Adequate management of replacement ewe lambs (F0) in dairy sheep farms during postnatal life may modify the germline cells, thus promoting transmission of intergenerational effects to the offspring (F1). To test this hypothesis, 18 newborn male lambs (F1), either born from methionine-supplemented ewe lambs (F0 ewe lambs being fed ad libitum with a milk replacer supplemented with 1 g methionine/kg DM) or not supplemented (F0 ewe lambs being fed ad libitum with the same milk replacer with no methionine added), were included in the present study. All the male F1 lambs were managed exactly in the same way along the whole lifespan in order to bring out the differences caused by methionine supplementation of F0 dams. Our data show that the methionine supplementation of dams (F0) during the suckling period did not promote significant (p > 0.05) changes on feed intake, growth rate, or feed efficiency of F1 male lambs during the fattening period. Moreover, the meat chemical composition (proximal, fatty acid profile, and volatile compounds) was similar for both groups (p > 0.05), but the meat of F1-MET lambs presented higher redness and hardness (p < 0.05) when compared to F1-CTRL lambs. The biochemical profile also highlighted significant (p < 0.05) differences in the serum creatinine and calcium content that may be at least partially related to the meat quality traits observed. Overall, all these results suggest that methionine supplementation of lambs (F0) during early postnatal life causes permanent changes in the offspring. This has positive effects, such as achieving a more attractive color of lamb meat (F1) for consumers, and negative effects, such as reduced meat tenderness. Full article

Objectives: This article presents a study evaluating the antibody levels against infectious bovine rhinotracheitis (IBR) and bovine viral diarrhea (BVD) in Kazakh Whiteheaded calves born to dams immunized with an experimental inactivated combined vaccine against these infections. The vaccine formulation includes the strains “R-93” (IBR) and “Oregon C24V” (BVD), which are preserved in the microorganism collection of the Research Institute for Biological Safety Problems. Methods: To assess the immune response in newborn calves, blood serum samples were collected before the first intake of colostrum, followed by weekly sampling for 28 weeks post-birth. The antibody response was determined using a virus neutralization assay on MDBK cell cultures and lamb testicle cell cultures. Results: The results demonstrated that the protective antibody level against the IBR virus (≥2 log2) persisted for up to 25 weeks, while the protective level against the BVD virus (≥3 log2) remained for 23 weeks. Based on these findings, the vaccine was deemed safe, as it did not induce abortions or clinical manifestations of the diseases. The overall duration of the colostral immunity in calves against the IBR and BVD viruses reached 23 weeks. Conclusions: Therefore, it is recommended that Kazakh Whiteheaded calves be vaccinated with the associated inactivated vaccine against infectious bovine rhinotracheitis and bovine viral diarrhea no earlier than 23 weeks of age. Background: Lamb health is crucial for producers; however, the percentage of lambs that die before weaning is still 15–20%. One factor that can contribute to lamb deaths is congenital diseases. A novel semi-lethal disease has been identified in newborn Polypay lambs and termed dozer lamb syndrome. This study aims to determine if there is a genetic predisposition to dozer lamb syndrome. These lambs are weak and unable to lift their heads, suckle, and swallow, resulting in nasal reflux. Methods: Genetic analyses, including a genome-wide association, runs of homozygosity, and fine mapping to determine haploblock within regions of interest, were utilized in determining genetic predispositions to dozer lamb syndrome. Results: The genome-wide association study identified a region of chromosome 15 with three significant SNPs (p-values of 6.81 × 10−6, 5.71 × 10−6, and 8.52 × 10−6). Genetic analysis identified a run of homozygosity on the same region of chromosome 15 with an odds ratio of 236.7. Fine mapping of this region identified three haploblocks associated with the dozer lamb syndrome (p-value = 2.41 × 10−5). Conclusions: The most significant and promising gene in this region is CELF1, which is known to play an important role in muscle development. Abnormal CELF1 abundance and cellular location are reported to result in abnormal muscle development. Identification of genetic aberrations associated with dozer lamb syndrome provides a tool for decreasing or eliminating the genotype and, thus, the associated phenotype(s) from Polypay sheep. Melatonin’s capacity to improve cashmere production and quality in goats is well established, but its underlying mechanisms, particularly those concerning the gastrointestinal microbiome, remain inadequately understood. This study aims to elucidate the effects of melatonin implantation on the production performance, blood biochemical parameters, nutrient digestibility, and gastrointestinal microbiome of Liaoning cashmere goats. Thirty newborn Liaoning cashmere goat lambs were selected and randomly assigned to control and melatonin groups using a paired test design. The melatonin group received three melatonin implantations at 15, 75, and 135 days of age, respectively, with a dosage of 2 mg/kg body weight, while the control group received no treatment. Digestive metabolism tests were conducted at 150 and 300 days of age; prior to these tests, blood, rumen fluid, and rectal feces were collected. Apparent nutrient digestibility and blood biochemical indexes were determined, and rumen fluid and rectal feces were analyzed using microbial 16S rRNA sequencing. The results indicated that melatonin significantly reduced daily weight gain and body weight at 60 days (p < 0.05) while significantly increasing daily weight gain at 300 days (p < 0.05). Additionally, it significantly increased cashmere length and reduced its fineness (p < 0.05). Melatonin significantly enhanced nitrogen deposition (p < 0.05), elevated plasma levels of T-AOC, CAT, GSH-PX, and BUN (p < 0.05), and reduced plasma levels of MDA, GOT, GPT, and AKP (p < 0.05). Moreover, melatonin significantly elevated the microbial Ace and Chao1 indices in rectal feces (p < 0.05), increasing genera beneficial for feed digestion and absorption, including Prevotella, Lachnospiraceae, Ruminococcus, and Synergistaceae (p < 0.05); the abundance of these beneficial genera were positively correlated with improved cashmere production performance, antioxidant activity, and liver and kidney function. In conclusion, melatonin enhances cashmere production by modulating gastrointestinal microbiota, antioxidant activity, liver and kidney function, and nitrogen metabolism in cashmere goats. This study provides a theoretical foundation for melatonin’s role in microbiota modulation, which is essential for promoting high-quality and sustainable development in the cashmere goat industry. Toxoplasmosis, caused by Toxoplasma gondii, poses a significant threat to sheep flocks, affecting reproductive performance and meat quality, and leading to economic losses. This study aimed to evaluate the correlation between anti-T. gondii IgG antibodies in the serum and colostrum of naturally infected ewes and to assess passive immunity in newborn lambs. Blood and colostrum samples were collected from 162 ewes and 182 lambs across 20 sheep farms in Paraíba, Brazil. Samples were tested for anti-T. gondii and anti-Neospora caninum IgG using indirect fluorescence antibody tests (IFATs), with titers ≥ 1:64 considered positive. Among the ewes, 45.1% tested positive for anti-T. gondii IgG in serum, with titers ranging from 1:64 to 1:16,384. The colostrum from 94.6% of the ewes also tested positive, although 74% had higher titers in their serum than in their colostrum. Concordance between serum and colostrum was high, with a kappa coefficient of 0.950. Lamb serum showed a perfect agreement with maternal colostrum (kappa = 0.962), demonstrating effective passive transfer of antibodies. This study confirms that colostrum is a reliable matrix for detecting anti-T. gondii antibodies and assessing passive immunity in lambs. The high concordance between serum, colostrum, and lamb titers suggests that IFATs on colostrum can be a practical tool for monitoring maternal antibody transfer, contributing to the better management of T. gondii infections in sheep flocks. Newborn lambs are susceptible to pathogenic bacterial infections leading to enteritis, which affects their growth and development and causes losses in sheep production. It has been reported that N6-methyladenosine (m6A) is closely related to innate immunity, but the effect of m6A on sheep small intestinal epithelial cells (IECs) and the mechanism involved have not been elucidated. Here, we investigated the effects of m6A on lipopolysaccharide (LPS)-induced inflammatory responses, apoptosis and oxidative stress in primary sheep IECs. First, the extracted IECs were identified by immunofluorescence using the epithelial cell signature protein cytokeratin 18 (CK18), and the cellular activity of IECs induced by different concentrations of LPS was determined by the CCK8 assay. Meanwhile, LPS could induce the upregulation of mRNA and protein levels of IECs cytokines IL1β, IL6 and TNFα and the apoptosis marker genes caspase-3, caspase-9, Bax, and apoptosis rate, reactive oxygen species (ROS) levels and mRNA levels of CAT, Mn-SOD and CuZn-SOD, and METTL3 were found to be upregulated during induction. It was hypothesized that METTL3 may have a potential effect on the induction of IECs by LPS. Overexpression and knockdown of METTL3 in IECs revealed that a low-level expression of METTL3 could reduce the inflammatory response, apoptosis and ROS levels in LPS-induced IECs to some extent. The results suggest that METTL3 may be a genetic marker for potential resistance to cellular damage. Full article

Cardiac congenital defects related to inheritance and teratogenesis have been reported in veterinary species and humans worldwide. Among these, ectopia cordis (EC), characterized by an externalized heart through a cleft, is extremely rare in sheep. This report presents the diagnostic features of two cases of complete thoracic EC in newborn lambs. Clinical findings in the lambs, aside from the EC, were unremarkable. Both animals exhibited exteriorized hearts without pericardial coverage, delineated in the thoracic cleft by a fibrous ring of the pericardium and adjacent skin. Histologically, the epicardium was thickened by fibrous tissue in both lambs, with one animal also showing marked edema, hemorrhage, and neutrophilic inflammatory infiltration. The prognosis of EC in the lambs of this study was poor, with fatal outcomes despite attempts at surgical correction. Full article

The neutral detergent fiber (NDF)/starch ratio is a key determinant of the carbohydrate composition in ruminant diets, which affects the development of the gastrointestinal tract and animal growth. In this study, we used a combination of 16S amplicon sequencing and metabolomics technologies to reveal changes in the microbiota and their metabolites associated with digestive matter in the colon of lambs between different starter NDF/starch ratios and before and after weaning. A total of 40 male lambs of Hu sheep with a newborn weight of 3.14 ± 0.05 kg were selected for the experiment and fed with breast milk until 10 days of age, and were randomly divided into 2 groups, which were fed ad libitum with a starter of NDF/starch of 0.5 (A) or 1.0 (B) for a period of 56 days, and then weaned off the milk powder at 35 days of age. Six lambs with weights close to the average weight of the group were selected for slaughter in each of the two groups before (35 days of age) and after weaning (56 days of age), and were accordingly named A35, A56, B35 and B56. The results showed that, before weaning, the concentrations of acetic acid, propionic acid, butyric acid and total volatile fatty acids (TVFA) in group B35 were significantly higher than those in group A35 (p < 0.05), while the concentrations of isobutyric acid, valeric acid and isovaleric acid were significantly lower than those in group A35 (p < 0.05). After weaning, the concentrations of all volatile acids and total acids in group B56 were significantly lower than those in group A56 (p < 0.01). At the phylum level, the dominant phyla identified were Firmicutes and Bacteroidetes; the relative abundances of Firmicutes and Desulfobacterota were significantly higher and lower in group B35 than in group A35, respectively (p < 0.05); the relative abundances of Euryarchaeota and Desulfobacterota were significantly higher and lower in group A56 than in group A35 (p < 0.05); and, at the genus level, the relative abundance of Lachnospiraceae_FCS020_group and Eubacterium nodatum group had higher relative abundance in group B35 before weaning (p < 0.05). Metabolomic results showed that feeding 1.0 NDF/starch ratio starter before weaning significantly up-regulated (p < 0.05) the concentrations of several anti-inflammatory-related metabolites such as lithocholic acid, oleanolic acid and LysoP. After weaning, the number of differential microorganisms and anti-inflammatory-related metabolites decreased between the two ratios. In summary, feeding a 1.0 NDF/starch ratio starter may be more effective in regulating microbial fermentation, leading to an increase in beneficial microbiota and metabolites, thus improving colonic environmental homeostasis in lambs before and after weaning. Contagious ecthyma is a highly transmissible eruptive viral disease of the skin and mucosa of sheep and goats distributed worldwide. The treatment of orf lesions is usually based on the use of antiseptics and antibiotics for the management of presumptive secondary infections, increasing risks of antimicrobial resistance. The wound dressing formulation Tri-Solfen® (TS) containing two local anaesthetics (lignocaine and bupivacaine), adrenaline and an antiseptic (cetrimide) in a gel formulation has been demonstrated to reduce suffering and enhance recovery in cattle and buffalo with oral and skin lesions due to foot-and-mouth disease virus infection and reduced the orf viral load in lambs. In the present study, experimental infection with the orf virus was conducted in 50 newborn lambs and 25 animals were treated after the presence of the first lesions with TS and repeated three days later. Daily clinical examination, haematological, serological, biomolecular and post-mortem analyses were conducted during 34 days after treatment. Results indicated that treatment had no effect on weight gain and clinical progression of the lesions. It was determined that seroconversion after experimental infection occurs 34 days after infection and suggested that the deep basal epithelial location of the orf lesions may have prevented the therapy from having altered the clinical course. Caffeine is widely used to improve neonatal health in animals with low vitality. Due to its pharmacokinetics and pharmacodynamics, caffeine stimulates the cardiorespiratory system by antagonism of adenosine receptors and alteration in Ca+2 ion channel activity. Moreover, the availability of intracellular Ca+2 also has positive inotropic effects by increasing heart contractibility and by having a possible positive effect on neonate vitality. Nonetheless, since neonatal enzymatic and tissular systems are immature at birth, there is a controversy about whether caffeine is an effective therapy for newborns. This review aims to analyze the basic concepts of caffeine in neonatal animal models (rat and mouse pups, goat kids, lambs, and piglets), and it will discuss the neuroprotective effect and its physiological actions in reducing apnea in newborns. The objective of this study was to compare the electrophoretic pattern of serum proteins in newborn calves, lambs, goat kids and piglets in precolostral blood samples. Blood samples were collected within 30 min of birth, prior to ingestion of first colostrum, and the concentrations of total proteins and protein fractions were analyzed using electrophoresis on agarose gel. The size and shape of the protein fractions on the electrophoretograms differed among the investigated animal species. Significant differences were found in the total protein values and all the separated protein fractions, as well as albumin to globulin ratio (p < 0.01 and p < 0.001). The mean total protein concentration in piglets was lower compared to ruminants. Albumin concentrations were markedly higher, and the concentrations of α1-globulins were lower, in neonatal ruminants compared to piglets. The values of α2-globulins were higher in lambs and goat kids, and lower in calves and piglets. An opposite tendency was found in the values of β-globulins: a detectable amount of γ-globulins was recorded in all evaluated animal species. Presented results suggest marked species related differences in the shape and size of protein fraction among neonates of farm animal species, and the importance of the evaluation of electrophoretograms with regard to these findings. Brown adipose tissue (BAT) plays a critical role in maintaining the body temperature in newborn lamb due to its unique non-shivering thermogenesis. Previous studies have found that BAT thermogenesis is regulated by several long non-coding RNAs (lncRNAs). Here, we identified a novel lncRNA, MSTRG.310246.1, which was enriched in BAT. MSTRG.310246.1 was localized in both the nuclear and cytoplasmic compartments. In addition, MSTRG.310246.1 expression was upregulated during brown adipocyte differentiation. Overexpression of MSTRG.310246.1 increased the differentiation and thermogenesis of goat brown adipocytes. On the contrary, the knockdown of MSTRG.310246.1 inhibited the differentiation and thermogenesis of goat brown adipocytes. However, MSTRG.310246.1 had no effect on goat white adipocyte differentiation and thermogenesis. Our results show that MSTRG.310246.1 is a BAT-enriched LncRNA that improves the differentiation and thermogenesis of goat brown adipocytes. Full article

The combination of perinatal acidemia with postnatal hyperoxia is associated with a higher incidence of hypoxic-ischemic encephalopathy (HIE) in newborn infants. In neonatal cardiac arrest, current International Liaison Committee on Resuscitation (ILCOR) and Neonatal Resuscitation Program (NRP) guidelines recommend increasing inspired O2 to 100% during chest compressions (CC). Following the return of spontaneous circulation (ROSC), gradual weaning from 100% O2 based on pulse oximetry (SpO2) can be associated with hyperoxia and risk for cerebral tissue injury owing to oxidative stress. We hypothesize that compared to gradual weaning from 100% O2 with titration based on preductal SpO2, abrupt or rapid weaning of inspired O2 to 21% after ROSC or use of 21% O2 during CC followed by upward titration of inspired O2 to achieve target SpO2 after ROSC will limit hyperoxia after ROSC. Nineteen lambs were randomized before delivery and asphyxial arrest was induced by umbilical cord occlusion. There was no difference in oxygenation during chest compressions between the three groups. Gradual weaning of inspired O2 from 100% O2 after ROSC resulted in supraphysiological PaO2 and higher cerebral oxygen delivery compared to 21% O2 during CC or 100% O2 during CC followed by abrupt weaning to 21% O2 after ROSC. The use of 21% O2 during CC was associated with very low PaO2 after ROSC and higher brain tissue lactic acid compared to other groups. Our findings support the current recommendations to use 100% O2 during CC and additionally suggest the benefit of abrupt decrease in inspired oxygen to 21% O2 after ROSC. Clinical studies are warranted to investigate optimal oxygen titration after chest compressions and ROSC during neonatal resuscitation. Full article

Chemometric Approaches to Analyse the Composition of a Ewe’s Colostrum

Colostrum is a major source of immunity in lambs and, in general, in all newborn ruminants. It allows the transfer of antibodies from the ewe to the lamb, and it becomes the exclusive source of nutrients for a newborn. Among the most significant Pearson correlations, the positive correlation between the Brix refractive index (Brix) and protein (0.90) should be noted. Both parameters (protein percentage and Brix) were then positively correlated with the percentage of fat (0.38 and 0.41), urea (0.81 and 0.67), calcium (0.39 and 0.29), and magnesium (0.58 and 0.59), as well as the yellowness (0.78 and 0.75). Somatic cell count (SCC) and pH, parameters which are indicators of subclinical mastitis, were positively correlated (0.49), and SCC was positively correlated with sodium (0.37) and negatively correlated with potassium (−0.28). Among the macroelements in colostrum—calcium, potassium, magnesium, and sodium—the correlations were largely positive. With respect to the colour parameters, yellowness was negatively correlated with lightness (−0.41) and redness (−0.45). The factor analysis split the total of variance into three latent factors. The first factor was named “Colostrum quality of grazing sheep” because it was positively correlated primarily with SCC, pH, Poly Unsatured Fatty Acids (PUFA), and the sum of the omega-3 Fatty Acids (FAs). The second factor, named “Good quality colostrum”, was positively correlated primarily with the refractive index, protein and fat percentages, urea content, phosphorus, Mono Unsatured Fatty Acids (MUFA), and yellowness. The third factor was positively correlated primarily with calcium, potassium, magnesium, and sodium, and it was therefore termed “Mineral component of colostrum”. Stepwise discriminant analysis showed that the protein percentage, calcium, and magnesium were able to explain more than 85% of the Brix refractive index, which remains a good parameter for simply evaluating the nutritional quality of sheep colostrum at the level of a farm. Previous research indicates that adequate choline nutrition during late gestation improves fetal development. However, there is a lack of studies describing choline’s role during early gestation. Thus, the current study hypothesizes that an herbal mixture as a source of choline (Biocholine) positively affects offspring development from ewes supplemented during early gestation. Therefore, the objectives were to evaluate the impact of biocholine on the programming of the offspring early in life through the evaluation of dams and newborn performance. Twenty-eight four-year-old Rambouillet ewes were assigned randomly to two treatments: non-supplementation and 4 gd−1 of biocholine during the early gestation. Compared with the dams without supplementation, the ewes supplemented using biocholine showed no increase in parameters such as birth and weaning weight (p > 0.05). Additionally, the milk yield and quality of colostrum and milk did not present statistical differences (p > 0.05). However, the placental membrane development was reduced in the ewes that received supplementation with biocholine; interestingly, those dams increased the weight of the newborns during the lambing period (p < 0.05). Finally, the current study proposes necessary elucidation of how placental size is programmed and if less placental development has potential benefits in the fetus’s development. Full article
