Dear Colleagues,
This Special Issue brings together current perspectives on the dynamics of DNA replication and repair in normal and malignant cells, and their broader consequences for genome stability and cancer therapy. Faithful DNA replication is essential for preserving genomic integrity, yet cancer cells frequently exploit or are constrained by alterations in replication timing, origin firing, and repair capacity. The contributions in this issue explore how defects in DNA replication and repair pathways create therapeutic vulnerabilities that can be exploited in targeted cancer treatment, and how ongoing replicative stress contributes to chromosomal instability, a hallmark of cancer that fuels tumor heterogeneity and evolution. Particular attention is given to the interplay between chromosomal instability and the tumor immune microenvironment, highlighting emerging links between genomic instability and response to immunotherapy. Additionally, this issue features work on the epigenetic mechanisms that regulate DNA replication and repair, underscoring how chromatin state and epigenetic marks shape genome maintenance and influence therapeutic outcomes. Together, these articles offer an integrated view of replication biology, repair mechanisms, and epigenetic regulation, with the aim of guiding future strategies for targeting genomically unstable tumors and improving patient outcomes.
