Background/Objectives: Puberty is a critical stage in sheep development when reproductive capability is established, but the hormonal mechanisms underlying this transition remain incompletely understood. This study aimed to investigate the dynamic changes in estradiol (E2) levels and the expression patterns of estrogen receptors (ERα and ERβ) during puberty in Duolang sheep, a breed characterized by early sexual maturity and high reproductive efficiency. Methods: A total of 18 female Duolang sheep were assigned to three developmental stages (n = 6 per group): prepuberty (145 days), puberty (within 0 h of first estrus), and postpuberty (+3 days). Serum E2 concentrations and the mRNA and protein levels of ERα and ERβ were assessed in the hypothalamus, pituitary, and ovary. Additionally, primary ovarian granulosa cells (GCs) were isolated and stimulated in vitro with increasing concentrations of E2 (0–1000 ng/mL) to evaluate the dose-dependent expression of ERα, ERβ, and gonadotropin-releasing hormone (GnRH). Results: E2 levels peaked at the onset of puberty and declined thereafter. ERα expression in the hypothalamus and pituitary decreased during puberty but rebounded postpuberty, indicating a role in negative feedback regulation. In contrast, ovarian ERα expression reached its highest level during puberty, while ERβ expression in the ovary gradually increased from prepuberty to postpuberty. In GCs, ERα exhibited a biphasic expression pattern, peaking at 250 ng/mL E2 and decreasing at higher concentrations. ERβ and GnRH expression levels increased in a dose-dependent manner. Conclusions: These findings suggest that ERα primarily mediates E2 feedback within the hypothalamus–pituitary axis, whereas ERβ is associated with ovarian development and may regulate GnRH expression during the pubertal transition. The study provides new insights into the hormonal regulation of puberty in Duolang sheep and offers potential biomarkers for improving reproductive efficiency through targeted breeding strategies. This study was conducted to investigate the effect of body condition (BC) and other characteristics on estrus responses post altrenogest treatment and on reproductive performance in 161 gilts, bred artificially, treated with 20 mg altrenogest orally for 18 consecutive days. Parameters recorded at the beginning of altrenogest treatment included age and body weight. Backfat (BFT) and muscle thickness (MT), both defining BC, puberty status and uterine size (UtS, based on cross-sectional areas of the uterine horns) were recorded by real-time-ultrasound (RTU), and BFT, along with MT together with ovary status, were recorded again at the end of altrenogest treatment. Estrus rate (ER), interval of last altrenogest treatment onset of estrus (ATOE), conception (CR) and farrowing (FR) rates, litter size characteristics (total, life, and still born piglets; TB, LB, SB) and individual piglet birth weights (IPWs) were recorded. Average age and weight were 252.6 ± 7.5 days and 153.0 ± 12.4 kg, respectively. BFT and MT were 0.9 ± 0.2 and 6.0 ± 0.4 cm, respectively, at the beginning of altrenogest treatment, and similar at the end. 147 gilts were determined as pubertal (PUB) and 14 prepubertal (PRE). All gilts only had follicles of 3.2 ± 0.4 mm on average at the end of altrenogest treatment. ER, ATOE, CR, FR, TB, LB, SB and IPW were 99.4%, 5.9 ± 0.5 days, 100%, 95.6%, 18.1 ± 2.6, 17.4 ± 2.5, 0.7 ± 1.0, 1.3 ± 0.2 kg, respectively. PUB and PRE did not differ, or only marginally differed, in all recorded parameters. BC or pubertal status did not have a great impact on the efficacy of altrenogest treatment or reproductive performance. However, UtS was significantly smaller in PRE than in PUB (0.51 ± 0.15 cm2 and 1.63 ± 0.48 cm2, respectively). There was a very slight significant relationship between UtS and LB (r = 0.19, p = 0.03) and a tendency towards TB (r = 0.15, p = 0.09). In conclusion, under the conditions of this study, BC, age and pubertal status did not have an effect on the efficacy of altrenogest or the reproductive performance of hyper-prolific gilts of Danish genetic origin, but uterine size may have had a positive effect. Fixed-time artificial insemination is an important technique in swine production that can improve reproductive efficiency and meat production quality through making better use of the genetic potential of breeding males and reducing the costs associated with double or multiple inseminations. Our goal was to evaluate the vaginal application of the GnRH agonist triptorelin acetate to synchronize ovulation in post-weaning sows and facilitate the implementation of a single, fixed-time insemination. In the first experiment, the efficacy of treatment with triptorelin in animals with or without signs of estrus was analyzed using a single insemination at a fixed time, compared to a control group following the standard insemination strategy. The farrowing rate was comparable between the triptorelin and control groups (100 vs. 87.50%), but triptorelin treatment without estrus had a lower rate (50%). Litter size did not differ between the groups. Estradiol and progesterone levels at 96 and 120 h post-weaning were similar in the control and triptorelin groups (p > 0.05). These results suggest that triptorelin has the potential to synchronize ovulation in pigs without affecting post-weaning hormonal profiles. In a second experiment, the objective was to evaluate the productive and economic impact of implementing a treatment with triptorelin acetate 96 h after weaning, compared to the standard insemination protocol. Sows were grouped according to treatment (control vs. triptorelin) and estrus onset (≤5 days and >5 days after weaning, which was considered late estrus). The farrowing rate was lower in the late-estrus control group than in the control and triptorelin groups, and similar to that in the late-estrus triptorelin group. No differences were found in litter size and live or dead piglets born (p > 0.05). We developed an estimation model to assess the cost/benefit of intravaginal triptorelin administration at 96 h post-weaning. The overall result was that the use of triptorelin increased the financial benefit per inseminated sow by EUR 15–20. This improvement was mainly related to an increase in the reproductive performance of the treated sows compared to the control sows and the reduction in the number of inseminations per sow. These results highlight the potential of triptorelin to optimize reproductive management in pigs, improving efficiency and economic viability. Full article

The left displacement of the abomasum (LDA) is a common condition in dairy cows that can significantly impact their welfare, productivity, and fertility. This study was carried out in Greek dairy farms over a period of 3 years. To ensure early detection, the farmers were trained to accurately identify the disease. The reproductive performance and milk production of 306 cows was assessed by considering the time to the first estrus, the calving-to-conception interval, and the number of artificial inseminations required for the establishment of pregnancy. Uterine health status, the timing of disease diagnosis, and the season of the year were also evaluated. In a separate study, the outcomes of 26 cases where cows suffered LDA and underwent surgical treatment with a delay of at least one week from disease onset, were compared to those of cases promptly treated. The results indicate that even early identification and treatment of LDA affects fertility and milk yield; these impacts worsen with the co-existence of uterine infections of affected. However, in late-treated cases, all reproductive and production indices show significant deterioration. Our findings suggest that timely diagnosis of the disease, preferably by the farmer, ensures minimal losses in cows affected by LDA. Full article

The use of exogenous hormones has long been of interest for improving reproductive performance in swine production. Enhancing litter size directly impacts the economic efficiency of pig production. Various strategies, including nutritional, genetic, and hormonal approaches, have been explored with varying degrees of success. Administering a gonadotropin-releasing hormone (GnRH) agonist, such as buserelin, at the onset of estrus can induce ovulation and reduce the variation in ovulation timing among sows. This study assessed the impact of GnRH agonist supplementation in boar semen doses on the litter size of inseminated gilts. The research was conducted on a commercial swine herd in northern Thailand. A total of 231 Landrace × Yorkshire crossbred gilts, aged 224.5 ± 16.2 days at the onset of estrus synchronization, participated in the experiment. The gilts’ estrus was synchronized with oral altrenogest supplementation at a dosage of 20 mg/day for 18 days. After exhibiting standing estrus, the gilts were randomly divided into three groups. Control group: gilts were inseminated at 0 and 12 h post standing estrus onset with a conventional semen dose (n = 94). Treatment 1: similar to the control group, but with an added 5 µg (1.25 mL) of buserelin acetate to the boar semen dose during the first insemination (n = 71). Treatment 2: similar to the control group, but with 10 µg (2.5 mL) of buserelin acetate added to the boar semen dose during the first insemination (n = 66). All gilts were inseminated twice during their standing estrus using the intrauterine artificial insemination method. Each semen dose contained 3.0 × 109 motile sperm in 80 mL. The farrowing rate averaged 78.8% and did not significantly differ between the groups (p = 0.141). The total number of piglets born per litter in the treatment 2 group was greater than in the control group (14.0 ± 0.3 vs. 13.2 ± 0.3, respectively, p = 0.049), but was not significantly different from the treatment 1 group (13.3 ± 0.3, p = 0.154). Similarly, the number of live-born piglets in the treatment 2 group was greater than in the control and treatment 1 groups (13.2 ± 0.4 vs. 12.3 ± 0.3 and 12.0 ± 0.4, respectively, p < 0.05). Moreover, the live-born piglets’ litter birth weight in the treatment 2 group was greater than in the control group (17.0 ± 0.4 vs. 15.6 ± 0.3 kg, respectively, p = 0.008) and the treatment 1 group (15.7 ± 0.4 kg, p = 0.025). In conclusion, adding a GnRH agonist to boar semen appears to enhance the litter size of gilts. Further research should focus on understanding the underlying mechanisms and determining the optimal dose and timing for GnRH agonist supplementation. Full article

Reproduction is classically controlled by gonadotropin-releasing hormone (GnRH-I) and its receptor (GnRHR-I) within the brain. In pigs, a second form (GnRH-II) and its specific receptor (GnRHR-II) are also produced, with greater abundance in peripheral vs. central reproductive tissues. The binding of GnRH-II to GnRHR-II has been implicated in the autocrine/paracrine regulation of gonadal steroidogenesis rather than gonadotropin secretion. Blood samples were collected from transgenic gilts, with the ubiquitous knockdown of GnRHR-II (GnRHR-II KD; n = 8) and littermate controls (n = 7) at the onset of estrus (follicular) and 10 days later (luteal); serum concentrations of 16 steroid hormones were quantified by high-performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). Upon euthanasia, ovarian weight (OWT), ovulation rate (OR), and the weight of each excised Corpus luteum (CLWT) were recorded; HPLC-MS/MS was performed on CL homogenates. During the luteal phase, serum progesterone concentration was reduced by 18% in GnRHR-II KD versus control gilts (p = 0.0329). Age and weight at puberty, estrous cycle length, and OWT were similar between lines (p > 0.05). Interestingly, OR was reduced (p = 0.0123), and total CLWT tended to be reduced (p = 0.0958) in GnRHR-II KD compared with control females. Luteal cells in CL sections from GnRHR-II KD gilts were hypotrophic (p < 0.0001). Therefore, GnRH-II and its receptor may help regulate OR, CL development, and progesterone production in gilts. This study investigates the non-invasive monitoring of the endocrine ovarian activities of captive female golden takins (Budorcas taxicolor bedfordi) based on long-term fecal sex steroid hormone metabolite dynamics. Fecal progesterone (P4) metabolite dynamics were monitored in nine females for 0.5–15 years between 2004 and 2022. Fecal estradiol-17β (E2) and estrone (E1) metabolites were measured during certain estrous cycles, and fecal E1 metabolite concentrations were measured during all gestation periods. The breeding season of the captive animals was mainly between May and December, and they were polyestrous animals whose breeding season begins during the long-day period. The onset of the breeding season occurred slightly earlier as age increased. The mean age (±SD) at puberty based on fecal P4 metabolite dynamics was 4.1 ± 2.9 years. The first conception ages ranged from 2.3–10.2 years. The mean estrous cycle period (±SEM) was 25.4 ± 1.1 days, and mounting and mating occurred in periods of low fecal P4 metabolite levels during the breeding season. The mean gestation period (±SD) from the estimated mating date to the calving date was 253.9 ± 5.7 days, and the fecal P4 metabolite distribution during pregnancy was bimodal. Fecal estrone metabolite levels gradually increased 21 weeks before delivery, peaked during the week of delivery, and then markedly decreased in the first week after delivery. Estrus resumed in the first April–August period after delivery (mean ± SD; 103.5 ± 40.9 days) or in May of the year after delivery (421.0 ± 16.5 days). This study revealed that the estrous cycle and pregnancy of female golden takins can be determined by fecal progesterone metabolite dynamics and that fecal estrone metabolite dynamics increases toward parturition and are useful for predicting the date of delivery. This endocrinological information is important for planned breeding efforts for the golden takins. Full article

Labor is in short supply in animal agriculture. One time-consuming task is estrus detection in gilts. Stimulation with a live boar causes the onset of puberty in young gilts. Typically, a live boar is used to stimulate and identify estrus in the gilts by exposing the gilts to him. Recently, a boar pheromone (BB) was developed to replace the use of a live boar for sows. Additionally, a novel automatic sprayer used as environmental enrichment (EE) by gilts for the self-administration of BB has been developed by this laboratory. A commercial study was conducted to determine whether the use of a live boar could be replaced with a simple EE sprayer, allowing gilts to self-administer BB. Our objective was to determine whether the number and percentage of gilts in estrus obtained using live boars was comparable to self-administration using an EE sprayer containing BB. A total of 242 gilts were randomly assigned to either a live boar (BOAR) or BB self-administration using the environmental enrichment (EE) sprayer. Gilts began simultaneous exposure to either the BOAR or the BB when they were about 4–5 months of age and this continued until they were found in estrus or were injured, died, or never cycled about 2 months later. A total of 83.3% of gilts with exposure to BOAR were identified in estrus and bred, while exposure to BB resulted in 92.9% of gilts reaching puberty and being bred (p < 0.05). The days to reach estrus were 11 days longer for gilts exposed to BB than BOAR. Eight percent more gilts were injured by the BOAR than by using BB (and no boar). The use of BB as a priming pheromone could prevent gilt injuries, save labor, and reduce costs for pig farmers while not inhibiting reproductive output. Full article

Investigations on the optimum timing of artificial insemination (AI) following automated activity monitoring (AAM) depending on different types of semen in heifers are limited and in part show controversial results. Therefore, the objective of this observational study was to determine the association between the timing of AI using different characteristics of estrus (i.e., the onset, peak, and end of estrus) and pregnancy per AI (P/AI) in Holstein heifers. Heifers were fitted with a neck-mounted AAM system and inseminated with frozen conventional and sexed semen. The pregnancy per AI (n = 4159) from 2858 heifers from six commercial dairy farms in Germany inseminated upon the alert of an AAM system was evaluated. Estrous intensity was classified based on peak activity into low (35 to 89 index value) and high (90 to 100 index value). We detected a quadratic association between the interval from the onset of estrus to AI and P/AI (p = 0.02). The greatest P/AI was observed for heifers inseminated from 9 to 32 h after the onset of estrus. The intervals from the peak of activity to AI and the end of estrus to AI were not associated with P/AI (p ≥ 0.05). Heifers inseminated with frozen conventional semen (50.1%) had a greater P/AI compared with heifers inseminated with frozen sexed semen (43.3%; p = 0.03). There were no interactions between the intervals from the onset, peak, or end of estrus to AI or the type of semen and the P/AI (p ≥ 0.05). The pregnancy per AI was not associated with estrous intensity (50.5% for low intensity vs. 53.0% for high intensity; p = 0.37). In conclusion, inseminating heifers between 9 and 32 h after the onset of estrus, as detected by the AAM, optimized the P/AI regardless of semen type. Full article

The combination of estrus synchronization and superovulation (SS) treatments causes alterations in ovarian and endometrial gene expression patterns, resulting in abnormal follicle and oocyte growth, fertilization, and embryo development. However, the impact of combined SS treatments on the transcriptome of the surviving embryos remains unidentified. In this study, we examined gene expression changes in day 6 blastocysts that survived a brief regimen of synchronization treatment combined with superovulation. The sows were included in one of three groups: SS7 group (n = 6), sows were administered Altrenogest (ALT) 7 days from the day of weaning and superovulated with eCG 24 h after the end of ALT treatment and hCG at the onset of estrus; SO group (n = 6), ALT nontreated sows were superovulated with eCG 24 h postweaning and hCG at the onset of estrus; control group (n = 6), weaned sows displaying natural estrus. Six days after insemination, the sows underwent a surgical intervention for embryo collection. Transcriptome analysis was performed on blastocyst-stage embryos with good morphology. Differentially expressed genes (DEGs) between groups were detected using one-way ANOVA with an un-adjusted p-value < 0.05 and a fold change </> 1.5. The effect of SO treatment on the number of altered pathways and DEGs within each pathway was minimal. Only four pathways were disrupted comprising only a total of four altered transcripts, which were not related to reproductive functions or embryonic development. On the other hand, the surviving blastocysts subjected to SS7 treatments exhibited moderate gene expression changes in terms of DEGs and fold changes, with seven pathways disrupted containing a total of 10 transcripts affected. In this case, the up-regulation of certain pathways, such as the metabolic pathway, with two up-regulated genes associated with reproductive functions, namely RDH10 and SPTLC2, may suggest suboptimal embryo quality, while the down-regulation of others, such as the glutathione metabolism pathway, with down-regulated genes related to cellular detoxification of reactive oxygen species, namely GSTK1 and GSTO1, could depress the embryos’ response to oxidative stress, thereby impairing subsequent embryo development. The gene expression changes observed in the present study in SS7 embryos, along with previous reports indicating SS7 can negatively affect fertilization, embryo production, and reproductive tract gene expression, make its use in embryo transfer programs unrecommendable. The aim of the present study was to assess whether the strategic supplementation of bypass LO can enhance reproductive indexes—fertility, lambing rate, and prolificacy—in dairy Sarda ewes at the end of lactation. To assess whether LO supplementation leads to the adsorptions of PUFAs and their subsequent utilization by the body tissues, milk composition and fatty acid content were analyzed. Forty-eight ewes were assigned to the following groups: the control group (CT; N = 24), fed with a control diet without LO; and the treatment group (LO; N = 24), fed with a diet supplemented with LO (10.8 g/ewe/day). Both diets had similar crude protein and energy levels and were offered for 38 days (−21 to +17 days after artificial insemination). The trial included an adaptation period (7 days) followed by a regular supplementation (31 days) period. Estrus synchronization was induced in all the ewes using an intravaginal sponge and equine chorionic gonadotropin. Fifty-five hours after pessaries withdrawal, all ewes were inseminated using the cervical route and fresh semen. Cholesterol (p < 0.01), high-density lipoprotein (p < 0.001), and triglyceride (p < 0.05) levels in plasma were higher in the LO group. Plasmatic levels of non-esterified fatty acids were lower in the LO group after the end of the supplementation period (p < 0.05). Milk unsaturated fatty acids (UFAs), monounsaturated fatty acids (MUFAs), total polyunsaturated fatty acids (PUFAs), PUFAs omega 3 (PUFAs-ω3) and 6 (PUFAs-ω6), and trans fatty acids were higher in the LO group (p < 0.001), while saturated fatty acids (SFAs) were higher in the CT group during the supplementation period (p < 0.001). Three days after the end of the supplementation period, the content of milk UFAs (p < 0.05), PUFAs (p < 0.001), MUFAs, and PUFAs-ω6 (p < 0.01) were still higher in the LO group. whereas SFA was higher in the CT group (p < 0.01). There was no difference between groups in terms of ovulation rate, progesterone levels in plasma, fertility rate, prolificacy, and total reproductive wastage. However, the total area of luteal tissue was higher in the LO group (p < 0.01). Results obtained demonstrated that LO supplementation exerts a positive role in corpus luteum size at the onset of the peri-implantation period in Sarda dairy ewes. Additionally, the results obtained in the present study showed that the use of dietary bypass LO affects lipid metabolites in plasma and milk fatty acid profiles, demonstrating the ALA uptake by body tissues. Cows show spontaneous estrus over 8–20 h but become refractory to the bull about 10–12 h before ovulation. This indicates that ovulation occurs 10–12 h after the end of estrus behavior, yet spermatozoa from the bull ejaculate need to undergo maturation and capacitation for 6 to 8 h in the female reproductive tract before they are capable of fertilization. Traditionally, the onset of estrus has been considered the best timing for artificial insemination (AI) in cattle, that is, 6 to 24 h from the first signs of estrus. However, recent findings suggest this interval should be reduced to 16 to 6 h before ovulation, bringing it closer to the end of estrus. In this review, the end of estrus rather than its onset is proposed as the best guide for AI timing in dairy cattle, and physiological indicators of late estrus are discussed such as relaxation of the intravaginal part of the uterus, a lower cervical mucus viscosity and a softer pre-ovulatory follicular consistency as simple cues indicating a cow is ready for service. In this study, we evaluated the effectiveness of using estrogen-induced prolonged luteal function followed by prostaglandin F2 alpha (PGF2α) treatment to synchronize estrus in gilts. On day12 of the estrus cycle (D0 = first day of standing estrus), 52 gilts were assigned at random to two experimental groups: non-treated gilts (CON, n = 22), serving as controls, and prolonged luteal function group (CYP, n = 30), receiving a single treatment with 10 mg of estradiol cypionate intramuscularly Starting on day 12, blood samples were collected for estradiol and progesterone assays. Estrus detection started on day 17. Gilts from the CON group were inseminated at the onset of natural estrus. On day 28 CYP gilts were treated with PGF2α to induce luteolysis and inseminated at the onset of estrus. Gilts were slaughtered 5 d after the last insemination. A single treatment with estradiol cypionate prolonged luteal function in 90% of treated gilts. The duration of the estrous cycle was longer (p < 0.0001) for CYP gilts compared to CON gilts. CYP gilts showed synchronized estrus 3.96 ± 0.19 d after induction of luteolysis. The conception rate was similar (p = 0.10) for CON and CYP gilts. No difference was observed in the embryo recovery rate (p = 0.18) and total number of embryos per female (p = 0.06). The percentage of unfertilized oocytes, fragmented embryos and viable embryos was similar among females from CON and CYP groups (p > 0.05). The treatment of gilts with a single application of 10 mg of estradiol cypionate on day 12 of the estrous cycle was effective in prolonging luteal function and treatment with PGF2α resulted in synchronized estrus. Additionally, the synchronization protocol had no deleterious effect on fertility and embryonic development. Full article

Over the past century, the average age for onset of puberty has declined. Several additives present in our food are thought to contribute significantly to this early puberty which is recognized to also affect people’s health in later life. On this basis, the impact of 40-days unique oral administration of the food dye tartrazine (7.5, 27, and 47 mg/kg BW doses) was evaluated on some sexual maturation parameters on immature female Wistar rats. Vaginal opening was evaluated during the treatment period. At the end of the treatments, animals were sacrificed (estrus phase) and the relative weight of reproductive organs, pituitary gonadotrophin and sexual steroids level, cholesterol level in ovaries and folliculogenesis were evaluated. Compared to the control group, animals receiving tartrazine (47 mg/kg BW) showed significantly high percentage of early vaginal opening from day 45 of age, and an increase in the number of totals, primaries, secondaries, and antral follicles; a significant increase in serum estrogen, LH and in uterine epithelial thickness. Our findings suggest that tartrazine considerably disturbs the normal courses of puberty. These results could validate at least in part the global observations on increasingly precocious puberty in girls feeding increasingly with industrially processed foods. Full article

Hormonal treatments used in the artificial insemination (AI) of sheep can cause several physiological problems that can affect negatively fertility and animal health; however, AI protocols based on the detection of natural estrus offer a more sustainable option and can achieve high fertility. In this study, an AI protocol at natural estrus in organic Churra sheep was performed. In the first phase (AI protocol development), 125 ewes were exocervically inseminated, and their fertility was assessed based on the following factors: number of AI, physiological state, body condition, estrus detection–AI interval, and vaginal fluids in cervix. That protocol was repeated for six consecutive years. In all individuals, fertilities based on the timing of insemination after estrus detection were very high. Lactating ewes produced better results than did dry ewes, which was probably because of the better feeding of the former. In addition, double insemination increased the fertility of ewes whose estrus was detected within 16 h of onset. Body condition and amount of vaginal fluid were correlated with fertility. Exocervical inseminations at natural estrus can produce acceptable fertility and prolificity in Churra ewes. Full article
