The present work aims to propose a new calibration strategy of the Hall–Thollet Body Force (BF) model to simulate the flow in multi-stage compressors and to capture inlet distortion effects within the machine. Both global (0D) and radial (1D) correction terms are introduced and calibrated to improve predictions in multi-stage compressors, accounting for highly interacting, highly loaded blades, falling outside the validity range of the model’s original coefficients. The modified model has been tested on the 3.5-stage high-pressure compressor CREATE, for which experimental data are available. The modified model is then employed to study different patterns of inlet distortion. The results show a very good agreement between Unsteady Reynolds-Averaged Navier–Stokes (URANS) simulations and Body Force calculations in terms of performance, key quantities along the radial and circumferential directions and distortion transfer across the compressor. Background: Among all studies describing COVID-19 clinical features during the first wave of the pandemic, only a few retrospective studies have assessed the correlation between olfac-tory dysfunction (OD) and the evolution of disease severity. The main aim was to assess whether OD is a predictive factor of COVID-19 severity based on the patient’s medical management (outpa-tient care, standard hospital admission, and ICU admission). Methods: A national, prospective, mul-ticenter cohort study was conducted in 20 public hospitals and a public center for COVID-19 screen-ing. During the first wave of the pandemic, from 6 April to 11 May 2020, all patients tested positive for COVID-19 confirmed by RT-PCR underwent two follow-up ENT consultations within 10 days of symptom onset. The main outcome measures were the evolution of medical management (out-patient care, standard hospital admission, and ICU admission) at diagnosis and along the clinical course of COVID-19 disease. Results: Among 481 patients included, the prevalence of OD was 60.7%, and it affected mostly female patients (74.3%) under 65 years old (92.5%), with fewer comor-bidities than patients with normal olfactory function. Here, 99.3% (290/292) of patients with OD presented with non-severe COVID-19 disease. Patients reporting OD were significantly less hospi-talized than the ones managed as outpatients, in either a standard medical unit or an ICU. Conclu-sions: As regards the clinical course of COVID-19 disease, OD could predict a decreased risk of hospitalization during the first wave of the pandemic. Full article

Research in the field of pharmacology aims to generate new treatments for pathologies. Nowadays, there are an increased number of chronic disorders that severely and durably handicap many patients. Among the most widespread pathologies, obesity, which is often associated with diabetes, is constantly increasing in incidence, and in parallel, neurodegenerative and mood disorders are increasingly affecting many people. For years, these pathologies have been so frequently observed in the population in a concomitant way that they are considered as comorbidities. In fact, common mechanisms are certainly at work in the etiology of these pathologies. The main purpose of this review is to show the value of anticipating the effect of baseline treatment of a condition on its comorbidity in order to obtain concomitant positive actions. One of the implications would be that by understanding and targeting shared molecular mechanisms underlying these conditions, it may be possible to tailor drugs that address both simultaneously. To this end, we firstly remind readers of the close link existing between depression and diabetes and secondly address the potential benefit of the pleiotropic actions of two major active molecules used to treat central and peripheral disorders, first a serotonin reuptake inhibitor (Prozac ®) and then GLP-1R agonists. In the second part, by discussing the therapeutic potential of new experimental antidepressant molecules, we will support the concept that a better understanding of the intracellular signaling pathways targeted by pharmacological agents could lead to future synergistic treatments targeting solely positive effects for comorbidities. Background: Intracranial solitary fibrous tumour (iSFT) is an exceptional mesenchymal tumour with high recurrence rates. We aimed to analyse the clinical outcomes of newly diagnosed and recurrent iSFTs. Methods: We carried out a French retrospective multicentre (n = 16) study of histologically proven iSFT cases. Univariate and multivariate Cox models were used to estimate the prognosis value of the age, location, size, WHO grade, and surgical extent on overall survival (OS), progression-free survival (PFS), and local recurrence-free survival (LRFS). Results: Eighty-eight patients were included with a median age of 54.5 years. New iSFT cases were treated with gross tumour resection (GTR) (n = 75) or subtotal resection (STR) (n = 9) and postoperative radiotherapy (PORT) (n = 32, 57%). The median follow-up time was 7 years. The median OS, PFS, and LRFS were 13 years, 7 years, and 7 years, respectively. Forty-two patients experienced recurrence. Extracranial metastasis occurred in 16 patients. Median OS and PFS after the first recurrence were 6 years and 15.4 months, respectively. A higher histological grade was a prognosis factor for PFS (p = 0.04) and LRFS (p = 0.03). GTR influenced LRFS (p = 0.03). Conclusion: GTR provided benefits as a first treatment for iSFTs. However, approximately 40% of patients experienced relapse, which remains a challenging state. The Chinese government introduced regulations to control emissions and reduce the level of NOx pollutants for the first time with the 12th Five-Year Plan in 2011. Since then, the changes in NOx emissions have been assessed using various approaches to evaluate the impact of the regulations. Complementary to the previous studies, this study estimates anthropogenic NOx emissions in 2015 and 2019 over Eastern China using as a reference the Hemispheric Transport of Air Pollution (HTAP) v2.2 emission inventory for 2010 and the new variational inversion system the Community Inversion Framework (CIF) interfaced with the CHIMERE regional chemistry transport model and OMI satellite observations. We also compared the estimated NOx emissions with the independent Multi-resolution Emission Inventory for China (MEIC) v1.3, from 2015. The inversions show a slight global decrease in NOx emissions (in 2015 and 2019 compared to 2010), mainly limited to the most urbanized and industrialized locations. In the locations such as Baotou, Pearl River Delta, and Wuhan, the estimations in 2015 compared to 2010 are consistent with the target reduction (10%) of the 12th Five-Year Plan. Comparisons between our emission estimates and MEIC emissions in 2015 suggest that our estimates likely underestimate the emission reductions between 2010 and 2015 in the most polluted locations of Eastern China. However, our estimates suggest that the MEIC inventory overestimates emissions in regions where MEIC indicates an increase of the emissions compared to 2010. Full article

Megafires occurred in Australia during the 2019/2020 bushfire season, leading to enhanced concentrations of many tropospheric pollutants. Here, we report on a fire plume with unusually high and persistent nitrous acid (HONO) levels that we could track during one day at free tropospheric levels over the Tasman Sea on 4 January 2020 using IASI and CrIS satellite observations. HONO concentrations up to about 25 ppb were retrieved during nighttime. Persistent HONO concentrations (>10 ppb) were still observed at sunrise, likely due to large aerosol concentrations within the plume, preventing HONO photodissociation. In addition, comparison with carbon monoxide (CO) measurements suggest a secondary production during the plume transport. Model simulations confirm that the plume is transported in the lower free troposphere with concentrations as high as 30 ppb at about 4 km. However, many uncertainties and unknowns remain in the plume aerosol load and in the chemical processes which may explain the model inability to reproduce elevated HONO concentrations at sunrise. The objective of this study is to assess and understand the NH3 recent trends and to identify the key components driving its concentrations. We have simulated the seasonal cycle, the interannual variability, and the trends in NH3 vertical column densities (VCD) from 2008 to 2015 over Europe, with the CHIMERE regional chemistry–transport model. We have also confronted the simulations against the Infrared Atmospheric Sounding Interferometer (IASI) satellite observations. IASI often shows a strong maximum in summer in addition to the spring peak, whereas CHIMERE only shows a slight peak in summer some years. This result could point to a misrepresentation of the temporal profile of the NH3 emissions, i.e., to missing emission sources during summertime either due to more than expected fertilizer use or to increased volatilization under warmer conditions. The simulated NH3 VCDs present an increasing trend over continental Europe (+2.7 ± 1.0 %/yr) but also at the national scale for Spain, Germany, UK, France, and Poland. Sensitivity tests indicate that these simulated positive trends are mainly due to (i) the trends in NH3 emissions, found heterogeneous in the EMEP NH3 emissions with strong disparities depending on the country, and (ii) the negative trends in NOx and SOx emissions. The impact of reductions in NO2 and SO2 emissions on NH3 concentrations should therefore be taken into account in future policies. This simulated NH3 VCD increase at the European scale is confirmed by IASI-v3R satellite observations in spring and summer, when ammonia emissions strongly contribute to the annual budget in accordance with crop requirements. Nevertheless, there are remaining differences about the significance and magnitude between the simulated and observed trends at the national scale, and it warrants further investigation. Full article

In the last decade there has been significant growth in the interest and application of using EEG (electroencephalography) outside of laboratory as well as in medical and clinical settings, for more ecological and mobile applications. However, for now such applications have mainly included military, educational, cognitive enhancement, and consumer-based games. Given the monetary and ecological advantages, consumer-grade EEG devices such as the Emotiv EPOC have emerged, however consumer-grade devices make certain compromises of data quality in order to become affordable and easy to use. The goal of this study was to investigate the reliability and accuracy of EPOC as compared to a research-grade device, Brainvision. To this end, we collected data from participants using both devices during three distinct cognitive tasks designed to elicit changes in arousal, valence, and cognitive load: namely, Affective Norms for English Words, International Affective Picture System, and the n-Back task. Our design and analytical strategies followed an ideographic person-level approach (electrode-wise analysis of vincentized repeated measures). We aimed to assess how well the Emotiv could differentiate between mental states using an Event-Related Band Power approach and EEG features such as amplitude and power, as compared to Brainvision. The Emotiv device was able to differentiate mental states during these tasks to some degree, however it was generally poorer than Brainvision, with smaller effect sizes. The Emotiv may be used with reasonable reliability and accuracy in ecological settings and in some clinical contexts (for example, for training professionals), however Brainvision or other, equivalent research-grade devices are still recommended for laboratory or medical based applications. We present a novel passive satellite remote sensing approach for observing the three-dimensional distribution of aerosols emitted from wildfires. This method, called AEROS5P, retrieves vertical profiles of aerosol extinction from cloud-free measurements of the TROPOMI satellite sensor onboard the Sentinel 5 Precursor mission. It uses a Tikhonov–Phillips regularization, which iteratively fits near-infrared and visible selected reflectances to simultaneously adjust the vertical distribution and abundance of aerosols. The information on the altitude of the aerosol layers is provided by TROPOMI measurements of the reflectance spectra at the oxygen A-band near 760 nm. In the present paper, we use this new approach for observing the daily evolution of the three-dimensional distribution of biomass burning aerosols emitted by Australian wildfires on 20–24 December 2019. Aerosol optical depths (AOD) derived by vertical integration of the aerosol extinction profiles retrieved by AEROS5P are compared with MODIS, VIIRS and AERONET coincident observations. They show a good agreement in the horizontal distribution of biomass burning aerosols, with a correlation coefficient of 0.87 and a mean absolute error of 0.2 with respect to VIIRS. Moderately lower correlations (0.63) were found between AODs from AEROS5P and MODIS, while the range of values for this comparison was less than half of that with respect to VIIRS. A fair agreement was found between coincident transects of vertical profiles of biomass burning aerosols derived from AEROS5P and from the CALIOP spaceborne lidar. The mean altitudes of these aerosols derived from these two measurements showed a good agreement, with a small mean bias (185 m) and a correlation coefficient of 0.83. Moreover, AEROS5P observations reveal the height of injection of the biomass burning aerosols in 3D. The highest injection heights during the period of analysis were coincident with the largest fire radiative power derived from MODIS. Consistency was also found with respect to the vertical stability of the atmosphere. The AEROS5P approach provides retrievals for cloud-free scenes over several regions, although currently limited to situations with a dominating presence of smoke particles. Cytokines are well known to play a central role in chronic rhinosinusitis with nasal polyps (CRSwNP), particularly in maintenance of the inflammatory response and the recruitment of eosinophils. The pathophysiological concepts concerning the involvement of inflammatory cytokines in CRSwNP have gradually evolved. Although the Th2 cytokines environment associated with an eosinophilic infiltration has retained a central role in the genesis of polyps, the role of other cytokine subpopulations has also and more recently been detailed, leading to a specific and complex signature in CRSwNP. The purpose of this review is to summarize the current state of knowledge about the cytokine signature in CRSwNP, the role of cytokines in the pathogenesis of this disease and in the intercellular dialog between epithelial cells, fibroblasts and inflammatory cells. Knowledge of this precise cytokine signature in CRSwNP is fundamental in the perspective of potential targeting biotherapies. Full article

Forty percent of somatotroph tumors harbor recurrent activating GNAS mutations, historically called the gsp oncogene. In gsp-negative somatotroph tumors, GNAS expression itself is highly variable; those with GNAS overexpression most resemble phenotypically those carrying the gsp oncogene. GNAS is monoallelically expressed in the normal pituitary due to methylation-based imprinting. We hypothesize that changes in GNAS imprinting of gsp-negative tumors affect GNAS expression levels and tumorigenesis. We characterized the GNAS locus in two independent somatotroph tumor cohorts: one of 23 tumors previously published (PMID: 31883967) and classified by pan-genomic analysis, and a second with 82 tumors. Multi-omics analysis of the first cohort identified a significant difference between gsp-negative and gsp-positive tumors in the methylation index at the known differentially methylated region (DMR) of the GNAS A/B transcript promoter, which was confirmed in the larger series of 82 tumors. GNAS allelic expression was analyzed using a polymorphic Fok1 cleavage site in 32 heterozygous gsp-negative tumors. GNAS expression was significantly reduced in the 14 tumors with relaxed GNAS imprinting and biallelic expression, compared to 18 tumors with monoallelic expression. Tumors with relaxed GNAS imprinting showed significantly lower SSTR2 and AIP expression levels. Altered A/B DMR methylation was found exclusively in gsp-negative somatotroph tumors. 43% of gsp-negative tumors showed GNAS imprinting relaxation, which correlated with lower GNAS, SSTR2 and AIP expression, indicating lower sensitivity to somatostatin analogues and potentially aggressive behavior. In France, apparently healthy dogs and cats that bite humans must undergo an observation period of 15 days with three veterinary visits to ascertain that they remain healthy, indicating that no zoonotic transmission of rabies virus occurred via salivary presymptomatic excretion. This surveillance protocol is mandatory for all pets that have bitten humans, despite France’s rabies-free status in non-flying mammals (i.e., a very low rabies risk). In this context, we aimed to perform a benefit–risk assessment of the existing regulatory surveillance protocol of apparently healthy biting animals, as well as alternative surveillance protocols. A scenario-tree modelling approach was used to consider the possible successions of events between a dog or cat bite and a human death attributed to either rabies or to lethal harm associated with the surveillance protocol (e.g., lethal traffic accidents when traveling to veterinary clinics or anti-rabies centers). The results demonstrated that the current French surveillance protocol was not beneficial, as more deaths were generated (traffic accidents) than avoided (by prompt post-exposure prophylaxis administration). We showed here that less stringent risk-based surveillance could prove more appropriate in a French context. The results in this study could allow policy-makers to update and optimize rabies management legislation. Full article

Bioindicators assess the mangroves ecological state according to the types of pressures but they differ with the ecosystem’s specificities. We investigated benthic meiofauna diversity and structure within the low human-impacted mangroves in French Guiana (South America) in response to sediment variables with various distances to the main city. Contaminant’s concentrations differed among the stations, but they remained below toxicity guidelines. Meiofauna structure (Foraminifera, Kinorhyncha, Nematoda) however varied accordingly. Nematode’s identification brought details on the sediment’s quality. The opportunistic genus Paraethmolaimus (Jensen, 1994) strongly correlated to the higher concentrations of Hg, Pb. Anoxic sediments were marked by organic enrichment in pesticides, PCB, and mangrove litter products and dominance of two tolerant genus, Terschellingia (de Man, 1888) and Spirinia (Gerlach, 1963). In each of these two stations, we found many Desmodora individuals (de Man, 1889) with the presence of epibionts highlighting the nematodes decreased fitness and defenses. Oxic sediments without contaminants were distinguished by the sensitive genera Pseudocella (Filipjev, 1927) and a higher diversity of trophic groups. Our results suggested a nematodes sensitivity to low contaminants concentrations. Further investigations at different spatio-temporal scales and levels of deterioration, would be necessary to use of this group as bioindicator of the mangroves’ ecological status. Full article

Background: Transcranial direct current stimulation (tDCS) is a safe, tolerable, and acceptable technique in adults. However, there is limited evidence for its safety in youth. Although limited, there are a handful of important empirical articles that have evaluated safety and tolerability outcomes in youth. However, a synthesis of pediatric safety studies is not currently available. Objective: To synthesize objective evidence regarding the safety and tolerability of pediatric tDCS based on the current state of the literature. Methods: Our search and report used PRISMA guidelines. Our method systematically examined investigations purposefully designed to evaluate the safety, tolerability, and acceptability of tDCS in healthy and atypical youth that were submitted to three databases, from the beginning of the database to November 2019. Safety considerations were evaluated by studies utilizing neuroimaging, physiological changes, performance on tasks, and by analyzing reported and objective side effects; tolerability via rate of adverse events; and acceptability via rate of dropouts. Results: We report on 203 sham sessions, 864 active sessions up to 2 mA, and 303 active hours of stimulation in 156 children. A total of 4.4% of the active sessions were in neurotypical controls, with the other 95.6% in clinical subjects. Conclusion: In spite of the fact that the current evidence is sporadic and scarce, the presently reviewed literature provides support for the safety, tolerability, and acceptability, of tDCS in youth for 1–20 sessions of 20 min up to 2 mA. Future pediatric tDCS research is encouraged. Full article

Dog and cat rabies cases imported from rabies enzootic countries represent a major threat for areas that have acquired rabies-free status and quantitative risk analyses (QRAs) are developed in order to assess this risk of rabies reintroduction through dog and cat movements. Herein we describe a framework to evaluate dog and cat rabies incidence levels in exporting countries along with the associated uncertainty for such QRAs. For enzootic dog rabies areas (EDRAs), we extended and adapted a previously published method to specify the relationship between dog rabies vaccination coverage and canine rabies incidence; the relationship between dog and cat rabies incidences; and then to predict annual dog and cat rabies incidences. In non-enzootic dog rabies areas (nEDRAs), we provided annual incidence based on declared dog and cat rabies cases. For EDRAs, we predicted an annual incidence potentially greater than 1.5% in dogs and about ten times lower in cats with a high burden in Africa and Asia but much lower in Latin America. In nEDRAs, the occurrence of rabies was lower and of similar magnitude in dogs and cats. However, wildlife could still potentially infect dogs and cats through spillover events. This framework can directly be incorporated in QRAs of rabies reintroduction. Full article
