Evaluation of Immunoprotective Activities of White Button Mushroom (Agaricus bisporus) Water Extract Against Major Pathogenic Bacteria (Aeromonas hydrophila or Vibrio fluvialis) in Goldfish (Carassius auratus)

The white button mushroom (Agaricus bisporus) is a widely cultivated edible and medicinal mushroom, which contains various active substances, and has application value against pathogenic bacteria in aquaculture. Firstly, A. bisporus water extract (AB-WE) was prepared. Through the detection kits, it was found that the polysaccharide, protein, and polyphenol components of AB-WE were 9.11%, 3.3%, and 1.5%, respectively. The 246 compounds were identified in AB-WE, and the major small-molecule components included L-Isoleucine, L-Tyrosine, L-Valine, and Linoleic acid by HPLC-Q Exactive-Orbitrap-MS. Secondly, the AB-WE was evaluated for its immunological activities through dietary administration and pathogen challenge (Aeromonas hydrophila and Vibrio fluvialis) in goldfish (Carassius auratus). The results showed that the levels of immune factors of acid phosphatase (ACP), alkaline phosphatase (AKP), and lysozyme (LZM) increased (p < 0.05) in goldfish, and the relative percentage survival of AB-WE against A. hydrophila and V. fluvialis were 80.00% (p < 0.05) and 81.82% (p < 0.05), respectively. The AB-WE reduced the bacterial content in renal tissue, enhanced the phagocytic activity of leukocytes, and exhibited antioxidant and anti-inflammatory effects by reducing the expression of antioxidant-related factors and inflammatory factors. Through histopathological and immunofluorescence techniques, it was found that AB-WE maintained the integrity of visceral tissues and reduced renal tissue apoptosis and DNA damage. Therefore, AB-WE exhibits immunoprotective activity against A. hydrophila and V. fluvialis infections in fish, and holds promise as an immunotherapeutic agent against major pathogenic bacteria in aquaculture. In this study, we focus on solving the nonlinear time-fractional Hirota–Satsuma coupled Korteweg–de Vries (KdV) and modified Korteweg–de Vries (MKdV) equations, using the Yang transform iterative method (YTIM). This method combines the Yang transform with a new iterative scheme to construct reliable and efficient solutions. Readers can understand the procedures clearly, since the implementation of Yang transform directly transforms fractional derivative sections into algebraic terms in the given problems. The new iterative scheme is applied to generate series solutions for the provided problems. The fractional derivatives are considered in the Caputo sense. To validate the proposed approach, two numerical examples are analysed and compared with exact solutions, as well as with the results obtained from the fractional reduced differential transform method (FRDTM) and the q-homotopy analysis transform method (q-HATM). The comparisons, presented through both tables and graphical illustrations, confirm the enhanced accuracy and reliability of the proposed method. Moreover, the effect of varying the fractional order is explored, demonstrating convergence of the solution as the order approaches an integer value. Importantly, the time-fractional Hirota–Satsuma coupled KdV and modified Korteweg–de Vries (MKdV) equations investigated in this work are not only of theoretical and computational interest but also possess significant implications for achieving global sustainability goals. Specifically, these equations contribute to the Sustainable Development Goal (SDG) “Life Below Water” by offering advanced modelling capabilities for understanding wave propagation and ocean dynamics, thus supporting marine ecosystem research and management. It is also relevant to SDG “Climate Action” as it aids in the simulation of environmental phenomena crucial to climate change analysis and mitigation. Additionally, the development and application of innovative mathematical modelling techniques align with “Industry, Innovation, and Infrastructure” promoting advanced computational tools for use in ocean engineering, environmental monitoring, and other infrastructure-related domains. Therefore, the proposed method not only advances mathematical and numerical analysis but also fosters interdisciplinary contributions toward sustainable development. Full article

Some Cretaceous ants belonging to the stem group of Formicidae exhibit bizarre morphology. This wide range of unusual adaptive features is primarily related to the mouthparts and clypeus. The researchers were perplexed by their specific ecology, as modern ant lineages do not exhibit anything similar. Here, we report and describe a new genus based on an extraordinary and mysterious alate ant from Late Eocene Baltic amber. Undoubtedly, the new ant is classified within the subfamily Formicinae (one of the crown groups), yet it displays a highly specialised morphology and an unusual array of features that are not observed in any extant ant lineages. Neither recent nor extinct ants have such a combination of features. While the exact phylogenetic placement of the new ant remains uncertain, we offer a discussion of its potential affinities based on our constrained phylogenetic analyses. We propose that †Eridanomyrma gen. n. should be considered in the new tribe †Eridanomyrmini trib. n. This new taxon highlights the adaptive diversity of a highly specialised, extinct lineage of Eocene crown-group ants. We also present a 3D model based on X-ray computed microtomography (µCT). Alzheimer’s disease (AD) is increasingly recognized as a multifactorial disorder driven by a combination of disruptions in proteostasis and organelle communication. The 2020 Lancet commission reported that approximately 10 million people worldwide were affected by AD in the mid-20th century. AD is the most prevalent cause of dementia. By early 2030, the global cost of dementia is projected to rise by USD 2 trillion per year, with up to 85% of that cost attributed to daily patient care. Several factors have been implicated in the progression of neurodegeneration, including increased oxidative stress, the accumulation of misfolded proteins, the formation of amyloid plaques and aggregates, the unfolded protein response (UPR), and mitochondrial–endoplasmic reticulum (ER) calcium homeostasis. However, the exact triggers that initiate these pathological processes remain unclear, in part because clinical symptoms often emerge gradually and subtly, complicating early diagnosis. Among the early hallmarks of neurodegeneration, elevated levels of reactive oxygen species (ROS) and the buildup of misfolded proteins are believed to play pivotal roles in disrupting proteostasis, leading to cognitive deficits and neuronal cell death. The accumulation of amyloid-β (Aβ) plaques and tau neurofibrillary tangles is a characteristic feature of AD. These features contribute to chronic neuroinflammation, which is marked by the release of pro-inflammatory cytokines and chemokines that exacerbate oxidative stress. Given these interconnected mechanisms, targeting stress-related signaling pathways, such as oxidative stress (ROS) generated in the mitochondria and ER, ER stress, UPR, and cytosolic chaperones, represents a promising strategy for therapeutic intervention. This review focuses on the relationship between stress chaperone responses and organelle function, particularly the interaction between mitochondria and the ER, in the development of new therapies for AD and related neurodegenerative disorders. Speaker burn-in is a controversial practice in the audio world, based on the belief that new devices reach optimal performance only after a certain period of use. Supporters claim it improves component flexibility, reduces initial distortion, and enhances sound quality—especially in the low-frequency range. Critics, however, emphasize the lack of scientific evidence for audible changes and point to the placebo effect in subjective listening tests. They argue that modern manufacturing and strict quality control minimize differences between new and “burned-in” devices. This study cites a standard describing a preliminary burn-in procedure, specifying the exact conditions and duration required. Objective tests revealed slight changes in speaker impedance and amplitude response after burn-in, but these differences are inaudible to the average listener. Notably, significant variation was observed between speakers of the same series, attributed to production line tolerances rather than use-related changes. The study also explored aging processes in speaker materials to better understand potential long-term effects. However, subjective listening tests showed that listeners rated the sound consistently across all test cases, regardless of whether the speaker had undergone burn-in. Overall, while minor physical changes may occur, their audible impact is negligible, especially for non-expert users. Full article

Endoreversible engine cycles are a cornerstone of finite-time thermodynamics. We show that endoreversible Stirling engines operating with a one-component plasma as a working medium run at maximal power output with the Curzon–Ahlborn efficiency. As a main result, we elucidate that this is actually a consequence of the fact that the caloric equation of state depends only linearly on temperature and only additively on volume. In particular, neither the exact form of the mechanical equation of state nor the full fundamental relation are required. Thus, our findings immediately generalize to a larger class of working plasmas, far beyond simple ideal gases. In addition, we show that for plasmas described by the photonic equation of state, the efficiency is significantly lower. This is in stark contrast to endoreversible Otto cycles, for which photonic engines have an efficiency larger than the Curzon–Ahlborn efficiency. Full article

Dietary patterns have been identified as one of the most important modifiable risk factors for several non-communicable diseases, inextricably linked to the health span of older people. Poor dietary choices may act as triggers for immune responses such as aggravated inflammatory reactions and oxidative stress contributing to the pathophysiology of several ageing hallmarks. Novel dietary interventions are being explored to restore gut microbiota balance and promote overall health in ageing populations. Probiotics and, most recently, postbiotics, which are products of probiotic fermentation, have been reported to modulate different signalling biomolecules involved in immunity, metabolism, inflammation, and oxidation pathways. This review presents evidence-based literature on the effects of postbiotics in promoting healthy ageing and mitigating various age-related diseases. The development of postbiotic-based therapeutics and diet-based interventions within a personalised microbiota-targeted approach is proposed as a possible direction for improving health in the elderly population. Despite growing evidence, the data regarding their exact mechanistic pathways for antioxidant and immunomodulating activities remain largely unexplored. Expanding our understanding of the mechanistic and chemical determinants of postbiotics could contribute to disease management approaches, as well as the development of and optimisation of biotherapeutics. Heart rate (HR) is strongly affected by the autonomic nervous system (ANS), while its spontaneous fluctuations, called heart rate variability (HRV), report about the dynamics of the complex, vegetative regulation of the heart rhythm. Hence, HRV is widely considered an important marker of the ANS effects on the cardiac system, and as such, a crucial diagnostic tool in cardiology. In order to obtain nontrivial results from HRV analysis, it would be desirable to establish exact, universal interrelations between the typical HRV parameters and HR itself. That, however, has not yet been fully accomplished. Hence, our aim was to perform a comparative statistical analysis of ECG recordings from a public database, with a focus on the HR dependence of typical HRV parameters. We revealed their fundamental connections, which were substantiated by basic mathematical considerations, and were experimentally demonstrated via the analysis of 24 h of ECG recordings of more than 200 healthy individuals. The large database allowed us to perform unique age-cohort analyses. We confirmed the HR dependence of typical time-domain parameters, such as RMSSD and SDNN, frequency-domain parameters such as the VLF, LF, and HF components, and nonlinear indices such as sample entropy and DFA exponents. In addition to shedding light on their relationship, we are the first, to our knowledge, to identify a new, diffuse structure in the VHF regime as an important indicator of SNS activity. In addition, the demonstrated age dependence of the HRV parameters gives important new insight into the long-term changes in the ANS regulation of the cardiac system. As a possible molecular physiological mechanism underlying our new findings, we suggest that they are associated with Piezo2 channel function and its age-related degradation. We expect our results to be utilized in HRV analysis related to both medical research and practice. Full article

Serotoninomics is an expanding field that focuses on the comprehensive study of the serotoninergic system, including serotonin’s biosynthesis, metabolism, and regulation, as well as related scientific methodologies 5-hydroxytryptamine (5-HT). This field explores serotonin’s complex roles in various physiological and pathological contexts. The essential amino acid tryptophan (Trp) is a precursor for several metabolic and catabolic pathways, with the kynurenine (KYN) pathway being particularly significant, representing about 95% of Trp metabolism. In contrast, only a small portion (1–2%) of dietary Trp enters the serotonin pathway. Anthranilic acid (AA), a metabolite in the KYN pathway, has emerged as a potential biomarker and therapeutic target for schizophrenia. Elevated serum AA levels in patients with schizophrenia have been associated with neurotoxic effects and disruptions in neurotransmission, suggesting AA’s critical role in the disorder’s pathophysiology. Furthermore, the 5-HT2A receptor’s involvement is particularly noteworthy, especially in relation to schizophrenia’s positive symptoms. Recent findings indicate that 5-HT2A receptor hyperactivity is linked to positive symptoms of schizophrenia, such as hallucinations and delusions. This study investigates serotoninomics’ implications for neuropsychiatric disorders, focusing on AA in schizophrenia and analysing recent research on serotonin signalling pathways and AA’s neurochemical effects. Understanding the roles of the 5-HT2A receptor and AA in neuropsychiatric disorders could lead to the development of more precise and less invasive diagnostic tools, specific therapeutic strategies, and improved clinical outcomes. Ongoing research is essential to uncover these pathways’ exact mechanisms and therapeutic potential, thereby advancing personalised medicine and innovative treatments in neuropsychiatry. Full article

Background/Objectives: Heart rate variability (HRV) is a marker of autonomic nervous system function, based on fluctuations in heartbeat intervals. Although several studies have investigated the association between frequency-domain HRV parameters and glaucoma, evidence based on large sample sizes remains limited. Therefore, the present study aimed to examine the relationship between frequency-domain HRV parameters and glaucoma subtypes, including primary open-angle glaucoma (PG) and exfoliation glaucoma (EG), using a larger sample size. Methods: Participants with primary open-angle glaucoma (PG), exfoliation glaucoma (EG), or no ocular disease other than cataract (controls) were recruited at Shimane University between June 2023 and July 2024. Frequency-domain HRV parameters (total power [TP], very-low-frequency [VLF], low-frequency [LF], high-frequency [HF], and LF/HF) were measured using a sphygmograph (TAS9 Pulse Analyzer Plus View). Group comparisons were conducted using unpaired t-tests, Fisher’s exact tests, and Tukey’s HSD test. Multivariate analyses were performed to identify factors associated with each HRV parameter. Results: A total of 809 participants were analyzed, including 522 with PG, 191 with EG, and 96 controls. The EG group showed significantly lower values across all frequency-domain HRV parameters compared to the PG group, and significantly lower LnLF values than the control group (p = 0.012). Multivariate analyses revealed that no significant associations were found between HRV measures and the presence of glaucoma or pseudoexfoliation material (PEM) deposition. Older age was significantly associated with lower values across all HRV parameters. Conclusions: In elderly glaucoma patients, age-related alterations in frequency-domain HRV parameters have been observed. Full article

Pharmacist-led transitions of care (TOC) services decrease preventable hospital readmission. TOC service implementation assessment can inform translation to real-world settings. The purpose of this study was to evaluate the implementation of a TOC service for patients with multiple admissions at a regional hospital using the RE-AIM framework. In this quasi-experimental, non-randomized study, individuals with ≥2 recent hospitalizations received pharmacist-led discharge medication reconciliation and counseling, management of drug-related problems, post-discharge telephonic visits, and social support. The reach, effectiveness, implementation, and maintenance RE-AIM dimensions were assessed using patient and service records. Outcomes included 30-day readmission rates for individuals completing ≥1 outpatient pharmacist visit (intervention) versus those unreachable in the outpatient setting (comparison), completed interventions, implementation features, and service adaptations. Chi-square and Fisher’s exact tests were used for comparison of categorical variables and the t-test was used for continuous variables. From February 2022 to August 2023, 72.7% of the 66 service participants participated in the intervention (reach). Additionally, 30-day readmission was 22.9% (intervention) versus 55.6% (comparison; p = 0.01). In total, 2279 interventions were documented (effectiveness). The service was adapted (implementation) and expanded to include additional populations (maintenance) to enhance sustainability. Based on RE-AIM evaluation, the pharmacist-led TOC intervention appears to be a sustainable solution for addressing readmission in multivisit patients. Full article

Early recognition and management of sepsis are essential in this population. This study evaluated the relationships between RDW, MPV, and MPV/PLT ratios and mortality in geriatric sepsis patients. Materials and Methods: This retrospective study was conducted between 2020 and 2024 in the Intensive Care Unit of the Department of Anesthesiology and Reanimation at a university hospital. Patients aged ≥ 65 years with a SOFA score of ≥2 were included. Demographic data (sex, age, height, weight, and BMI), hemogram parameters (RDW, MPV, and PLT), blood gas, and biochemical values were analyzed. Furthermore, their comorbidities; site of infection; ICU length of stay; vital signs; and SOFA, APACHE II, and SAPS II scores, recorded within the first 24 h following ICU admission, were evaluated. Statistical analysis was performed using the chi-square test, Student’s t-test, the Mann–Whitney U test, the Monte Carlo exact test, and ROC analysis. A p-value of <0.05 was considered statistically significant. Results: A total of 247 patients were included, with 46.2% (n = 114) classified as non-survivors during the 28-day follow-up period. Among them, 64.9% (n = 74) were male, with a mean age of 78.22 ± 8.53 years. Significant differences were also found in SOFA, APACHE-II, and SAPS-II scores between non-survivors and survivors (SOFA: 7.64 ± 3.16 vs. 6.78 ± 2.78, p = 0.023; APACHE-II: 21.31 ± 6.36 vs. 19.27 ± 5.88, p = 0.009; SAPS-II: 53.15 ± 16.04 vs. 46.93 ± 14.64, p = 0.002). On days 1, 3, and 5, the MPV/PLT ratio demonstrated a statistically significant predictive value for 28-day mortality. The optimal cut-off values were >0.03 on day 1 (AUC: 0.580, 95% CI: 0.516–0.642, sensitivity: 72.81%, specificity: 65.91%, p = 0.027), >0.04 on day 3 (AUC: 0.602, 95% CI: 0.538–0.663, sensitivity: 60.53%, specificity: 60.61%, p = 0.005), and >0.04 on day 5 (AUC: 0.618, 95% CI: 0.554–0.790, sensitivity: 66.14%, specificity: 62.88%, p = 0.001). Conclusions: The MPV and MPV/PLT ratios demonstrated statistically significant but limited predictive value for 28-day mortality in geriatric patients with sepsis. In contrast, the limited prognostic value of RDW may be related to variability in the inflammatory response and other underlying conditions. The correlations found between SOFA, APACHE II, and SAPS II scores highlight their importance in mortality risk prediction. Full article

This article discusses the transformation of a continuous-time model of the fractional system into a discrete-time model of the fractional system. For the continuous-time model, the exact solution of the nonlinear equation with fractional derivatives (FDs) that has the form of the damped rotator type with power non-locality in time is obtained.This equation with two FDs and periodic kicks is solved in the general case for the arbitrary orders of FDs without any approximations. A three-stage method for solving a nonlinear equation with two FDs and deriving discrete maps with memory (DMMs) is proposed. The exact solutions of the nonlinear equation with two FDs are obtained for arbitrary values of the orders of these derivatives. In this article, the orders of two FDs are not related to each other, unlike in previous works. The exact solution of nonlinear equation with two FDs of different orders and periodic kicks are proposed. Using this exact solution, we derive DMMs that describe a kicked damped rotator with power-law non-localities in time. For the discrete-time model, these damped DMMs are described by the exact solution of nonlinear equations with FDs at discrete time points as the functions of all past discrete moments of time. An example of the application, the exact solution and DMMs are proposed for the economic growth model with two-parameter power-law memory and price kicks. It should be emphasized that the manuscript proposes exact analytical solutions to nonlinear equations with FDs, which are derived without any approximations. Therefore, it does not require any numerical proofs, justifications, or numerical validation. The proposed method gives exact analytical solutions, where approximations are not used at all. Disruption Events in the HPV18 E1 and E2 Genes in Precancerous Cervical Lesions

Human papillomavirus 18 (HPV18) is the second most oncogenic high-risk HPV genotype, after HPV16, and is responsible for about 15% of cervical cancer cases worldwide. The integration of high-risk HPV DNA into the host genome leads to the disruption of the E1 and/or E2 genes, which is considered a risk factor for viral-induced carcinogenesis. This study examined the disruption events of HPV18 E1 and E2 genes in precancerous cervical lesions to investigate the rates and sites of gene disruption in the Greek population. The complete E1 and E2 genes were amplified using three and four overlapping primer sets, respectively. Extensive analysis revealed that the disruption/deletion events of the E1 and E2 genes were detected in all grades of cytology-determined lesions, with high frequency. E2 gene disruption was significantly related to LSIL+ cases (Fisher’s exact test, p = 0.022). No significant association was found in the analysis of the E1 gene. Additionally, no preferential sites of E1/E2 gene disruption were detected. This is the first study to provide evidence of disruption events of the HPV18 E1 gene. The data from the current analysis suggest that disruption of the E2 gene could be a significant marker for the progression of cytology-determined cervical dysplasia. However, future studies are required to evaluate whether different geographic populations have particular profiles regarding the rates and sites of gene disruption to further determine population-specific biomarkers. Wormholes are non-trivial topological structures that arise as exact solutions to Einstein’s field equations, theoretically connecting distinct regions of spacetime via a throat-like geometry. While static traversable wormholes necessarily require exotic matter that violates the classical energy conditions, subsequent studies have sought to minimize such violations by introducing time-dependent geometries embedded within cosmological backgrounds. This review provides a comprehensive survey of evolving wormhole solutions, emphasizing their formulation within both general relativity and alternative theories of gravity. We explore key developments in the construction of non-static wormhole spacetimes, including those conformally related to static solutions, as well as dynamically evolving geometries influenced by scalar fields. Particular attention is given to the wormholes embedded into Friedmann–Lemaître–Robertson–Walker (FLRW) universes and de Sitter backgrounds, where the interplay between the cosmic expansion and wormhole dynamics is analyzed. We also examine the role of modified gravity theories, especially in hybrid metric–Palatini gravity, which enable the realization of traversable wormholes supported by effective stress–energy tensors that do not violate the null or weak energy conditions. By systematically analyzing a wide range of time-dependent wormhole solutions, this review identifies the specific geometric and physical conditions under which wormholes can evolve consistently with null and weak energy conditions. These findings clarify how such configurations can be naturally integrated into cosmological models governed by general relativity or modified gravity, thereby contributing to a deeper theoretical understanding of localized spacetime structures in an expanding universe.
