Round 1

Reviewer 1 Report

This is a well written review. minor edits are needed.

1. Please define urothelial cancer in the abstract. Author talked about bladder cancer also in the text. The scope of the review needs to be cleared in the abstract and in the introduction.

2. In table 1, it is better if the authors can put more detail about other outcome and immune response columns. Such as, hoe radiotherapy alters expression f CD80, CD86, CCR5, CCR7? Up or down regulation? The author said, "Vaccine effects observer?" What type of effect?

3. What are the candidates among the ICD inducers described in table 1, are in clinical trials? what are the side effects? If there issues can be presented in a table form that will be easy to follow.

A: Thank you, we have edited accordingly.

  1. In table 1, it is better if the authors can put more detail about other outcome and immune response columns. Such as, hoe radiotherapy alters expression f CD80, CD86, CCR5, CCR7? Up or down regulation? The author said, "Vaccine effects observer?" What type of effect?

A: In table 1, the word “Expression” is changed to “upregulation”. An explanation of vaccination effect is added to table1 legend. Vaccination effect was already explained in the results under the subtitle 4.2 (line 213).

  1. What are the candidates among the ICD inducers described in table 1, are in clinical trials? what are the side effects? If there issues can be presented in a table form that will be easy to follow.

A: We separated clinical trials and made a new table (table 2). Side effects are described in table 2.

Reviewer 2 Report

The authors performed a scoping review on immunogenic death role in bladder cancer.

Bladder cancer is heterogeneous disease with largely different treatment in case of muscle invasive (radical cystectomy).

Overall, the manuscript sounds scientifically well. Notable, PRISMA guidelines were used to perform this review. Thus, a summary on the current knowledge  has been reported.

My suggestion is to underline the clinical impact of this disease in discussion. In particular, opportunity to develop new target of therapy (i.e. immunogenic cell death) able to avoid radical cystectomy may be of interest in the field of bladder cancer due to surgical risk and the common occurrence of the disease in octogenarians. Thus, can you cite the following studies:

              -  Multicenter Analysis of Postoperative Complications in Octogenarians After Radical Cystectomy and Ureterocutaneostomy: The Role of the Frailty Index - PMID: 31402279 DOI: 10.1016/j.clgc.2019.07.002.

A: Added to the first paragraph of discussion.
