Multiscale Geometric Characterization and Discrimination of Dermatoglyphs (Fingerprints) on Hardened Clay—A Novel Archaeological Application of the GelSight Max

A relatively new GelSight Max measurement instrument was applied to the microtopographies of experimental hardened clay surfaces, both with and without fingerprint (dermatoglyph) impressions, and the surface of an archaeological pottery handle fragment with a preserved fingerprint (paleodermatoglyph). The experimental clay surfaces were documented in order to determine the instrument’s ability to capture these surfaces in three-dimensions by imprinting them onto an elastomeric tactile membrane. Fingerprints on the experimental hardened clay and the archaeological pottery fragment were mathematically documented to test this instrument’s ability to capture these impressions. The surface texture measurements of the hardened clay and the pottery fragment were mathematically compared using conventional topographic characterization parameters (height and hybrid), fractal dimensions (Das) with associated coefficients of determination (R2), and multiscalar geometric characterization parameters, particularly relative area (Srel), area-scale complexity (Asfc), relative length (RL), and length-scale complexity (Lsfc). The surfaces of the experimental hardened clay with and without fingerprints and the archaeological pottery handle fragment with a fingerprint can be discriminated using some conventional height parameters, as well as some multiscale geometric topographic characterization parameters. Specifically, relative area (Srel), area-scale complexity (Asfc), relative length (RL), and length-scale complexity (Lsfc) could all discriminate between the hardened clay block with and without fingerprints and the fingerprint on the archaeological pottery handle fragment at different scales of measurement. Mean square ratios (MSRs) above 90% and 95% confidence levels indicated that the discrimination of these multiscale geometric characterizations was significant. Background/Objectives: Skin regeneration is a rapidly advancing field with significant implications for regenerative medicine, particularly in treating wounds and burns. This study explores the potential of hydrogels functionalized with fibroblast-derived extracellular vesicles (EVs) to enhance skin regeneration in vivo. Being immunoprivileged, EVs minimize immune rejection, offering an attractive alternative to whole-cell therapies by replicating fibroblasts’ key roles in tissue repair. Methods: To promote EVs’ versatility and effective application across different conditions, a lyophilization method with lyoprotectants was optimized. Then, EVs were used to functionalize a hydrogel to perform experiments on murine cutaneous wound models. Results: Gelatin methacrylate (GelMA) was selected as the polymeric hydrogel due to its biocompatibility, tunable mechanical properties, and ability to support wound healing. Mechanical tests confirmed GelMA’s strength and elasticity for this application. Fibroblast-derived EVs were characterized using Western blot, Transmission Electron Microscopy, and NanoSight analysis, proving their integrity, size distribution, and stability. miRNome profiling identified enriched biological pathways related to cell migration, differentiation, and angiogenesis, emphasizing the critical role of EV cargo in promoting wound repair. In a murine model, hydrogels loaded with fibroblast-derived EVs significantly accelerated wound healing compared to controls (mean wound area 0.62 mm2 and 4.4 mm2, respectively), with faster closure, enhanced epithelialization, increased vascularization, and reduced fibrosis. Notably, the lyoprotectants successfully preserved the EVs’ structure and bioactivity during freeze-drying, reducing EVs loss by 35% compared to the control group and underscoring the feasibility of this approach for long-term storage and clinical application. Conclusions: This study introduces a novel scalable and adaptable strategy for regenerative medicine by combining fibroblast-derived EVs with GelMA, optimizing EVs’ stability and functionality for enhanced wound healing in clinical settings, even in challenging contexts such as combat zones or large-scale natural disasters. Transferring knowledge learned from standard GelSight sensors to other visuotactile sensors is appealing for reducing data collection and annotation. However, such cross-sensor transfer is challenging due to the differences between sensors in internal light sources, imaging effects, and elastomer properties. By understanding the data collected from each type of visuotactile sensors as domains, we propose a few-sample-driven style-to-content unsupervised domain adaptation method to reduce cross-sensor domain gaps. We first propose a Global and Local Aggregation Bottleneck (GLAB) layer to compress features extracted by an encoder, enabling the extraction of features containing key information and facilitating unlabeled few-sample-driven learning. We introduce a Fourier-style transformation (FST) module and a prototype-constrained learning loss to promote global conditional domain-adversarial adaptation, bridging style-level gaps. We also propose a high-confidence guided teacher–student network, utilizing a self-distillation mechanism to further reduce content-level gaps between the two domains. Experiments on three cross-sensor domain adaptation and real-world robotic cross-sensor shape recognition tasks demonstrate that our method outperforms state-of-the-art approaches, particularly achieving 89.8% accuracy on the DIGIT recognition dataset. Congenital clinical anophthalmos and blind microphthalmos describe the absence of an eye or the presence of a small eye in the orbit. Between 1999 and 2013, 97 children with anophthalmos or microphthalmos were treated with self-inflating, hydrophilic gel expanders at the Rostock Eye Clinic. More than a decade later, this study investigated the perspective of patients and parents regarding the treatment, the surgical outcome, and the emotional and social well-being of the patients. A total of 22 families with 16 patients sighted in the other eye and six patients blind in both eyes participated. Questionnaires were developed, including items on physical, emotional, social, and medical aspects. The patients felt emotionally stable and integrated into their social environment, with no major limitations reported by the majority. These statements were confirmed by most of the parents. Parents (67%) indicated that the success of the operation was already apparent after the first intervention and that the current situation did not play a role in the patients’ social environment. The study provided new insights into the therapy results, the postoperative care, and the social and emotional stability of the prosthesis-wearing patients, indicating the chosen expander methods as promising in terms of positive postoperative care. In this review, several cost-effective thin-film coating methods, which include dip-coating, spin-coating, spray-coating, blade-coating, and roll-coating, are presented. Each method has its own set of advantages and disadvantages depending on the proposed application. Not all of them are appropriate for large-scale production due to their certain limitations. That is why the coating method should be selected based on the type and size of the substrate, including the thickness and surface roughness of the required thin films. The sol–gel method offers several benefits, such as simplicity in fabrication, excellent film uniformity, the capacity to cover surfaces of any size and over vast areas, and a low processing temperature. Nevertheless, these coating methods are somewhat economical and well managed in low-budget laboratories. Moreover, these methods offer thin films with good homogeneity and low-surface roughness. Furthermore, some other thin-film deposition methods, for instance, physical vapor deposition (PVD) and chemical vapor deposition (CVD), are also discussed. Since CVD is not restricted to line-of-sight deposition, a characteristic shared by sputtering, evaporation, and other PVD methods, many manufacturing methods favor it. However, these techniques require sophisticated equipment and cleanroom facilities. We aim to provide the pros and cons of thin-film coating methods and let the readers decide the suitable coating technique for their specific application. Full article

Transfer of learning or leveraging a pre-trained network and fine-tuning it to perform new tasks has been successfully applied in a variety of machine intelligence fields, including computer vision, natural language processing and audio/speech recognition. Drawing inspiration from neuroscience research that suggests that both visual and tactile stimuli rouse similar neural networks in the human brain, in this work, we explore the idea of transferring learning from vision to touch in the context of 3D object recognition. In particular, deep convolutional neural networks (CNN) pre-trained on visual images are adapted and evaluated for the classification of tactile data sets. To do so, we ran experiments with five different pre-trained CNN architectures and on five different datasets acquired with different technologies of tactile sensors including BathTip, Gelsight, force-sensing resistor (FSR) array, a high-resolution virtual FSR sensor, and tactile sensors on the Barrett robotic hand. The results obtained confirm the transferability of learning from vision to touch to interpret 3D models. Due to its higher resolution, tactile data from optical tactile sensors was demonstrated to achieve higher classification rates based on visual features compared to other technologies relying on pressure measurements. Further analysis of the weight updates in the convolutional layer is performed to measure the similarity between visual and tactile features for each technology of tactile sensing. Comparing the weight updates in different convolutional layers suggests that by updating a few convolutional layers of a pre-trained CNN on visual data, it can be efficiently used to classify tactile data. Accordingly, we propose a hybrid architecture performing both visual and tactile 3D object recognition with a MobileNetV2 backbone. MobileNetV2 is chosen due to its smaller size and thus its capability to be implemented on mobile devices, such that the network can classify both visual and tactile data. An accuracy of 100% for visual and 77.63% for tactile data are achieved by the proposed architecture. Full article

Resorcinol-formaldehyde/titanium dioxide composite (RF/TiO2) gel was prepared simultaneously by acid catalysis and then dried to aerogel with supercritical fluid CO2. The carbon/titanium dioxide aerogel was obtained by carbonization and then converted to nanoporous titanium carbide/carbon composite aerogel via 800 °C magnesiothermic catalysis. Meanwhile, the evolution of the samples in different stages was characterized by X-ray diffraction (XRD), an energy-dispersive X-ray (EDX) spectrometer, a scanning electron microscope (SEM), a transmission electron microscope (TEM) and specific surface area analysis (BET). The results showed that the final product was nanoporous TiC/C composite aerogel with a low apparent density of 339.5 mg/cm3 and a high specific surface area of 459.5 m2/g. Comparing to C aerogel, it could also be considered as one type of highly potential material with efficient photothermal conversion. The idea of converting oxide–carbon composite into titanium carbide via the confining template and low-temperature magnesiothermic catalysis may provide new sight to the synthesis of novel nanoscale carbide materials. Bacterial endophthalmitis (BE) is a potentially sight-threatening inflammatory reaction of the intraocular fluids or tissues caused by bacteria. Ciprofloxacin (CIP) eye drops are prescribed as first-line therapy in BE. However, frequent administration is necessary due to precorneal loss and poor ocular bioavailability. The objective of the current research was to prepare CIP containing nanostructured lipid carriers (CIP-NLCs) loaded an in situ gel system (CIP-NLC-IG) for topical ocular administration for enhanced and sustained antibacterial activity in BE treatment. CIP-NLCs were prepared by the hot homogenization method and optimized based on physicochemical characteristics and physical stability. The optimized CIP-NLC formulation was converted into CIP-NLC-IG with the addition of gellan gum as a gelling agent. Furthermore, optimized CIP-NLC and CIP-NLC-IG were evaluated for in vitro release and ex vivo transcorneal permeation studies, using commercial CIP ophthalmic solution (CIP-C) as the control. The optimized CIP-NLC formulation showed particle size, polydispersity index, zeta potential, assay and entrapment efficiency of 193.1 ± 5.1 nm, 0.43 ± 0.01, −32.5 ± 1.5 mV, 99.5 ± 5.5 and 96.3 ± 2.5%, respectively. CIP-NLC-IG with 0.2% w/v gellan gum showed optimal viscoelastic characteristics. The in vitro release studies demonstrated sustained release of CIP from CIP-NLC and CIP-NLC-IG formulations over a 24 h period. Transcorneal flux and permeability increased 4 and 3.5-fold, and 2.2 and 1.9-fold from CIP-NLC and CIP-NLC-IG formulations, respectively, when compared to CIP-C. The results demonstrate that CIP-NLC-IG could be considered as an alternate delivery system to prolong the residence time on the ocular surface after topical administration. Thus, the current CIP ophthalmic formulations may exhibit improved ocular bioavailability and prolonged antibacterial activity, which may improve therapeutic outcomes in the treatment of BE. Tactile sensing is an important perception mode for robots, but the existing tactile technologies have multiple limitations. What kind of tactile information robots need, and how to use the information, remain open questions. We believe a soft sensor surface and high-resolution sensing of geometry should be important components of a competent tactile sensor. In this paper, we discuss the development of a vision-based optical tactile sensor, GelSight. Unlike the traditional tactile sensors which measure contact force, GelSight basically measures geometry, with very high spatial resolution. The sensor has a contact surface of soft elastomer, and it directly measures its deformation, both vertical and lateral, which corresponds to the exact object shape and the tension on the contact surface. The contact force, and slip can be inferred from the sensor’s deformation as well. Particularly, we focus on the hardware and software that support GelSight’s application on robot hands. This paper reviews the development of GelSight, with the emphasis in the sensing principle and sensor design. We introduce the design of the sensor’s optical system, the algorithm for shape, force and slip measurement, and the hardware designs and fabrication of different sensor versions. We also show the experimental evaluation on the GelSight’s performance on geometry and force measurement. With the high-resolution measurement of shape and contact force, the sensor has successfully assisted multiple robotic tasks, including material perception or recognition and in-hand localization for robot manipulation. Significant progress in the manufacturing of biopharmaceuticals has been made by increasing the overall titers in the USP (upstream processing) titers without raising the cost of the USP. In addition, the development of platform processes led to a higher process robustness. Despite or even due to those achievements, novel challenges are in sight. The higher upstream titers created more complex impurity profiles, both in mass and composition, demanding higher separation capacities and selectivity in downstream processing (DSP). This creates a major shift of costs from USP to DSP. In order to solve this issue, USP and DSP integration approaches can be developed and used for overall process optimization. This study focuses on the characterization and classification of host cell proteins (HCPs) in each unit operation of the DSP (i.e., aqueous two-phase extraction, integrated countercurrent chromatography). The results create a data-driven feedback to the USP, which will serve for media and process optimizations in order to reduce, or even eliminate nascent critical HCPs. This will improve separation efficiency and may lead to a quantitative process understanding. Different HCP species were classified by stringent criteria with regard to DSP separation parameters into “The Good, the Bad, and the Ugly” in terms of pI and MW using 2D-PAGE analysis depending on their positions on the gels. Those spots were identified using LC-MS/MS analysis. HCPs, which are especially difficult to remove and persistent throughout the DSP (i.e., “Bad” or “Ugly”), have to be evaluated by their ability to be separated. In this approach, HCPs, considered “Ugly,” represent proteins with a MW larger than 15 kDa and a pI between 7.30 and 9.30. “Bad” HCPs can likewise be classified using MW (>15 kDa) and pI (4.75–7.30 and 9.30–10.00). HCPs with a MW smaller than 15 kDa and a pI lower than 4.75 and higher than 10.00 are classified as “Good” since their physicochemical properties differ significantly from the product. In order to evaluate this classification scheme, it is of utmost importance to use orthogonal analytical methods such as IEX, HIC, and SEC. Full article
