developing metastases enriched for the aggressive MetB subtype. The development of the contrasting MetA subtype, showing the best response to ADT, could be predicted by the opposite staining pattern. We conclude that outcome after ADT and metastasis subtype can, at least to some extent, be predicted by analysis of primary tumor characteristics, such as tumor cell proliferation and PSA expression, and AR expression in stromal cells. Technological meat quality and sensory attributes of fresh and frozen lamb meat were compared. Samples were collected from two abattoirs (one small-scale, one large-scale) that use different slaughter methods in terms of chilling regime and electrical stimulation. The fresh and frozen meat samples included products from both slaughter systems. Ten twin pairs of ram lambs were used in the study, with one of each twin slaughtered at each abattoir. Fresh meat was analysed after chilling and frozen meat was stored frozen for three months and analysed after thawing. The Musculus longissimus thoracis et lumborum was analysed for colour, cooking loss, sensory attributes, Warner-Bratzler shear force (WBSF) and distribution of water and lipid within each meat sample. Meat samples analysed after frozen storage were darker, less red and more yellow than the fresh meat. Freezing and frozen storage increased fluid loss and WBSF compared with the fresh meat, due to protein denaturation. Frozen storage affected sensory attributes by increasing fatty odour, frying flavour, sour flavour, fatty flavour and liver flavour, and by reducing juicy texture and mushy texture. Full article

Assessment of dietary intake is challenging. Traditional methods suffer from both random and systematic errors; thus objective measures are important complements in monitoring dietary exposure. The study presented here aims to identify serum metabolites associated with reported food intake and to explore whether combinations of metabolites may improve predictive models. Fasting blood samples and a 4-day weighed food diary were collected from healthy Swedish subjects (n = 119) self-defined as having habitual vegan, vegetarian, vegetarian + fish, or omnivore diets. Serum was analyzed for metabolites by 1H-nuclear magnetic resonance spectroscopy. Associations between single and combined metabolites and 39 foods and food groups were explored. Area under the curve (AUC) was calculated for prediction models. In total, 24 foods or food groups associated with serum metabolites using the criteria of rho > 0.2, p < 0.01 and AUC ≥ 0.7 were identified. For the consumption of soybeans, citrus fruits and marmalade, nuts and almonds, green tea, red meat, poultry, total fish and shellfish, dairy, fermented dairy, cheese, eggs, and beer the final models included two or more metabolites. Our results indicate that a combination of metabolites improve the possibilities to use metabolites to identify several foods included in the current diet. Combined metabolite models should be confirmed in dose–response intervention studies. Full article

Effect of N-3 Polyunsaturated Fatty Acids on Lipid Composition in Familial Hypercholesterolemia: A Randomized Crossover Trial

Individuals with familial hypercholesterolemia (FH) have an increased risk of cardiovascular disease. Treatment is mainly low-density lipoprotein cholesterol (LDL-C) reduction. How omega-3 polyunsaturated fatty acids (n-3 PUFAs) supplements affect lipoproteins in FH subjects is unknown. We hypothesized that a high-dose n-3 PUFA supplement would reduce atherogenic lipoproteins and influence the high-density lipoprotein cholesterol (HDL-C) function. We performed a randomized, double-blinded crossover study with 34 genetically verified FH individuals (18–75 years, clinically stable, statin treatment > 12 months). Treatment was 4 g n-3 PUFAs (1840 mg eicosapentaenoic acid and 1520 mg docosahexaenoic acid daily) or four capsules of olive oil for three months in a crossover design with a washout period of three months. The defined outcomes were changes in triglycerides, lipoproteins, lipoprotein subfractions, apolipoproteins, and HDL-C function. After treatment with n-3 PUFAs, total cholesterol, LDL-C, and triglycerides were reduced compared to placebo (p ≤ 0.01 for all). Total HDL-C levels were unchanged, but the subfraction of large HDL-C was higher (p ≤ 0.0001) after n-3 PUFAs than after placebo, and intermediate HDL-C and small HDL-C were reduced after n-3 PUFAs compared to placebo (p = 0.02 and p ≤ 0.001, respectively). No changes were found in apolipoproteins and HDL-C function. N-3 PUFAs supplements reduced atherogenic lipoproteins in FH subjects, leaving HDL-C function unaffected. Full article

Criminal recidivism is a major global concern. There is a well-known association between substance use disorders and offending. Yet, little is known about crime-specific recidivism. The aim of this study was to investigate the relationship between specific substance use and crime-specific recidivism. The study is based on 4207 Swedish prison clients with substance use assessed with Addiction Severity Index interviews between 2001 and 2006. Clients were followed for an average of 2.7 years. Risk factors for criminal recidivism were assessed with the Cox regression analysis. Sixty-eight percent of the clients returned to the criminal justice system. Apart from well-known risk factors such as male gender and young age, amphetamine, injection drug use, prior prosecution for violent and property crime, as well as homelessness and psychiatric problems, were risk factors for criminal recidivism. Sedatives and cannabis were, in this setting, negative risk factors for general recidivism. Age, heroin and injection drug use elevated the risks of recidivism to property and drug crime. Alcohol was associated with violent recidivism. When analysing different categories of crime separately, risk factors differed substantially. This further highlights the need for crime-specific research. Identifying crime-specific risk factors should be an important part of improving rehabilitation into society after imprisonment and hopefully decrease recidivism. Full article
