Protein and Amino Acid Supplementation Among Recreational Gym Goers and Associated Factors—An Exploratory Study

Objective: The present study investigated the relationship between protein and amino acid supplementation and various associated aspects among recreational gym goers at 2 gymnasiums in Oradea (Romania). Methods: A total of 165 gym goers (110 men and 55 women, most of them 18–30 years old) with high educational levels were included in the present study, which was conducted as face-to-face interviews. Results: Participants were divided into 4 groups: protein supplement users (PSUs, 42/165), creatine supplement users (CSUs, 38/165), L-carnitine supplement users (LcSUs, 37/165), and protein + creatine + L-carnitine supplement users (PCLcSUs, 48/165). Most consumers were young (18–30 years) and preferred the triple combination. Females consumed PS and CS (38.2% and 34.5%, respectively), while the most-used NSs by males were PCLcS (36.4%) and LcS (27.3%). Obese gym goers opted for LcS consumption (r = 0.999, p < 0.05). Creatine and L-carnitine were consumed for force training (65.79 and 62.16%), while PCLcS and PS were used in cardio + force and force training in equal measures (42.86 and 47.92%, respectively). Most PSUs were gym goers for 7–12 months and more than 1 year (r = 0.999 and r = 0.952, respectively, p < 0.05), while PCLcSUs had a training frequency of at least 5 times a week (r = 0.968, p < 0.05). Muscle mass growth was the primary training focus for all NS users (57.89%), followed by muscular tonus (40.54%, p < 0.05). Almost 30% of one-only NS users reported various side effects, whereas all PCLcSUs claimed side effects (p < 0.05). Conclusions: Age and gender were key factors in diet type, training type, frequency, duration, scope, NS type, and dose intake. The frequency of side effects substantially depended on the kind of NS and the dose consumed. The present study’s results highlight the need for health professionals’ advice and monitoring in personalized diets and protein and amino acid supplementation in recreational gym goers. Full article

Background and Objectives: The effects of face yoga, which continues to be popular as an anti-aging technique, on facial muscles in relation to aging represent an area of interest. The aim of this study was to investigate the effect of 8 weeks of intensive face yoga on facial muscles’ tonus, stiffness, and elasticity in middle-aged women. Materials and Methods: Twelve female participants with a mean age of 49.75 ± 3.76 were included in this pre-experimental clinical trial. Face yoga was applied to the participants for 8 weeks, 2 days a week face-to-face, and 5 days a week as a home program. At the beginning and the end of 8 weeks, the tonus, stiffness, and elasticity of the participants’ facial muscles were evaluated with the Myoton®PRO device. Results: Following the face yoga program, the tonus and stiffness of the frontalis (p = 0.008, p = 0.002), corrugator supercilii (p = 0.008, p = 0.019), orbicularis oculi (p = 0.023, p = 0.034), and orbicularis oris (p = 0.007, p = 0.015) muscles decreased significantly, while the tonus and stiffness of the buccinator (p = 0.008, p = 0.002) and digastric (p = 0.008, p = 0.023) muscles increased. Elasticity values increased in all evaluated facial muscles (p = 0.045, p = 0.045, p = 0.034, p = 0.023, p = 0.028, p = 0.005, respectively). Conclusions: The results reveal that face yoga has different effects depending on the physiological structure and function of the muscles and positively affects connective tissue. Future studies should reproduce the results presented here to further our understanding of the effects of face yoga. Full article

Pathologies in neck and masticatory muscles affect muscle tone and biomechanical and viscoelastic properties, necessitating precise assessment for treatment. This study evaluated the impact of two guided interventions—relaxing exercises targeting the neck and masticatory muscles (“Exercise”) and heavy chewing using six chewing gums (“Chewing”)—on the masseter and upper trapezius muscles. Twelve participants (aged 19–40 years) underwent myotonometric assessment pre- and post-intervention, measuring tone, stiffness, decrement, relaxation time, and creep. The results showed significant changes in the masseter muscle after exercise, with increased stiffness (14.46%, p < 0.001) and tone (7.03%, p < 0.001) but decreased creep (−9.71%, p < 0.001) and relaxation time (−11.36%, p < 0.001). Conversely, chewing decreased stiffness (−8.82%, p < 0.001) and tone (−5.53%, p < 0.001), while it increased creep (9.68%, p < 0.001) and relaxation time (9.98%, p < 0.001). In the trapezius muscles, tone decreased after both interventions (Exercise: −7.65%, p < 0.001; Chewing: −1.06%, p = 0.003), while relaxation increased (Exercise: 1.78%, p < 0.001; Chewing: 2.82%, p < 0.001). These findings reveal the distinct effects of exercise and chewing on muscle properties, emphasising the complexity of their therapeutic potential and the need for further investigation. Full article

Recombinant human growth hormone therapy (rhGH) has been widely accepted as the safe treatment for short stature in children with such genetic syndromes as Prader–Willi syndrome and Turner or Noonan syndrome. Some patients with short stature and rare genetic syndromes are treated with rhGH as growth hormone-deficient individuals or as children born small for their gestational age. After years of experience with this therapy in syndromic short stature, it has been proved that there are some aspects of long-term rhGH treatment beyond growth promotion, which can justify rhGH use in these individuals. This paper summarizes the data of a literature review of the effects of rhGH treatment beyond growth promotion in selected genetic syndromes. We chose three of the most common syndromes, Prader–Willi, Turner, and Noonan, in which rhGH treatment is indicated, and three rarer syndromes, Silver–Russel, Kabuki, and Duchenne muscular dystrophy, in which rhGH treatment is not widely indicated. Many studies have shown a significant impact of rhGH therapy on body composition, resting energy expenditure, insulin sensitivity, muscle tonus, motor function, and mental and behavioral development. Growth promotion is undoubtedly the primary benefit of rhGH therapy; nevertheless, especially with genetic syndromes, the additional effects should also be considered as important indications for this treatment. There is a growing body of evidence that ER stress and the unfolded protein response (UPR) play a key role in numerous diseases. Impaired liver perfusion and ER stress often accompany each other in liver diseases. However, the exact impact of ER stress and UPR on the hepatic perfusion is not fully understood. The aim of this study was to disclose the effect of ER stress and UPR on the size of liver vessels and on the levels of Ca2+ and nitric oxide (NO), critical regulators of vascular tonus. This study was carried out in precisely cut liver tissue slices. Confocal microscopy was used to create 3D images of vessels. NO levels were determined either using either laser scan microscopy (LSM) in cells or by NO-analyser in medium. Ca2+ levels were analysed by LSM. We show that tunicamycin, an inducer of ER stress, acts similarly with vasodilator acetylcholine. Both exert a similar effect on the NO and Ca2+ levels; both induce significant vasodilation. Notably, this vasodilative effect persisted despite individual inhibition of UPR pathways—ATF-6, PERK, and IRE1—despite confirming the activation of UPR. Experiments with HUVEC cells showed that elevated NO levels did not result from endothelial NO synthase (eNOS) activation. Our study suggests that tunicamycin-mediated ER stress induces liver vessel vasodilation in an NO-dependent manner, which is mediated by intracellular nitrodilator-activatable NO store (NANOS) in smooth muscle cells rather than by eNOS. There is limited evidence comparing the effects of manual and instrumented-assisted manipulations among adults with neck pain. Our purpose was to determine the effects of a multisession regime of manual and instrument-assisted cervical manipulation on pain, disability, perception of change, and muscle properties in subjects with nonspecific neck pain. We conducted a double-blind, randomized, placebo-controlled study in 32 subjects with nonspecific neck pain. Two groups received three sessions of cervical (C3/C4) manipulation, one group manual and the other instrument-assisted, a third group received three sessions of sham manipulation, and a fourth group served as a control. Self-reported pain, pressure pain thresholds, neck disability, patient perception of change, and properties (tonus, stiffness, and elasticity) of the upper trapezius and biceps brachii were assessed at baseline, immediately after the first session and 15 days after the end of the intervention. After the end of the intervention, the percentage of changes in the visual analogue scale score, Neck Disability Index, and Patient Global Perception of Change score were significantly higher in the manual group in comparison with the other groups (p < 0.05). No between-group differences were observed in the percentage of changes in tonus, stiffness, and elasticity of the four muscles at the end of the intervention. We concluded that three sessions of C3/C4 manual manipulation improved pain and disability in subjects with nonspecific neck pain. Full article

Facial hemiplegia happens when the seventh cranial nerve is inflamed, causing a dysfunction of the facial nerve in specific regions. This case report brings a complex case of facial hemiplegia, a non-temporary lesion caused by a traumatic accident, which had a more conservative approach, treating the patient with botulinum toxin. After explanation of treatment outcomes, the patient favored treatment on a unilateral side with botulinum toxin applied locally to the muscles. It was applied on her left side, in order to change the muscles tonus and improve the esthetic. The patient adhered to immediate and short-term instructions following the procedure, including movement limitation and skin exposure avoidance. At 2 weeks, the patient returned to follow-up, and the result was checked. After around 6-month follow-up, the patient was reassessed, and a new application was done. The patient tried to contract the procerus and corrugator muscles which were treated, and periorbicular region that was corrected. After contracting the frontal muscle, a satisfactory result was also seen in the frontal area. While limited to a single case presentation, botulinum toxin may be an effective short-term tool for treatment of facial hemiplegia to establish an effective esthetic result. Full article

Facial nerve paralysis interferes with mimetic muscle function. To reconstruct natural facial movement, free muscle flaps are transplanted as new muscles. However, it is difficult to maintain resting tonus. A dual innervation technique in which other nerves such as the hypoglossal nerve or contralateral facial nerve are added is often applied. Using 10-week-old rats (n = 10), the masseteric and hypoglossal nerves were cut, and the distal stump of the masseteric nerve and the proximal stump of the hypoglossal nerve were then sutured (suture group). In the other group, the masseteric nerve was cut and cauterized (cut group). Immunohistochemistry and microarray were performed on the extracted masseter muscle. The immunohistochemistry results suggested that the muscles in the suture group obtained oxidative characteristics. The microarray showed the genes involved in mitochondrial function, including Perm1. In summary, our data support the validity of the dualinnervation technique for facial paralysis treatment. Evidence has supported the use of leucine as a promising agent for the maintenance of muscle tone. This study aimed to assess the combined effect of leucine and cream-based lactic acid (novel cosmetic product), associated with the use of surface electrical stimulation to improve contour and facial tonus in women. A total of 23 women were randomly allocated into two groups: Experimental (EG)—use of the leucine-based cream and lactic acid + electrostimulation for facial toning (mean intensity 13 Hz and protocol in progression); and placebo (GP)—use of the placebo cream (without the addition of leucine and lactic acid) + stimulation with the same protocol as the EG. Each group used their cream daily and underwent the intervention protocol three x/week with stimulation for 40 min, for a total of 8 weeks. Three main outcomes were reported: angular variation of facial contour by means of photogrammetry, muscle tone through the electromyographic activity of the masseter and zygomatic muscles during rest and functional tasks of biting and smiling. A significant effect of the intervention and between the groups was obtained for the experimental group against the placebo group for facial contour and muscle tone. An increased muscular activity of the masseter (average 28%) when smiling, and a reduction of zygomatic activity (in average 41%) when biting were found. The use of cream containing leucine and lactic acid combined with electrostimulation contributes to the improvement of facial contour and muscle tone when biting and smiling. Full article

During the COVID-19 pandemic, the use of masks has been recommended as a containment measure. The mask is a hindrance to normal breathing that causes discomfort. This could put more work on the respiratory accessory muscles, and, consequently, these muscles could see their tone increase. For this reason, during this clinical trial (registered in clincaltrials.gov, number: NCT04789603), it was observed whether the use of the mask produced changes in the distance traveled, in the heart rate, in the oxygenometry, in the self-perceived dyspnea and in the tone of accessory respiratory muscles during a 6-min walk test (6MWT). Fifty healthy volunteers were recruited and carried out the 6MWT on three occasions. They carried out the 6MWT in various situations: using an FFP2/N95 mask, using a surgical mask, and without using a mask. The distance walked, the heart rate, the oxygen therapy, the tone of the accessory respiratory muscles, and the self-perceived dyspnea were recorded in each situation. Significant differences were found between the three situations in terms of self-perceived dyspnea FFP2/N95 > surgical mask > no mask. However, there are no differences between the experimental situations during the 6MWT in terms of distance travelled, heart rate, oxygenometry or respiratory muscle tone. Full article

In children, the quality and muscle function are altered in many pathologic conditions, including temporomandibular disorders. Although several methods have been used to evaluate muscle tonus, none became a golden standard. Moreover, the masseter muscle characteristics in children have not been investigated to date. This study aimed to measure the stiffness of the masseter muscle using shear-wave elastography in healthy children. We enrolled 30 healthy children (mean age 10.87 ± 3.38 years). The stiffness of masseter muscles was measured with shear wave elastography. Stiffness for the total sample was 6.37 ± 0.77 kPa. A comparison of the measurements did not show significant differences between the right and the left masseter muscles (left—6.47 ± 0.78 kPa; right—6.24 ± 0.76 kPa; p = 0.3546). A significant difference was seen between boys and girls (boys—5.94 ± 0.50 kPa; girls—6.63 ± 0.80; p = 0.0006). Shear-wave elastography is a promising diagnostic tool. It may help to detect changes in the stiffness of the masseter muscle and draw attention to pathological processes within the jaw muscles. Directions for further research shall include determining stiffness values in pathological conditions and the impact of biological and functional factors on the stiffness of the masseter muscle. Background: Axial spondyloarthritis (axSpA) affects spinal muscles, due to inflammation and structural damage. The mechanical properties of the muscles, such as tone or stiffness, could be altered in axSpA. The aim of this work is to analyze the mechanical properties of cervical and lumbar spine muscles in axSpA patients and their relationship with metrology measures, function, disease activity, structural damage and quality of life. Methods: axSpA patients and age/gender/BMI matched healthy controls were recruited. The muscle mechanical properties (MMPs), such as tone or frequency, stiffness, decrement (linear elastic properties), relaxation and creep (viscoelastic properties), of cervical (semispinalis capitis) and lumbar (erector spinae) muscles were bilaterally measured at rest using myotonometry. Additionally, conventional metrology, BASMI (metrology index), BASDAI (disease activity index), mSASSS (radiological structural damage index) and SF-12 (health-related quality of life questionnaire) were used in the axSpA group. Between-groups comparison, intra-group correlations and multivariable regression analyses were performed to achieve the study aims. Results: Thirty-four axSpA patients (mean age: 46.21 ± 8.53 y) and 34 healthy volunteers (mean age: 43.97 ± 8.49 y) were recruited. Both in cervical and lumbar spine, linear elastic parameters were significantly higher in axSpA patients in comparison with controls, while viscoelastic parameters were significantly lower. Lumbar muscle frequency, stiffness, relaxation, creep and cervical muscle elasticity were fair to strongly correlated (|0.346| < r < |0.774|) with age, functional status, activity of disease, structural damage and quality of life in axSpA patients. Furthermore, moderate to good fitted multivariate models (0.328 < R2 < 0.697) were obtained combining age, conventional metrology, activity of the disease and function for the estimation of cervical and lumbar MMPs. Conclusion: Mechanical properties of spinal muscles of axSpA patients differ from controls. Lumbar and cervical muscles exhibit greater linear elastic properties and lower viscoelastic properties, which are related with age, clinical and psychophysiological features of axSpA. Full article

Background: Pre-competition massage is usually used to improve athletic performance and reduce risk of injury. Despite its usual use, the effects of pre-competition massage on neuromuscular function have barely been studied. The aim of this study is to evaluate the effects of the pre-competition massage over the gastrocnemius neuromuscular function. Method: The study is a quasi-experimental clinical trial thirty healthy athletes were enrolled in the study. Subjects received an intervention in one leg (experimental), consisting of a massage, and no intervention in the opposite leg (control). From all values of neuromuscular function, the following were analyzed: contraction time (Tc) and maximal displacement (Dm) by tensiomyography, and stiffness and tone by myotonometry. Results: Main effects of pre-competition massage on neuromuscular function include a significant (p < 0.05) increase in Tc and Dm variables, as well as a reduction in stiffness and tone. Conclusion: Data shows an increase in Tc and maximal radial displacement (Dm) variables, as well as a reduction in stiffness and tone. More quality studies are needed to draw clear conclusions about the effects of pre-competition massage. Full article

The pathophysiology underlying painful intercourse is challenging due to variability in manifestations of vulvar pain hypersensitivity. This study aimed to address whether the anatomic location of vestibular-provoked pain is associated with specific, possible causes for insertional dyspareunia. Women (n = 113) were assessed for “anterior” and “posterior” provoked vestibular pain based on vestibular tenderness location evoked by a Q-tip test. Pain evoked during vaginal intercourse, pain evoked by deep muscle palpation, and the severity of pelvic floor muscles hypertonicity were assessed. The role of potential confounders (vestibular atrophy, umbilical pain hypersensitivity, hyper-tonus of pelvic floor muscles and presence of a constricting hymenal-ring) was analyzed to define whether distinctive subgroups exist. Q-tip stimulation provoked posterior vestibular tenderness in all participants (6.20 ± 1.9). However, 41 patients also demonstrated anterior vestibular pain hypersensitivity (5.24 ± 1.5). This group (circumferential vestibular tenderness), presented with either vestibular atrophy associated with hormonal contraception use (n = 21), or augmented tactile umbilical-hypersensitivity (n = 20). The posterior-only vestibular tenderness group included either women with a constricting hymenal-ring (n = 37) or with pelvic floor hypertonicity (n = 35). Interestingly, pain evoked during intercourse did not differ between groups. Linear regression analyses revealed augmented coital pain experience, umbilical-hypersensitivity and vestibular atrophy predicted enhanced pain hypersensitivity evoked at the anterior, but not at the posterior vestibule (R = 0.497, p < 0.001). Distinguishing tactile hypersensitivity in anterior and posterior vestibule and recognition of additional nociceptive markers can lead to clinical subgrouping. Full article

Ultraviolet (UV) filters are chemicals widely used in personal care products (PCPs). Due to their effect as endocrine disruptor compounds (EDCs), the toxicity of UV filters is a current concern for human health. EDC exposure may be correlated to cardiovascular diseases (CVD), but to our knowledge, no studies assessed the UV filters effects as human EDCs at the vascular level. Octylmethoxycinnamate (OMC) is the world’s most widely used UV-B filter, present in more than 90% of PCPs. Due to its demonstrated multiple hormonal activities in animal models, this substance is also suspected to be a human EDC. The purpose of this study was to assess the rapid/short-term effects of OMC on arterial tonus and analyse its mode of action (MOA). Using human umbilical arteries, the endocrine effects of OMC were evaluated in in vitro (cellular and organ) experiments by planar cell surface area (PCSA) and organ bath, respectively. Our data show that OMC induces a rapid/short-term smooth muscle relaxation acting through an endothelium-independent MOA, which seems to be shared with oestrogens, involving an activation of soluble guanylyl cyclase (sGC) that increases the cyclic guanosine monophosphate (cGMP) intracellular levels and an inhibition of L-type voltage-operated Ca2+ channels (L-Type VOCC).
