This review is of interest, updated and well written. 

The only suggestion concerns the title. Since half of the review describes the role of sclerostin in regulating bone mineralization, I would suggest to modify the title including both bone and ectopic mineralization (two sides of the same coin).

This review on sclerostin is a comprehensive, extensive and thorough overview of the up-to-date knowledge on this important molecule. The authors have gathered loads of information on the topic, illustrated by the number of refs, yet made the article clear, comprehensible, and, taking into consideration the complexity of the featured issue, relatively easy to follow. The text is accompanied by figures that, although simple, sufficiently illustrate the information provided in the manuscript. I have read it with pleasure.
