Mycotoxins are toxic secondary metabolites produced by filamentous fungi which are commonly found as natural contaminants in food and feed worldwide. In recent years, aquaculture research has increasingly focused on changing fish feed by replacing traditional protein sources with plant-based and insect ingredients to promote sustainability. However, this shift has raised concerns about mycotoxin contamination in farmed fish, fish products, and processing by-products. As a result, the pursuit of sustainable aquaculture practices may inadvertently increase the risk of mycotoxin exposure. To date, studies on freshwater fish have focused primarily on regulated mycotoxins, and the findings have demonstrated their occurrence in muscle, liver, intestine, ovaries, and hepatopancreas. Most studies have investigated aflatoxin B1, and its presence has been confirmed in the muscle tissue of several fish species. In marine fish, research has encompassed a broader spectrum of mycotoxins, including emerging and masked forms, across multiple tissues and organs. However, across various studies, conflicting results have been reported regarding the occurrence of emerging mycotoxins, particularly enniatins and beauvericin. This paper reviews current research on mycotoxin contamination in farmed fish, summarising detected levels across freshwater and marine species and in derived products, and also discusses future perspectives on mycotoxin risks in sustainable aquaculture. Mycotoxins are toxic secondary metabolites produced by various fungal species, posing significant food safety concerns due to their health impacts and economic burden. Accurate structural elucidation and quantitative analysis are essential for effective risk assessment and regulatory control. This review highlights recent advances in the application of nuclear magnetic resonance (NMR) spectroscopy for the structural and quantitative analysis of major mycotoxins, including aflatoxins, ochratoxins, fumonisins, trichothecenes, and zearalenone. One- and two-dimensional NMR techniques enable precise molecular characterization, positional isomer identification, including modified forms such as masked or conjugated mycotoxins, and toxicity-related molecular interaction investigation. NMR spectroscopy offers superior structural resolution, high reproducibility, and nondestructive analysis, making it invaluable in mycotoxin research. Quantitative NMR spectroscopy has emerged as a robust and accurate method for determining the absolute concentration and purity of mycotoxins, without requiring analyte-specific reference standards, an advantage particularly important for modified toxins lacking commercially available standards. The integration of NMR-based approaches strengthens analytical reliability, supports reference material development, and contributes to enhanced food safety assessment. This review also discusses ongoing analytical challenges and future directions, including the application of artificial intelligence to improve the automation and interpretation of NMR data in mycotoxin research. Plant-based materials are increasingly being used as ingredients of aquaculture feed. These materials are prone to mycotoxin contamination, as mycotoxigenic fungi infest crop plants in the field and agricultural products during storage. As mycotoxins can cause toxic effects in aquatic animals, their occurrence in feedstuffs should be monitored. To this end, we performed an extensive global survey of mycotoxin contamination in aquaculture feed and plant-based feed raw materials. We collected samples of compound feed for fish (n = 226) and shrimps (n = 61), maize (n = 3448), maize DDGS (n = 149), wheat (n = 1578), soybean (n = 428), and rice (n = 65). We analyzed concentrations of 51 mycotoxins, emerging mycotoxins, masked mycotoxins, and mycotoxin metabolites. Mycotoxins were almost ubiquitously present in compound feed, as >90% of samples were contaminated with at least one mycotoxin. Feed raw materials exhibited distinct mycotoxin occurrence patterns consistent with known susceptibility to fungal pathogens and with their production process. Unsafe concentrations of aflatoxin B1 exceeding the EU maximum level were detected in 7.2% of fish feed samples. While most feedstuffs complied with EU guidance values for deoxynivalenol, zearalenone, and fumonisins, a comparison of detected concentrations with dietary concentrations reported to cause adverse effects in fish and shrimps in published studies indicated that significant fractions of samples contained potentially harmful levels of these mycotoxins. In addition to regulated mycotoxins, several emerging mycotoxins (e.g., enniatins, beauvericin, alternariol, moniliformin) were prevalent. Feed was frequently co-contaminated with multiple mycotoxins indicating a risk of combined effects. In conclusion, mycotoxin contamination was common in aquaculture feed and fractions of samples were contaminated with mycotoxin levels known to exert adverse effects in aquaculture species. Results of this survey highlight the necessity for targeted studies on the effects of frequently detected mycotoxin mixtures and emerging mycotoxins in fish and shrimp. Full article

Deoxynivalenol (DON) is an important mycotoxin produced by Fusarium spp., typically found in cereals, which has garnered considerable research attention. However, the risk assessment of DON exposure to muti-cereal is partial and biased, especially lacking the evaluation of different coarse grains. In this study, we synthesized and compared the presence of the total deoxynivalenol (free, acetylated, and masked) of a total of 372 grain samples (17 different types) based on high-performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS), as well as assessed the chronic and acute risks of total DON exposure in the Chinese population. DON was found at the highest frequency with an occurrence of 85.8% (319/372), followed by D3G at 17.2% (64/372). In total, 88.7% (330/372) of the grains were co-contaminated with mycotoxins. The DON and D3G contamination correlation coefficient was 0.68 in wheat flour. Moreover, different DON contamination levels were found in black beans (133.5 µg/kg), soybeans (128.7 µg/kg), and black rice (122.1 µg/kg). The DON/D3G/15A/tDON contamination level was significant differently among different coarse grains. Notably, the Monte Carlo model showed that 3.2–5.9% of adolescents consuming wheat flour and noodles suffered a chronic tDON risk. Full article

Mycotoxins are well-known secondary metabolites produced by several fungi that grow and occur in different crops during both pre-harvest and post-harvest conditions. The contamination and occurrence of mycotoxins currently represent some of the major issues in the entire agri-food system. The quantification of mycotoxins in different feeds and foodstuffs is extremely difficult because of the low concentration ranges; therefore, both sample collection and preparation are essential to providing accurate detection and reliable quantification. Currently, several analytical methods are available for the detection of mycotoxins in both feed and food products, and liquid chromatography coupled with high-resolution mass spectrometry (LC-HRMS) represents the most reliable instrumental approach. In particular, the fast development of high-throughput methods has made it possible to screen and analyze, in the same analytical run and with high accuracy, multiple mycotoxins, such as those regulated, masked, or modified, and emerging ones. Therefore, the aim of this review is to provide an overview of the state of the art of mycotoxins occurrence, health-related concerns, and analyses, discussing the need to perform multi-screening approaches combined with omics technologies to simultaneously analyze several mycotoxins in different feed and food matrices. This approach is expected to provide more comprehensive information about the profile and distribution of emerging mycotoxins, thus enhancing the understanding of their co-occurrence and impact on the entire production chain. Trichothecenes produced by Fusarium species are commonly detected in oats. However, the ratios of the concentrations of free trichothecenes and their conjugates and how they are impacted by different interacting environmental conditions are not well documented. This study aims to examine the effect of water activity (0.95 and 0.98 aw) and temperature (20 and 25 °C) stress on the production of T-2 and HT-2 toxins, deoxynivalenol and their conjugates, as well as diacetoxyscirpenol (DAS). Multiple mycotoxins were detected using liquid chromatography–tandem mass spectrometry from 64 contaminated oat samples. The highest concentrations of HT-2-glucoside (HT-2-Glc) were observed at 0.98 aw and 20 °C, and were higher than other type A trichothecenes in the natural oats’ treatments. However, no statistical differences were found between the mean concentrations of HT-2-Glc and HT-2 toxins in all storage conditions analysed. DAS concentrations were generally low and highest at 0.95 aw and 20 °C, while deoxynivalenol-3-glucoside levels were highest at 0.98 aw and 20 °C in the naturally contaminated oats. Emerging mycotoxins such as beauvericin, moniliformin, and enniatins mostly increased with a rise in water activity and temperature in the naturally contaminated oats treatment. This study reinforces the importance of storage aw and temperature conditions in the high risk of free and modified toxin contamination of small cereal grains. Full article

The presence of mycotoxins and their masked forms in chicken feed poses a significant threat to both productivity and health. This review examines the multifaceted impacts of mycotoxins on various aspects of chicken well-being, encompassing feed efficiency, growth, immunity, antioxidants, blood biochemistry, and internal organs. Mycotoxins, toxic substances produced by fungi, can exert detrimental effects even at low levels of contamination. The hidden or masked forms of mycotoxins further complicate the situation, as they are not easily detected by conventional methods but can be converted into their toxic forms during digestion. Consequently, chickens are exposed to mycotoxin-related risks despite apparently low mycotoxin levels. The consequences of mycotoxin exposure in chickens include reduced feed efficiency, compromised growth rates, impaired immune function, altered antioxidant levels, disturbances in blood biochemical parameters, and adverse effects on internal organs. To mitigate these impacts, effective management strategies are essential, such as routine monitoring of feed ingredients and finished feeds, adherence to proper storage practices, and the implementation of feed detoxification methods and mycotoxin binders. Raising awareness of these hidden hazards is crucial for safeguarding chicken productivity and health. Full article

Mycotoxins, as natural products of molds, are often unavoidable contaminants of food and feed, to which the increasingly evident climate changes contribute a large part. The consequences are more or less severe and range from economic losses to worrying health problems to a fatal outcome. One of the best preventive approaches is regular monitoring of food and feed for the presence of mycotoxins. However, even under conditions of frequent, comprehensive, and conscientious controls, the desired protection goal may not be achieved. In fact, it often happens that, despite favorable analytical results that do not indicate high mycotoxin contamination, symptoms of their presence occur in practice. The most common reasons for this are the simultaneous presence of several different mycotoxins whose individual content does not exceed the detectable or prescribed values and/or the alteration of the form of the mycotoxin, which renders it impossible to be analytically determined using routine methods. When such contaminated foods enter a living organism, toxic effects occur. This article aims to shed light on the above problems in order to pay more attention to them, work to reduce their impact, and, eventually, overcome them. Full article

Mycotoxins are frequent toxic contaminants in foods and beverages, causing a significant health threat. Interactions of mycotoxins with biotransformation enzymes (e.g., cytochrome P450 enzymes, sulfotransferases, and uridine 5′-diphospho-glucuronosyltransferases) may be important due to their possible detoxification or toxic activation during enzymatic processes. Furthermore, mycotoxin-induced enzyme inhibition may affect the biotransformation of other molecules. A recent study described the strong inhibitory effects of alternariol and alternariol-9-methylether on the xanthine oxidase (XO) enzyme. Therefore, we aimed to test the impacts of 31 mycotoxins (including the masked/modified derivatives of alternariol and alternariol-9-methylether) on XO-catalyzed uric acid formation. Besides the in vitro enzyme incubation assays, mycotoxin depletion experiments and modeling studies were performed. Among the mycotoxins tested, alternariol, alternariol-3-sulfate, and α-zearalenol showed moderate inhibitory actions on the enzyme, representing more than tenfold weaker impacts compared with the positive control inhibitor allopurinol. In mycotoxin depletion assays, XO did not affect the concentrations of alternariol, alternariol-3-sulfate, and α-zearalenol in the incubates; thus, these compounds are inhibitors but not substrates of the enzyme. Experimental data and modeling studies suggest the reversible, allosteric inhibition of XO by these three mycotoxins. Our results help the better understanding of the toxicokinetic interactions of mycotoxins. Full article

Small grain cereals are frequently infected with mycotoxigenic Fusarium fungi. Oats have a particularly high risk of contamination with type A trichothecene mycotoxins; their glucoside conjugates have also been reported. Agronomy practices, cereal variety and weather conditions have been suggested to play a role in Fusarium infection in oats. The current study investigates concentrations of free and conjugated Fusarium mycotoxins in organic and conventional oats grown in Scotland. In 2019, 33 milling oat samples (12 organic, 21 conventional) were collected from farmers across Scotland, together with sample questionnaires. Samples were analysed for 12 mycotoxins (type A trichothecenes T-2-toxin, HT-2-toxin, diacetoxyscirpenol; type B trichothecenes deoxynivalenol, nivalenol; zearalenone and their respective glucosides) using LC-MS/MS. The prevalence of type A trichothecenes T-2/HT-2 was very high (100% of conventional oats, 83% of organic oats), whereas type B trichothecenes were less prevalent, and zearalenone was rarely found. T-2-glucoside and deoxynivalenol-glucoside were the most prevalent conjugated mycotoxins (36 and 33%), and co-occurrence between type A and B trichothecenes were frequently observed (66% of samples). Organic oats were contaminated at significantly lower average concentrations than conventional oats, whereas the effect of weather parameters were not statistically significant. Our results clearly indicate that free and conjugated T-2- and HT-2-toxins pose a major risk to Scottish oat production and that organic production and crop rotation offer potential mitigation strategies. Fusarium head blight (FHB) is a disease of small grains caused by the fungus Fusarium graminearum. In this study, we explored the use of hyperspectral imaging (HSI) to evaluate the damage caused by FHB in wheat kernels. We evaluated the use of HSI for disease classification and correlated the damage with the mycotoxin deoxynivalenol (DON) content. Computational analyses were carried out to determine which machine learning methods had the best accuracy to classify different levels of damage in wheat kernel samples. The classes of samples were based on the DON content obtained from Gas Chromatography–Mass Spectrometry (GC-MS). We found that G-Boost, an ensemble method, showed the best performance with 97% accuracy in classifying wheat kernels into different severity levels. Mask R-CNN, an instance segmentation method, was used to segment the wheat kernels from HSI data. The regions of interest (ROIs) obtained from Mask R-CNN achieved a high mAP of 0.97. The results from Mask R-CNN, when combined with the classification method, were able to correlate HSI data with the DON concentration in small grains with an R2 of 0.75. Our results show the potential of HSI to quantify DON in wheat kernels in commercial settings such as elevators or mills. Full article

Mycotoxins are toxic metabolites of molds. Chronic exposure to alternariol, zearalenone, and their metabolites may cause the development of endocrine-disrupting and carcinogenic effects. Alternariol-3-glucoside (AG) and alternariol-9-monomethylether-3-glucoside (AMG) are masked derivatives of alternariol. Furthermore, in mammals, zearalenone-14-glucuronide (Z14Glr) is one of the most dominant metabolites of zearalenone. In this study, we examined serum albumins and cyclodextrins (CDs) as potential binders of AG, AMG, and Z14Glr. The most important results/conclusions were as follows: AG and AMG formed moderately strong complexes with human, bovine, porcine, and rat albumins. Rat albumin bound Z14Glr approximately 4.5-fold stronger than human albumin. AG–albumin and Z14Glr–albumin interactions were barely influenced by the environmental pH, while the formation of AMG–albumin complexes was strongly favored by alkaline conditions. Among the mycotoxin–CD complexes examined, AMG–sugammadex interaction proved to be the most stable. CD bead polymers decreased the mycotoxin content of aqueous solutions, with moderate removal of AG and AMG, while weak extraction of Z14Glr was observed. In conclusion, rat albumin is a relatively strong binder of Z14Glr, and albumin can form highly stable complexes with AMG at pH 8.5. Fusarium graminearum and Fusarium meridionale are primary contaminants of barley, capable of producing several mycotoxins, mainly type B trichothecenes and zearalenone. Cold plasma decontamination has been gaining prominence, seeking to control the fungal and mycotoxin contamination of food and feed and to improve product quality. To reach this objective, the present study was divided into two parts. In the first part, F. meridionale and F. graminearum strains were exposed to gliding arc plasma jet (GAPJ). Cell viability tests showed the inactivation of F. meridionale after 15-min treatment, whereas F. graminearum showed to be resistant. In the second part, barley grains were treated by GAPJ for 10, 20, and 30 min, demonstrating a reduction of about 2 log CFU/g of the barley’s mycobiota, composed of yeasts, strains belonging to the F. graminearum species complex, Alternaria, and Aspergillus. A decrease in DON levels (up to 89%) was observed after exposure for 20 min. However, an increase in the toxin Deoxynivalenol-3-glucoside (D3G) was observed in barley grains, indicating a conversion of DON to D3G. Full article

Three-Year Survey of Fusarium Multi-Metabolites/Mycotoxins Contamination in Wheat Samples in Potentially Epidemic FHB Conditions

Fusarium head blight (FHB) is a fungal disease of cereals including wheat, which results in significant economic losses and reductions in grain quality. Additionally, the presence of Fusarium spp. results in productions of mycotoxins/metabolites, some of which are toxic in low concentrations. The liquid chromatography with tandem mass spectrometry (LC-MS/MS) method was applied to 216 wheat samples from field conditions diseased with FHB. Data obtained show that out of 28 metabolites detected, deoxynivalenol (DON), deoxynivalenol-3-glucoside (D3G), enniatin B (ENN B), enniatin B1 (ENN B1), culmorin, 15-hydroxyculmorin, and aurofusarin were the most prevalent mycotoxins/metabolites over three years (2014–2016). In 2014–2016, 100, 100 and 96% of the samples were contaminated with zearalenone (ZEN). Of the masked mycotoxins, D3G occurred at a high incidence level of 100% in all three investigated years. Among emerging mycotoxins, moniliformin (MON), beauvericin (BEA) and enniatins (ENNs) showed high occurrences ranging from 27 and 100% during three investigated years. Co-occurrence of Fusarium mycotoxins/metabolites was high and almost all were highly correlated to each other but their possible synergistic, additive, or antagonistic effects of toxicity, should be taken into consideration. Our results demonstrated that modified and emerging mycotoxins/metabolites contributed substantially to the overall contamination of wheat grains. To avoid disparagement, it is necessary to analyse these forms in future mycotoxin monitoring programs and to set their maximum levels. Full article

Alternaria mycotoxins, including alternariol (AOH), alternariol-9-monomethylether (AME), and their masked/modified derivatives (e.g., sulfates or glycosides), are common food contaminants. Their acute toxicity is relatively low, while chronic exposure can lead to the development of adverse health effects. Masked/modified metabolites can probably release the more toxic parent mycotoxin due to their enzymatic hydrolysis in the intestines. Previously, we demonstrated the complex formation of AOH with serum albumins and cyclodextrins; these interactions were successfully applied for the extraction of AOH from aqueous matrices (including beverages). Therefore, in this study, the interactions of AME, alternariol-3-sulfate (AS), and alternariol-9-monomethylether-3-sulfate (AMS) were investigated with albumins (human, bovine, porcine, and rat) and with cyclodextrins (sulfobutylether-β-cyclodextrin, sugammadex, and cyclodextrin bead polymers). Our major results/conclusions are the following: (1) The stability of mycotoxin–albumin complexes showed only minor species dependent variations. (2) AS and AMS formed highly stable complexes with albumins in a wide pH range, while AME–albumin interactions preferred alkaline conditions. (3) AME formed more stable complexes with the cyclodextrins examined than AS and AMS. (4) Beta-cyclodextrin bead polymer proved to be highly suitable for the extraction of AME, AS, and AMS from aqueous solution. (5) Albumins and cyclodextrins are promising binders of the mycotoxins tested. Full article
