Special Issue Information

Dear Colleagues,

Valvular heart diseases (VHDs) represent a significant cause of cardiovascular morbidity and mortality worldwide, traditionally attributed to rheumatic, degenerative, or infectious processes. However, advances in cardiovascular genetics have revealed that genetic factors play a pivotal role in the development, progression, and clinical heterogeneity of many valvular disorders. Both inherited and sporadic forms of VHDs, including bicuspid aortic valve, arrhythmic mitral valve prolapse, and calcific aortic valve disease, have been linked to specific genetic variants and molecular pathways. Mutations affecting extracellular matrix remodeling, signaling pathways such as NOTCH, TGF-β, and WNT, as well as genes regulating valve morphogenesis and calcification, have provided new insights into disease mechanisms. Understanding the genetic basis of VHDs not only improves knowledge of pathophysiology but also opens avenues for early diagnosis, risk stratification, and personalized management. As genomic technologies continue to evolve, integrating genetics into clinical cardiology holds promise for improving prevention strategies and developing targeted therapies for valvular heart diseases.

This Special Issue invites contributions that explore this exciting frontier. We seek original research, comprehensive reviews, and insightful perspectives that connect genetic discoveries to molecular mechanisms, clinical diagnostics, and novel therapeutic strategies in the field of VHDs.
