Sequential drought and heat stress pose a growing threat to forest ecosystems in the context of climate change, yet systematic evaluation methods for woody plants remain limited. This study aimed to develop a comprehensive screening platform for identifying woody plant species tolerant to sequential drought and heat stress among 27 native species growing in Korea. A sequential stress protocol was applied: drought stress for 2 weeks, followed by high-temperature exposure at 45 °C. Physiological indicators, including relative water content (RWC) and electrolyte leakage index (ELI), were used for preliminary screening, supported by phenotypic observations, Evans blue staining for cell death, and DAB staining to assess oxidative stress and recovery ability. The results revealed clear differences among species. Chamaecyparis obtusa, Quercus glauca, and Q. myrsinaefolia exhibited strong tolerance, maintaining high RWC and low ELI values, while Albizia julibrissin was highly susceptible, showing severe membrane damage and low survival. DAB staining successfully distinguished tolerance levels based on oxidative recovery. Additional species such as Camellia sinensis, Q. acuta, Q. phillyraeoides, Q. salicina, and Ternstroemia japonica showed varied responses: Q. phillyraeoides demonstrated high tolerance, T. japonica showed moderate tolerance, and Q. salicina was relatively sensitive. The integrated screening system effectively differentiated tolerant species through multiscale analysis—physiological, cellular, and morphological—demonstrating its robustness and applicability. This study provides a practical and reproducible framework for evaluating sequential drought and heat stress in trees and offers valuable resources for urban forestry, reforestation, and climate-resilient species selection. The increasing prevalence of cancer necessitates the development of novel and effective therapeutic agents. This study evaluates the anticancer potential of biosynthesized copper oxide nanoparticles (CuO NPs) using Camellia sinensis extract against human colon and breast cancer cells. The CuO NPs were characterized using various techniques to confirm their structure, size, morphology, and functional groups. The average size of CuO NPs synthesized was 20–60 nm, with spherical shape. The cytotoxic effects of these CuO NPs reveal a dose-dependent reduction in cell viability with 50% inhibitory concentration (IC50) at 58.53 ± 0.13 and 53.95 ± 1.1 μg/mL, respectively. Further investigation into the mechanism of action was conducted using flow cytometry and apoptosis assays, which indicated that CuO NPs induced cell cycle arrest and apoptosis in cancer cells. Reactive oxygen species (ROS) generation, caspase activity assay, and comet assay were also performed to elucidate the underlying pathways, suggesting that oxidative stress and DNA damage play pivotal roles in the cytotoxicity observed. Overall, our findings demonstrate that biosynthesized CuO NPs exhibit notable anticancer activity against colon and breast cancer cells, with moderate selectivity over normal cells, highlighting their potential as a therapeutic agent due to their biocompatibility. However, further studies are required to validate their selectivity and safety profile. Full article

This study investigates the effect of precursor pH (1.3, 2, 4, 6, 8, and 10) on the synthesis of gold nanoparticles (AuNPs) via a green synthesis approach using an aqueous extract of green tea (Camellia sinensis) leaves. The formation of AuNPs was monitored using UV-vis spectrophotometry and confirmed using transmission electron microscopy (TEM). The results confirmed that the morphology and size of the AuNPs are strongly dependent on the pH of the reaction medium. Based on spectral features, the color of the colloids, and TEM analysis, the synthesized samples were classified into three groups. The first (pH 8 and 10) contained predominantly spherical nanoparticles with an average diameter of ~18 nm, the second (pH 1.3 and 2) contained different shaped nanoparticles (20–250 nm in diameter), and the third (pH 4 and 6) contained flower-like nanostructures with a mean diameter of ~60 nm. UV-vis analysis revealed good stability of all AuNP colloids, except at pH 1.3, where a significant decrease in absorbance intensity over time was observed. These findings confirm that tuning the precursor pH allows for controlled manipulation of nanoparticle morphology and stability in green synthesis systems. Full article

Green tea, derived from the unoxidized leaves of Camellia sinensis (L.) Kuntze, is one of the least processed types of tea and is rich in antioxidants and polyphenols. Among these, catechins—particularly epigallocatechin gallate (EGCG)—play a key role in regulating cell signaling pathways associated with various chronic conditions, including cardiovascular diseases, neurodegenerative disorders, metabolic diseases, and cancer. This review presents a comprehensive analysis of recent clinical studies focused on the therapeutic benefits and potential risks of interventions involving green tea extracts or EGCG. A systematic literature survey identified 17 relevant studies, classified into five key areas related to catechin interventions: toxicity and detoxification, drug pharmacokinetics, cognitive functions, anti-inflammatory and antioxidant properties, and obesity and metabolism. Findings from these clinical studies suggest that the health benefits of green tea catechins outweigh the potential risks. The review highlights the importance of subject genotyping for enzymes involved in catechin metabolism to aid in interpreting liver injury biomarkers, the necessity of assessing drug–catechin interactions in clinical contexts, and the promising effects of topical EGCG in reducing inflammation. This analysis underscores the need for further research to refine therapeutic applications while ensuring the safe and effective use of green tea catechins. Full article

Background/Objectives: Ultraviolet B (UVB) radiation contributes significantly to skin aging and skin disorders by promoting oxidative stress, inflammation, and collagen degradation. Natural antioxidants such as theaflavins and thearubigins from Camellia sinensis L. (black tea) have shown photoprotective effects. This study aimed to optimize the extraction of theaflavins and thearubigins from black tea leaves and evaluate the efficacy of the extract against UVB-induced damage using a transfersome-based topical formulation. Methods: Extraction of theaflavins and thearubigins was optimized via response surface methodology (Box-Behnken Design), yielding an extract rich in active polyphenols. This extract was incorporated into transfersomes that were characterized for size, polydispersity, zeta potential, storage stability, and entrapment efficiency. Human dermal fibroblasts (NHDF) were used to assess cytotoxicity, protection against UVB-induced viability loss, collagen degradation, and expression of inflammatory (IL6, COX2, iNOS) and matrix-degrading (MMP1) markers. Cellular uptake of the extract’s bioactive marker compounds was measured via LC-MS/MS. Results: The transfersomes (~60 nm) showed a good stability and a high entrapment efficiency (>85%). The transfersomes significantly protected NHDF cells from UVB-induced cytotoxicity, restored collagen production, and reduced gene expression of MMP1, IL6, COX2, and iNOS. Cellular uptake of key extract’s polyphenols was markedly enhanced by the nanoformulation compared to the free extract. Conclusions: Black tea extract transfersomes effectively prevented UVB-induced oxidative and inflammatory damage in skin fibroblasts. This delivery system enhanced bioavailability of the extract and cellular protection, supporting the use of the optimized extract in cosmeceutical formulations targeting photoaging and UV-induced skin disorders. Dietary advanced glycation end-products (dAGEs) contained in high-sugar/fat and ultra-processed foods of the “Western diet” (WD) pattern predispose to several diseases by altering protein function or increasing oxidative stress and inflammation via RAGE (receptor for advanced glycation end-products). Although elevated endogenous AGEs are associated with loss of muscle mass and functionality (i.e., muscle wasting; MW), the impact of dAGEs on MW has not been elucidated. Here, we show that the most common dAGEs or their precursor, methylglyoxal (MGO), induce C2C12 myotube atrophy as endogenous AGE-derived BSA. ROS production, mitochondrial dysfunction, mitophagy, ubiquitin–proteasome activation, and inhibition of myogenic potential are common atrophying mechanisms used by MGO and AGE-BSA. Although of different origins, ROS are mainly responsible for AGE-induced myotube atrophy. However, while AGE-BSA activates the RAGE-myogenin axis, reduces anabolic mTOR, and causes mitochondrial damage, MGO induces glycolytic stress and STAT3 activation without affecting RAGE expression. Among thirty selected natural compounds, Vaccinium macrocarpon (VM), Camellia sinensis, and chlorophyll showed a surprising ability in counteracting in vitro AGE formation. However, only the standardized VM, containing anti-glycative metabolites as revealed by UHPLC-HRMS analysis, abrogates AGE-induced myotube atrophy. Collectively, our data suggest that WD-linked dAGE consumption predisposes to MW, which might be restricted by VM food supplements. Full article

Tea is one of the most consumed beverages in the world, belonging to the category of compounds known as tannins and flavonoids. One of the polyphenols found in large amounts in green tea leaves (Camellia sinensis) is epigallocatechin-3-O-gallate (EGCG). Though EGCG has shown some pharmacological effects, to date, it has not been utilised as a therapeutic agent. This is attributed to the fact that EGCG lacks adequate stability, and it is known to degrade through epimerization or auto-oxidation processes, especially when it is exposed to light, temperature fluctuations, some pH values, or the presence of oxygen. Consuming green tea with EGCG can alleviate the effects of bone diseases, such as osteoporosis, and support faster bone regeneration in the case of fractures. Therefore, this review focuses on the current state of research, highlighting the effects of EGCG on bone biology, such as enhancing osteoblast differentiation, promoting bone mineralisation, improving bone microarchitecture, and inhibiting osteoclastogenesis through the modulation of the RANK/RANKL/OPG pathway. Additionally, EGCG exerts antioxidant, anti-inflammatory, and dose-dependent effects on bone cells. It also downregulates inflammatory markers (TNF-α, IL-1β, and COX-2) and reduces oxidative stress via the inhibition of reactive oxygen species generation and the activation of protective signalling pathways (e.g., MAPK and NF-κB). Studies in animal models confirm that EGCG supplementation leads to increased bone mass and strength. These findings collectively support the further exploration of EGCG as an adjunct in the treatment and prevention of metabolic bone diseases. The authors aim to present the relationship between EGCG and bone health, highlighting issues for future research and clinical applications. Full article

The excessive use of chemical fertilizers in tea cultivation threatens soil health, environmental sustainability, and long-term crop productivity. This study explores the application of plant growth-promoting bacteria (PGPB) as an eco-friendly alternative to conventional fertilizers. A bacterial consortium was developed using selected rhizobacterial isolates—Lysinibacillus fusiformis, five strains of Serratia marcescens, and two Bacillus spp.—based on their phosphate and zinc solubilization abilities and production of ACC deaminase, indole-3-acetic acid, and siderophores. The consortium was tested in both pot and field conditions using two tea clones, S3A3 and TS491, and compared with a chemical fertilizer treatment. Plants treated with the consortium showed enhanced growth, biomass, and antioxidant activity. The total phenolic contents increased to 1643.6 mg GAE/mL (S3A3) and 1646.93 mg GAE/mL (TS491), with higher catalase (458.17–458.74 U/g/min), glutathione (34.67–42.67 µmol/gfw), and superoxide dismutase (679.85–552.28 units/gfw/s) activities. A soil metagenomic analysis revealed increased microbial diversity and the enrichment of phyla, including Acidobacteria, Proteobacteria, Actinobacteria, Chloroflexi, and Firmicutes. Functional gene analysis showed the increased abundance of genes for siderophore biosynthesis, glutathione and nitrogen metabolism, and indole alkaloid biosynthesis. This study recommends the potential of a PGPB consortium as a sustainable alternative to chemical fertilizers, enhancing both the tea plant performance and soil microbial health. Full article

Soil salinization poses a significant threat to tea plant (Camellia sinensis) production by compromising its bioactive compounds, such as polyphenols, L-theanine, and caffeine, which are key contributors to the plant’s health benefits and economic value. This study investigates the Salt Overly Sensitive 1 (SOS1) gene family, a critical salt-tolerance regulator in tea plants, to elucidate its role in maintaining quality under environmental stress. Genome-wide analysis identified 51 CsSOS1 genes, with phylogenetic and synteny analyses revealing strong evolutionary conservation with Populus trichocarpa and Arabidopsis thaliana. Promoter analysis detected stress- and hormone-responsive cis-elements, indicating adaptive functions in abiotic stress. Expression profiling demonstrated tissue-specific patterns, highlighting significant upregulation of CsSOS1-15 and CsSOS1-41 under salt and drought stress. Co-expression network analysis further linked CsSOS1 genes to carbohydrate metabolism, implicating their roles in stress resilience and secondary metabolite synthesis. Our findings provide molecular insights into CsSOS1-mediated salt tolerance, proposing potential targets for preserving bioactive compounds. This work facilitates developing salt-resistant tea plant cultivars to ensure sustainable production and quality stability amid environmental challenges. Full article

The tea tree root system is an important tissue for nutrient uptake, accumulation, and transport, and pH is an important environmental factor regulating the growth of tea tree (Camellia sinensis). However, the physiological and molecular mechanisms of how the tea tree root system responds to pH are unclear. In this study, Tieguanyin tea tree was used as the research object, and treated with different pH values to determine the morphological indexes of the tea plant root system and systematically study the physiological and molecular mechanisms of the effect of pH on the growth of the tea plant root system using transcriptomics in combination with metabolomics. The results showed that total root length, root surface area, root volume, total root tips, root fork number, and root crossing number of root crosses of the tea plant root system increased significantly (p < 0.05) with increasing pH. Transcriptome analysis showed that a total of 2654 characteristic genes were obtained in response to pH regulation in the root system of the tea plant, which were mainly enriched in six metabolic pathways. Metabolomics analysis showed that the metabolites with the highest contribution in differentiating tea plant responses to different pH regulations were mainly heterocyclic compounds, amino acids and derivatives, alkaloids, and flavonoids. Interaction network analysis showed that pH positively regulated the metabolic intensity of the MAPK signaling pathway (plant, plant hormone signal transduction, and RNA degradation pathway), positively regulated the content of the heterocyclic compound, amino acids and derivatives, and alkaloids, and positively regulated tea plant root growth. However, it negatively regulated ribosome, protein processing in the endoplasmic reticulum, and phenylpropanoid biosynthesis pathway intensity, and negatively regulated the flavonoid content. This study reveals the physiological and molecular mechanisms of the tea plant root system in response to pH changes and provides an important theoretical basis for the cultivation and management of tea plants in acidified tea plantations. The land-use conversion of Masson pine forests to tea fields is extensively practiced across subtropical China, primarily driven by its economic benefit. However, the effects of this conversion on soil fungal communities and functional guilds are poorly understood. Herein, a field experiment was conducted in a Masson pine forest (F), a 5-year-old tea plantation without (FT-CK) fertilization or with (FT-N), and a 30-year-old tea plantation (FT-O) to assess the impact of Masson pine forest-to-tea conversion on soil fungal abundance, community structure, and functional guilds by using qPCR and high-throughput sequencing. Compared to F, fungal abundance significantly decreased by 95%, 68%, and 79% in FT-CK, FT-N, and FT-O, respectively, probably caused by the decreased total nitrogen content and habitat disruption. Fungal alpha diversity significantly increased in FT-N and FT-O compared to FT-CK. FT-O presented the highest percentages of Mortierella among treatments, which favours soil organic carbon accumulation. FUNGuild-based predictions showed that FT-CK and FT-N had higher relative abundances of plant pathogens than F and FT-O. FT-O presented the highest percentages of litter and soil saprotrophs but exhibited the lowest percentages of ectomycorrhizal fungi among treatments, likely driven by increased soil organic carbon, total nitrogen, and total phosphorus content. Our findings demonstrate that Masson pine forest-to-tea conversion significantly degrades soil fungal community and function, highlighting the urgent need for soil management strategies (e.g., organic amendments) to enhance soil health in tea agroecosystems. Kujingcha (KJC) is a widely consumed substitute tea due to its unique flavor quality and health benefits. However, the biochemical basis for the formation of KJC’s unique flavor quality and health benefits remain unclear. In this study, using Camellia sinensis green tea and its processed fresh leaves as a control, we systematically analyzed the non-volatile and volatile components in KJC and its processed fresh leaves. The results indicate that high levels of flavonoids and water-soluble sugars, and low levels of amino acids and water-soluble proteins, are important biochemical foundations for the formation of taste quality in KJC. High aldehyde, alkene and heterocyclics contents contribute significantly to the aroma of KJC, among which heterocyclics are the key components for the formation of KJC’s rich pan-fried bean-like aroma. Flavonoids such as neohesperidin, hyperoside, rutin, astilbin and morin are important components for the formation of KJC’s health benefits. Full article

Fruit rot seriously damages pomelo production. Given concerns regarding the safety of chemical agents, biological alternatives are becoming more preferable. Therefore, the experiment aimed to (i) identify the pathogens causing pomelo fruit rot disease and (ii) select Trichoderma spp. strains controlling the determined pathogens in Ben Tre, Vietnam. Three pathogenic fungal strains isolated from diseased pomelo fruits were selected. The three pathogenic fungal strains were randomly injected into 9 healthy pomelo fruits. The strain PCP-B02-A2 led to a completely rotten fruit on day 17 after infection, while strains PCP-B02-B2 and PCP-B03-A1 had infected spots whose lengths were 17.5 and 28.1 mm, became larger, and eventually led to the whole fruit rot. The pathogens were identified by the internal transcribed spacer (ITS) technique as Colletotrichum gloeosporioides PCP-B02-A2, Pseudopestalotiopsis camelliae sinensis PCP-B03-A1, and P. chinensis PCP-B02-B2. Twenty-five Trichoderma spp. strains were isolated. The ITS technique identified four strains, including Trichoderma asperellum TP-B01, T. harzianum TP-B08, T. harzianum TP-B09, and T. asperellum TP-C25. The PCP-B02-A2 strain had antagonism at 66.7–68.7%, while those of PCP-B02-B2 and PCP-B03-A1 were 64.2–71.1% and 55.7–57.4%, respectively. Full article

The undertaking of distant hybridization holds paramount significance for the innovation of tea germplasm resources and the cultivation of superior, specialized tea varieties. However, challenges manifest during the process of tea plant distant hybridization breeding, with reproductive barriers impeding the successful acquisition of hybrid progeny; the precise stages at which these barriers occur remain unclear. In this study, utilizing Camellia sinensis cv. Jinxuan as the maternal parent, as well as C. gymnogyna Chang and C. sinensis cv. Yinghong No.9 as the paternal parents, interspecific distant hybridization (DH) and intraspecific hybridization (IH) were conducted. The investigation involved the observation of pollen germination and pollen tube behavior on the stigma, the scrutiny of the developmental dynamics of the ovary post-hybridization, and the examination of the stages and reasons for reproductive disorders during tea tree distant hybridization. The findings indicate that both IH and DH exhibit pre-fertilization barriers. The pre-embryonic development of hybrids obtained from DH is normal, but there is a significant fruit drop during the stage of fruit development. The germination rate of mature seeds obtained from DH is low, and there are pronounced post-fertilization disorders, which are the primary reasons for the difficulty in achieving successful tea plant distant hybridization. An analysis of the genetic variation in phenotypes and chemical components in the progeny after distant hybridization revealed widespread variation and rich genetic diversity. The identification of progeny with a high amino acid and caffeine content holds promise for future production and breeding, providing valuable theoretical references for the selection of parents in the creation of low-caffeine-content tea germplasm resources. This study systematically investigated the effects of dietary tea polyphenols (TPs, major bioactive polyphenols from Camellia sinensis with potent antioxidant and anti-inflammatory properties) on the growth performance and intestinal health of hybrid crucian carp HCC2 under chronic crowding stress. A low-density control group (44.4 fish/m3, basal diet without TPs) and four high-density crowding stress groups (222.2 fish/m3) were established, one fed the basal diet without TPs (CS) and three fed basal diets supplemented with 100 (CSLTP), 200 (CSMTP), or 400 (CSHTP) mg/kg TPs. We analyzed the impacts of TPs on growth performance, serum biochemical parameters, antioxidant capacity, expression of lipid metabolism-related genes, and intestinal microbiota composition. The results demonstrated that chronic crowding stress significantly suppressed the final body weight, weight gain rate, and specific growth rate of HCC2, while increasing serum lactate LDH, TG, and ALB and decreasing GLU, LDL-C, ALT, AST, and ALP levels. Dietary TPs supplementation enhanced antioxidant capacity (T-AOC, SOD, CAT, and GSH) and alleviated lipid metabolic disorders by activating the Nrf2/Keap1 and PPARα signaling pathways, thereby upregulating the expression of liver antioxidant genes (CAT and SOD) and fatty acid oxidation genes (CPT1 and acox1). Furthermore, intestinal microbiota analysis revealed that chronic crowding stress significantly increased the abundance of Proteobacteria and decreased the proportion of Firmicutes compared to the low-density control. Dietary TPs intervention, particularly at higher doses, partially restored the Firmicutes abundance and reduced the enrichment of potential pathogenic bacteria associated with stress. This study is the first to comprehensively elucidate the mechanism by which TPs alleviate crowding stress through enhanced antioxidant capacity, metabolic regulation, and microbiota remodeling, providing robust theoretical support for the application of plant-based additives in aquaculture.
