African, Caribbean, and Black (ACB) communities face increased COVID-19 morbidity and mortality, coupled with significant barriers to vaccine acceptance and uptake. Addressing these challenges requires innovative, multifaceted strategies. Peer-led interventions, grounded in critical health literacy (CHL) and critical racial literacy (CRL), and integrating collaborative equity learning processes, can enhance community capacity, empowerment, and health outcomes, contributing to long-term health equity. This paper describes and presents the evaluative outcomes of a peer-led intervention aimed at enhancing COVID-19 vaccine confidence and acceptance. The Peer-Equity Navigator (PEN) intervention consisted of a specialized training curriculum grounded in CHL and CRL. Following training, PENs undertook a 5-month practicum in community or health settings, engaging in diverse outreach and educational activities to promote vaccine literacy in ACB communities. The evaluation utilized a modified Reach, Effectiveness, Adoption, Implementation, and Maintenance (RE-AIM) Framework, using quantitative and qualitative methods to collect data. Sources of data included tracking records with community feedback, and a PEN focus group, to assess program feasibility, outreach, and effectiveness. From 16 September 2022, to 28 January 2023, eight trained PENs conducted 56+ community events, reaching over 1500 community members. Both PENs and community members reported high engagement, endorsing peer-led, community-based approaches and increased vaccine literacy. The PEN approach proves feasible, acceptable, and effective in promoting positive health behaviors among ACB communities. This intervention has clear implications for health promotion practice, policy, and research in equity-deserving communities, including immigrants and refugees, who also face multiple and intersecting barriers to health information and care. This research investigated the impact of corn–soybean meal-based fermented feed on the growth performance, pork quality, and fatty acid profiles of lean-type finishing pigs. A total of 80 lean-type growing DLY (Duroc × Landrace–Yorkshire) pigs were randomly assigned to 2 groups, with 5 replicates of 8 pigs per pen. The pigs in control group (CON group) were fed a basal diet, while the pigs in fermented feed group (FF group) were fed a diet supplemented with 10% fermented feed. The experimental period lasted 70 days. Results exhibited that pigs in FF group had a significant increase in final body weight and average daily gain (ADG) (p < 0.05) and had a significant decrease in the feed-to-gain ratio (F/G) (p < 0.05). The FF group also exhibited significant promotion in muscle intramuscular fat content, marbling score, and meat color and significantly reduced the meat shear force and drip loss (p < 0.05). Serum analysis indicated that fermented feed significantly elevated blood glucose, total cholesterol, triglyceride levels, and serum hormones such as insulin, leptin, and IGF-1 (p < 0.05). Additionally, fermented feed significantly elevated the levels of polyunsaturated fatty acids (PUFAs) and monounsaturated fatty acids (MUFAs), whereas it decreased the saturated fatty acids (SFAs) contents (p < 0.05). The fermented feed also significantly enhanced pork nutritional values (p < 0.05). The fermented feed increased the expression of IGF-1, SREBP1c, PDE3, PPARγ, SCD5, and FAT/CD36 mRNA (p < 0.05). Furthermore, microbial 16S rDNA analysis uncovered that FF supplementation significantly reduced the Campilobacterota phylum abundance, while increasing the genus abundances of Clostridium_sensu_stricto, norank_f_Oscillospiraceae, unclassified_c_Clostridia, and V9D2013 (p < 0.05). In summary, the results indicated that the microbial fermented feed exhibited the regulation effects on pork quality and nutritional values of lean-type pigs through regulating lipid metabolism and gut microbial composition. Camelina sativa (CS) is an emerging sustainable oilseed crop with interesting feed application potentialities. The research assessed the potentiality of Camelina sativa (CS) in the diet for free-range laying hens, aiming at reaching a nutritional claim. To this purpose, two feeding groups of hens (n = 100 Livorno hens, n = 25 hens/pen) were farmed outdoor and received either a Control diet, which was a commercial diet for laying hens, or a CS diet. The latter was formulated to include the 5% CS cake and 1% CS oil, replacing conventional feedstuffs. Diets were isoprotein and isoenergy and were available ad libitum throughout the laying period (February–September). At day 1, n = 12 eggs/diet were sampled to assess the initial proximate composition and FA profile. Every 7 days the sampling was repeated to analyze the eggs’ FA profile, up to day 35. At the end of the laying season, n = 6 hens/dietary treatment were slaughtered and subjected to meat quality evaluations. Results highlighted that a 28-day feeding was the period required to obtain 227 mg of C18:3 n-3 and 81 mg of C20:5 n-3 + C22:6 n-3/100 g egg, whereas a further 7 days of feeding trial were ineffective in further enhancing the omega-3 FA content of eggs. CS eggs were comparable to Control ones for overall physical traits, proximate composition, and shelf-life. In addition, at the end of the laying period, the meat of CS hens was also found to be healthier than that of Control ones, thanks to a higher omega-3 FA proportion (p < 0.01), which was, however, not sufficient to reach any nutritional claim. Instead, the proximate composition of CS meat was overall comparable to Control hens. In conclusion, the present research demonstrated that feeding Camelina sativa meal and oil to laying hens is feasible and allows to reach the nutritional claim in eggs “rich in omega-3 FA” after a feeding period of 28 days, without any negative effects on other eggs’ quality trials. Camelina sativa can thus be defined as a promising sustainable feedstuff for the poultry sector for diversification purposes and to enhance the nutritional quality of eggs. Full article

Long-term storage may induce lipid oxidation in brown rice and impact its utilization in animal diets. One-day-old male Ross 308 broiler chickens (with an initial body weight of 20 g) were randomly divided into three groups: corn-based diet (Corn), fresh brown rice-based diet (BR1) and stored brown rice-based diet (BR6), with 8 replicates of 10 birds per pen, in a 42-day feeding trial. The results showed that lipid oxidation indexes increased and fatty acid composition changed significantly in BR6 (p < 0.05). The dietary replacement of corn with brown rice showed no effects on growth performance of broilers (p > 0.05). However, palmitic acid and oleic acid increased, and stearic acid, linoleic acid and docosadienoic acid decreased in the broiler breast muscle of the BR1 and BR6 groups (p < 0.05). Ileum antioxidant enzyme activities increased in the BR1 and BR6 groups compared to the Corn group (p < 0.05), and the activities of α-amylase, trypsin, chymotrypsin and lipase decreased in the BR6 group compared to the BR1 and Corn groups (p < 0.05). Also, compared to the BR1 group, the overall expression of metabolites involved in drug metabolism—cytochrome P450, GnRH secretion and the estrogen signaling pathway in broiler ileum were down-regulated in the BR6 group (p < 0.05). In conclusion, the lipid oxidation of stored brown rice decreased digestive enzyme activities and changed metabolic characteristics in the ileum of broilers. While replacing corn with brown rice did not affect broiler growth performance, it reduced the contents of unsaturated and essential fatty acids in breast muscle and enhanced the ileal antioxidant functions of broilers. Optimizing the metabolizable protein level in ruminant diets represents a promising strategy to increase nitrogen use efficiency and mitigate environmental pollution. This study explored the impacts of varying metabolizable protein (MP) levels on amino acid (AA) balance, nitrogen (N) utilization, and the ruminal microbiota in Hu lambs. Fifty-four female Hu lambs of 60 d old, with an average body weight (BW) of 18.7 ± 2.37 kg, were randomly allocated to three dietary MP groups: (1) low MP (LMP, 7.38% of DM), (2) moderate MP (MMP, 8.66% of DM), and (3) high MP (HMP, 9.93% of DM). Three lambs with similar BW within each group were housed together in a single pen, serving as one experimental replicate (n = 6). The feeding trial lasted for 60 days with 10 days for adaptation. The final BW of lambs in the MMP and HMP groups increased (p < 0.05) by 5.64% and 5.26%, respectively, compared to the LMP group. Additionally, lambs fed the MMP diet exhibited an 11.6% higher (p < 0.05) average daily gain than those in the LMP group. Increasing dietary MP levels enhanced (p < 0.05) N intake, urinary N, retained N, and percent N retained, but decreased apparent N digestibility (p < 0.05). Urinary uric acid, total purine derivatives, intestinally absorbable dietary protein, microbial crude protein, intestinally absorbable microbial crude protein, and actual MP supply all increased (p < 0.05) with higher MP values in the diet. The plasma concentrations of arginine, lysine, methionine, phenylalanine, threonine, aspartic acid, proline, total essential AAs, and total nonessential AAs were the lowest (p < 0.05) in the LMP group. In the rumen, elevated MP levels led to a significant increase (p < 0.05) in the ammonia N content. The relative abundances of Candidatus_Saccharimonas, Ruminococcus, and Oscillospira were the lowest (p < 0.05), whereas the relative abundances of Terrisporobacter and the Christensenellaceae_R-7_group were the highest (p < 0.05) in the MMP group. In conclusion, the moderate dietary metabolizable protein level could enhance growth performance, balance the plasma amino acid profiles, and increase nitrogen utilization efficiency in Hu lambs, while also altering the rumen bacterial community by increasing beneficial probiotics like the Christensenellaceae_R-7_group. Beef production in Greece is a sector that has been characterized by a decline in both the output and the number of beef-producing animals over the last decades. The major challenge is low beef self-sufficiency; only 19.1% of demand is met by domestic production. The latter leads to a growing reliance on imports of both live animals and carcasses. Hence, the fattening of young bulls from dairy breeds could be an option to address this challenge subject to improving the quality of produced meat. The objective of the present study was to investigate the effects of extruded linseed in the diet of young bulls on their performance and meat quality. Sixty-eight young Holstein bulls were equally assigned in two experimental groups: the control group (CON, n = 34) and Linseed Group (LS, n = 34). Bulls in the CON group received a basal total mixed ration while LS young bulls were offered the same basal ration supplemented with linseed (5% on dry matter basis) during the final fattening stage. All bulls were subjected to three individual weightings at the beginning, the middle and the end of the trial. The feed offered was recorded daily and feed refusals were weighed for each pen to calculate feed intake. After slaughter, the Longissimus dorsi muscle from each carcass was collected to evaluate meat pH, color, chemical composition, tenderness and fatty acid profile. Analysis of variance was used to evaluate the effect of dietary intervention on performance and examined meat parameters, with significance set at p < 0.05, using SPSS software (version 29.0). Average daily gain, dry matter intake and feed conversion ratio were not affected by the dietary intervention (p > 0.05). Similarly, carcass yield and dressing percentage remained unaffected (p > 0.05). Adding extruded linseed did not result in differences in meat quality traits (p > 0.05), except for meat pH, which was significantly decreased in the LS group (p < 0.05), indicating more efficient post-mortem glycolysis. Finally, the inclusion of extruded linseed resulted in higher levels of α-linolenic acid in the meat (p < 0.05). These results suggest that including 5% extruded linseed (on a DM basis) in the diet of young Holstein bulls increased meat n-3 content, improved beef pH and maintained production performance. Environmentally modified housing [EMH; windowless, insulated sidewalls and ceiling, thermostatically controlled ventilation fans) versus conventional housing [CVH; cross-ventilated, insulated ceiling, ceiling fans) improved broiler performance in the summer. The objective of this investigation was to determine whether social behaviors differed between two population densities (0.06 m2/chick [HD] or 0.07 m2/chick [LD]) in these houses. We used a randomized block statistical design, involving houses, population densities, observation times, and bird age. Behaviors were observed weekly, during the morning and the afternoon. Individual observers focused on the group of broilers in one of three defined 26.76 m2 areas in each of the four pens in each house. Aggressive encounters, tail and back pecking, feather eating, thermoregulatory, preening, and flock mobility were recorded. Feather pecking, eating and aggressive encounters were expressed at greater rates in HD birds in CVH. A salt-deficient diet caused increased feather pecking and aggressive encounters, which decreased after correction of the mistake. Increased heat indices (HIs), HD, and greater light intensity in CVH influenced behaviors and mortality more severely than in EMH. In CVH and EMH, burrowing/thermoregulatory/resting activity increased with increasing HIs. Afternoon preening was elevated significantly in EMH. It was concluded that broilers reared in EMH were more comfortable and experienced improved welfare compared to those reared in CVH. Full article

Background: Reliable and non-invasive methods for assessing welfare in poultry are essential for improving evidence-based welfare monitoring and advancing management practices in commercial production systems. The iris-to-pupil (IP) ratio, previously validated by our group in primates and cattle, reflects autonomic nervous system balance and may serve as a physiological indicator of stress in laying hens. This study evaluated the utility of the IP ratio under field conditions across diverse commercial layer housing systems. Materials and Methods: In total, 296 laying hens (Lohmann Brown, n = 269; White Leghorn, n = 27) were studied across four locations in Canada housed under different systems: Guelph (indoor; pen), Spring Island (outdoor and scratch; organic), Ottawa (outdoor, indoor and scratch; free-range), and Toronto (outdoor and hobby; free-range). High-resolution photographs of the eye were taken under ambient lighting. Light intensity was measured using the light meter app. The IP ratio was calculated using NIH ImageJ software (Version 1.54p). Statistical analysis included one-way ANOVA and linear regression using GraphPad Prism (Version 5). Results: Birds housed outdoors had the highest IP ratios, followed by those in scratch systems, while indoor and pen-housed birds had the lowest IP ratios (p < 0.001). Subgroup analyses of birds in Ottawa and Spring Island farms confirmed significantly higher IP ratios in outdoor environments compared to indoor and scratch systems (p < 0.001). The IP ratio correlated weakly with ambient light intensity (r2 = 0.25) and age (r2 = 0.05), indicating minimal influence of these variables. Although White Leghorn hens showed lower IP ratios than Lohmann Browns, this difference was confounded by housing type; all White Leghorns were housed in pens. Thus, housing system but not breed was the primary driver of IP variation. Conclusions: The IP ratio is a robust, non-invasive physiological marker of welfare assessment in laying hens, sensitive to housing environment but minimally influenced by light or age. Its potential for integration with digital imaging technologies supports its use in scalable welfare assessment protocols. Full article

This study investigated the effects of dietary niacinamide supplementation on blood parameters, antioxidant status, and intestinal health in finishing pigs fed low-protein diets. Sixty-four pigs (Duroc × Landrace × Yorkshire; 80.4 ± 0.1 kg) were randomly allocated to four dietary treatments supplemented with 30 (NAM30), 130 (NAM130), 230 (NAM230), and 330 (NAM330) mg/kg niacinamide for 30 days. Each treatment had four replicate pens and four pigs per pen. Growth performance was not significantly affected. However, the NAM130 group showed higher (p < 0.05) hemoglobin levels, reduced (p < 0.05) serum malondialdehyde and interleukin-1β (IL-1β) concentrations, and altered intestinal microbiota composition, including lower Streptococcus abundance (p < 0.05). Serum alanine aminotransferase levels increased quadratically (p < 0.05) with niacinamide supplementation, with a breakpoint at approximately 221 mg/kg. These results suggest that dietary supplementation with 130 mg/kg niacinamide improves antioxidant status, modulates inflammation, and supports intestinal microbial balance, with a safety threshold to avoid hepatic stress. This study evaluated the effect of mixed meal replacement of soybean meal on growth conditions, carcass traits, and meat quality of finishing pigs by partially and entirely replacing soybean meal with equal proportions of rapeseed, cotton, and sunflower meal. A total of fifty-four pigs with an average initial weight of 97.60 ± 0.30 kg were selected and randomly divided into three groups according to their initial weight, with six pens in each group and three pigs in each pen. The experimental groups were as follows: control group (CON), fed corn–soybean meal type basal diet; corn–soybean mixed meal group (CSM), using equal proportions of rapeseed meal, cotton meal, and sunflower meal (3.52% each) to replace 9.06% of soybean meal in the basal diet; and corn mixed meal group (CMM), using equal proportions of rapeseed meal, cotton meal, and sunflower meal (6.46% each) to replace soybean meal in the basal diet completely. According to the results, the use of mixed meal as a replacement for soybean meal did not have a significant impact (p > 0.05) on the average daily weight gain, average daily feed intake, feed-to-weight ratio, body size, carcass traits, and meat quality of finishing pigs. The entire replacement of soybean meal with a mixed meal resulted in a significant increase (p < 0.05) in leaf fat weight. The use of mixed meal as a substitute for soybean meal had no significant effect (p > 0.05) on the antioxidant capacity and fatty acid composition of the longissimus thoracis in finishing pigs. However, longissimus thoracis muscle fiber diameter was reduced in the mixed meal partially replaced soybean meal group compared to the mixed meal completely replaced soybean meal group (p < 0.05). In addition, mixed meal replacing soybean meal did not significantly affect (p > 0.05) the expression of the longissimus thoracis muscle fiber type genes MYHC1 and MYHC2. Mixed meal replacement of soybean meal did not significantly affect (p > 0.05) the expression of ACACA, FASN, and PPARG genes in the longissimus thoracis. This study showed that mixed meal as an alternative to soybean meal in diets did not have significant negative effects on the growth performance and meat quality of finishing pigs. These results can help develop further mixed meals as a functional alternative feed ingredient for soybean meals in pig diets. Full article

Weaning stress leads to intestinal dysfunction and impaired growth performance and intestinal development in piglets. This study aims to investigate the effects of Lactobacillus reuteri LR1 on growth performance and amino acid metabolism in the gut–liver axis of weaned piglets. A total of 48 weaned piglets (Duroc × Landrace × Yorkshire, 21 days old) were randomly assigned to the CON group (fed a basal diet) and the LR1 group (fed the basal diet supplemented with 5 × 1010 CFU/kg of Lactobacillus reuteri LR1) with six pens per group and 4 piglets each pen. The results demonstrated that LR1 significantly increased average daily gain (ADG), average daily feed intake (ADFI), and final body weight (p < 0.05). Additionally, LR1 significantly enhanced the villus height of the ileum (p < 0.05) and upregulated the expression of SLC6A19 in the jejunum, as well as SLC6A19, SLC7A1, and SLC38A9 in the ileum (p < 0.05). Amino acid analysis revealed that LR1 elevated the serum concentrations of glycine and hydroxyproline, along with increased taurine in the liver. Masson staining indicated LR1 reduced ileum fiber deposition, with COL3A1 identified as a key component. Furthermore, untargeted metabolomic analysis identified 27 amino acid-related differential metabolites and 11 significantly up-regulated in the plasma of the hepatic portal vein, including L-asparagine, L-citrulline, His-Cys, N-acetyltryptophan, 4-hydroxy-l-isoleucine, Gly-Arg, creatine, ornithine, ectoine, 3-methyl-l-histidine, and stachydrine. Correlation analysis suggested that COL1A2 and COL3A1 were closely associated with these metabolic changes. Overall, these findings suggest that LR1 supplementation promotes growth, improves intestinal morphology, reduces fiber deposition, and enhances amino acid metabolism in the gut–liver axis of weaned piglets. Background/Objectives: The use of black soldier fly (BSF) larvae meal in poultry nutrition is gaining attention as a sustainable protein source with a high nutritional value, an efficient bioconversion of organic waste, and potential functional benefits for intestinal health. This study evaluated the dietary effects of including 5% BSF larvae meal on the growth performance, nutrient digestibility, and energy utilization as well as on the intestinal integrity, gene expression, lipid profile, and short-chain fatty acid (SCFA) production of broilers under an intestinal challenge. Methods: Eight hundred one-day-old male broilers were assigned to four dietary treatments with eight replicates (25 birds/pen) and reared until day 40. Birds were fed either a Basal corn–soy diet or a BSF diet (5% BSF larvae meal replacing energy- and protein-yielding ingredients). Diets were provided to a non-challenged group and a challenged group, which was orally gavaged with Eimeria spp. on day 1 and Clostridium perfringens on days 11 and 14. The growth performance was evaluated up to day 40, while the nutrient digestibility, meat lipid profile, intestinal histomorphology, and gene expression were assessed at 21 days. The SCFAs were determined at both 21 and 40 days. Results: It was observed that the intestinal challenge induced dysbiosis and negatively affected growth performance, whereas the BSF meal inclusion partially mitigated these adverse effects. Broilers fed the BSF larvae meal showed increased cecal SCFA concentrations and a lower interleukin-6 gene expression, along with higher lauric and myristic acid levels in breast muscle (p ≤ 0.05). Conclusions: The inclusion of 5% BSF larvae meal improved performance without impairing nutrient digestibility or intestinal histomorphology, while increasing cecal concentrations of butyric and acetic acids and promoting a beneficial lipid deposition. Colibacillosis in nursery pigs, caused by Escherichia coli (ETEC, EPEC, and STEC pathotypes), remains a major economic concern in the swine industry. This study evaluated the effects of in-feed or in-water chlortetracycline (CTC) administration on the fecal prevalence of virulence genes and pathotypes associated with colibacillosis. A total of 1296 weaned piglets (21 days old) were allocated to 48 pens (16 pens/treatment; 27 piglets/pen) and assigned randomly to no CTC, in-feed CTC, or in-water CTC groups. CTC was administered from days 0 to 14. Fecal samples from five piglets per pen on days 0, 14, and 28 were enriched, screened by 11-plex PCR, cultured for pathotypes, and tested for CTC susceptibility and tetracycline resistance genes. None of the 360 fecal samples or 3267 E. coli isolates were positive for bfpA or aggA. Prevalence of estB (96.9%) and astA (92.8%) was highest. ETEC was the dominant pathotype (41.2%), with astA (29%) and estB (21.9%) as predominant enterotoxin genes. CTC administration had no significant effect on fecal prevalence of virulence genes or pathotypes (p > 0.05). stx2 and STEC were detected only at day 28, all harboring stx2e. All pathotypes were CTC-resistant, with tetA as the predominant resistance gene. Full article

This study was conducted to investigate the effects of the addition of dried mealworm, hydrolyzed mealworm, fermented poultry by-product meal (FPBM), and hydrolyzed fish soluble protein (HFSP) on the growth performance, nutrient digestibility, fecal score, and blood profiles of weaning pigs. A total of 40 (21-day-old) weaned piglets ([Yorkshire × Landrace] × Duroc) with an initial average body weight (BW) of 7.14 ± 1.29 kg were randomly assigned to one of four treatments for 35 days. There were four treatment groups with five replicates and two pigs (male and female) per pen. The dietary treatment included TRT1, a basal diet + 10% dried mealworm; TRT2, a basal diet + 10% hydrolyzed mealworm; TRT3, a basal diet + 10% fermented poultry by-product meal (FPBM); and TRT4, a basal diet + 10% hydrolyzed fish soluble protein (HFSP). The inclusion of FPBM in weaning pig diets had a significant effect on the blood profile with blood urea nitrogen (BUN) levels (p < 0.05), but not on other blood profiles of serum IGF-1, IgG, and IgA concentrations. BUN concentration decreased in response to the dietary inclusion of FPBM (p = 0.019). BUN concentration decreased and was at its lowest with FPBM (6.3 mg/dL), followed by hydrolyzed mealworm (7.3 mg/dL), while increasing with dried mealworm and HFSP (8.5 mg/dL). There was no significant difference in the growth performance, nutrient digestibility, and fecal score of piglets fed a basal diet with dried mealworm, hydrolyzed mealworm, FPBM, and HFSP. Hence, the addition of fermented poultry by-products in weaning pigs’ diets decreases the blood urea nitrogen, without any detrimental effect on performance, nutrient digestibility, or fecal score. Octocorals have proven to be a prolific source of bioactive natural products, exhibiting a wide spectrum of pharmacological activities. Among octocorals, Pennatulaceans, commonly known as sea pens, are among the most dominant soft coral species living in benthic communities. Nonetheless, reports on bioactivity and chemical investigations of this genus are scarce. This prompted us to shed light on the pharmacological potential of the extracts of the sea pen Pennatula phosphorea, Linneus 1758, and gain an overview of its metabolome. Crude octocoral extracts, obtained with a modified Kupchan extraction protocol, were assessed for their bioactivity potential, revealing the hexanic extract to exert anti-inflammatory effects and interesting protective properties in an in vitro model of sarcopenia and in auditory HEI-OC1 cisplatin-treated cells, while the chloroformic extract was active in reducing A375 melanoma cell viability in a concentration-dependent manner. An untargeted metabolomic analysis unveiled that P. phosphorea collects a wide array of glycerophospholipids and phosphosphingolipids belonging to the ceramide phosphoinositol class, which were exclusive or more abundant in the hexanic extract. Their proven anti-inflammatory and cytoprotective effects could demonstrate the activity shown by the P. phosphorea hexanic extract. In addition, a group of prostaglandins, eluted mainly in the chloroformic extract, were putatively annotated. Since prostanoids from marine origin have been demonstrated to exert cytotoxic and anti-proliferative properties against various cancer cell lines, the presence of PGs in the P. phosphorea chloroform extract could justify its anti-melanoma activity. This is the first report on the presence of glycerophospholipids, phosphosphingolipids, and prostaglandins, along with the identification of novel congeners, in sea pens. Full article
