IgE Occupancy and Antigen Valency Cooperate to Control FcεRI Aggregation Geometry and Signaling Efficiency

The crosslinking of IgE-bound FcεRI by multivalent allergens initiates mast cell and basophil signaling underlying Type 1 allergy. Yet, how allergen properties and IgE occupancy impact receptor aggregation and downstream signaling remains unclear. We used Phl p 1-specific IgE and recombinant fusion proteins presenting a Phl p 1-derived peptide in defined valencies and positions (MB1N, MB1N1C, MB2N, and MB4N) to probe antigen-dependent signaling. Tetravalent MB4N evoked stronger degranulation and Ca2+ response than bivalent antigens MB1N1C and MB2N. MB4N was also capable of signaling at low IgE occupancy and in Lyn-deficient cells. Monte Carlo simulations predicted that MB4N forms larger, complex receptor aggregates, while MB1N1C and MB2N produce linear chains. Consistently, the addition of MB4N showed larger aggregates by electron microscopy and slower mobility by single-particle tracking, compared to bivalent antigens. Thus, allergen valency and epitope spatial arrangement dictate FcεRI aggregate organization and subsequent effector cell activation. Full article

Indoor mobile robots equipped with low-cost and sparse sensors often suffer from limited vertical perception and dynamic residual artifacts in the final map. This paper presents a lightweight 2.5D simultaneous localization and mapping (SLAM) framework using a single-line laser distance sensor (LDS), time-of-flight (ToF) sensing, wheel odometry, and an inertial measurement unit (IMU). In this work, 2.5D refers to a 2D grid map with discretized vertical occupancy bins for each grid cell, rather than a full continuous 3D reconstruction. The system integrates multi-sensor synchronization, motion correction, error-state Kalman filter (ESKF)-based state estimation, normal distributions transform (NDT) registration, and pose graph optimization to reconstruct a pose-consistent global map. Based on this map, an offline dynamic refinement module estimates temporal voxel support across keyframes, extracts low-support candidate regions, and applies geometric clustering and isolated-point filtering to suppress transient residual artifacts while preserving stable structures. A 24-bit RGB occupancy encoding is further proposed to store the discretized vertical occupancy state in a compact three-channel image format. The proposed framework emphasizes system-level deployment value by combining sparse multi-sensor mapping, conservative offline refinement, and compact height-aware map export on a low-cost indoor robot platform. Experiments on public datasets, embedded hardware, and self-collected indoor sequences evaluate odometry reference performance, resource usage, platform-specific 2.5D mapping, dynamic refinement, and height-aware encoding. Background/Objectives: Glycation, a non-enzymatic post-translational modification, can increase structural heterogeneity in therapeutic monoclonal antibodies (mAbs) and is considered a critical quality attribute in biosimilar (BS) development. Despite extensive studies on glycation in individual mAbs, comparative reports on originator (OR) and BS mAbs under forced glycation conditions remain limited. This study aimed to comparatively evaluate the glycation profiles of one OR and three BS anti-tumor necrosis factor-alpha (anti TNF-α) mAb products using integrated mass spectrometry-based methods. Methods: Forced glycation was induced by incubating mAbs with 200 mM D-glucose at 37 °C for 7 days. Intact mass analysis and peptide mapping were used to assess glycation extent and site distribution, respectively. Results: Intact mass analysis revealed a consistent mass increase of approximately 486 Da across all major glycoform species for each mAb product, indicating predominant formation of the tri-glycated mAb population under the applied stress conditions. The overall glycation levels were comparable at the intact level, ranging from 67% to 73% among the OR and BS mAb products. Peptide mapping identified nine glycated lysine (K)-containing peptides, among which three major glycation hotspots (LC:V5 K145/K149, LC:V7 K183, and HC:V7 K250/K252) showed elevated occupancies (~10–16%). These sites collectively accounted for the dominant intact level mass shift. The remaining glycated peptides exhibited only minimal modification levels (<2%). Despite their distinct manufacturing processes, all tested mAb products showed nearly identical site-specific glycation profiles. Conclusions: Forced glycation susceptibility in the studied mAbs was driven by a limited set of structurally preferred K hotspots, leading to highly comparable glycation profiles across OR and BS mAb products. The combined intact mass and peptide mapping strategy provides a robust analytical platform for comparative glycation assessment in BS characterization. Full article

Agriculture remains one of the most hazardous industries in the United States, yet nonfatal surveillance remains incomplete. Prior studies have shown that trauma registries can support agricultural injury surveillance, but many approaches rely on text review, state-specific indicators, or coding frameworks that are not easily transferable across systems. In this study, Florida Trauma Registry records from 2016 to 2021 were screened using a reproducible case definition based on International Classification of Diseases, Tenth Revision (ICD-10) external cause of injury codes, registry occupation and industry fields, and the work-related indicator. Agricultural injuries were summarized descriptively and mapped to Occupational Injury and Illness Classification System (OIICS) categories. Injury severity scores (ISS) were modeled using generalized additive models (GAMs). The case definition identified 963 agricultural injuries requiring trauma care, including 17 fatalities. Most injuries were minor, and the leading event categories were contact incidents, falls, slips, and trips, and transportation incidents. Body region was the strongest correlate of severity, with head and abdomen/pelvic contents injuries showing the highest adjusted ISS. These findings suggest that the National Trauma Data Standard (NTDS)-aligned trauma registry fields may support reproducible agricultural injury surveillance when narrative text is unavailable. Biomedical scientists/researchers are critical to advancing human health, but their experiences of occupational stress, burnout, and subsequent consequences are surmised but understudied. The current narrative review aimed to search and summarize existing literature and scientific discourse specific to biomedical scientist burnout to better understand the prevalence and unique drivers of stress in this population and highlight critical areas that require further research and intervention efforts. The authors searched six primary databases from August 2024 to December 2025 using defined search terms to identify peer-reviewed articles and scientific discourse related to occupational burnout of academic medicine biomedical faculty researchers, postdoctoral researchers, or lab personnel. Prevalence and primary drivers of burnout, stress mediators, and mitigation/intervention recommendations were extracted from the included works. A total of 16 empirical articles and 4 non-empirical works were included. Over 30% of biomedical scientists exhibit high occupational burnout, nearly 45% report high distress, and just over 34% experience high emotional exhaustion. Seven primary drivers of biomedical scientist distress were identified including intense workloads/expectations, funding pressures, and lack of perceived organizational support/recognition. Biomedical scientist burnout prevalence appears comparable to clinical faculty but is driven, in part, by distinct stressors. The attributable organization and societal cost of biomedical scientist burnout remain unquantified. The current review establishes a significant need for further research and tailored efforts to address the distinct needs of biomedical scientists across academic medical centers. Full article

Salt stress limits tomato productivity, yet how translational regulation contributes to root salt adaptation remains poorly understood. We integrated RNA-seq and ribosome profiling in wild-type (WT) and FERONIA (FER) mutant (fer) tomato roots under control and 150 mM NaCl conditions. In WT roots, the salt response was predominantly transcript-driven, but a 29-gene ribosome-associated module showed reduced RNA abundance alongside increased translational efficiency, indicating selective translational buffering. FER loss-of-function disrupted this balance, constitutively elevating ribosome occupancy of ribosome-associated genes while reducing basal expression of stress- and ion-transport-related genes; under salt treatment, fer also showed stronger ion-transport transcriptional responses but weaker translational efficiency responses of this module. WT salt stress further shifted ribosome allocation from the 5′ untranslated region (UTR) toward the coding sequence (CDS), an effect attenuated in fer, alongside positive coupling between uORF and CDS translational efficiency. Feature modeling identified sequence and structural predictors of uORF translation, including weaker local RNA folding near the start codon and specific amino acid and stop codon preferences. Together, these results reveal FER-associated changes in ribosome-associated translational buffering during tomato root salt responses. Full article

Psychosocial job characteristics are important components of the work environment and are closely associated with occupational health, particularly among nurses working in demanding healthcare settings. However, evidence from primary healthcare settings in low- and middle-income countries remains limited. This study aimed to describe psychosocial job characteristics and job strain among nurses in urban primary healthcare centers in Vietnam and to examine factors associated with psychosocial job characteristics and job strain. A multicenter cross-sectional study was conducted among 768 nurses across four district-level healthcare centers in Ho Chi Minh City, Vietnam, using a structured questionnaire including the validated Vietnamese Job Content Questionnaire. Nurses reported moderate-to-high job demands, high social support, and limited decision latitude. Most participants reported low strain (45.6%) or active jobs (42.4%), while fewer were in the high-strain (6.3%) and passive (5.7%) categories. Sociodemographic and work-related factors were significantly associated with job strain (p < 0.001). Although the prevalence of high strain was relatively low, differences between perceived job demands and specific dimensions of decision latitude were observed. These findings highlight the importance of job control, workload, and workplace support as factors associated with psychosocial job characteristics and job strain and may help inform future workplace interventions and longitudinal research. Indoor temperature forecasting in large public buildings is affected by heating, ventilation, and air-conditioning (HVAC) control, occupancy, airflow, and zone use, not only by recent temperature histories. This study asks whether operationally derived functional-zone proxy labels—room groupings derived heuristically from HVAC statistics rather than from field-verified building metadata—can provide a useful learnable context for temporal convolutional network (TCN)-based room-level forecasting when verified semantic metadata are unavailable. Using the public BEAR (Building Efficiency and Renewables) multizone building HVAC dataset, we construct a reproducible operational proxy scenario: eighty rooms are grouped from occupancy, airflow, damper, control-command, and temperature-variability statistics, and Niagara-inspired point-naming conventions are used only as structured metadata descriptors, not as tags exported from a deployed system. The proposed SemanticTCN augments a TCN backbone with learnable room, temperature-variability proxy embeddings. Experiments compare persistence, Long Short-Term Memory (LSTM), Gated Recurrent Unit (GRU), TCN, DLinear, PatchTST, a TCNOnly variant, SemanticTCN, and ablations for 1 h, 6 h, and 24 h forecasts, evaluated by mean absolute error (MAE), root mean squared error (RMSE), mean absolute percentage error (MAPE), and the coefficient of determination (R2). Persistence is strongest at 1 h and 24 h, while DLinear is strongest at 6 h, indicating strong thermal inertia and daily regularity. Within the TCN family, SemanticTCN improves over TCNOnly in the locked run, shows a statistically significant within-distribution MAE improvement across multiple seeds (most consistently at 6 h), and reduces large-error rates; however, a leave-one-zone-out experiment shows that this advantage does not transfer to rooms unseen during training, and a five-seed operational-versus-random-proxy control shows that the specific operational grouping rule is not statistically distinguishable from arbitrary grouping. The bounded conclusion is that providing room/group context to the TCN backbone helps predict rooms whose identity was observed during training, especially at 6 h, but the gain reflects room-identity context rather than generalizable operational semantics: the proxy labels are heuristic operational constructs rather than verified building semantics; they do not outperform arbitrary grouping under multi-seed control, and they do not generalize to unseen rooms under this split. Occupational burnout has become a major topic in occupational health psychology because of its implications for employee well-being, mental health, and organizational functioning. Although the scientific literature has grown steadily, the field still requires an integrated picture of its development, collaboration patterns, and thematic structure. This study examined the evolution, collaboration patterns and thematic structure of occupational burnout research through a bibliometric analysis of publications indexed in the Web of Science Core Collection between 2000 and 2025. A total of 3227 publications were retrieved using a topic-based search focused specifically on occupational burnout terminology. Bibliometric analyses used VOSviewer to map and visualize trends in scientific production, co-authorship networks, country/region collaboration, keyword co-occurrence, and journal citation patterns. The findings indicate an accelerating increase in publication output, from 17 publications in 2000 to 454 in 2025. The period 2020–2025 alone accounted for 2142 publications, representing 66.4% of the corpus. Collaboration analyses revealed an increasingly international field involving 122 countries/regions, with the United States occupying a highly connected position and collaboration clusters emerging in Europe and Asia. Spain also appeared to connect European and Latin American research communities. Thematic mapping showed that burnout is closely associated with organizational variables, psychological distress, healthcare settings, and measurement-related issues. Overall, occupational burnout research has become an internationalized and multidisciplinary field, shaped by the growing relevance of psychosocial risks, mental health, and work-related well-being. Free generative AI (GenAI) services appear to democratize access, but freemium platform models can create post-access stratification after basic connectivity and initial use have been achieved. This article conceptualizes paid commitment to GenAI as an emergent outcome of a socio-technical system composed of platform tiers, user capabilities, task environments, economic constraints, affective feedback, and institutional access. Using a cross-sectional 2025 survey of 2000 South Korean adults aged 18–69, the study models the observed states of non-use, free use, and paid commitment through sequential logit models. Descriptive results show that 70.6% of respondents used GenAI in the previous year, yet only 20.6% of users, or 14.6% of the total sample, paid for access. Practical AI literacy is positively associated with both initial use and paid commitment, whereas critical AI awareness is associated with initial use but not payment. Negative emotions are negatively associated with initial use but positively associated with paid commitment among users, a pattern consistent with vigilant and intensive engagement. The premium digital divide is defined not as a fourth level of the digital divide, but as a tier-mediated extension of second-level inequality that may condition access to later educational, occupational, or civic benefits. Because the data are cross-sectional and combine different GenAI platforms, the estimates are ecosystem-level associations and do not establish causal direction or platform-specific effects. Personal health and safety are significant concerns across agricultural education, specifically agricultural mechanics. While hearing safety has been discussed for numerous years as a shortcoming in overall safety actions, it has been identified that attitudes and perceptions of sound and noise within agricultural settings are not aligned. This research continues work to focus on students’ understanding of and application of safety culture within an agricultural mechanics environment. Through direct and indirect exposure to noise levels, students reflected on and identified their use and intended use of hearing personal protective equipment (PPE). Pre- and post-instructional data aided in establishing students’ thresholds to use proper personal protective equipment and identify their perceptions of the decibel (dB) outputs for commonly used power tools in an agricultural laboratory setting. Course reflections alarmingly show a disconnect between intention and practice as many students do not utilize hearing PPE. While indicated threshold values and tool-specific PPE use align with the National Institute for Occupational Safety and Health’s recommended hearing PPE guidelines, the self-reported hearing PPE use results are concerning. While knowledge and application gaps still need to be addressed, based on this exploratory study, bounded in a single instructional context, continued exposure through direct and indirect instruction leads to increased knowledge of sound levels. Background/Objectives: Children with cancer admitted to the pediatric intensive care unit (PICU) are at increased risk for post-intensive care syndrome (PICS-p) due to prolonged immobility, deep sedation, and severe illness. The ABCDEF bundle offers a framework for enhancing ICU care and patient recovery, but implementing all components in pediatric oncology patients is challenging. This study assesses the development and implementation of the BRAVE (Beginning Restorative Activities Very Early) initiative, specifically BRAVE2, to integrate the comprehensive ABCDEF bundle and a nurse-led mobility program, in collaboration with rehabilitation specialists, within a pediatric oncology intensive care unit. Methods: BRAVE2 was a quality improvement project conducted in a single pediatric ICU from 2022 to 2023. We analyzed ICU data to assess patient demographics, frequency of physical and occupational therapy (PT/OT) consultations, time to initial mobilization, and delirium screening rates (CAPD score of 9 or higher) for patients with ICU stays over 48 h. BRAVE2 addressed all elements of the ABCDEF bundle, including regular pain assessments, evaluation of spontaneous breathing readiness, sedation adjustments, delirium screening, early mobilization, and family engagement. Outcomes were monitored using statistical process control methods. Results: Of 140 patients, 117 (84%) remained in the ICU for more than 48 h. The delirium screening rate was 15.5%, consistently below the target of 30%. PT/OT consultations within 72 h occurred in 80.7% of patients, and early mobilization in 49.7% of patients, both below the 80% goal. However, 90.4% of patients with tracked mobility were able to ambulate during their ICU stay. No mobility-related safety incidents were reported. Conclusions: Rolling out a full ICU liberation plan in a pediatric oncology ICU is possible, and implementing a comprehensive one is feasible and sustainable despite challenges. Although therapist-led early mobilization did not meet targets, incorporating nurse-led mobility strategies and routine delirium screening has established a scalable model to enhance ICU care and support long-term recovery for these patients. The acoustic quality of a school’s indoor environment affects the health, performance and comfort of the teachers who work in it, yet assessment often reduces that environment to a single overall level. In a cross-sectional case study of one secondary school in China, all 148 teachers rated eleven school sound-source categories on audibility, annoyance and interference with teaching and office concentration, and reported perceived noisiness, voice raising, voice strain, fatigue, emotional distress and noise sensitivity, while selected-room acoustic measurements provided site context not matched to individual respondents. Ratings across the eleven sources were dominated by one component, but the resulting composite score was not associated with all teacher responses alike. Source load was associated with emotional distress, voice raising and perceived noisiness but not fatigue, which instead tracked voice strain, whereas emotional distress was associated most strongly with noise sensitivity. In exploratory source-specific models, traffic and other external sources had the largest associations with perceived noisiness, whereas playground activity and student talking had the largest associations with voice raising. No tested subgroup interaction survived false-discovery-rate correction. The selected-room measurements documented conditions compatible with source intrusion and raised vocal effort. The findings support an occupant-centred, source-resolved approach to school indoor acoustics and provide hypotheses for future multi-school testing. Full article

Wearable and environmental sensors are increasingly used to monitor human performance and health in real time, but their practical value remains limited when signals are interpreted in isolation or through context-independent algorithms. This Perspective focuses on real-time physiological monitoring in elite sport, using exertional heat illness risk and physiological strain as a high-stakes worked example. Our work further illustrates how such monitoring could move from device-level measurement toward fit-for-purpose sensor ecosystems. We argue that such ecosystems should be defined not by multimodal data collection alone, but by five core pillars: time-synchronised sensing, context-specific interpretation, multi-level validation, uncertainty-aware modelling and embedded governance—each evaluated against a defined high-stakes decision rather than device accuracy alone. Accurate decision-making in this context may require integration of thermoregulatory, cardiovascular, biomechanical and environmental data streams. Emerging artificial intelligence approaches may support prediction and decision support, but only if models are transparent, calibrated, prospectively validated and robust to individual and contextual variability. We also examine unresolved risks, including algorithmic bias, false reassurance, athlete surveillance, proprietary opacity, data asymmetry and regulatory constraints. While the framework and evidence presented here are anchored in elite sport, we discuss where its core validation and governance principles may extend to clinical, occupational and military settings, and where the underlying evidence remains sport-specific. The proposed framework is intended to complement, rather than replace, existing digital health, athlete management and cyber–physical systems by specifying the validation and governance requirements needed for high-stakes human performance monitoring. This study examines the critical relationship between geoenvironmental changes and human occupation patterns in the central Arabian Peninsula, focusing on the Al-Aflaj region. By integrating geospatial modeling with preliminary archeological excavation results that indicated the site is dated to 5th century BCE to 6th century CE, we evaluate how climatic stressors dictated human adaptation and eventual site abandonment. Late Quaternary climatic fluctuations, particularly the Early Holocene pluvial phases, initially created favorable conditions for settlement through sustained freshwater resources. Subsequent aridification triggered significant migration and settlement contraction, demonstrating a high degree of human resilience. The site of Qusayrat Aad serves as a compelling case study of sophisticated adaptation, evidenced by mudbrick architecture and advanced irrigation systems. Integration of geological, topographic, and paleoclimatic factors indicates that the porous sedimentary layers of the Heet and Al Biyadh Formations were essential for groundwater recharge and spring formation. The geospatial analysis reveals that the settlement was strategically localized on a stable surface with a mean slope of 1.51°. Furthermore, the Topographic Wetness Index (TWI) identifies the site as a significant hydrological function, with a mean value of 8.79 (reaching a 90th percentile of 12.37), which is markedly higher than the regional average of 7.96. These quantitative findings establish a causal necessity for the site’s advanced subsurface canal systems as an engineered response to minimize evaporative losses in high-potential moisture zones. Ultimately, the correlation between the archeological record and climatic proxies suggests that the intensification of late Holocene aridification depleted these specific water resources beyond adaptive capacity, serving as the primary driver for the site’s abandonment and the migration of populations toward the eastern and southern parts of the Peninsula.
