This study investigates the species composition and distribution of blackflies (Diptera: Simuliidae) in Kazakhstan, with a focus on two species newly recorded for the country: Simulium murmanum (Enderlein, 1935) and Simulium reptans (Linnaeus, 1758). The presence of S. murmanum in Kazakhstan is reported for the first time, supported by morphological and molecular genetic analyses. Diagnostic features of the larva, pupa, and adult stages are described in detail, including the structure and coloration of the larval head capsule, pupal cocoon, and genitalia of both sexes. Habitat preferences and pupal substrate attachment patterns are illustrated, with observations on variations in cocoon branching across different flow regimes. Species identification was conducted using the morphological keys of Rubtsov and Yankovsky, and taxonomic classification was confirmed using the framework proposed by Adler. Molecular confirmation of S. murmanum was performed via DNA analysis. The species was found to be restricted to the foothill regions of East Kazakhstan, suggesting a distribution closely associated with the Altai mountain systems and adjacent regions in Mongolia and China. Unlike its status as a dominant hematophagous species in parts of Russia, S. murmanum has not demonstrated biting activity in Kazakhstan, Mongolia, or China. Additionally, the study provides the first records of S. reptans within the fauna of Kazakhstan, initially identified in the Irtysh River (Pavlodar Region). Subsequent sampling conducted in June 2024 revealed a continuous distribution of S. reptans along the Irtysh River through to the mountain streams of East Kazakhstan. The species was found in mountainous, foothill, and lowland environments, highlighting its wide ecological plasticity. Full article

The genus Ajuga (Lamiaceae family) comprises approximately 300 species, which are widely used in traditional medicine for their diaphoretic, antiseptic, hemostatic, and anti-inflammatory properties, but scarcely in official ones. Therefore, the study of Ajuga reptans holds promise for developing new medicinal products. In aqueous and aqueous-alcoholic soft extracts of the A. reptans herb, 16 amino acids, 20 phenolics, and 10 volatile substances were identified by HPLC and GC/MS. The assays of the main substances’ groups were also determined by spectrophotometry (vitamin K1, polyphenols, tannins, flavonoids, and hydroxycinnamic acids) and titrometry (ascorbic and organic acids). A. reptans herb extracts are practically non-toxic, exhibit hepatoprotective activity (dose 25 mg/kg) in experimental carbon tetrachloride-induced hepatitis, moderate anti-inflammatory activity (dose 100 mg/kg) in carrageenan-induced edema models, and possess significant local hemostatic (reducing bleeding time by 40.6%) and wound-healing properties (complete wound healing after 9 days). The aqueous-ethanolic soft A. reptans extract (extractant 50% ethanol) demonstrated the most pronounced hepatoprotective and anti-inflammatory effects. A. reptans extracts are capable of inhibiting the growth of microorganisms and showing higher activity against Gram-positive bacteria. A. reptans herb extracts are promising agents for implementation in official medicine as wound healing and hepatoprotective remedies after further preclinical and clinical studies. Full article

Disease’s severity, mortality rates, and common failures to achieve clinical improvement during the unprecedented COVID-19 pandemic exposed the emergency need for new antiviral therapeutics with higher efficacy and fewer adverse effects. This study explores the potential to encapsulate multi-component plant extracts in liposomes as optimized delivery systems and to verify if they exert inhibitory effects against human seasonal betacoronavirus OC43 (HCoV-OC43) in vitro. The selection of Sambucus nigra, Potentilla reptans, Allium sativum, Aesculus hippocastanum, and Glycyrrhiza glabra L. plant extracts was based on their established pharmacological and antiviral properties. The physicochemical characterization of extract-loaded liposomes was conducted by DLS and electrokinetics. Encapsulated amounts of the extract were evaluated based on the total flavonoid content (TFC) and total polyphenol content (TPC) by colorimetric methods. The BALB 3T3 neutral red uptake (NRU) phototoxicity/cytotoxicity assay was used to estimate compounds’ safety. Photo irritation factors (PIFs) of the liposomes containing extracts were <2 which assigned them as non-phototoxic substances. The antiviral capacities of liposomes containing medicinal plant extracts against HCoV-OC43 were measured by the cytopathic effect inhibition test in susceptible HCT-8 cells. The antiviral activity increased by several times compared to “naked” extracts’ activity reported previously. A. hippocastanum extract showed 16 times higher inhibitory properties reaching a selectivity index (SI) of 58.96. Virucidal and virus-adsorption effects were investigated using the endpoint dilution method and ∆lgs comparison with infected and untreated controls. The results confirmed that nanoparticles do not directly affect the viral surface or cell membrane, but only serve as carriers of the active substances and the observed protection is due solely to the intracellular action of the extracts. Mesquite (Prosopis spp.) is one of the main plant representatives in regions with a dry climate, and is a fundamental part of the flora of the Mexican arid, with an indisputable importance from ecological, economic and industrial points of view. However, the restrictive factors of dry climates, as well as genetic variability, are sources of the great diversity of mesquite, so its taxonomy is not yet well defined. The present study seeks to determine the diversity of mesquite in the Mexican semi-desert based on the morphometric characterization of its leaves. Methods: Different leaf parameters such as the number, length and width of the leaflets were recorded in 31 well-marked sites in the area, to obtain measures of central tendency and dispersion, and to determine the differences and similarities between the sites, as well as the groups of homogeneous and heterogeneous taxa. Results: Five taxa were identified, of which there were two varieties (Prosopis glandulosa var. glandulosa and Prosopis reptans var. cinerascens), a pure or typical species (Prosopis laevigata) and two hybrids (Prosopis glandulosa var. glandulosa x Prosopis laevigata and Prosopis laevigata x Prosopis glandulosa var. glandulosa). The discriminant analysis indicated that five variables presented the highest percentage of separation or best separated the taxa, so the study was based on the phenogram with the combination of these five variables: (a) length of middle leaflets, (b) length of upper leaflets, (c) spacing of middle leaflets, (d) length/width relationship of middle leaflets and (e) length/width relationship of middle leaflets. Conclusions: Both the traditional taxonomic classification and the cluster and discriminant analyses reflected the same taxa (Prosopis glandulosa var. glandulosa, Prosopis laevigata and Prosopis reptans var. cinerascens) and the existence of hybridization between Prosopis glandulosa var. glandulosa and Prosopis laevigata. In this work, the phytochemical composition and the biological activity of the ethanolic extracts obtained from Ajuga reptans L. (Lamiaceae) leaves and roots (growing in Lesser Poland Voivodeship, Poland) were compared. The phytochemical composition of the extracts were determined by the high-performance liquid chromatography with diode-array detection (HPLC–DAD) method. The dominant compounds in both extracts were verbacoside, isoverbacoside, 3,4-dihydroxyphenylacetic acid and rosmarinic acid. The antioxidant capacity of the extracts was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP) and 2,2’-azobis(3-ethylbenzothiazoline-6-sulfonate (ABTS) tests. The Folin–Ciocalteu assay was used to determine the total polyphenolic content. Additionally, enzyme (tyrosinase, collagenase) inhibition tests and metal chelating ability were studied to assess the anti-aging properties of the extracts. Moreover, the A. reptans extracts’ capacity to absorb the whole range of ultraviolet radiation and high-energy visible (HEV) light was evaluated. The skin irritation test (SIT) EpiDerm was applied to evaluate the safety of the bugle extracts. The noteworthy point is that there is a lack of literature on the assessment of A. reptans root extract activity. Our study is the only one that compares the quantitative composition and biological activity of extracts from the root and leaves of A. reptans. The obtained results indicated that both of the extracts exhibit high antioxidant, chelating and photoprotective activity, but the extract from A. reptans roots showed a higher enzyme inhibition effect for mature skin. The A. reptans root extract, similarly to the leaf extract, could be applied as potentially multi-functional, safe and sensitive cosmetic raw materials, especially in anti-aging and anti-pollution cosmetics. Nature-based solutions are promising for climate adaptation and environmental management in urban areas, but urban conditions are stressful for vegetation. In particular, the interaction of drought and high temperatures may be detrimental. Guiding plant selection for urban greening with native species requires a far better knowledge of plant adaptations and stress acclimation. We tested the physiological responses of four candidate calcareous grassland species for green roofs and walls to the combined effects of drought and high temperatures under controlled conditions. The tested species proved relatively resistant to stress despite different strategies to protect the photosynthetic apparatus, maintain water balance, and repair damages. Based on the physiological responses, we rank the species in descending order of resistance to the stress factors tested: Trifolium medium > Festuca ovina > Carex flacca > Potentilla reptans, but all four can serve as potential candidates for green walls and roofs. Physiological stress screening of plant species for use on green roofs and walls supplements the habitat template approach to provide a stronger and wider base for prioritizations. Full article

Edaphic properties have been widely shown to influence community composition and distribution. However, the degree to which edaphic factors can affect grasses in semi-arid lowlands is still little researched. We assessed the significance of nine edaphic factors to explain the distributions of 65 grass species with various ecological traits (i.e., the ecological indicator values for their preferred habitat) in the semi-arid lowlands of Pakistan. To record information on species composition and related ecological conditions, we selected 10 random sampling locations between 2020 and 2021. For each species, we determined the important value index (IVI) and looked at the primary indicator species that were identified using the indicator species analysis approach. The major genera were Setaria, Brachiaria, and Cenchrus with 6.15% species in each followed by Aristida, Panicum, and Eragrostis with 4.61% wild grass species, Bothriochloa, Bromus, Phragmites, Polypogon, Saccharum, Poa, Echinochloa, and Dactyloctenium with 3.07% species, whereas other genera had a single species each. In total, 80% of the species were native, while only 20% were introduced species. Microphylls accounted for 49.23% of the leaf size spectra of the grass flora in the study area. The other frequent traits included macrophylls (21.53%), nanophylls (20%), and leptophylls (9.23%). The major life forms were therophytes (56.92%) followed by hemicryptophytes (38.46%) and geophytes (4.61%). The results of an ordination analysis indicated that the distribution of grasses was significantly (p ≤ 0.002) influenced by several edaphic parameters, with pH having the greatest impact on species distribution. The analyses of indicator species showed that pH and EC were the most powerful and important edaphic factors for determining the composition of plant communities and indicator species. The significant indicator species in various ecosystems were Cynodon dactylon (L.) Pers. rainer.bussmann@smnk.de and Leptochloa chinensis (L.) Nees (agroecosystem), Brachiaria reptans (L.) C.A. Gardner and C.E. Hubb, Dichanthium annulatum (Forssk.) Stapf, and Saccharum spontaneum L. (forest ecosystem), Cenchrus biflorus Roxb., Cenchrus ciliaris L., and Desmostachya bipinnata (L.) Stapf (urban ecosystem), Arundo donax L., Echinochloa crus-galli, and Phragmites australis (wetland ecosystem), and Saccharum spontaneum and Echinochloa crus-galli (L.) P. Beauv. (riparian ecosystem). We discovered that different species groupings had different habitat preferences and that soil pH had a significant beneficial effect on plant variety. These results provide a scientific roadmap for soil and plant restoration in semi-arid lowland habitats. Full article

Polyploidization in plants, which involves doubling or further multiplying of genome, has the potential to improve the constituents that make medicinal plants, like Ajuga reptans, attractive to the pharmaceutical, cosmetics, and food production industries; botanical pesticide effects could also be derived. The aim of this study was to determine how artificial polyploidization in A. reptans plants affected the composition and quantity of biologically active substances from the glycoside and phytoecdysone families. Diploids and artificial tetraploids of A. reptans were analyzed. Changes in the contents of trans-teupolioside, trans-verbascoside, and 20-hydroxyecdysone were evident in the aboveground parts of the cultivated plants (e.g., leaves and flowers). The tetraploid lines of Ajuga plants displayed variability in, and increased levels of, trans-teupolioside and trans-verbascoside content. The 20-hydroxecdysone content was slightly higher in tetraploids. These findings indicated that Ajuga tetraploids could be used in breeding programs to enhance the yield of substances with potential medicinal and industrial applications. Full article

The administration of natural antioxidants is considered to be a prevention strategy for chronic diseases and a useful tool for the healthcare system to reduce the administration of expensive and often not effective treatments. The chemical characterization of a methanolic extract (AJ) of Ajuga reptans L. was performed, and its antioxidant activity was evaluated. AJ and the major compounds, characterized by chromatographic techniques as phenylpropanoids and iridoids, were able to reduce the Reactive Oxygen Species levels in cancer cell lines (melanoma, A375, cervical cancer, HeLa, and alveolar adenocarcinoma, A549), stimulated by E. coli lipopolysaccharide. However, a clinical translation of these results encountered a significant limitation represented by the poor water solubility and bioavailability of the extract and compounds. Consequently, a hydro-soluble powder system (AJEP3) was developed by spray-drying encapsulating AJ into a multi-component solid matrix that is based on L-proline and hydroxyethylcellulose as loading and coating agents, and lecithin as solubility enhancer. The technological approach led to a satisfactory process yield (71.5%), encapsulation efficiency (99.9%), and stability. The in vitro water dissolution rate of the bioactive compounds appeared to be improved with respect to the extract, suggesting higher feasibility in the manufacturing and administration; even the in vitro biological activity of the produced multi-component AJEP3 was clearly enhanced. Several Ajuga species are used in Romanian folk medicine for their antioxidant, antimicrobial and anti-inflammatory properties, to treat pain, fever or arthritis. Still, the active compounds responsible for these effects and their mechanism of action are scarcely known. This research was designed to investigate the phytochemical profile (e.g. iridoids, polyphenolic compounds, phytosterols), as well as the biological potential (antioxidant, antibacterial, antifungal, anti-inflammatory properties) of two selected Ajuga species collected from different regions of Romanian spontaneous flora. The main compounds identified in A. reptans aerial parts extracts were 8-O-acetylharpagide, isoquercitrin and β-sitosterol, whilst in A. genevensis were 8-O-acetylharpagide, luteolin and campesterol. The extracts were screened for their antioxidant potential using different methods (DPPH, TEAC, EPR) and the results showed a good activity, in accordance with the polyphenol content (18–26 mg GAE/g dw). The antifungal activity on the tested strains was good. The determination of few parameters linked with the inflammatory mechanism allowed the assessment of in vivo anti-inflammatory potential. Ajuga reptans and A. genevensis ethanol extracts had anti-inflammatory activity through lowering the oxidative stress, phagocytosis, PMN and total leukocytes. The best anti-oxidative and anti-inflammatory activity was observed for the Ajuga reptans 100 mg dw/mL extract when compared with diclofenac, thus the dose could be correlated with the pharmacological effect. These findings provide substantial evidence that both selected Ajuga species have the potential to be valued as sources of phytochemicals in effective anti-inflammatory herbal preparations. Phytotoxic Activity of the 6-acetyl-5-hydroxy-2isopropenyl-2,3-dihydrobenzofurane (1) isolated from Trichocline reptans (Asteraceae) was investigated in two weed species. Results indicate that the best growth inhibition effect ocurres on Chenopodium album weed. Phythotoxic effect of the T. reptans chloroformic extract and of the benzofurane are discussed and compared in the two weed species. Full article
