This paper presents an analysis of helminth diversity in green frogs of the Pelophylax esculentus complex from twelve provinces of the Middle Volga region (European Russia). The study relies on the occurrence dataset recently published in GBIF in the form of a Darwin Core archive. The database contains current information on helminth occurrences in these anurans, comprising records from our long-term helminthological survey conducted in 1997–2025. Our database includes 13,634 helminth occurrence records for three Pelophylax species residing in the Middle Volga region. Each helminth occurrence record is linked to georeferenced data. A total of 43 parasite species are documented in the dataset, including 29 species of trematodes, one cestode, 11 nematodes, and two acanthocephalans. The greatest helminth diversity was recorded in Pelophylax ridibundus (42 species), which is the most widespread and abundant amphibian species in European Russia. The helminth fauna is less diverse in Pelophylax lessonae (32 species) and their hybrid, Pelophylax esculentus (25). Most helminth species found in green frogs within the studied area belong to the Palearctic faunal complex (29 species). Five helminth species each have European and Holarctic distributions, while four species are cosmopolitan. Of the 43 species of helminths found, three (Alaria alata, Spirometra erinacei, Ascarops strongylina) are of medical and veterinary importance as causative agents of parasitic zoonoses, posing a threat to domestic animals and humans. Gametogenesis is a fundamental biological process that ensures both genetic recombination and the continuity of successive generations. Interspecific hybrids can reproduce through modified mechanisms, such as hybridogenesis, by transmitting clonal, unrecombined genomes of only one of the parental species via their gametes. Pelophylax grafi (RP) is a natural hybrid frog composed of mixed genomes (subgenomes) of two related species, Pelophylax perezi (P) and Pelophylax ridibundus (R), and coexists in populations with P. perezi. This study tested the involvement of programmed genome elimination in gamete production of P. grafi, providing new insight into reproductive mechanisms of hybrid vertebrates. Using comparative genomic hybridization (CGH) and fluorescent in situ hybridization (FISH), we examined the genomic constitution of germline cells in tadpoles and adult male and female P. grafi. Controlled crosses between P. perezi and P. grafi produced F1 hybrid tadpoles, whose genotypes confirmed that P. grafi parents transmitted the R subgenome through their gametes. In the early germline cells (gonocytes) of these tadpoles, P chromosomes were selectively eliminated via micronuclei formation during interphase. The occasional presence of the R genome and mixed R/P genome micronuclei suggests variability and imperfect fidelity in the elimination process. In adult hybrids, the majority of diplotene oocytes, spermatogonial stem cells (SSC) and spermatocytes carried R subgenomes. We demonstrated that programmed genome rearrangement in Pelophylax hybrids is an evolutionarily conserved mechanism underlying this unique reproductive strategy. Full article

Extensive research on amphibians has focused on areas such as morphological and molecular taxonomy, ecology, embryology, and molecular phylogeny. However, the structure and biotechnological potential of egg jelly—which plays a protective and nutritive role for embryos—have remained largely unexplored. This study presents, for the first time, a detailed physicochemical analysis of the egg jelly of Pelophylax ridibundus, an amphibian species, using Fourier Transform Infrared Spectroscopy, Thermogravimetric Analyzer, X-ray Diffraction, and elemental analysis. The carbohydrate content was determined via High-Performance Liquid Chromatography analysis, and the protein content was identified using Liquid Chromatography-Tandem Mass Spectrometry analysis. Additionally, it was revealed that this jelly exhibits a significant cytotoxic effect on melanoma cells (viability < 30%) while showing no cytotoxicity on healthy dermal fibroblast cells (viability > 70%). Consequently, this non-toxic, biologically derived, and cultivable material is proposed as a promising candidate for cancer applications, paving the way for further research in the field. An analysis of the age structure of three water frog species (P. lessonae, P. ridibundus, P. esculentus) in the Middle Volga region of Russia was conducted. The skeletochronological method determined their maximum lifespans to be 7, 6, and 4 years, respectively. This study revealed specific characteristics of the age structure of Pelophylax frogs in relation to their taxonomic composition when cohabiting (population system type) and the degree of habitat transformation. Differences in body size and lifespan were observed among cryptic forms exhibiting mitochondrial DNA replacement and the presence of various types of heterozygous and homozygous alleles. Water frogs of the genus Pelophylax have a complex taxonomic structure, including hybrid species and morphologically cryptic forms, as well as massive mitochondrial DNA introgression. The aim of our paper was to analyze the age structure of three species of water frogs (P. lessonae, P. ridibundus, and P. esculentus) in the Middle Volga River region (European Russia). Based on the skeletochronological method, we determined their maximum lifespans of seven, six, and four years, respectively. The onset of sexual maturity in the marsh frogs occurred after two and four winterings in males and females (respectively), in the pond frog—one to two years, and in the edible frog—one to three years. This study revealed specific characteristics of the age structure of Pelophylax frogs in relation to their taxonomic composition when cohabiting (population system type) and the degree of habitat transformation. Analyses of the hematological statuses of animals inhabiting areas of anthropogenic pollution may provide valuable insights into the extent of disturbance of their living conditions and the mechanisms of adaptation to various environmental stressors. The current work compares the erythrogram and erythrocyte-metric parameters of marsh frogs (Pelophylax ridibundus) inhabiting the polluted sedimentation lake of Brikel TPP in southern Bulgaria to those in frogs inhabiting a relatively clean habitat (reference population). The study includes a total of 120 individuals (30 females and 30 males from each site). For all of them, total erythrocyte count, hemoglobin concentration, hematocrit, MCV, MCH, and MCHC were evaluated via standard laboratory techniques. All erythrocyte metrics were determined microscopically in two blood smears from each animal. Our study reveals alterations in the erythrogram parameters and the erythrocyte sizes in marsh frogs living in conditions of chronic pollution with industrial wastewater compared to the animals from the reference site. The mean values for all erythrocyte morphology parameters are significantly lower in both female and male individuals inhabiting the polluted area compared to those originating from the reference one. Conversely, three erythrogram parameters—erythrocyte count, hemoglobin, and hematocrit—appeared significantly higher in females and males from the polluted site. The observed changes in the erythrogram parameters and erythrocyte sizes result from the deteriorated water quality of the sedimentation lake. Full article

This study aimed to describe the morphological characteristics and age structure of a P. ridibundus population sampled from a high altitude. For this purpose, a total of 54 adult frogs (33 males and 21 females) were collected from Şavşat district, Artvin province, Türkiye. The samples were aged based on skeletochronology method. Furthermore, von Bertalanffy growth curve models were constructed using SVL and weight data. To assess sexual size and shape dimorphism, the measurements of 23 different morphological characters were analyzed using univariate and multivariate statistical techniques. The mean SVL and weight were found to be 78.05 mm and 60.87 g for males and 93.95 mm and 109.61 g for females. The mean age was 3.67 years and 4.05 years for males and females, respectively. The maximum age was found to be 6 years for both sexes. Growth curve models revealed a significant relationship between age, SVL, and weight. A clear female-biased sexual size dimorphism was found in all morphological characters. However, SVL-adjusted values indicated that males had larger head and limb structures resulting from evolutional and sexual pressures associated with reproduction and male–male competition. The shape dimorphism was found only in forelimbs, and it was male-biased and was related to clasping in amplexus. Full article

Studying the distribution of morphologically cryptic animal species is always a very difficult task. Because most marsh frog species (the Pelophylax ridibundus complex) are cryptic, we used molecular markers to identify them. Three marsh frog species (P. ridibundus, P. kurtmuelleri and P. cf. bedriagae) inhabit the northern part of Western Palearctic. We created a database of localities and built models of their modern distribution. These models showed that the most suitable habitats are on the north of the Mediterranean region for P. cf. bedriagae, temperate Europe for P. ridibundus, and the Balkan coastal areas for P. kurtmuelleri. The projection of the modern ecological niches under the late-Quaternary climatic conditions showed that the range of P. kurtmuelleri remained largely unchanged during the period, whereas the ranges of P. cf. bedriagae and especially P. ridibundus changed greatly over time. During the Last Glacial Maximum, the presumed range of P. cf. bedriagae covered a relatively large area in the north of the Mediterranean region and the south of European Russia. Glacial refugia of P. ridibundus were apparently located in the northern Balkans, the northern coast of the Black and Azov seas, and possibly in Western Europe. The northward long-distance post-glacial dispersal of P. ridibundus occurred from refugia in the northeastern Balkans and the Black-Azov seas region. Since the Late Pleistocene, suitable habitats for P. cf. bedriagae in southern Russia began to decline, but local habitats for P. ridibundus become more suitable. Therefore, a mosaic of populations consisting of these both species and their hybrids has now been found here. Full article

Agriculture can have different effects on wildlife depending on land-use type and extensive/intensive practices. The aim of this study was to establish the significance of rice fields in Central Bulgaria as anuran habitats. We used Audiomoth acoustic loggers to record calling activity at three sites near the city of Plovdiv—one shallow pond and two rice fields—from June to September in 2022 and 2023. We registered the three most widespread species in the region—Bufotes viridis, Hyla orientalis and Pelophylax ridibundus—and created pattern-matching models for them using the free web interface Arbimon, which enabled us to perform presence/absence counts and abundance estimates. At the beginning and the end of the rice vegetation period, water samples were collected and analysed for 62 substances. Substance concentrations were compared between ponds and with LC50 data from the available literature. We registered 19 and 21 substances in 2022 and 2023, respectively, with concentrations within the accepted limits, and although some metals were near risk levels, this was not reflected in the presence counts or the abundance estimates. The results indicated that frog activity was not related to any of the registered substances, but that it was positively correlated with daily rainfall and was higher in the rice fields. Full article

Informed conservation management requires exact knowledge on the species diversity of the taxon group in focus within a geographic area, specifically on the occurrence of endemics. In Turkey, there are three water frog taxa of the genus Pelophylax; one is the widespread P. bedriagae, the other two are geographically restricted to either Thrace (P. ridibundus) or to the Anatolian Lake district (P. caralitanus). The species status of the Beyşehir frog P. caralitanus has been questioned since its proposal in 2001. We recorded and analyzed advertisement and release vocalizations at representative populations of Pelophylax taxa to assess the degree of inter-taxon differentiation and the potential for premating isolation. We found that P. bedriagae and P. caralitanus have much more similar vocalizations than both have compared to P. ridibundus. A functional bioacoustic premating barrier between bedriagae and caralitanus in syntopy does not exist according to our study. The low degree of bioacoustic differentiation mirrors earlier genetic, karyological, and morphological findings. We conclude that the Beyşehir frogs do not deserve species status and that P. caralitanus should be considered a junior synonym of P. bedriagae. Therefore, the number of endemic anuran species in Turkey decreases to one. Full article

In this paper, we assess the environmental impact of urbanization in three freshwater biotopes, using the levels of fluctuating asymmetry (FA) in 10 meristic morphological traits in the Marsh Frog (Pelophylax ridibundus (Pallas, 1771)). Two of the studied biotopes are located in the boundaries of the city of Plovdiv (one in the central part, the other in a suburban residential area), and the third is located in the vicinity of the village of Orizare. Our working hypothesis is based on the assumption that urban and suburban sites are more severely affected by human activities than rural sites. However, according to our results, the population of P. ridibundus inhabiting Maritsa River in the central part of Plovdiv City, and that in the suburban zone, have found relatively good living conditions. Contrary to our expectations, the worst environmental conditions were observed in the rural zone, where anthropogenic stress related to intensive pastoral animal husbandry and crop farming was present. The absence of adult individuals in the rural site is also an indicator of unfavorable living conditions. Full article

In order to study the diversity, phylogenetic relationships and distribution of marsh frogs of the Pelophylax ridibundus complex in Kazakhstan and northwest China, we conducted phylogeographic analyses of 125 samples from 53 localities using the mtDNA ND2 and COI genes and the SAI nuclear gene sequences. Phylogenetic inference of mtDNA revealed three main lineages—sister lineages Balkhash and Syrdarya (as the Central Asian P. sp. novum), and the Anatolian P. cf. bedriagae, while from nDNA data, we additionally detected the western form, P. ridibundus. According to mtDNA data, the mean genetic distances between P. sp. novum and two other forms of marsh frogs was more than 5%. Genetic homogeneity within populations of the Syrdarya lineage and P. cf. bedriagae is characterized by low nucleotide diversity and high haplotype diversity. Demographic analyses of the lineages showed past population expansions of the Balkhash and the Syrdarya forms. Divergence from the most recent ancestor had occurred in the Early Pleistocene period (2.46 Mya) for the Balkhash and the Syrdarya lineages, and 1.27 Mya for the P. cf. bedriagae. Our findings provide a first investigation of the lineage diversification and population dynamics of the Central Asian marsh frogs and will be useful for further taxonomic implications and conservational actions. The toxic action of the Actara 25 WG insecticide (it contains 25% thiamethoxam as an active substance) in non-lethal doses was studied in two species of aquatic organisms—the Prussian carp (Carassius gibelio) and the marsh frog (Pelophylax ridibundus)—at two thermal levels, 6–8 °C (low temperature) and 18–20 °C (room temperature), respectively. In the Prussian carp, we recorded decreases in oxygen consumption and stimulation of the respiratory rhythm, changes that were more pronounced in the case of intoxicated fish and when the species were kept at room temperature. The histopathology of the lung in the frog illustrated the thickening of the conjunctival septum, an increase in the number of mucous cells, and an increase in the ratio between the diameter of the nucleus and the diameter of the pneumocyte. All of these changes were more pronounced in the animals kept at higher temperature. Our study looks at the extent to which temperature changes can influence the ability of poikilothermic organisms to withstand the presence of toxic substances in the environment as a result of the impact of the use of insecticides in agriculture. The two tested organisms are a common presence for the study area, which was affected in the last decade by climate change. Full article

Anurans have been introduced in many parts of the world and have often become invasive over large geographic areas. Although predation is involved in the declines of invaded amphibian populations, there is a lack of quantitative assessments evaluating the potential risk posed to native species. This is particularly true for Pelophylax water frogs, which have invaded large parts of western Europe, but no studies to date have examined their predation on other amphibians in their invaded range. Predation of native amphibians by marsh frogs (Pelophylax ridibundus) was assessed by stomach flushing once a month over four months in 21 ponds in southern France. Nine percent of stomachs contained amphibians. Seasonality was a major determinant of amphibian consumption. This effect was mediated by body size, with the largest invaders ingesting bigger natives, such as tree frogs. These results show that invasive marsh frogs represent a threat through their ability to forage on natives, particularly at the adult stage. The results also indicate that large numbers of native amphibians are predated. More broadly, the fact that predation was site- and time-specific highlights the need for repeated samplings across habitats and key periods for a clear understanding of the impact of invaders. Hybridogenesis in an interspecific hybrid frog is a coupling mechanism in the gametogenic cell line that eliminates the genome of one parental species with endoduplication of the remaining genome of the other parental species. It has been intensively investigated in the edible frog Pelophylax kl. esculentus (RL), a natural hybrid between the marsh frog P. ridibundus (RR) and the pool frog P. lessonae (LL). However, the genetic mechanisms involved remain unclear. Here, we investigated the water frogs in the western Russian territory. In three of the four populations, we genetically identified 16 RL frogs living sympatrically with the parental LL species, or with both parental species. In addition, two populations contained genome introgression with another species, P. bedriagae (BB) (a close relative of RR). In the gonads of 13 RL frogs, the L genome was eliminated, producing gametes of R (or R combined with the B genome). In sharp contrast, one RL male eliminated the L or R genome, producing both R and L sperm. We detected a variation in genome elimination within a population. Based on the genetic backgrounds of RL frogs, we hypothesize that the introgression of the B genome resulted in the change in choosing a genome to be eliminated. Full article

Multidirectional Changes in Parameters Related to Sulfur Metabolism in Frog Tissues Exposed to Heavy Metal-Related Stress

The investigations showed changes of the cystathionine γ-lyase (CTH), 3-mercaptopyruvate sulfurtransferase (MPST) and rhodanese (TST) activity and gene expression in the brain, heart, liver, kidney, skeletal muscles and testes in frogs Pelophylax ridibundus, Xenopus laevis and Xenopus tropicalis in response to Pb2+, Hg2+ and Cd2+ stress. The results were analyzed jointly with changes in the expression of selected antioxidant enzymes (cytoplasmic and mitochondrial superoxide dismutase, glutathione peroxidase, catalase and thioredoxin reducatase) and with the level of malondialdehyde (a product of lipid peroxidation). The obtained results allowed for confirming the role of sulfurtransferases in the antioxidant protection of tissues exposed to heavy metal ions. Our results revealed different transcriptional responses of the investigated tissues to each of the examined heavy metals. The CTH, MPST and TST genes might be regarded as heavy metal stress-responsive. The CTH gene expression up-regulation was confirmed in the liver (Pb2+, Hg2+, Cd2+) and skeletal muscle (Hg2+), MPST in the brain (Pb2+, Hg2+), kidney (Pb2+, Cd2+), skeletal muscle (Pb2+, Hg2+,Cd2+) and TST in the brain (Pb2+) and kidney (Pb2+, Hg2+, Cd2+). Lead, mercury and cadmium toxicity was demonstrated to affect the glutathione (GSH) and cysteine levels, the concentration ratio of reduced to oxidized glutathione ([GSH]/[GSSG]) and the level of sulfane sulfur-containing compounds, which in case of enhanced reactive oxygen species generation can reveal their antioxidative properties. The present report is the first to widely describe the role of the sulfane sulfur/H2S generating enzymes and the cysteine/glutathione system in Pb2+, Hg2+ and Cd2+ stress in various frog tissues, and to explore the mechanisms mediating heavy metal-related stress.
