Dear Authors, 

thank you very much for your manuscript, the topic you've tackled is indeed very important, however less often raised in the literature, therefore worth investigating. I hope that my comments below will be helpful. In the entire text there are some typos, capital and small letters mixed etc. Especially some abbreviations need to be explained, especially if they are used for the very first time in the text. In many places source information are missing, please complete it. Several items are understandable for experience readers, but might be new to younger ones, thus must be explained - e.g. HPV 16. 

Line 27 --> what is SEER?

Line 34 --> any cancer, not just anus cancer. Also, sources are missing from here onwards. 

Lines 35 - 38 ---> sources needed; also, this sentence needs grammar corrections

Line 38 --> What is it HPV 16 and 18? 

Figure 1 --> I suggest small rearrangements of the graph components, e.g. the central circle with HPV oncoproteins should be on the top, having lines equally connected it with other 4 circles above - in this way all 4 results can be taken as equally important and possibly paralel. 

Line 222 --> 1-2 sentences of introduction would be welcomed here, in order to briefly explain the concept of tumour vaccines - similarly as it has been done in ACT chapter.

This review is addressed to clinical trials of  immunotherapy in anal cancer  used as  monotherapy  or in combination with other therapies. In general, the MS is interesting and useful both from a scientific and practical point of view.

 My comments and suggestions.

 1.      The abstract is not informative enough. It would be appropriate to briefly list the main immunotherapeutic approaches to the treatment of anal cancer.

 2.      At the end of each paragraph (except for the introduction and conclusions), it would be useful to add a concluding sentence(s) in order to briefly summarize the data presented in the paragraph.

 3.      The figures have titles but no captions. Captions could improve understanding of the figures.
