Oxidative stress contributes significantly to chronic disease burden, necessitating identification of accessible dietary antioxidant sources. Pigeon peas (Cajanus cajan L.) contain substantial bioactive compounds, yet most exist in bound forms with limited bioavailability. This study evaluated wild fermentation combined with systematic extraction optimization to enhance antioxidant recovery from pigeon peas. Seeds underwent wild fermentation in brine solution, followed by extraction under varying conditions (seven solvent systems, three temperatures, and three-time durations). Multiple complementary assays assessed antioxidant capacity (total phenolic content, DPPH radical scavenging, ferric reducing power, and ABTS activity). Fermentation substantially improved antioxidant properties across all parameters, with particularly pronounced effects on radical scavenging activities. Extraction optimization identified 70% methanol at 40 °C for 24 h as optimal, demonstrating marked improvements over conventional protocols. Strong intercorrelations among assays confirmed coordinated enhancement of multiple antioxidant mechanisms rather than isolated changes. The findings demonstrate that both biotechnological processing and analytical methodology critically influence antioxidant characterization in pigeon peas. This integrated approach offers practical guidance for developing antioxidant-rich functional foods, particularly relevant for resource-limited settings where pigeon peas serve as dietary staples. The study establishes foundation for translating fermentation technology into nutritional interventions, though further research addressing bioavailability, microbiological characterization, and bioactive compound identification remains essential. Isoflavones are the main phenolic compounds of soybean that affect its biological activity. The quantity of these valuable compounds extracted from plant material can significantly vary, influenced by the chosen extraction method and the specific extractants employed. Moreover, in cosmetics and pharmacy, the application of non-toxic, eco-friendly solvents is very important. This study aimed to develop the best mixture of extractants to maximize the recovery of individual isoflavones from soybean seeds by optimization of the proportion of three components: ethanol, water, and propanediol. The design of experiments (DOE) method was strategically employed. The extracts were obtained through accelerated solvent extraction and meticulously analyzed for isoflavone content using advanced electrospray ionization–time of flight–mass spectrometry (ESI-TOF-MS) profiling. The predominant isoflavones were daidzin, genistin, malonylgenistin, malonyldaidzin, and malonylglycitin. Our experiment demonstrated that employing three extractants in a balanced 1:1:1 v/v/v ratio resulted in the highest isolation of isoflavones compared to all other mixtures tested. Nevertheless, a detailed exploration of approximate values and utility profiles revealed a more effective composition for extraction efficiency. This optimal mixture features 32.8% ethanol, 39.2% water, and 27.8% propanediol, maximizing the yield of isoflavones from soybean seeds. The innovative use of mixture design and triangular response surfaces has proven to be a powerful approach for developing this superior three-component extraction mixture. This innovative approach not only enhances extraction efficiency but also paves the way for improved processing methods in the industry. Full article

Phenolic compounds present in plants and foods are receiving increasing attention for their bioactive and sensory properties, accompanied by consumers’ interest in products with health benefits derived from natural rather than artificial sources. This, together with the sustainable development goals for the 21st century, has driven the development of green extraction techniques that allow obtaining these compounds with the safety and quality required to be applied in the food, cosmetic and pharmaceutical industries. Green extraction of natural products involves practices aiming at reducing the environmental impact of the preparation processes, based on using natural or less-polluting solvents, lower energetic requirements and shorter extraction times, while providing greater efficiency in the recovery of target compounds. In this article, the principles of sustainable extraction techniques and the advances produced in recent years regarding green isolation of polyphenols from plants, food and food waste are reviewed. Different solvents water, ethanol and ethanol/water (6:4 v/v), were compared in the extraction of pomegranate peels and seeds (PPS) in terms of recovery yields, antioxidant properties, and antimicrobial action against typical spoilage bacterial and fungal species. The best performing extract (ethanol/water (6:4 v/v) was shown to contain mostly ellagic acid and punicalagin as phenolic compounds (5% overall) and hydrolysable tannins (16% as ellagic acid equivalents) and was able to inhibit the growth of the acidophilic Alicyclobacillus acidoterrestris at a concentration as low as 1%. The preservation of the organoleptic profile of A. acidoterrestris-inoculated apple juice with extract at 1% over 20 days was also observed thanks to the complete inhibition of bacterial growth, while the extract at 0.1% warranted a significant (40%) inhibition of the enzymatic browning of apple smoothies over the first 30 min. When incorporated in whey proteins’ isolate (WPI) at 5% w/w, the hydroalcoholic extract conferred well appreciable antioxidant properties to the resulting coating-forming hydrogel, comparable to those expected for the pure extract considering the amount present. The WPI coatings loaded with the hydroalcoholic extract at 5% were able to delay the browning of cut fruit by ca. 33% against a 22% inhibition observed with the sole WPI. In addition, the functionalized coating showed an inhibition of lipid peroxidation of Gouda cheese 2-fold higher with respect to that observed with WPI alone. These results open good perspectives toward sustainable food preservation strategies, highlighting the potential of PPS extract for the implementation of WPI-based active packaging. Full article

A biorefinery process was developed to isolate phycobiliproteins, sulfated polysaccharides, and phenolic compounds from Palmaria palmata. The extraction process was carried out in three stages using ultrasound-assisted extraction (UAE) and pressurized liquid extraction (PLE) integrated with different natural deep eutectic solvents (NaDESs). In general, PLE provided higher phycobiliprotein contents than UAE in the first step of the process. In fact, the hydrolysis product of the PLE-NaDES extracts achieved a higher antioxidant capacity than that of the UAE-NaDES extracts. Particularly, glycerol:glucose (2:1) with 50% water in combination with PLE was the most suitable NaDES to recover the highest phycobiliprotein, protein, and sulfated polysaccharide contents from Palmaria palmata in the first and second steps of the biorefinery process. Finally, a PLE-NaDES using choline chloride:glycerol (1:2) with 60% water as the NaDES was employed for the recovery of antioxidant and neuroprotective phenolic compounds from the residue of the second step, obtaining a higher total phenolic content than employing PLE with ethanol/water (70:30, v/v) as the extraction solvent. Moreover, a forced stability study revealed that the NaDESs provided a protective effect compared to the water extracts against the degradation of phycobiliproteins, preserving their color over time. This study contributes to the recovery of high-value components from an undervalued biomarine source through a sustainable biorefinery process. Full article

Parkinson’s disease (PD) is a neurodegenerative disorder characterized by the loss of dopaminergic neurons responsible for unintended or uncontrollable movements. Mutations in the leucine-rich repeat kinase 2 locus contribute to genetic forms of PD. The fruit fly Drosophila melanogaster carrying this mutation (LRRK2-Dm) is an in vivo model of PD that develops motor impairment and stands for an eligible non-mammalian paradigm to test novel therapeutic approaches. Dehydrozingerone (DHZ) is a natural phenolic compound isolated from ginger and presents anti-inflammatory, antioxidant and neuroprotective properties, making it a potential therapeutic target for PD. We administered DHZ and its C2-symmetric dimer (DHZ-DIM) at 0.5 and 1 mM for 14 and 21 days in the LRRK2-Dm, with the aim of assessing changes in rescuing motor behavior, brain dopaminergic neurons, mitochondria and synapses (T-bars). The shorter treatment with both molecules revealed efficacy at the higher dose, improving climbing behavior with a prevention of dopaminergic neuronal demise. After 21 days, a recovery of the motor disability, dopaminergic neuron loss, mitochondrial damage and T-bars failure was observed with the DHZ-DIM. Our data indicate that the DHZ-DIM exerts a more potent neuroprotective effect with respect to the monomer in LRRK2-Dm, prompting further investigation of these compounds in rodent models of PD. Grapes, especially those of the red varieties, have a high content of polyphenolic compounds. After the removal of the juice during the winemaking process, the grape marc (peels and seeds) remains as waste, making it a promising source for the isolation of polyphenols. The separation, recovery, and enrichment of samples of phenolic compounds offers the possibility of their subsequent utilization in the food, pharmaceutical, and cosmetics industries. In this paper, results are presented on both laboratory and pilot scales, including the effect of basic extraction parameters such as the solvent (water, ethanol, acetone, polyethylene glycol, ethyl acetate, and their respective 50% aqueous solutions), the solid/liquid ratio, the extraction time, and the temperature. The enrichment of the extracts in phenolic compounds was performed with the help of a series of membrane processes and rotary evaporation. The experiments showed the presence of almost all known compounds reported in the literature with pro-anthocyanidins (dimers-trimers) and flavan-3-ols together with various metabolites accompanied by a significant reduction in the values of total organic load. High-performance counter-current chromatography (HPCCC) was used as a tool for the isolation and fractionation of phenolic compounds (PCs) in extracts from wine lees (WL) and grape pomace (GP). The biphasic solvent systems applied for HPCCC separation were n-butanol:methyl tert-butyl ether:acetonitrile:water (3:1:1:5) with 0.1% trifluoroacetic acid (TFA) and n-hexane:ethyl acetate:methanol:water (1:5:1:5). After refining the ethanol:water extracts of GP and WL by-products by ethyl acetate extraction, the latter system yielded an enriched fraction of the minor family of flavonols. Recoveries of 112.9 and 105.9 mg of purified flavonols (myricetin, quercetin, isorhamnetin, and kaempferol) in GP and WL, respectively, from 500 mg of ethyl acetate extract (equivalent to 10 g of by-product) were obtained. The HPCCC fractionation and concentration capabilities were also exploited for the characterization and tentative identification of constitutive PCs by ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS). In addition to the isolation of the enriched flavonol fraction, a total of 57 PCs in both matrixes were identified, 12 of which were reported for the first time in WL and/or GP. The application of HPCCC to GP and WL extracts may be a powerful approach to isolate large amounts of minor PCs. The composition of the isolated fraction demonstrated quantitative differences in the individual compound composition of GP and WL, supporting the potential exploitation of these matrixes as sources of specific flavonols for technological applications. The plants in the Sideritis genus are postulated to exhibit several important medicinal properties due to their unique chemical composition. To isolate the targeted phytochemical compounds, the selection of a suitable extraction method is of primary importance. In this work, a comparative study on the phytochemical profiles of various Sideritis raeseri and Sideritis scardica extracts has been carried out. An untargeted metabolomics approach based on ultra-high performance liquid chromatography coupled with high-resolution mass spectrometry was applied to investigate the metabolic differences between extracts obtained by conventional extraction and extractions assisted by microwaves, ultrasounds and high pressure. Additionally, the influence of extraction solvents on HPLC antioxidant profiles obtained following the derivatization of analytes with ABTS reagent was evaluated. A total of 102 metabolites have been putatively identified. The major secondary metabolites groups were classified as flavonoids, terpenoids, phenylethanoid glycosides and phenolic acids. The main antioxidants in the extracts were isoscutellarein and hypolaetin derivatives as well as verbascoside and chlorogenic acid. The results showed that 70% ethanol was the most effective extractant for different classes of phytochemicals including antioxidants. In addition, extraction supported with microwaves, ultrasounds or high pressure improved the overall recovery of metabolites by about 3 times compared to the conventional extraction method. Aniseeds (Pimpinella anisum) have gained increasing attention for their nutritional and health benefits. Aniseed extracts are known to contain a range of compounds, including flavonoids, terpenes, and essential oils. These compounds have antimicrobial properties, meaning they can help inhibit the growth of nasty bacteria and other microbes. The purpose of this study was to determine if aniseed extracts have potential antioxidant, phytochemical, and antimicrobial properties against multidrug-resistant (MDR) bacteria. A disc diffusion test was conducted in vitro to test the aniseed methanolic extract’s antibacterial activity. The MIC, MBC, and inhibition zone diameters measure the minimum inhibitory concentration, minimum bactericidal concentration, and size of the zone developed when the extract is placed on a bacterial culture, respectively. HPLC and GC/MS are analytical techniques used for identifying the phenolics and chemical constituents in the extract. DPPH, ABTS, and iron-reducing power assays were performed to evaluate the total antioxidant capacity of the extract. Using HPLC, oxygenated monoterpenes represented the majority of the aniseed content, mainly estragole, cis-anethole, and trans-anethole at 4422.39, 3150.11, and 2312.11 (g/g), respectively. All of the examined bacteria are very sensitive to aniseed’s antibacterial effects. It is thought that aniseed’s antibacterial activity could be attributed to the presence of phenolic compounds which include catechins, methyl gallates, caffeic acid, and syringic acids. According to the GC analysis, several flavonoids were detected, including catechin, isochiapin, and trans-ferulic acid, as well as quercitin rhamnose, kaempferol-O-rutinoside, gibberellic acid, and hexadecadienoic acid. Upon quantification of the most abundant estragole, we found that estragole recovery was sufficient for proving its antimicrobial activity against MDR bacteria. Utilizing three methods, the extract demonstrated strong antioxidant activity. Aniseed extract clearly inhibited MDR bacterial isolates, indicating its potential use as an anti-virulence strategy. It is assumed that polyphenolic acids and flavonoids are responsible for this activity. Trans-anethole and estragole were aniseed chemotypes. Aniseed extracts showed higher antioxidant activity than vitamin C. Future investigations into the compatibility and synergism of aniseed phenolic compounds with commercial antibacterial treatments may also show them to be promising options. Full article

Increasing intensity and frequency of droughts are leading to forest dieback, growth decline and tree mortality worldwide. Reducing tree-to-tree competition for water resources is a primary goal for adaptive climate silviculture strategies, particularly in reforested areas with high planting density. Yet, we need better insights into the role of stand type (i.e., natural forests versus plantations) on the resilience of pine forests to droughts across varying time scales. In this study, we combined dendrochronological data and stable C (δ13C) and H (δ2H) isotopes measured in tree-ring wood as well as in specific wood chromatographically isolated compounds to investigate contrasting responses to drought of natural versus planted stands of two representative pine species, i.e., Pinus pinaster and Pinus nigra in southeastern Europe. Natural stands exhibited about two-fold increase in tree-ring growth in average (basal area at 20 years-BAI20) as compared to planted stands. A response function analysis showed contrasting seasonal growth patterns for both species, which were related to monthly mean temperature and precipitation. Both stand type and species variables influenced growth resilience indices. Both pine species revealed contrasting resilience patterns among forest types; whereas planted stands seemed to be less sensitive to yearly droughts as determined by a higher recovery index (CRc) for P. pinaster, the contrary was found in the case of P. nigra. On the other hand, while resistance CRT and resilience CRS indices were higher for planted than natural forests in the case of P. pinaster, little differences were found for P. nigra. Beyond comparisons, carbon stable isotopes shed lights on the role of forest types in dry sites, being δ13C consistently lower in natural than in planted forests for both pine species (p < 0.05). We concluded that planted forest assimilated more carbon as per unit of water used than natural stands in response to droughts. Both δ13C and δ2H isotopic signals were positively correlated for both species for planted forests. However, a lack of correlation was evidenced for natural stands. Consistent with δ13C observations, δ2H concentrations in woody phenolic compounds (guaiacol and oleic acid) revealed contrasting patterns among forest types. This puts forward that δ2H concentrations in woody phenolic compounds (rather than in woody tree ring) accounts for other confounding factors in tree ring formation that can be associated with forest type. Our results highlight the value of stable isotope approaches versus conventional dendrochronological tools in drought studies and call for the consideration of forest type as an endogenous aspect defining the vulnerability of pine forests to climate. The chestnut tree (Castanea sativa, Mill.) is a widespread plant in Europe whose fruits and wood has a relevant economic impact. Chestnut wood (CW) is rich in high-value compounds that exhibit various biological activities, such as antioxidant as well as anticarcinogenic and antimicrobial properties. These metabolites can be mainly divided into monomeric polyphenols and tannins. In this piece of work, we investigated a sustainable protocol to isolate enriched fractions of the above-mentioned compounds from CW residues. Specifically, a sequential extraction protocol, using subcritical water, was used as a pre-fractionation step, recovering approximately 88% of tannins and 40% of monomeric polyphenols in the first and second steps, respectively. The optimized protocol was also tested at pre-industrial levels, treating up to 13.5 kg CW and 160 L of solution with encouraging results. Ultra- and nanofiltrations were used to further enrich the recovered fractions, achieving more than 98% of the tannin content in the heavy fraction, whilst the removed permeate achieved up to 752.71 mg GAE/gext after the concentration (75.3%). Samples were characterized by means of total phenolic content (TPC), antioxidant activity (DPPH· and ABTS·), and tannin composition (hydrolysable and condensed). In addition, LC-MS-DAD was used for semiqualitative purposes to detect vescalagin/castalagin and vescalin/castalin, as well as gallic acid and ellagic acid. The developed valorization protocol allows the efficient fractionation and recovery of the major polyphenolic components of CW with a sustainable approach that also evaluates pre-industrial scaling-up. The aim of this study was to investigate the influence of cryogrinding pretreatment on the recovery of essential oil, phenolics and pigments from myrtle leaves. The duration of cryogrinding (3, 6 and 9 min) in combination with the duration of hydrodistillation (30, 60 and 90 min) for the isolation of essential oils and the duration of hydroethanolic extraction (5, 10 and 15 min) for the isolation of phenols and pigments were studied as independent factors in a full factorial design. The major volatile components detected in myrtle leaf essential oil were myrtenyl acetate, 1,8-cineole, α-pinene and linalool. The most abundant phenols detected were myricetin derivatives (myricetin 3-O-galactoside and myricetin 3-O-rhamnoside), galloylquinic acid, myricetin and digalloylquinic acid, while the major pigments were chlorophyll b, pheophytin a and lutein. A 3 min cryogrinding pretreatment significantly increased the yield and concentrations of essential oil volatile compounds and reduced the distillation time to 30 min. A 9 min cryogrinding pretreatment and 15 min extraction resulted in at least 40% higher concentrations of phenolic compounds and pigments in the extracts when compared to the untreated control. According to the results obtained, cryogrinding can significantly increase the yield of myrtle EO and extracts and also modulate their composition. Full article

This study investigated the efficacy of roasting pre-treatment by air frying to enhance the extraction and recovery of the predominant sinapic acid derivatives (SADs) from roasted canola meal and the antioxidant potential of the methanolic extracts. Canola meal was obtained by air frying canola seed at 160, 170, 180 or 190 °C for 5, 10, 15 or 20 min. Oil was extracted using the Soxhlet method, and the de-oiled meal fraction was air-dried. Phenolic compounds were isolated using ultrasound-assisted extraction with 70% (v/v) methanol and then quantified by high-performance liquid chromatography-diode array detection. The antioxidant potential of the defatted meal methanolic extracts was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and metal ion-chelating activity (MIC) assays. The highest total phenolic content of 3.15 mg gallic acid equivalent/g dry weight was recorded in the defatted meal extract from seeds pre-treated with air frying at 190 °C for 15 min. Sinapine, sinapic acid and an unknown compound at a retention time (RT) of 26.6 min were the major sinapates identified in the defatted meal with the highest concentrations of 7572 ± 479.2 µg/g DW, 727 ± 43.45 µg/g DW and 1763 ± 73.5 µg/g DW, respectively, obtained at 160 °C for 5 min. Canolol (151.35 ± 7.65 µg/g DW) was detected after air frying at a temperature of 170 °C for 20 min. The FRAP and MIC correlated positively (r = 0.85) and generally decreased with increased air frying temperature-time conditions. The highest FRAP and MIC values of 0.53 mM and 80% were obtained at 160 °C for 5 and 20 min, respectively. The outcome of this study will contribute new knowledge that could improve the value addition and by-product utilization of canola seeds. Full article

Currently, the consumption of natural extracts from medicinal plants and fruits present in nature, and the discovery of the many benefits and positive impacts on the human body connected to their intake have favored and promoted research in the field of extraction. For this purpose, in recent years, the extracts of plant parts (leaves, branches, seeds, stem, roots, fruits, etc.) of the genus of Asteriscus, Atriplex, Haloxylon, Ruta, Ficus, Olea, and Zizyphus have been investigated for their several biological properties and therapeutic activities including those as an antioxidant, antitumoral, antiproliferative, hepatoprotective, and antimicrobial. The shortcoming of using conventional methods, such as maceration, Soxhlet solvent extraction, and pressurized hot water, has promoted the development of novel, efficient, economical, and safe extraction techniques to recover the bioactive compounds without losing their quality and properties. Innovative methodologies, such as supercritical fluid technology and pulsed electric field-assisted extraction, have been proposed as sustainable methodologies to isolate nutraceuticals and pharmaceuticals from natural matrices, such as herbs, spices, and aromatic and medicinal plants. This study aimed to obtain extracts from fruits, leaves, and roots of Ziziphus lotus by Supercritical Fluid Extraction (SFE, P = 25 MPa, T = 40 °C, Time = 60 min, Flow = 10 mL/min, Solvent = 90 CO2:10 Ethanol) and Pulsed Electric Field (PEF, E = 3 kV/cm, Ws = 100 kJ/kg, Solvent = water, followed by 60 min of agitation) assisted extraction and characterize the extracts in terms of total antioxidant capacity (Oxygen Radical Absorbance Capacity (ORAC) and Total Equivalent Antioxidant Capacity (TEAC)), total phenolics, chlorophyll, and carotenoids content. PEF extracts presented an interesting content of total phenolic compounds (TPC): Fruits: 14.8 ± 0.1; Leaves: 31.1 ± 1.8; Roots: 28.8 ± 1.7 mg GAE/g DW. On the other hand, the concentration of TPC in SFE extracts was: Fruits: 5.91 ± 0.2; Leaves: 1.184 ± 0.2; Roots: 5.72 ± 0.8 mg GAE/g DW. Total chlorophyll content (TCC) was: Fruits: 366 ± 1; Leaves: 471 ± 12; Roots: 192 ± 16 mg/g DW in the case of PEF extracts and 5.0 ± 0.3; 97 ± 1; 8.5 ± 0.1 mg/g DW, respectively, in the case of SFE. Finally, the content of carotenoids was 128.0 ± 0.5; 62 ± 1; 169 ± 1 mg/g DW for PEF-extracted fruits, leaves, and roots, respectively, and 5.92 ± 0.04; 19.76 ± 0.02; 4.97 ± 0.04 mg/g DW for SFE extracted fruits, leaves, and roots, respectively. PEF-assisted extraction allowed the recovery of more phenolics, chlorophyll, and carotenoid compounds than SFE. The TEAC was higher for PEF extracts but ORAC was higher for SFE extracts. This could be related to the different polarities of the solvent used for extraction in each technique. Green extraction methodologies represent an efficient and safe extraction technique to recover bioactive compounds without losing their quality and properties. Full article
