Enhancing Sheep Vitality Through Diverse Pastures and Seaweed-Based Bio-Stimulants: Effects on Performance, Health, and Product Quality

This on-farm study explored the effects of diverse pasture systems and seaweed-based bio-stimulants (AgriSea NZ Seaweed Products, Paeroa, New Zealand) on sheep performance, metabolic health, milk composition, and carcass characteristics. A 3 × 2 factorial design was used to compare three pasture systems; ryegrass-white clover (RW), a 23-species diverse mix (DI), and functionally diverse strip swards (ST), with (SW) or without (CO) a seaweed-based bio-stimulant. Ninety pregnant ewes were stratified by live weight and allocated across six treatment groups (15 ewes per treatment). Lambing occurred on treatment paddocks. At weaning, 90 lambs (15 per treatment) were selected based on body weight and sex balance to continue through to finishing. Pasture chemical composition differed among treatments: ST had lower fibre (neutral detergent fibre, NDF; acid detergent fibre, ADF) than RW and DI, while SW increased dry matter digestibility (DMD) and metabolisable energy (ME), and reduced NDF and ADF (p < 0.05). Strip pastures improved lamb average daily gain (ADG) by 17% from lambing to weaning compared to DI, and by 14% from weaning to finishing compared to RW (p < 0.05). Seaweed-based bio-stimulant treatment enhanced lamb ADG by up to 12% and improved carcass traits, including loin and shoulder yields (p < 0.05). Ewes and lambs on seaweed-treated pastures exhibited lower serum non-esterified fatty acid (NEFA) concentrations (p < 0.05), indicating better energy balance. Milk from ST and/or SW treated ewes had elevated omega-6 fatty acids and essential amino acids, suggesting enhanced nutritional value. These findings demonstrate that combining botanical diversity with natural bio-stimulants can improve animal growth, metabolic health, and product quality, offering a promising strategy for sustainable and welfare-oriented sheep production systems. Due to the increased restrictions on antibiotic growth promoters (AGPs), natural substitutes such as organic selenium and prebiotics are gaining increased attention to enhance the gut health and performance of broilers. This study aimed at assessing the effects of organic selenium separately and in combination with mannan-oligosaccharides (MOSs) on gut development, immunity, carcass traits and overall growth performance in broiler chickens. For this study, 528-day-old straight-run broiler chicks (Ross-308) were assigned to four dietary treatments (six replicates of 22 birds each), in a completely randomized design under a 2 × 2 factorial layout, including: (1) Se0.2 (0.20 mg/kg organic selenium), (2) Se0.2 + MOS (0.20 mg/kg selenium + 1 g/kg MOS), (3) Se0.4 (0.40 mg/kg selenium) and (4) Se0.4 + MOS (0.40 mg/kg selenium + 1 g/kg MOS). The results showed that the lower level of organic selenium, along with MOS (Se0.2 + MOS), resulted in significantly enhanced (p ≤ 0.05) feed conversion ratio, body weight gain and livability. Whereas the higher level of organic selenium, along with prebiotic (Se0.4 + MOS), remarkably (p ≤ 0.05) improved carcass traits, immunity and villus height in broilers. In conclusion, the combined provision of 0.4 mg/kg organic Se and 1 g/kg MOS optimally enhances broiler performance, immunity and gut health, offering a promising substitute to AGPs. Full article

Background: Load carriage is an activity of daily living, can be an occupational risk, and is a popular method of resistance training. Type of carry could be related to vascular and blood pressure (BP) responses and may be related to cardiovascular risk. As physical activity is recommended by clinicians and exercise physiologists, understanding the vascular responses in relation to type of carry is important to consider in terms of risk. The purpose of this study was to compare the vascular health and BP responses to the farmer’s handle (load at the side of the body) and zercher (load in front of the body) carry and to examine sex differences in the responses. Methods: A total of 38 adults (22 females and 16 males) completed farmer’s handle and zercher load carriage with pre- and post-vascular measurements using pulse wave analysis. Results: We found that load carriage with weight in front of the body (zercher) elicits a larger increase in arterial stiffness (AIx@75), and load carriage with the load on the side of the body (farmer’s handle) elicits a larger increase in peripheral systolic BP. There were no sex differences in the responses to carry. Females exercise (3.1 vs. 4.8) and weight train (1.5 vs. 2.6) fewer times per week and had a lower resting systolic BP (121.5 ± 10.3 vs. 131.3 ± 8.3 mmHg) (p < 0.05). Both types of load carriage increased peripheral systolic BP, central systolic BP, and vascular stiffness (p < 0.05). Conclusions: Location of load carriage is important to consider based on potential cardiac risk of patients. Full article

The global burden of ischemic stroke requires a search for new factors that affect its risk and outcomes. Recent studies have shown that triiodothyronine could potentiate favorable stroke outcomes, but the reason for this is still unclear. To clarify the effects of the free triiodothyronine (fT3) level on stroke outcomes, we adopted a Mendelian randomization (MR) approach to evaluate their causal relationship. The genetic variants associated with the free triiodothyronine (fT3) level were obtained from the ThyroidOmics Consortium. Thirteen single-nucleotide polymorphisms, genetically predicting the fT3 level with a significance of p < 10−7, were adopted for MR analysis. Summary-level data for ischemic stroke outcomes (reported as a continuous variable, namely the modified Rankin score 3 months after stroke) was obtained from the GISCOME network. MR analyses were performed using the TwoSampleMR framework. The inverse-variance weighting method of MR analysis showed that a genetically predicted increase in fT3 level is associated with a reduction in ordinal Modified Rankin Scale scores (OR = 0.581, 95% CI 0.37–0.92, p = 0.0183). This study showed that higher fT3 levels could be causally associated with more favorable ischemic stroke outcomes and provides more evidence for the possibility of using thyroid hormone (TH) analogs to improve stroke outcomes. Background/Objectives: Intraoperative frozen biopsies are essential during surgery for Hirschsprung’s disease (HD). However, this method has several limitations with the need for a faster and real-time diagnostic alternative. For this, consistent histoanatomical and morphometric differences between aganglionic and ganglionic bowel must be established. The primary objective was to compare dimensions of bowel wall layers between aganglionic and ganglionic segments histopathologically in resected rectosigmoid specimens from children with HD. Secondary objectives were to design a diagnostic algorithm to distinguish aganglionosis from ganglionosis and assess whether full bowel wall thickness correlates with patient weight and age. Methods: Each histoanatomic bowel wall layer—mucosa, submucosa, and muscularis propria’s layers—was delineated manually on histopathological images. Mean thicknesses were calculated automatically using an in-house image analysis software. Paired parametric tests compared measurements in aganglionic and ganglionic segments. Results: Resected specimens from 30 children with HD were included. Compared to aganglionic bowel, ganglionic bowel showed a thicker muscularis interna (mean 0.666 mm versus 0.461 mm, CI −0.257–(−0.153), p < 0.001), and a higher muscularis interna/muscularis externa ratio (2.047 mm versus 1.287 mm, CI −0.954–(−0.565), p < 0.001). An algorithm based on these features achieved 100% accuracy in distinguishing aganglionosis from ganglionosis. No significant difference in full bowel wall thickness was found between aganglionic and ganglionic segments, nor any correlation with patient weight or age. Conclusions: Histoanatomic layer thickness differs between aganglionic and ganglionic bowel, forming the basis of a diagnostic algorithm. First-Trimester Clinical Characteristics and Pregnancy Outcomes in Women with Recurrent Pregnancy Loss

Objective: To describe first-trimester maternal, biochemical, biophysical, and ultrasound characteristics in women with recurrent pregnancy loss (RPL) compared to women without RPL. Methods: This was a retrospective cohort study analyzing data from 4440 pregnant women, including 142 women with previous RPL. Maternal and pregnancy characteristics, first-trimester biochemical markers, biophysical assessments, early-onset preeclampsia (EOPE) risk, and perinatal outcomes were compared. Results: Women with RPL were older (37.8 vs. 34.0 years, p < 0.001) and had higher rates of antiphospholipid syndrome (4.9% vs. 0.9%, p < 0.001), other thrombophilias (5.6% vs. 0.8%, p < 0.001), and thyroid disorders (14% vs. 7.5%, p = 0.010) than women without RPL. First-trimester uterine artery pulsatility index (UtA-PI) values, pregnancy-associated plasma protein-A (PAPP-A) levels, mean arterial pressure, and final risk for EOPE were comparable between groups. However, the RPL group had higher rates of very high risk for PE (10.6 vs. 5.1, p = 0.011). Likewise, second-trimester UtA-PI was higher in this group (1.10 vs. 1.01, p = 0.045). Aspirin and low molecular weight heparin prophylaxis were more frequent in women with RPL (23.8% vs. 9.6%, p < 0.001; 14.7% vs. 0.1%, p < 0.001). Regarding perinatal outcomes, we found a higher incidence of second-trimester intrauterine demise in the RPL group (6.4% vs. 1.4%, p = 0.011), with no other differences observed in the remaining outcomes. Conclusions: Women with RPL exhibit distinct maternal characteristics and worse pregnancy outcomes, although first-trimester markers do not seem to significantly differ from findings in women without RPL. These findings underscore the importance of tailored screening and intervention protocols to improve perinatal outcomes in this high-risk population. Full article

Background: Pre-pregnancy overweight and obesity are established risk factors for adverse maternal and perinatal outcomes in the general obstetric population. However, data regarding their impact in female liver transplant recipients remain limited. This study aimed to evaluate the association between pre-pregnancy body mass index (BMI) and pregnancy-related complications and neonatal outcomes in this high-risk cohort. Methods: A retrospective cohort analysis was conducted on pregnancies in liver transplant recipients who delivered between 2001 and 2022 at a single tertiary referral center. Participants were stratified into two groups based on pre-pregnancy BMI: normal weight (18.5–24.9 kg/m2) and overweight/obese (≥25 kg/m2). Maternal characteristics, pregnancy complications, and perinatal outcomes were compared using appropriate statistical methods, with significance set at p < 0.05. Results: Among 72 pregnancies included in the analysis, 48 (66.7%) were in women with normal BMI, and 24 (33.3%) were in those with an elevated BMI. No statistically significant differences were observed in gestational age at delivery, neonatal birth weight, Apgar scores, or incidence of preterm birth. Although pregnancy-induced hypertension and cesarean delivery were more prevalent among overweight/obese individuals, these differences did not reach statistical significance (PIH: 28% vs. 10.4%, p = 0.112; cesarean delivery: 76% vs. 64.6%, p = 0.465). Conclusions: In conclusion, pre-pregnancy overweight and obesity were not significantly associated with adverse obstetric or neonatal outcomes in liver transplant recipients. Nevertheless, the observed trends suggest a potential predisposition to hypertensive disorders (PIH: 28% vs. 10.4%, p = 0.112), underscoring the importance of individualized preconception counseling and weight optimization strategies in this high-risk patient population. Objective: To assess gestational diabetes mellitus (GDM) awareness among women in the Northern Borders Province of Saudi Arabia and identify factors associated with knowledge levels. Methods: A cross-sectional study was conducted among 461 women using a structured, validated online questionnaire assessing demographics, medical history, and knowledge of GDM’s effects on maternal and neonatal outcomes. Associations between demographic factors and GDM awareness were analyzed using chi-square tests. Results: Most participants (98.5%) were Saudi nationals, with a mean age of 34.5 ± 10.0 years; 82.6% had university-level education. Overall, 42.3% demonstrated fair awareness of GDM, 34.3% had good awareness, and 23.4% had poor awareness. Knowledge of GDM’s maternal complications varied: 70% recognized the risk of emergency cesarean section, but only 8.2% were aware of increased preeclampsia risk. For neonatal outcomes, 58.4% identified high birth weight as a risk, while only 31.9% recognized the risk of congenital anomalies. Higher awareness was significantly associated with education level (p = 0.02), pregnancy status (p = 0.001), trimester (p = 0.002), and family history of relevant conditions (p = 0.04). Conclusion: Although many women showed fair-to-good awareness of GDM, notable gaps remain, especially regarding specific complications. Targeted educational interventions, particularly for less-educated and non-pregnant women, are recommended to improve GDM awareness and pregnancy outcomes. Full article

Background: Rice bran proteins and their hydrolysates exhibit anticancer activity. Our previous study demonstrated that Riceberry glutelin and its hydrolysates possessed potent in vitro antioxidant and antimutagenic properties. However, their cancer chemopreventive effects in animals remain unclear. Methods: This study investigated chemopreventive mechanisms in diethylnitrosamine (DEN)- and 1,2-dimethylhydrazine (DMH)-induced preneoplastic lesions, including glutathione S-transferase placental form (GST-P)-positive foci in the liver and aberrant crypt foci (ACF) in the colon of rats. Rats received GTL, GTLH, and total protein hydrolysate (TPH) at 500 mg/kg body weight, five days per week for ten weeks. Results: GTLH significantly reduced GST-P-positive foci in the liver and ACF in the colon, while GTL decreased GST-P-positive foci only in the liver. However, TPH did not affect preneoplastic lesions in both the liver and the colon. GTLH suppressed cell proliferation by reducing proliferating cell nuclear antigen (PCNA)-positive cells and promoted apoptosis, as indicated by an increase in terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL)-positive cells in both organs. GTL also decreased PCNA-positive cells in the liver and colon. Moreover, GTLH significantly upregulated BAX and CASP3 in the liver, while only BAX in the colon was observed. Conclusions: This study highlighted the cancer chemopreventive potential of Riceberry GTLH with its underlying mechanism to reduce the number of preneoplastic lesions in the liver and colon through cell proliferation inhibition and apoptosis induction. These findings suggested that this protein hydrolysate might be used as a functional food ingredient or dietary supplement for cancer prevention. Full article

Background: Food addiction is a new clinical entity that is beginning to be linked to obesity and eating disorders. The present study aims to investigate the association between the risk of food addiction in children and the presence of obesity. It also explores the relationship between food addiction, the development of eating disorders, and body image dissatisfaction. Material and methods: A matched case-control study was conducted in a Spanish pediatric population (cases have obesity, and controls have normal weight). The main outcome measures were evaluation of food addiction (S-YFAS-C), child feeding attitudes (ChEAT), and evaluation of body image (CDRS). Additionally, sociodemographic and anthropometric data were gathered. Results: A total of 62 children were evaluated (31 cases with age 11 ± 0.7 years and BMI Z-score 2.89 ± 1.33; 31 controls with age 10.7 ± 0.8 years and BMI Z-score −0.05 ± 0.52). For all items on the S-YFAS-C scale, significant differences were observed between the two groups (∧ = 0.252, p = 0.002). Food addiction was diagnosed in 32.3% of cases (2.06 ± 1.7 symptoms) and 22.6% of controls (1.61 ± 1.6 symptoms), although no statistically significant differences were observed between groups. A statistically significant correlation exists between all the scores of the scales studied in the children. Conclusions: Children with obesity have a higher number of food addiction symptoms compared to those with normal weight. In general, as food addiction scores increase, higher scores are observed for the risk of developing eating disorders and body image dissatisfaction. Full article

Developing a more productive, resource-efficient, and climate-smart rubber agroforestry model is essential for the sustainable growth of natural rubber cultivation. In this study, we evaluated whether a double-row rubber plantation intercropped with the medicinal crop Ficus hirta Vahl. (DR-F) could achieve this goal, using a single-row rubber plantation (SR) as the control. We assessed the feasibility of the DR-F system based on productivity, solar utilization efficiency (SUE), partial factor productivity of applied nitrogen (PFPN), carbon efficiency (CE), net ecosystem carbon balance (NECB), and carbon footprint (CF). No significant difference was observed in rubber tree biomass between the DR-F (10.49 t·ha−1) and SR (8.49 t·ha−1) systems. However, the DR-F system exhibited significantly higher total biomass productivity (23.34 t·ha−1) than the SR systems due to the substantial contribution from intercropped Ficus hirta Vahl., which yielded 12.84 t·ha−1(p < 0.05). The root fresh weight yield of Ficus hirta Vahl. reached 17.55 t·ha−1, generating an additional profit of 20,417 CNY ha−1. The DR-F system also exhibited higher solar radiation interception and greater availability of soil nutrients. Notably, the roots of rubber trees and Ficus hirta Vahl. did not overlap at a 4 m distance from the rubber trees. The DR-F system achieved higher SUE (0.64%), PFPN (51.40 kg·kg−1 N), and CE (6.93 kg·kg−1 C) than the SR system, with the SUE and PFPN differences being statistically significant (p < 0.05). Although the NECB remained unaffected, the DR-F system demonstrated significantly higher productivity and a substantially lower CF (0.33 kg CO2·kg−1, a 56% reduction; p < 0.05). In conclusion, the DR-F system represents a more sustainable and beneficial agroforestry approach, offering improved productivity, greater resource use efficiency, and reduced environmental impact. In chickens, meat yield is a crucial trait in breeding programs. Identifying key molecular markers associated with increased muscle yield is essential for breeding strategies. This study applied transcriptome sequencing and machine learning methods to examine gene expression and alternative splicing (AS) events in muscle tissues of commercial broilers and local chickens. On the basis of differentially expressed genes (DEGs) and differentially spliced transcripts (DSTs) significantly related to breast muscle weight percentage (BrP), high-accuracy prediction models were developed by evaluating 10 machine learning models (e.g., eXtreme Gradient Boosting (XGBoost), Generalized Linear Model Network (Glmnet)). Feature importance was assessed using the Shapley Additive exPlanations (SHAP) method. The results revealed that 50 DEGs and 95 DSTs contributed significantly to BrP prediction. The XGBoost model achieved over 90% accuracy when using DEGs, and the Glmnet model reached 95% accuracy when using DSTs. Through Shapley evaluation, genes and AS events (e.g., ENSGALG00010012060, HINTW, and VIPR2-201) were identified as having the highest contributions to BrP prediction. Additionally, the breed effect was effectively mitigated. This study introduces new candidate genes and AS targets for the molecular breeding of poultry breast muscle traits, offering a paradigm shift from traditional gene mining approaches to artificial intelligence-driven predictive methods. Apparent diffusion coefficient (ADC) values, derived from diffusion-weighted magnetic resonance imaging (DW-MRI), increase with age, reflecting microstructural changes in white matter integrity. However, factors beyond chronological aging may influence cerebral diffusion characteristics. We investigated whether anticoagulant use is associated with favorable white matter ADC profiles, suggesting preserved microvascular health. ADC values were analyzed in cerebral white matter across four age-defined adult cohorts (20–59 years). Minimum, mean, and maximum ADC values were extracted. Patients at the lowest and highest ends of the ADC spectrum within each group were identified. The prevalence of anticoagulant use was compared between groups, and a logistic regression model adjusted for age was used to assess the independent association between anticoagulant use and lower ADC values. Across all cohorts (n = 892), anticoagulated patients (n = 89) were significantly overrepresented among individuals with low ADC values consistent with younger diffusion profiles. Of the anticoagulated patients, 93.3% had ADC values below the lower cut-off limit. In contrast, only 30% of non-anticoagulated patients exhibited such profiles. Anticoagulant use was independently associated with low ADC values after adjusting for age (OR = 4.89, p < 0.0001). Anticoagulation is strongly associated with lower, more favorable ADC values in cerebral white matter, independent of age. These findings support the potential neuroprotective role of anticoagulants and suggest that diffusion MRI may serve as a surrogate marker for early microvascular brain health. Full article

Background: Muscle-specific strength (MSS), defined as muscle strength relative to muscle mass, is a key indicator in the assessment of sarcopenia and functional status, especially in patients with cancer. Dual-energy X-ray absorptiometry (DXA) is the reference method for estimating muscle mass, but its limited accessibility hinders routine use. This study aimed to evaluate the correlation between DXA-based MSS and MSS estimated through more accessible techniques. Methods: A cross-sectional study was conducted in 205 adult oncology outpatients. Muscle strength was assessed by handgrip dynamometry, and muscle mass was estimated using DXA, bioimpedance (BIA), ultrasound, and anthropometry. MSS was calculated by dividing grip strength by each muscle mass parameter. Results: MSS calculated with BIA, body weight, and calf circumference showed very strong correlations with MSS-DXA (r > 0.90). Ultrasound-derived MSS showed only a moderate correlation (r = 0.55; p < 0.01). Similar patterns were observed in both men and women. Conclusions: BIA and anthropometric methods, particularly those using body weight and calf circumference, are reliable and accessible alternatives to DXA for estimating MSS in oncology patients. These tools may help improve the identification and monitoring of sarcopenia in clinical settings with limited resources. Full article

Background/Objectives: Normal-weight obesity describes those with a normal body mass index (BMI) and high body fat percent. Older adults with normal-weight obesity (NWO-O) are at increased risk for cardiovascular disease (CVD), but underlying mechanisms remain unclear. This pilot study examined whether NWO-O had an unfavorable cardiometabolic response to acute high-fat meal intake compared to normal BMI, low body fat percent individuals that were both older (NWL-O) and younger (NWL-Y). Methods: Participants (N = 29) with a normal BMI were grouped as follows: NWL-Y (18–35 years, low body fat percent; n = 12), NWL-O (≥60 years, low body fat percent; n = 9), and NWO-O (≥60 years, high body fat percent; n = 8). All participants completed an abbreviated fat tolerance test (75 g fat). Fasting and 4 h blood samples were collected to measure lipids (triglycerides and high-density lipoprotein cholesterol [HDL-C]), biomarkers of intestinal permeability (lipopolysaccharide binding protein [LBP] and soluble cluster of differentiation [sCD14]), and the inflammatory marker interleukin (IL)-6. Results: NWO-O had higher percent, absolute, and trunk fat compared to NWL-Y and NWL-O (p’s ≤ 0.01). Conversely, percent lean mass was lower in NWO-O versus both NWL groups (p’s ≤ 0.01). NWO-O had higher fasting triglycerides than NWL-Y (p < 0.05), but all groups were in the clinically normal range on average (≤107 mg/dL). However, NWO-O had higher 4 h triglycerides (239.4 ± 101.0 mg/dL) compared to NWL-Y and NWL-O (p < 0.01), consistent with an adverse response. The absolute change in triglycerides was higher in NWO-O relative to NWL-Y (p < 0.01), but not compared to NWL-O (p = 0.06). Fasting IL-6 was higher in NWO-O relative to NWL-Y (p < 0.05). Fasting and 4 h sCD14 were similarly higher in NWL-O and NWO-O versus NWL-Y (p’s < 0.01). Conclusions: NWO-O had an exaggerated postprandial triglyceride response compared to younger and similar-aged NWL individuals, which could reflect hepatic very low-density lipoprotein overproduction or impaired triglyceride clearance. Future work should continue to investigate the role of postprandial dyslipidemia in NWO-O’s reported CVD risk. Full article
