The properties of Hardware-in-the-Loop (HIL) for the development of controllers, together with electronic emulation of physical process by Digital Twins (DT) significantly enhance the optimization of design and implementation in nonlinear control applications. The study emphasizes the use of the Raspberry Pi (RBP), a low-cost and portable electronic board for two interrelated goals: (a) the Electronic Control Unit (ECU-RBP) implementing a Lyapunov-based Controller (LBC) for nonlinear trajectory tracking of P3DX wheeled robots, and (b) the Digital Twin (DT-RPB) emulating the real robot behavior, which is remotely connected to the control unit. ECU-RBP, DT-RBP and real robot are connected as nodes within the same wireless network, enhancing interaction between the three physical elements. The development process is supported by the Matlab/Simulink environment and the associated packages for the specified electronic board. Following testing of the real robot from the ECU-RBP in an open loop, the model is identified and integrated into the DT-RBP to replicate its functionality. The LBC solution, which has also been validated through simulation, is implemented in the ECU-RBP to examine the closed-loop control according to the HIL strategy. Finally, the study evaluates the effectiveness of the HIL approach by comparing the results obtained from the application of the LBC, as implemented in the ECU-RBP to both the real robot and its DT. Antimicrobial resistance (AMR) has emerged as one of the most pressing global health challenges of our time. Alarming projections of increasing mortality from resistant infections highlight the urgent need for innovative solutions. While many candidates have shown promise in preliminary studies, they often encounter challenges in terms of efficacy and safety during clinical translation. This review examines cutting-edge approaches to combat AMR, with a focus on engineered antimicrobial peptides, functionalized nanoparticles, and advanced genomic therapies, including Clustered Regularly Interspaced Short Palindromic Repeats-associated proteins (CRISPR-Cas systems) and phage therapy. Recent advancements in these fields are critically analyzed, with a focus on their mechanisms of action, therapeutic potential, and current limitations. Emphasis is given to strategies targeting biofilm disruption and quorum sensing interference, which address key mechanisms of resistance. By synthesizing current knowledge, this work provides researchers with a comprehensive framework for developing next-generation antimicrobials, highlighting the most promising approaches for overcoming AMR through rational drug design and targeted therapies. Ultimately, this review aims to bridge the gap between experimental innovation and clinical application, providing valuable insights for developing effective and resistance-proof antimicrobial agents. Tanacetum parthenium (Asteraceae) has been traditionally used worldwide for medicinal purposes, and some of its therapeutic uses have been attributed to the pharmacological effects of its secondary metabolites. The root culture of this species might represent a sustainable source of several pharmacologically active compounds. The biomass of a root T. parthenium culture was extracted with methanol and fractionated using column chromatography. Three selected fractions (4TP, 5TP, and 8TP) were analyzed via spectrophotometric, chromatographic, and mass spectrometry techniques and in vitro pharmacological assays. The greatest values for total phenolic and phenolic acid contents and antibacterial activity against Escherichia coli were determined for 4TP. The highest values for total flavonoid and sesquiterpene lactone contents, antioxidant potential, and α-amylase inhibitory effect were determined for 8TP. The antibacterial effect against Staphylococcus aureus was not significantly different among the three fractions. The root culture of T. parthenium is a potential source of several metabolites, such as phenolic acids, fatty acids, coumarins, sesquiterpenoids, and triterpenoids, which are capable of exerting α-amylase inhibition and antioxidant, antibacterial, and cytotoxic effects. Among eight phenolic compounds detected and quantified in the fractions, chlorogenic acid was the most abundant. Full article

Background/Objectives: The incidence of cardiac morbimortality in patients with relapsed/refractory (R/R) acute myeloid leukemia (AML) is unknown. Methods: We analyze the characteristics, incidence, risk factors, and outcomes of cardiac events in AML patients treated for second-line (2L) or third-line (3L) episodes. Results: Among 327 2L AML patients (median age 62 years old), 135 experienced cardiac events, with an incidence of 38.6% non-fatal and 1.3% fatal events at 6 months. The grade 1–2 incidence was 16.8%, and the grade 3–4 incidence was 23.5% at 6 months. Overall, 207 cardiac events occurred in the 2L cohort, the most frequent being hypertension (n = 45), bradycardia (n = 39), QTc prolongation (n = 35), heart failure (n = 33), syncope/presyncope (n = 22), arrhythmia (n = 18), and myocardial ischemia (n = 8). Median OS in the 2L cohort was 9.4 months, 21.4 months in patients with grade 1–2, 8.8 months in patients without a cardiac event, 7.6 months in grade 3–4 patients, and 2.1 months with in 5 patients (p = 0.0035). The multivariate analysis showed prior cardiologic antecedents (p = 0.013), intensive 2L chemotherapy (p = 0.01), and inclusion in a 2L clinical trial (p < 0.001) as independent risk factors for non-fatal cardiac events. Among 189 patients of the 3L cohort, the incidence of non-fatal and fatal cardiac events was 49.2% and 0% at 6 months, respectively. Non-fatal cardiac events were more frequent in patients with prior cardiac antecedents (p = 0.004). Conclusions: In summary, cardiotoxicity is a frequent and challenging complication in R/R AML patients. We identified the risk factors that could be relevant to implementing risk-adapted management guidelines, aiming to reduce morbi-mortality in this difficult-to-treat setting. Full article

Osteoarthritis (OA) is a progressive joint disease that is commonly managed with palliative drugs, many of which are associated with undesirable side effects. This study investigated the therapeutic potential of a novel supplementation with guarana, selenium, and L-carnitine (GSC) in a rat model of chemically induced OA. Forty male Wistar rats (8–9 weeks old) received intra-articular sodium monoiodoacetate (Mia) to induce OA, and were subsequently treated with GSC. Inflammatory and oxidative stress parameters were analyzed at the end of the experiment. GSC supplementation enhanced endogenous antioxidant defenses, suggesting systemic antioxidant activity. However, no histological improvement was observed. In silico analyses indicated that Mia-induced OA may involve a complex molecular environment that GSC, at the tested dose, failed to modulate at the site of injury. Despite the limited local effects, these findings support the systemic benefits of GSC and highlight the potential of natural compound-based strategies in OA management. Given the adverse effects of conventional pharmacotherapy, the development of alternative, naturally derived treatments remains a promising avenue for future research. Full article

To study behavioral and productive factors to detect changes that may indicate and predict clinical mastitis cure, Holstein dairy cows (n = 60), in an automatic milking system (AMS) and equipped with behavioral monitoring collar, were monitored from the diagnosis of clinical mastitis (D0) until clinical cure. The parameters collected through sensors were feeding activity, milk electrical conductivity (EC), milk yield, Mastitis Detection Index (MDi), milk flow, and number of gate passages. Clinical mastitis cases (n = 22) were monitored and divided into cured cases (n = 14) and non-cured cases within 30 days (n = 8), paired with a control case group (n = 28). Cows were assessed three times per week, and cure was determined when both clinical assessment and California Mastitis Test (CMT) results were negative in three consecutive evaluations. Mixed generalized linear regression was used to assess the relationship between parameters and clinical mastitis results. Mixed generalized logistic regression was used to create a predictive model. The average clinical cure time for cows with clinical mastitis was 11 days. Feeding activity, gate passages, milk yield, milk flow, EC, and the MDi were associated with cure. The predictive model based on data from D0 showed an Area Under the Curve of 0.89 (95% CI = 0.75–1). Sensitivity and specificity were 1 (95% CI = 1–1) and 0.63 (95% CI = 0.37–0.91), respectively. The predictive model demonstrated to have good internal sensitivity and specificity, showing promising potential for predicting clinical mastitis cure within 14 days based on data on the day of clinical mastitis diagnosis. Three different vegetable oils, namely coconut oil, palm olein and olive oil, were analyzed for mineral oil hydrocarbons (MOHs) in our laboratory and in five commercial laboratories well recognized for their expertise in this field. The analysis consisted of a preliminary quantitative estimation of MOH content by hyphenated liquid chromatography–gas chromatography with flame ionization detection (LC-GC-FID), followed by a confirmatory analysis of MOH components by two-dimensional gas chromatography with time-of-flight mass spectrometry (GCxGC-ToF). The results provided by the six laboratories were compared to check their consistency, which would have led to a hypothetical commercial agreement or dispute scenarios, for instance. The comparison was based merely on information provided by the laboratories in their analytical reports (i.e., the methodology was not challenged, and chromatograms were not reviewed). Additionally, some of the laboratories were willing to provide some more information or details of the analysis. Similar quantitative results were provided by all six laboratories, emphasizing the utility of the current available harmonized guidelines and official standards for this method. However, as regards confirmatory results, discrepancies were observed among some laboratories in terms of the detection of MOH markers at low levels and the interpretation of GCxGC-ToF information. Even taking into account the limitation of this study as regards the reduced number of laboratories included, it highlights the need for harmonizing the GCxGC-ToF confirmatory method for MOHs in order to increase the alignment of results between laboratories for this kind of analysis. Deficits in social interaction and communication characterize Autism Spectrum Disorder (ASD). Although widely recognized by its symptoms, diagnosing ASD remains challenging due to its wide range of clinical presentations. Methods: In this study, we propose a method to assist in the early diagnosis of autism, which is currently primarily based on clinical assessments. Our approach aims to develop an early differential diagnosis based on electroencephalogram (EEG) signals, seeking to identify patterns associated with ASD. In this study, we used EEG data from 56 participants obtained from the Sheffield dataset, including 28 individuals diagnosed with Autism Spectrum Conditions (ASC) and 28 neurotypical controls, applying numerical techniques to handle missing data. Subsequently, after a detailed analysis of the signals, we applied three different starting approaches: one with the original database and the other two with selection of the most significant attributes using the PSO and evolutionary search methods. In each of these approaches, we applied a series of machine learning models, where relatively high performances for classification were observed. Results: We achieved accuracies of 99.13% ± 0.44 for the dataset with original signals, 99.23% ± 0.38 for the dataset after applying PSO, and 93.91% ± 1.10 for the dataset after the evolutionary search methodology. These results were obtained using classical classifiers, with SVM being the most effective among the first two approaches, while Random Forest with 500 trees proved more efficient in the third approach. Conclusions: Even with all the limitations of the base, the results of the experiments demonstrated promising findings in identifying patterns associated with Autism Spectrum Disorder through the analysis of EEG signals. Finally, we emphasize that this work is the starting point for a larger project with the objective of supporting and democratizing the diagnosis of ASD both in children early and later in adults. Human T-lymphotropic virus 1 (HTLV-1) infection has been associated with inflammatory, autoimmune, and lymphoproliferative diseases with a wide spectrum of clinical manifestations. Among patients with inflammatory rheumatological disease manifestations, cases of rheumatoid arthritis, Sjögren’s syndrome, polymyositis, and fibromyalgia, among others, have been reported. Another common feature of rheumatological diseases is the presence of joint manifestations, such as arthralgia and arthritis. In the present study, we sought to determine the laboratory profile and clinical rheumatological manifestations of people living with HTLV-1/2 residing in a metropolitan area in the Brazilian Amazon. A total of 957 individuals were screened for HTLV-1/2 infection by enzyme-linked immunosorbent assay (ELISA), and samples from seropositive individuals were subjected to infection confirmation by Western blotting or quantitative polymerase chain reaction (qPCR). Individuals with confirmed HTLV-1 and HTLV-2 infection were clinically evaluated for signs and symptoms of rheumatological diseases. Of the 957 individuals tested, 69 were positive for HTLV-1/2 infection, with 56 confirmed cases of HTLV-1 infection (5.9%), 12 of HTLV-2 infection (1.2%), and 1 classified as undetermined (0.1%). After clinical screening, 15 infected individuals with complaints suggestive of rheumatological disease were selected for evaluation by a rheumatologist (11 with HTLV-1 infection (1.1%) and 4 with HTLV-2 infection (0.4%)). The predominant pain pattern was symmetrical polyarthralgia, with large joints predominantly being affected. The diseases diagnosed were psoriatic arthritis, osteoarthritis, fibromyalgia, and regional pain syndromes. Antinuclear antibody (ANA) positivity was observed in two patients. Our findings confirm that HTLV-1 infection is associated with rheumatological disease manifestations and highlight the novel finding of cases of HTLV-2 infection in patients with rheumatoid arthritis symptoms. Methods: Patients with a confirmed diagnosis of acute decompensated heart failure were retrospectively enrolled in the study; admission NT-proBNP/chloride ratio was used to stratify patients above or below the median (>/<83). The primary composite endpoint consisted of cardiovascular mortality, decompensated HF readmission, and unplanned emergency department visits. Results: A total of 197 individuals were included, of whom 100 (50.7%) were classified above and 97 (49.2%) below the median. Patients showing a high ratio had a lower LVEF (31 vs. 39%), a higher proportion of previous MI (30 vs. 15%), a lower diastolic blood pressure (73 vs. 80 mmHg), and higher BUN (38 vs. 23 mg/dL) and creatinine (1.6 vs. 1.1 mg/dL). After a follow-up period of 92 ± 3 days, 46 patients (23%) presented the primary endpoint; those with a high NT-proBNP/chloride ratio showed an increased risk (HR 3.18, 95% CI 1.55–6.52, p = 0.0015) of the primary endpoint. After multivariate analysis, only serum NT-proBNP/chloride ratio (p = 0.02) and diastolic pressure (0.037) remained significant. The area under the ROC curve for the NT-proBNP/chloride ratio for predicting the primary composite endpoint was significantly superior when compared with AUC for NT-proBNP or chloride alone. Conclusions: The serum NT-proBNP/chloride ratio is a novel, easy to use predictor of short- and medium-term cardiovascular events in patients with acute decompensated HF. Full article

The popularity of small-scale and local fruit and vegetable production has increased in recent years due to perceived economic, environmental, and social benefits. However, these operations face contamination risks that both consumers and small-scale producers may underestimate. The present study aimed to assess the microbiological and chemical hazards on fruit, vegetables, soil, and water samples from small-scale farms in north-western Portugal during pre-harvest activities. Additionally, the study investigated farmers’ non-compliance with food safety regulations and good agricultural practices (GAPs), exploring how their behaviour might contribute to the identified hazards. A before-and-after analysis of non-compliant behaviours was conducted to determine the impact of training on improving food safety practices. The analysis identified the presence of pathogenic bacteria, pesticides, flame retardant residues, nitrates, and heavy metals. Lead (Pb) concentrations exceeded EU limits in organic carrots from one producer (0.156 ± 0.043 mg/kg) and in chard from another (0.450 ± 0.126 mg/kg). Cadmium (Cd) levels were also above regulatory thresholds in bell peppers (0.023 ± 0.009 mg/kg) and organic tomatoes (0.026 ± 0.015 mg/kg) from two different producers. Elevated levels of heavy metals were detected in irrigation water from two sites, with zinc (Zn) at 0.2503 ± 0.0075 mg/L and Pb at 0.0218 ± 0.0073 mg/L. Among food samples, the most prevalent microorganisms were Pseudomonas spp. (88.2%), Bacillus cereus (76.5%), and aerobic mesophilic bacteria (100%). Phosphorus flame retardants (PFRs), particularly tris(2-butoxyethyl) phosphate (TBEP), were detected in all food and soil samples. Some EU-banned pesticides were detected in food and soil samples, but at levels below the maximum residue limits (MRLs). Chlorpyrifos (35.3%) and p,p’-DDD (23.5%) were the most detected pesticides in food samples. After the training, GAP behaviour improved, particularly that related to hygiene. However, issues related to record-keeping and soil and water analyses persisted, indicating ongoing challenges in achieving full compliance. Full article

Rotavirus alphagastroenteritidis (R. alphagastroenteritidis) remains the leading cause of pediatric diarrhea. Although Angola introduced Rotarix®, the human monovalent R. alphagastroenteritidis vaccine since 2014 as part of its routine childhood immunization program, no follow-up study has been conducted. The aim of this study was to evaluate the distribution of R. alphagastroenteritidis genotypes among children under five years of age, hospitalized with acute gastroenteritis (AGE), after the introduction of the rotavirus vaccine. To achieve this goal, stool samples collected between 2021 and 2022 from children under 5 years of age diagnosed with AGE at six hospitals in Luanda Province were analyzed. The R. alphagastroenteritidis-antigen immunochromatographic test (SD Bioline™, Abbott, Chicago, IL, USA) was performed, and 121 positive samples were genotyped. Ten samples were randomly selected for further Sanger sequencing. The results showed that the G9P[6] was the most prevalent genotype (17.3%), followed by G9P[8] (16.5%), G2P[4] (14.9%), G3P[6] (13.2%), G8P[6] (11.5%), and less frequently G12P[8] (9.1%), G1P[6] (4.1%), and G1P[8] (2.5%). The genotype combinations G3P[6], G8P[6], and G12P[8] were detected for the first time in Luanda Province. In conclusion, the emergence of new genotype combinations supports the need for continuous surveillance to identify the trend in R. alphagastroenteritidis infection and the emergence of new strains circulating in Luanda Province in the post-vaccination period. Severe Acute Respiratory Syndrome (SARS) represents a significant cause of morbidity and mortality in children under one year of age, a particularly vulnerable population due to immunological and respiratory immaturity. The diverse etiology includes multiple respiratory viruses such as Respiratory Syncytial Virus (RSV), influenza, rhinovirus, and SARS-CoV-2, each with distinct potential to cause severe illness and death. Understanding the specific incidence and lethality by etiological agents in the recent Brazilian context (2024), after the COVID-19 pandemic, is essential to guide surveillance and public health strategies. This study aimed to analyze the risk of incidence and lethality by specific etiology of SARS in children under one year of age hospitalized in Brazil during the year 2024. A descriptive cross-sectional study was performed using secondary data from the 2024 Influenza Epidemiological Surveillance Information System (SIVEP-Gripe), obtained via OpenDataSUS. Reported cases of SARS hospitalized in children <1 year of age in Brazil were included. Distribution by final classification and epidemiological week (EW) was analyzed; the incidence rate by Federative Unit (FU) (cases/100,000 < 1 year) with risk classification (Low/Moderate/High) was assessed; and, for cases with positive viral RT-PCR, the etiological frequency and virus-specific lethality rate (deaths/total cases of etiology ×100), also with risk classification, were extracted. A multivariate logistic regression model was performed for the risk factors of death. A total of 66,170 cases of SARS were reported in children under 1 year old (national incidence: 2663/100,000), with a seasonal peak between April and May. The majority of cases were classified as “SARS due to another respiratory virus” (49.06%) or “unspecified” (37.46%). Among 36,009 cases with positive RT-PCR, RSV (50.06%) and rhinovirus (26.97%) were the most frequent. The overall lethality in RT-PCR-positive cases was 1.28%. Viruses such as parainfluenza 4 (8.57%), influenza B (2.86%), parainfluenza 3 (2.49%), and SARS-CoV-2 (2.47%) had higher lethality. The multivariate model identified parainfluenza 4 (OR = 6.806), chronic kidney disease (OR = 3.820), immunodeficiency (OR = 3.680), Down Syndrome (OR = 3.590), heart disease (OR = 3.129), neurological disease (OR = 2.250), low O2 saturation (OR = 1.758), SARS-CoV-2 (OR = 1.569) and respiratory distress (OR = 1.390) as risk factors for death. Cough (OR = 0.477) and RSV (OR = 0.736) were associated with a lower chance of death. The model had good calibration (Hosmer–Lemeshow p = 0.693) and overall significance (p < 0.001). SARS represented a substantial burden of hospitalizations, with marked seasonal and geographic patterns. RSV and rhinovirus were the main agents responsible for the volume of confirmed cases but had a relatively low to moderate risk of lethality. In contrast, less frequent viruses such as parainfluenza 4, influenza B, parainfluenza 3, and SARS-CoV-2 were associated with a significantly higher risk of death. These findings highlight the importance of dissociating frequency from lethality and reinforce the need to strengthen etiological surveillance, improve diagnosis, and direct preventive strategies (such as immunizations) considering the specific risk of each pathogen for this vulnerable population. Why Do Adolescents Skip Breakfast? A Study on the Mediterranean Diet and Risk Factors

Background/Objectives: Skipping breakfast is increasingly common among adolescents and has been associated with adverse health and academic outcomes. The average prevalence of breakfast skipping among adolescents is around 16%, although worldwide, it varies greatly across studies, ranging from 1.3 to 74.7%. This study aimed to assess the frequency of daily breakfast consumption and explore the factors associated with its omission in a stratified sample of Spanish adolescents. Methods: A cross-sectional study was conducted among 547 third-year secondary school students (aged 14–15) from both urban and rural areas in Castilla-La Mancha. Self-reported questionnaires were used to gather sociodemographic, psychosocial, and lifestyle data, including adherence to the Mediterranean diet (via the Kidmed questionnaire) and breakfast habits during school days. Descriptive, bivariate (Chi-square), and multivariate (binary logistic regression) analyses were conducted separately for boys and girls. Results: Findings showed a high prevalence of breakfast skipping one or more days (33.46%), with a significantly higher rate among girls (43.27%) than among boys (24.42%). Also, girls were more likely than boys to skip breakfast every day (14.18% vs. 6.87%, p < 0.001). In both groups, low adherence to the Mediterranean diet was strongly associated with skipping breakfast, along with higher screen time, shorter sleep duration, and being overweight/obese. Among girls, low olive oil consumption (OR 0.145 (CI 0.03–0.67) p 0.014) and poor Mediterranean diet adherence (OR 0.140 (CI 0.06–0.34) p < 0.001) were significant predictors. For boys, being overweight/obese (OR 2.185 (CI 1.06–4.52) p 0.035), low Mediterranean diet adherence (OR 0.136 (CI 0.06–0.32) p < 0.001), and not eating industrial pastries were associated factors (OR 0.413 (CI 0.20–0.88) p 0.022). Predictive models demonstrated good discriminatory power (AUC = 0.807 for girls; 0.792 for boys). Conclusions: Skipping breakfast is prevalent among adolescents, particularly girls, and is linked to poor dietary patterns and excess weight. These findings underscore the need for gender-specific nutritional interventions to promote regular breakfast consumption and improve dietary habits in adolescents. Full article

The Oropouche virus (OROV) is an arbovirus (Peribunyaviridae: Orthobunyavirus) that traditionally causes febrile outbreaks in Latin America’s Amazon region. Previously, OROV was not associated with severe pregnancy outcomes. During the 2022–2024 outbreak in Brazil, OROV expanded geographically, revealing links to adverse pregnancy outcomes. This study describes six cases with varied fetal outcomes, including miscarriage, antepartum, intrauterine fetal demise (IFD), and normal development, correlating with maternal symptoms but not symptom severity. Vertical transmission was confirmed by detecting OROV through RT-qPCR, ELISA, and immunohistochemistry in fetal tissues. Genome sequencing from an IFD case identified a novel reassortment pattern reported in the 2022–2024 outbreak. Severe encephalomalacia, meningoencephalitis, vascular compromise, and multi-organ damage were evident, underscoring the significant risk OROV poses to fetal development and emphasizing the need for further investigation.
