This article describes the immunomodulation capabilities of mesenchymal stem cells from perirenal adipose tissue. This paper is quite well written, it is clear and the experiences correct although the technologies used for pcr are old. It would be interesting to know the advantages of this tissue compared to other sources of MSC.
A minor revision would be to mention the regulatory capacity of the mixed lymphocyte reaction as a test to demonstrate the immunomodulatory activity of MSCs and to justify why the authors did not use this test (refer to the article by Nasef et al. Int J Lab Hematol. 2009 Feb;31(1):9-19. doi: 10.1111). Another point that needs to be discussed is the ability of MSCs to secrete HLA-G as an immunomodulator and why the authors did not measure HLA-G (refer to the article by Nasef et al,
Transplantation. 2007 Jul 27;84(2):231-7).

The authors perform an interesting study about the isolation, characterization and differentiation of fat stem cells taken from perirenal fat. This study is interesting and above all very clear and well conducted. The undersigned reviewer does not see any major problems in conducting the study and the results highlighted. The only question that this reviewer asks the authors concerns the potential clinical use of this study and these results. Authors can better explain this in the discussion.

We absolutely share the concern of the reviewer due to the clinical use of prASCs because of the limited access to perirenal tissue. We have now added two sentences in the discussion part (page 13), whereas we think that this issue is hard to explain.
