Evaluation of Immunoprotective Effects of DNA Vaccine Based on Eimeria maxima EF-1α Antigen and Chicken XCL1 Chemokine

The moderate pathogenicity coupled with high host susceptibility of Eimeria maxima has precipitated substantial economic losses in the poultry industry. Addressing challenges such as emerging drug resistance underscores the imperative for innovative vaccine strategies. This study developed a novel DNA vaccine to solve this challenge by fusing E. maxima elongation factor-1α (EmEF1α) with chicken chemokine XCL1 (ChXCL1) in the pVAX1 vector. The recombinant plasmid, designated pVAX1-ChXCL1-EmEF1α, was successfully constructed and confirmed to express the ChXCL1-EmEF1α fusion protein in vitro. Immunization of chickens with this DNA vaccine elicited a robust and balanced immune response, characterized by significantly increased proportions of CD4+ (11.76%) and CD8+ (5.58%) T lymphocytes, elevated levels of Th1-associated cytokines (IFN-γ and IL-12), and strong antigen-specific IgG and IgA antibody responses. Following experimental challenge with E. maxima, vaccinated birds exhibited substantial protection: a 66.4% reduction in oocyst shedding, a 71.7% improvement in relative weight gain, marked attenuation of intestinal lesions, and an anticoccidial index (ACI) of 170. These findings demonstrate that the ChXCL1-EmEF1α DNA vaccine effectively enhances both cellular and humoral immunity. Collectively, this study validates ChXCL1 as a potent molecular adjuvant and establishes the “antigen–adjuvant” fusion DNA platform as a promising strategy for developing next-generation vaccines against avian coccidiosis. Zn-based ternary deep eutectic solvents (TDESs) have attracted significant attention due to their good biodegradability, stability, and sustainability. In this work, TDESs composed of choline chloride:urea (ChCl:U) and zinc salts, ZnCl2, Zn(CH3COO)2, and ZnSO4 were synthesized and characterized by Fourier transform infrared (FTIR) spectroscopy and laser desorption ionization mass spectrometry (LDI MS). Their antibacterial activity against cariogenic Streptococcus species isolates was determined by microdilution assay, while their cytotoxic potential and effect on the intracellular reactive oxygen species (ROS) induction were analyzed on the MRC-5 fibroblast cell line by XTT, trypan blue, and DCF assays, respectively. FTIR confirmed that hydrogen bonds prevail in the molecular structure of ChCl:U:Zn salts, while LDI MS revealed the interactions between zinc salts and ChCl:U. The antibacterial TDES potential was high, especially against Streptococcus sanguinis, with ChCl:U:ZnCl2 displaying the most promising effects (MICs 1.13–18.12 µg/mL). Cytotoxicity assessment showed that concentrations up to 100 µg/mL of all TDESs did not display significant cytotoxicity, while higher concentrations significantly reduced cell viability by increasing ROS production and cell membrane damage, outlining the safety window of up to 100 µg/mL. Strong antibacterial activity of low TDESs concentrations combined with their good biocompatibility highlights their potential as innovative candidates for biomedical application. Full article

Coccidiosis is a major parasitic disease that suppresses poultry productivity and causes significant global economic losses. Currently, controlling Eimeria parasites relies primarily on the use of anticoccidial drugs or live vaccines. However, these conventional control strategies face the dual constraints of escalating drug resistance and unsustainable economic expenditures. In this study, the efficacy of a chimeric subunit vaccine comprising Eimeria maxima Elongation Factor-1α (EmEF1α) and chicken chemokine Ligand-1 (ChXCL1) was assessed for protection against experimental Eimeria maxima infection. The synthetic gene fragment ChXCL1-EmEF1α was ligated to the pET28a vector and expressed in vitro. Western blot analysis confirmed the successful expression of the recombinant ChXCL1-EmEF1α protein. Chickens immunized with the ChXCL1-EmEF1α exhibited a significantly stronger IgY response and higher secretion of IL-2 and IL-17 compared to those vaccinated with recombinant ChXCL1 alone or challenged solely with E. maxima. Furthermore, the ChXCL1-EmEF1α group demonstrated enhanced anticoccidial effects, including reduced intestinal lesions, higher body weight gain, and lower oocyst shedding compared to control groups. Following E. maxima challenge, the EmEF1α and ChXCL1-EmEF1α groups demonstrated robust protective efficacy, achieving high ACI values of 182 and 178, respectively. In contrast, the ChXCL1 and UC groups exhibited significantly lower ACI values (150 and 149, respectively), indicating minimal protection. This improvement was also reflected in the immune response, with significantly elevated levels of CD4+ and CD8+ T cells in the ChXCL1-EmEF1α-treated chickens. Moreover, ChXCL1 acts as an effective adjuvant when fused with EmEF1α, enhancing the vaccine’s anticoccidial efficacy. These results suggest that the ChXCL1-EmEF1α chimeric immunogen is a promising candidate for developing subunit vaccines against E. maxima infections. Full article

Organic–inorganic CH3NH3PbCl3 perovskite nanocrystals (PNCs) doped with Mn2+, CH3NH3PbxMn1−xCl3, have been successfully prepared using a reprecipitation method at room temperature. Structural and morphological characterizations reveal that the CH3NH3PbxMn1−xCl3 PNCs with cubic phase transforms from particles to cubes and increases in size from 16.2 ± 4.4 nm in average diameter to 25.3 ± 7.2 nm in cubic length after the addition of Mn2+ precursor. The CH3NH3PbxMn1−xCl3 PNCs exhibit a weak exciton emission at ~405 nm with a low absolute quantum yield (QY) of around 0.4%, but a strong Mn2+ dopant emission at ~610 nm with a high QY of around 15.2%, resulting from efficient energy transfer from the PNC host to the Mn2+ dopant via the 4T1→6A1 transition. In addition, the thermal and air stability of CH3NH3PbxMn1−xCl3 PNCs are improved due to the passivation with (3-aminopropyl) triethoxysilane (APTES), which is important for applications such as light emitting diodes (LEDs). Dimers of lithium formamidinate, CH(NH)2Li, and halogenated formamidines, HN=CHNHX, (X=Cl, Br, or I) are used as model systems to investigate simultaneous N-X···N and N-Li···N interactions, in tandem with orthogonal Li···X interactions. Geometry optimizations and energy calculations for the dimers are examined with the MP2 method and the M06-2X hybrid functional and the aug-cc-pVTZ basis set (the aug-cc-pVTZ-PP basis set is used for the iodine atom). Both methods predict the formation of a planar structure of C2v symmetry, regardless of the identity of the halogen atom. In this structure, the identities of the constituent monomers are essentially lost. Accordingly, the N-X···N interactions emerge as a rather symmetric quasi-linear N···X···N, where the covalent N-X bond in the halogenated formamidine is replaced by a partly covalent N···X interaction. Formation of the C2v structure is also driven by a fairly linear N···Li···N interaction parallel to the N···X···N interaction, and a Li···X interaction orthogonal to both the N···X···N and N···Li···N interactions. The strength of the interactions increases with the size of the halogen. The robustness of the interactions suggests that the dimers studied here or suitable analogues may find diverse applications including their use as novel polymeric synthons. Full article
