Infertility is a complex and common problem in reproductive medicine consultations. Three factors must be examined during the preconception phase: breeding management, the fertility of the bitch, and the fertility of the stud dog. Among these factors, improper breeding management remains the main cause of reproductive failure, with accurate recognition of ovulation being crucial for successful mating. Artificial insemination allows for a thorough evaluation of semen quality compared to natural mating. In addition, genetic selection, nutritional factors, and reproductive health management can either impair or improve the fertility of females and males. Idiopathic infertility can occur in bitches, but it is important to rule out other possible causes first. In bitches with irregular estrus cycles, ovarian dysfunction and endocrine imbalances should be investigated. In bitches with regular cycles, uterine disorders such as cystic endometrial hyperplasia, endometritis or congenital anomalies may be the cause. Both mating-related and chronic endometritis are recognized as contributing factors to infertility. Infectious agents, particularly Brucella spp. and Mycoplasma spp., should also be evaluated, although interpretation of Mycoplasma test results requires caution. In males presenting with poor semen quality, potential causes include infectious diseases (with brucellosis always requiring exclusion), hormonal imbalances, and the impact of exogenous treatments. The article underscores the critical role of comprehensive diagnostic protocols, proactive health surveillance, and data-driven breeding strategies in systematically addressing this multifaceted challenge. In the present study, we examine the possibilities of planned generation in snakes by controlling the two most important factors for their estrus—ambient temperature and daylight hours. As a result of controlling these environmental parameters in an increasing gradient until reaching optimal values for the species, we observed copulations in late March and early April between a female Vipera ammodytes transcaucasiana and a male V. a. ammodytes. After three months of “pregnancy”, we obtained viable offspring in early July, which is about two months earlier compared to wild populations. The species used in the experimental setup, in natural conditions, usually produce offspring in late August to early October. Another aspect considered in the publication and followed in the experimental setup was to possibly test if in evolutionary and developmental aspect, both subspecies are closely related and interspecies breeding is possible (which might indicate mutual ancestry). The hybrid individuals were monitored during their entire development from newborns to subadults for pathological traits during development to roll-out crossbreeding incompatibility. In our pilot investigation, no acquired or inherited pathological traits have been observed. The individuals were consistent with feeding and exhibited excellent individual development. Future research coupled with genetic investigation can give valuable insight in the field, whether it is valid to regard the genera as a Vipera ammodytes complex or as different subspecies groups. Sheep have evolved remarkable phenotypic diversity through artificial and natural selection, with reproductive traits being pivotal for breeding economics. As a unique genetic resource, Pishan red sheep exhibit exceptional advantages, including perennial estrus, high fecundity, and stable hereditary characteristics, establishing them as an optimal model for investigating reproductive genetics. In this study, we performed whole-genome resequencing of Pishan red sheep, generating 9084.81 Gb of raw data and identifying 53,968,686 high-quality single-nucleotide polymorphisms (SNPs). Through selective sweep analysis, 92 genomic regions under selection were detected, containing 90 positional candidate genes significantly associated with growth, reproduction, and immune functions. Notably, we revealed BMPRIB, UNC5C, PDLIM5, GRID2, and HPGDS as core positional candidate genes influencing litter size, operating through the TGF-beta and Thyroid hormone signaling pathways. A genome-wide association study (GWAS) further identified 59 trait-related SNPs, including 39 loci linked to growth traits (affecting positional candidate genes such as PROM1, TAPT1, LDB2, and KIF16B) and 20 loci of positional candidate genes associated with reproductive traits (involving ASPA, RAP1GAP2, PHIP, and WDR82).These findings not only elucidate the molecular basis of superior reproductive performance in Pishan red sheep, but also provide functional markers for precision breeding. Background: Accurate estrus identification in dairy cows is essential for enhancing reproductive efficiency and economic performance. The dispersed nature of estrus data and individual cow differences pose significant challenges for accurate identification. Methods: This study gathered cow estrus data from 812 literature sources using Python 3.9 crawler technology. The data were then preprocessed using CiteSpace 6.4. We constructed a knowledge graph depicting physiological, behavioral, and appearance changes during estrus through entity and relationship extraction. To uncover potential relationships within the graph, we applied and compared two association rule algorithms: FP-Growth and Apriori. We utilized Boolean functions derived from association rule learning to validate the ability of the rules to identify normal estrus. Additionally, we employed an enhanced Iforest-OCSVM anomaly detection model to assess the performance of the association rules in detecting abnormal estrus. Furthermore, we optimized the Incremental FP-Growth Algorithm for Dynamic Knowledge Expansion. Results: Based on the initial knowledge graph with 86 entities and 9 relationships, mining added 17 new strong association relationships marked by ‘with’, enhancing its completeness and providing deeper insights into estrus behaviors and physiological changes. Furthermore, these strong association rules exhibited notable effectiveness in both normal and abnormal estrus detection, validating their robustness in practical applications. The algorithm’s optimization bolstered its scalability, making it more adaptable to future data expansions and complex knowledge integrations. Conclusions: By constructing a knowledge graph that integrates physiological, behavioral, and appearance changes during estrus, we established a comprehensive framework for understanding cow estrus. Association rule mining, particularly with the FP-Growth algorithm, added 17 new strong association relationships to the graph, enriching its content and offering deeper insights into estrus behaviors and physiological changes. The strong association rules derived from FP-Growth demonstrated notable effectiveness in both normal and abnormal estrus detection, validating their robustness and practical utility in enhancing estrus identification accuracy, and providing a robust foundation for future multi-dimensional estrus research. A key feature in sheep biology is reproduction seasonality which concerns the cyclical occurrence of natural breeding, which therefore does not take place throughout the year. Since sheep are short-day breeders, the amount of daylight has an impact on their reproductive activity. The melatonin receptor subtype 1A (MTNR1A) gene is the primary gene that has been linked with seasonality. Nonetheless, information regarding the potential genetic association between other loci and the seasonality of sheep reproduction is scarce. Genome-wide association study (GWAS) is considered a cutting-edge methodology for comprehending the genetic architecture of complex traits since it enables the discovery of many markers linked to different features. In the present study, three indigenous Greek sheep breeds were investigated using GWAS—two of which presented strict patterns of reproduction seasonality, i.e., the Florina and Karagkouniko breeds, while the third one, i.e., the Chios breed had the ability to exhibit estrus throughout the year—in an attempt to detect the genetic loci linked with reproduction seasonality. All three breeds of investigated animals were purebred with Chios and Florina breeds originating from the Greek national stationary stock, whereas Karagkouniko originated from a commercial farm. Interestingly, a significant genetic differentiation of the national stationary stock groups was suggested by principal component analysis, phylogenetic analysis, and admixture and spatial point patterns, with these two breeds being less heterogeneous. This finding highlights the value of stationary stocks towards the maintenance of genetic integrity in indigenous sheep, demonstrating the Greek station’s critical role in the conservation of native sheep breeds. On the other hand, according to the GWAS data analysis, no genetic loci were correlated with reproduction seasonality, emphasizing the MTNR1A gene as the main determinant of the seasonality in native non genetically improved breeds. This study evaluated the effects of serum (egpS) and follicular fluid (egpFF) from estrus guinea pigs as in vitro maturation (IVM) supplements for natural cycle guinea pig oocytes. Two experiments were conducted: the first assessed three IVM media, a commercial medium (CMOM) and two homemade media (HMOM-P and HMOM-S), for oocyte in vitro maturation using 615 bovine oocytes, while the second evaluated the incorporation of 5%, 10%, and 20% egpFF or egpS into IVM media for 1744 guinea pig oocytes. Initially, we optimized the IVM base medium using bovine oocytes to determine the most suitable culture conditions. The results obtained from these experiments served as a critical foundation for subsequent supplementation trials conducted with guinea pig oocytes. The oocytes were cultured in 70 μL drops under controlled atmospheric conditions, and maturation rates were assessed based on cumulus cell expansion and nuclear progression. The HMOM-S medium significantly enhanced cumulus cell expansion (72.5 ± 2.88%) compared with the CMOM medium (57.7 ± 5.33%; p < 0.05). Supplementation with egpFF at 5% (68.8 ± 6.22%), 10% (76.3 ± 5.39%), and 20% (80.9 ± 6.22%) significantly improved both cumulus cell expansion and nuclear progression in high-quality oocytes (types A and B), compared to the control group (43.3 ± 4.82%). Conversely, supplementation with egpS did not have a significant effect on cumulus cell expansion (p > 0.05); however, it notably improved nuclear maturation in low-quality oocytes (type C) at concentrations of 10% and 20% (p < 0.05). This resulted in an overall improvement in maturation outcomes, particularly for oocytes with compromised initial quality. These findings demonstrated that the IVM of guinea pig oocytes using HMOM-S medium was significantly enhanced by the presence of egpFF, whereas egpS supplementation exhibited a less pronounced effect on IVM outcomes. The main objective of this study was to determine the influence of the recipient dairy cows’ breed, lactation number, estrus condition, the type, location and volume of the corpus luteum (CL) and the time of year that the embryo transfer (ET) was performed on the pregnancy rates of a large, fresh in vitro fertilization–embryo transfer program for dairy cows in a commercial herd in China. The recipients were from a herd of dairy cows in Ningxia, a province in northwest China, and we statistically analyzed the data of 495 cows from 2021 to 2023. Cumulus oocyte complexes (COCS) were isolated from follicular fluid obtained through ovum pick-up (OPU) and oocytes were incubated 20–22 h for in vitro maturation (IVM). Embryos were obtained after 10–12 h of in vitro fertilization (IVF) and six days of in vitro culture (IVC). Embryos at the morula or blastocyst stage were transferred to randomly chosen recipients (n = 495). The influence of recipients’ breed (Holstein or other), recipients’ lactation number (heifers or cows), estrus type (natural or synchronized), CL type (homogeneous, CLhom or cavitary, CLcav), CL side (left or right), volume of the CL and season of transfer (spring, autumn or winter) on pregnancy rates were determined. The pregnancy rates were analyzed by binomial logistic regression with IBM SPSS statistics software, version 26. Pregnancy rates after ET to Holstein cows and other breeds were 43.49% and 42.68%, respectively (p > 0.05). Regarding age, pregnancy rates were 45.56% for heifers and 30.77% for cows (p < 0.05). Pregnancy rates following ET during natural and synchronized estrus were 44.41% and 41.5%, respectively (p > 0.05). Pregnancy rates with a left- or right-side CL were 40.18% and 45.65%, respectively (p > 0.05). The pregnancy rates achieved with a CLhom and CLcav were 44.44% and 39.68%, respectively (p < 0.05). The rates obtained in spring, autumn and winter were 49.26%, 46.02% and 34.64%, respectively (p < 0.05). Moreover, it was found that pregnancy rates were higher in recipients with a CL volume measuring greater than 10 cm3 compared with those with a CL volume measuring less than 10 cm3 (p < 0.05). The comparisons showed that recipients’ breed, estrus type or side of the CL had no effect, but the recipients’ lactation number, ET season and the type and volume of the CL have significant effects on pregnancy rates during ET. The combination of estrus synchronization and superovulation (SS) treatments causes alterations in ovarian and endometrial gene expression patterns, resulting in abnormal follicle and oocyte growth, fertilization, and embryo development. However, the impact of combined SS treatments on the transcriptome of the surviving embryos remains unidentified. In this study, we examined gene expression changes in day 6 blastocysts that survived a brief regimen of synchronization treatment combined with superovulation. The sows were included in one of three groups: SS7 group (n = 6), sows were administered Altrenogest (ALT) 7 days from the day of weaning and superovulated with eCG 24 h after the end of ALT treatment and hCG at the onset of estrus; SO group (n = 6), ALT nontreated sows were superovulated with eCG 24 h postweaning and hCG at the onset of estrus; control group (n = 6), weaned sows displaying natural estrus. Six days after insemination, the sows underwent a surgical intervention for embryo collection. Transcriptome analysis was performed on blastocyst-stage embryos with good morphology. Differentially expressed genes (DEGs) between groups were detected using one-way ANOVA with an un-adjusted p-value < 0.05 and a fold change </> 1.5. The effect of SO treatment on the number of altered pathways and DEGs within each pathway was minimal. Only four pathways were disrupted comprising only a total of four altered transcripts, which were not related to reproductive functions or embryonic development. On the other hand, the surviving blastocysts subjected to SS7 treatments exhibited moderate gene expression changes in terms of DEGs and fold changes, with seven pathways disrupted containing a total of 10 transcripts affected. In this case, the up-regulation of certain pathways, such as the metabolic pathway, with two up-regulated genes associated with reproductive functions, namely RDH10 and SPTLC2, may suggest suboptimal embryo quality, while the down-regulation of others, such as the glutathione metabolism pathway, with down-regulated genes related to cellular detoxification of reactive oxygen species, namely GSTK1 and GSTO1, could depress the embryos’ response to oxidative stress, thereby impairing subsequent embryo development. The gene expression changes observed in the present study in SS7 embryos, along with previous reports indicating SS7 can negatively affect fertilization, embryo production, and reproductive tract gene expression, make its use in embryo transfer programs unrecommendable. This study was performed to monitor estrous patterns and, more importantly, changes in anti-Müllerian hormone (AMH) concentrations during the peri-ovulatory period in deslorelin-induced estrous bitches. Healthy anestrous bitches (n = 4) were used. Estrus and ovulation were monitored after deslorelin implantation. Blood samples were collected for analysis of progesterone, estradiol-17ß and AMH concentrations before implantation (day 0) and on days 6, 8, 10, 12, 14, 16, 18, 20 and 22 after implantation. Six days following treatment, all bitches showed estrus signs. Ovulation took place between days 12 and 15. Circulating AMH concentrations varied among bitches from 0.12 to 3.08 ng/mL. However, no significant differences in AMH levels (mean ± SD) were observed between day 0 and days following post-implantation (p > 0.05). There were no significant correlations between AMH and estradiol or AMH and progesterone (p > 0.05). Ultrasonographically, the number of clearly identifiable ovarian follicles was higher before ovulation and the area of ovaries increased after ovulation (p < 0.05). Except for AMH, changes in vaginal cytology, estradiol-17ß and progesterone levels observed in our study were similar to naturally occurring estrus. Large intra- and inter-individual variation in AMH were observed suggesting that AMH is currently not suitable as a canine fertility marker to monitor ovarian response to deslorelin treatment for estrus induction. In this study, we evaluated the effectiveness of using estrogen-induced prolonged luteal function followed by prostaglandin F2 alpha (PGF2α) treatment to synchronize estrus in gilts. On day12 of the estrus cycle (D0 = first day of standing estrus), 52 gilts were assigned at random to two experimental groups: non-treated gilts (CON, n = 22), serving as controls, and prolonged luteal function group (CYP, n = 30), receiving a single treatment with 10 mg of estradiol cypionate intramuscularly Starting on day 12, blood samples were collected for estradiol and progesterone assays. Estrus detection started on day 17. Gilts from the CON group were inseminated at the onset of natural estrus. On day 28 CYP gilts were treated with PGF2α to induce luteolysis and inseminated at the onset of estrus. Gilts were slaughtered 5 d after the last insemination. A single treatment with estradiol cypionate prolonged luteal function in 90% of treated gilts. The duration of the estrous cycle was longer (p < 0.0001) for CYP gilts compared to CON gilts. CYP gilts showed synchronized estrus 3.96 ± 0.19 d after induction of luteolysis. The conception rate was similar (p = 0.10) for CON and CYP gilts. No difference was observed in the embryo recovery rate (p = 0.18) and total number of embryos per female (p = 0.06). The percentage of unfertilized oocytes, fragmented embryos and viable embryos was similar among females from CON and CYP groups (p > 0.05). The treatment of gilts with a single application of 10 mg of estradiol cypionate on day 12 of the estrous cycle was effective in prolonging luteal function and treatment with PGF2α resulted in synchronized estrus. Detection Method of Cow Estrus Behavior in Natural Scenes Based on Improved YOLOv5

Natural breeding scenes have the characteristics of a large number of cows, complex lighting, and a complex background environment, which presents great difficulties for the detection of dairy cow estrus behavior. However, the existing research on cow estrus behavior detection works well in ideal environments with a small number of cows and has a low inference speed and accuracy in natural scenes. To improve the inference speed and accuracy of cow estrus behavior in natural scenes, this paper proposes a cow estrus behavior detection method based on the improved YOLOv5. By improving the YOLOv5 model, it has stronger detection ability for complex environments and multi-scale objects. First, the atrous spatial pyramid pooling (ASPP) module is employed to optimize the YOLOv5l network at multiple scales, which improves the model’s receptive field and ability to perceive global contextual multiscale information. Second, a cow estrus behavior detection model is constructed by combining the channel-attention mechanism and a deep-asymmetric-bottleneck module. Last, K-means clustering is performed to obtain new anchors and complete intersection over union (CIoU) is used to introduce the relative ratio between the predicted box of the cow mounting and the true box of the cow mounting to the regression box prediction function to improve the scale invariance of the model. Multiple cameras were installed in a natural breeding scene containing 200 cows to capture videos of cows mounting. A total of 2668 images were obtained from 115 videos of cow mounting events from the training set, and 675 images were obtained from 29 videos of cow mounting events from the test set. The training set is augmented by the mosaic method to increase the diversity of the dataset. The experimental results show that the average accuracy of the improved model was 94.3%, that the precision was 97.0%, and that the recall was 89.5%, which were higher than those of mainstream models such as YOLOv5, YOLOv3, and Faster R-CNN. The results of the ablation experiments show that ASPP, new anchors, C3SAB, and C3DAB designed in this study can improve the accuracy of the model by 5.9%. Furthermore, when the ASPP dilated convolution was set to (1,5,9,13) and the loss function was set to CIoU, the model had the highest accuracy. The class activation map function was utilized to visualize the model’s feature extraction results and to explain the model’s region of interest for cow images in natural scenes, which demonstrates the effectiveness of the model. Therefore, the model proposed in this study can improve the accuracy of the model for detecting cow estrus events. Additionally, the model’s inference speed was 71 frames per second (fps), which meets the requirements of fast and accurate detection of cow estrus events in natural scenes and all-weather conditions. Hormonal treatments used in the artificial insemination (AI) of sheep can cause several physiological problems that can affect negatively fertility and animal health; however, AI protocols based on the detection of natural estrus offer a more sustainable option and can achieve high fertility. In this study, an AI protocol at natural estrus in organic Churra sheep was performed. In the first phase (AI protocol development), 125 ewes were exocervically inseminated, and their fertility was assessed based on the following factors: number of AI, physiological state, body condition, estrus detection–AI interval, and vaginal fluids in cervix. That protocol was repeated for six consecutive years. In all individuals, fertilities based on the timing of insemination after estrus detection were very high. Lactating ewes produced better results than did dry ewes, which was probably because of the better feeding of the former. In addition, double insemination increased the fertility of ewes whose estrus was detected within 16 h of onset. Body condition and amount of vaginal fluid were correlated with fertility. Exocervical inseminations at natural estrus can produce acceptable fertility and prolificity in Churra ewes. Full article

This research aimed to evaluate whether the administration of hCG at the time of removal of a progesterone device may be effective at inducing estrus and ovulations in sheep during the natural seasonal anestrous, by comparing reproductive outputs (induction and duration of estrus, follicle development, ovulation, ovulation rate, and pregnancy rates) in ewes treated with eCG or only saline solution at the time of CIDR removal. Whereas results demonstrated no response in the control non-treated group, the largest rate of narrowly synchronized estrus signs and ovulations occurred in the eCG-group. The administration of hCG was effective at inducing estrus, promote follicular growth and a delayed yet significant ovulation (>84%) on day 10 after CIDR removal; moreover, an increased embryonic implantation rate was also observed. Moreover, if the hCG ewes remain exposed to active males for some days, said strategy could be adopted, aiming to induce pregnancies by natural mating in a short period of time. Full article

To improve reproductive performance in cattle, the accurate detection of estrus and optimization of insemination relative to ovulation are necessary. However, poor heat detection by farm staff leads to a decreased conception rate, thus inflicting economic damage to the beef and dairy industries. This study aimed to develop monoclonal antibodies (mAb) that can specifically bind to the bovine lactoferrin (bLF) protein, which we have previously demonstrated to be overexpressed in bovine cervical mucus during estrus. Female rats were intraperitoneally immunized with bLF protein as the antigen. Anti-bLF mAbs were then purified by affinity chromatography, and their binding affinity for the bLF antigen was examined using ELISA. We found a high binding affinity between mAbs and bLF. Finally, we developed a rapid bovine heat detection kit using the anti-bLF mAbs that we generated and tested on cervical mucus from 12 cows (estrous synchronization, n = 2; natural cycling, n = 10). We found that the kits accurately detected estrus. Overall, our fabricated heat detection kit based on rat anti-bLF mAbs could pave the way for the development of potent tools for heat detection devices for dairy cattle, thereby preventing economic loss. The aim of this study was to evaluate the possible effect of two doses of hCG (100 and 300 IU) applied at two different times (7 and 14 d) after a fixed-time artificial insemination protocol (FTAI) upon some variables involved in the embryonic implantation rate in goats during the natural deep anestrous season (April, 25° north). The experimental units considered crossbred, multiparous, anovulatory goats (n = 69, Alpine, Saanen, Nubian x Criollo), with average body weight (43.6 ± 5.7 kg) and body condition score (1.86 ± 0.28 units) located in northern–semiarid Mexico (25° N, 103° W). Once the goat’s anestrus status was confirmed, goats were subjected to an estrus induction protocol. Upon estrus induction confirmation, goats (n = 61) were subjected to a FTAI procedure. Immediately after the FTAI, the goats were randomly distributed to five experimental groups: (1). G100-7 (n = 13) 100 IU, hCG 7 d post-FTAI, (2). G100-14 (n = 12) 100 IU hCG, 14 d post-FTAI, (3). G300-7 (n = 12) 300 IU, hCG, 7 d post-FTAI, (4). G300-14 (n = 12) 300 IU hCG 14 d post-FTAI, and (5). Control group, CONT (n = 12) 0.5 mL saline, 7 and 14 d post-FTAI. The response variables conception rate (39.36 ± 0.23), fertility rate (27.96%), prolificacy rate (1.1 ± 0.29 kids), ovulation rate (0.74 ± 0.20 corpus luteum) corpus luteum diameter (10.15 ± 0.59 mm), embryo number (1.58 ± 0.20), and embryo implantation rate (48.96%), did not differ between treatments. However, while the variables fecundity rate (67%), embryo efficiency index-1 (33.99 ± 0.20%), and embryo efficiency index-2 (27.94 ± 0.30%) were favored by the G300-14 treatment, the corpus luteum area was favored (p < 0.05) by both G300-7 (113.30 ± 0.19 mm2) and G300-14 (103.04 ± 0.17 mm2). Such reproductive strategy emerges as an interesting approach, not only to enhance the out-of-season reproductive outcomes, but also to boost one of the main rulers defining the global reproductive efficiency of a heard, namely, the embryo implantation efficiency.
