Aquatic alien species (AAS) have had a major impact on freshwater ecosystems, including Lake Naivasha in Kenya. Here, the ecosystem has undergone tremendous changes and multiple species introductions over the past 100 years, and molluscs have experienced a major decline in species diversity. The East African Rift Lakes have experienced a steady rise in lake levels since 2010. We investigated the impact of recent ecosystem changes on the lakes’ molluscs by determining the current mollusc diversity and its composition. We also reconstruct the history of faunal change and turnover over the last 100 years and discuss the future of molluscs in the lake and the implications in a One Health context. The combined effects of rising water levels and the decline of the crayfish Procambarus clarkii are likely to be responsible for the resurgence of Lake Naivasha’s mollusc fauna. The current fauna consists of three global invaders of American origin, one of which is new to East Africa (Pseudosuccinea columella) and another of which has only recently begun to spread (Pomacea canaliculata). A further three species are native to Africa, two of which are known from historical records, while one is new to Lake Naivasha (Bulinus forskalii). All native species are of public health (Biomphalaria sudanica) and veterinary health (Bulinus tropicus, Bulinus forskalii) concern. The current biodiversity of molluscs has reached the same level as in historical times, but the species composition is remarkably different. Full article

Metabolism, mainly driven by oxygen consumption, plays a key role in life, as it is one of the main ways to respond to extreme temperatures through internal processes. Theba pisana, a widespread Mediterranean land snail, is exposed to a wide range of ambient temperature. In this species the oxygen consumption was tested as a response variable by multiple regression modelling on the “explanatory” variables shell-free mass, temperature, and relative humidity. Our results show that the oxygen consumption of T. pisana can be well described (73.1%) by these three parameters. In the temperature range from 23 °C to 35 °C the oxygen consumption decreased with increasing temperature. Relative humidity, in the range of 67% to 100%, had the opposite effect: if it increases, oxygen consumption will increase as well. Metabolism is proportional to an individual’s mass to the power of the allometric scaling exponent α, which is between 0.62 and 0.77 in the mentioned temperature range. CT scans of shells and gravimetry revealed the shell-free mass to be calculated by multiplying the shell diameter to the third power by 0.2105. Data were compared to metabolic scaling exponents for other snails reported in the literature. Full article

The problems associated with the greenhouse effect have increased the desire to limit carbon dioxide (CO2) emissions into the atmosphere, including emissions produced by shipboard electrical power systems. This has led to a further search for solutions in this area, such as shipboard hybrid electric power systems (SHEPSs). These systems do not yet have a wide application compared with traditional shipboard electrical power systems for several reasons, including the lack of studies establishing the fault tolerance of such systems. Therefore, in this work, problems related to generated power quality deterioration under large disturbances are studied. To achieve the fault-free operation of SHEPS, an emergency prevention control (EPC) system based on controlled parameter forecasting, along with a system structure and operation algorithm, is developed. The goals of improving electrical power quality are achieved by increasing the control efficiency of the power system’s generating sources. To validate the feasibility of the proposed control system, a computer simulation was carried out after developing a mathematical model of the SHEPS under study. The results of the study show that the use of the proposed EPC system will improve power quality when the controlled parameters are within acceptable limits. At the same time, further research is needed, as the problem of false control action as a consequence of EPC system hardware or software faults remains unstudied. Full article

This multiproxy work presents the archeozoological analysis of fish and microvertebrate remains from the Middle Bronze Age tell site of Borsodivánka (Borsod Plain, North-eastern Hungary). The fish faunal assemblage provides valuable data on the choice of exploited consumption patterns, taphonomy, and aquatic paleoenvironmental conditions at the site during the Bronze Age. Only freshwater taxa are present in the assemblage, for example, northern pike (Esox lucius); cyprinids: roach (Rutilus rutilus), common carp (Cyprinus carpio), common chub (Squalius cephalus) and common nase (Chondrostoma nasus); and percids: European perch (Perca fluviatilis) and pikeperch (Sander lucioperca). Herpetofaunal and micromammal remains are also part of this study, improving our knowledge of the site’s freshwater ecosystem. The grass snake (Natrix cf. natrix) and the European pond terrapin (Emys orbicularis), typical of aquatic ecosystems, are associated with the Aesculapian ratsnake (Zamenis longissimus), more typical of forest, shrubland, and grassland. The presence of amphibians such as toads (Bufo/Bufotes sp.) and frogs (Rana sp.) complete the herpetofaunal list. The microvertebrates also support a mature fluvial system, as represented by taxa like the European water vole (Arvicola amphibius). Other micromammals are present, such as the wood mouse (Apodemus sylvaticus), the group of the common/field vole (Microtus arvalis/agrestis), the European mole (Talpa europaea), and the house mouse (Mus musculus). All of them are common in forests, shrubland, and grassland. However, the commensal house mouse is more commonly associated with anthropogenic areas. In conclusion, Borsodivánka is characterized by a diverse landscape mosaic, displayed by the co-existence of a well-developed forest and a freshwater inland ecosystem with agricultural land in the wider area. Finally, the Tisza River and its flood plain represented the main water source close to the site, distinguished by the dominance of fish species from deep and slow-flowing waters. Full article

Background: At altitudes above 2500 m, the risk of developing high altitude pulmonary edema (HAPE) grows with the increases in pulmonary arterial pressure. HAPE is characterized by severe pulmonary hypertension, though the incidence and relevance of individual risk factors are not yet predictable. However, the systolic pulmonary pressure (SPAP) and peak in tricuspid regurgitation velocity (TVR) are crucial factors when diagnosing pulmonary hypertension by echocardiography. Methods: The SPAP and TVR of 27 trekkers aged 20–65 years en route to the Solu Khumbu region of Nepal were assessed. Echocardiograph measurements were performed at Lukla (2860 m), Gorak Shep (5170 m), and the summit of Kala Patthar (5675 m). The altitude profile and the participants’ characteristics were also compiled for correlation with the measured data. Results: The results showed a highly significant increase in SPAP and TVR after ascending Kala Patthar. The study revealed a lower increase of SPAP and TVR in the group of older participants, although the respective initial measurements at Gorak Shep were significantly higher for this group. A similar finding occurred in those using Diamox® as prophylaxis. There was an inverse relationship between TVR and SPAP, the peripheral capillary oxygen saturation, and heart rate. Conclusions: The echocardiograph results indicated that older people are an at-risk group for developing HAPE. A conservative interpretation of the basic tactical rules for altitudes should be followed for older trekkers or trekkers with known problems of altitude acclimatization (“slow acclimatizer”) as SPAP elevates with age. The prophylactic use of Acetazolamide (Diamox®) should be avoided where not necessary for acute medical reasons. Acetazolamide leads to an increase of SPAP, and this may potentially enhance the risk of developing HAPE. Arterial oxygen saturation measurements can provide an indicator for the self-assessment for the risk of developing HAPE and a rule of thumb for the altitude profile, but does not replace a HAPE diagnosis. Backpack weight, sex, workload (actual ascent speed), and pre-existing diseases were not statistically significant factors related to SPAP and TVR (p ≤ 0.05). Full article

Neuroblastoma is a rare disease. Rare are also the possibilities to test new therapeutic options for neuroblastoma in clinical trials. Despite the constant need to improve therapy and outcomes for patients with advanced neuroblastoma, clinical trials currently only allow for testing few substances in even fewer patients. This increases the need to improve and advance preclinical models for neuroblastoma to preselect favorable candidates for novel therapeutics. Here we propose the use of a new patient-derived 3D slice-culture perfusion-based 3D model in combination with rapid treatment evaluation using isothermal microcalorimetry exemplified with treatment with the novel carbonic anhydrase IX and XII (CAIX/CAXII) inhibitor SLC-0111. Patient samples showed a CAIX expression of 18% and a CAXII expression of 30%. Corresponding with their respective CAIX expression patterns, the viability of SH-EP cells was significantly reduced upon treatment with SLC-0111, while LAN1 cells were not affected. The inhibitory effect on SH-SY5Y cells was dependent on the induction of CAIX expression under hypoxia. These findings corresponded to thermogenesis of the cells. Patient-derived organotypic slice cultures were treated with SLC-0111, which was highly effective despite heterogeneity of CAIX/CAXII expression. Thermogenesis, in congruence with the findings of the histological observations, was significantly reduced in SLC-0111-treated samples. In order to extend the evaluation time, we established a perfusion-based approach for neuroblastoma tissue in a 3D perfusion-based bioreactor system. Using this system, excellent tissue quality with intact tumor cells and stromal structure in neuroblastoma tumors can be maintained for 7 days. The system was successfully used for consecutive drug response monitoring with isothermal microcalorimetry. The described approach for drug testing, relying on an advanced 3D culture system combined with a rapid and highly sensitive metabolic assessment, can facilitate development of personalized treatment strategies for neuroblastoma. Neuroblastoma, like other cancer types, has an increased need for energy. This results in an increased thermogenic profile of the cells. How tumor cells optimize their energy efficiency has been discussed since Warburg described the fact that tumor cells prefer an anaerobic to an aerobic metabolism in the 1920s. An important question is how far the energy efficiency is influenced by the substrate. The aim of this study was to investigate how the metabolic activity of neuroblastoma cells is stimulated by addition of glucose or fructose to the medium and if this can be measured accurately by using isothermal microcalorimetry. Proliferation of Kelly and SH-EP Tet-21/N cells was determined in normal medium, in fructose-enriched, in glucose-enriched and in a fructose/glucose-enriched environment. Heat development of cells was measured by isothermal microcalorimetry. The addition of fructose, glucose or both to the medium led to increases in the metabolic activity of the cells, resulting in increased proliferation under the influence of fructose. These changes were reflected in an enhanced thermogenic profile, mirroring the results of the proliferation assay. The tested neuroblastoma cells prefer fructose metabolism over glucose metabolism, a quality that provides them with a survival benefit under unfavorable low oxygen and low nutrient supply when fructose is available. This can be quantified by measuring thermogenesis. Full article

Phytochemicals, such as monoterpenes, polyphenols, curcuminoids, and flavonoids, are known to have anti-inflammatory, antioxidant, neuroprotective, and procognitive effects. In this study, the effects of several polyhydroxy flavonoids, as derivatives of differently substituted 5,7-dihydroxy-4H-chromen-4-one including apigenin, genistein, luteolin, kaempferol, quercetin, gossypetin, and phloretin with different lipophilicities (cLogP), as well as topological polar surface area (TPSA), were tested for induction of Ca2+ transients by α7 human nicotinic acetylcholine (α7 nACh) receptors expressed in SH-EP1 cells. Apigenin (10 μM) caused a significant potentiation of ACh (30 μM)-induced Ca2+ transients, but did not affect Ca2+ transients induced by high K+ (60 mM) containing solutions. Co-application of apigenin with ACh was equally effective as apigenin preincubation. However, the effect of apigenin significantly diminished by increasing ACh concentrations. The flavonoids tested also potentiated α7 nACh mediated Ca2+ transients with descending potency (highest to lowest) by genistein, gossypetin, kaempferol, luteolin, phloretin, quercetin, and apigenin. The specific binding of α7 nACh receptor antagonist [125I]-bungarotoxin remained unchanged in the presence of any of the tested polyhydroxy flavonoids, suggesting that these compounds act as positive allosteric modulators of the α7-nACh receptor in SH-EP1 cells. These findings suggest a clinical potential for these phytochemicals in the treatment of various human diseases from pain to inflammation and neural disease. Full article

Surrogate modeling techniques are widely used to replace the computationally expensive black-box functions in engineering. As a combination of individual surrogate models, an ensemble of surrogates is preferred due to its strong robustness. However, how to select the best quantity and variety of surrogates for an ensemble has always been a challenging task. In this work, five popular surrogate modeling techniques including polynomial response surface (PRS), radial basis functions (RBF), kriging (KRG), Gaussian process (GP) and linear shepard (SHEP) are considered as the basic surrogate models, resulting in twenty-six ensemble models by using a previously presented weights selection method. The best ensemble model is expected to be found by comparative studies on prediction accuracy and robustness. By testing eight mathematical problems and two engineering examples, we found that: (1) in general, using as many accurate surrogates as possible to construct ensemble models will improve the prediction performance and (2) ensemble models can be used as an insurance rather than offering significant improvements. Moreover, the ensemble of three surrogates PRS, RBF and KRG is preferred based on the prediction performance. The results provide engineering practitioners with guidance on the superior choice of the quantity and variety of surrogates for an ensemble. Full article

Agricultural technology transfer plays a chief role in transforming agricultural productivity in rural areas especially in the current setting where food demand surpasses the production capacity. Technology transfer facilitates the movement of soft and hard skills essential for improving farm production. Yet, the technical cooperation projects in Africa have been suffering from effectiveness and sustainability challenges while lacking responsiveness to local demand. This study applies a system dynamic method and a literature review to bring lessons from Japan and China’s experiences in agricultural technology transfer projects to Africa. Three cases in agricultural technical cooperation projects are presented: China—(Agricultural Technology Demonstration Center (ATDC) in Tanzania), Japan—(Smallholder Horticulture Empowerment Project (SHEP), and Rice Industry Development Support (RIDS) in Kenya and Tanzania, respectively). Japanese and Chinese agricultural technology has the potential to improve productivity and the livelihood of rural households. Nevertheless, strong linkages, commitment, and participation of all stakeholders in the design and implementation of agricultural technology transfer projects play an important role in enhancing project sustainability in the recipient countries. Further studies are recommended such as, to explain the agricultural technology transfer mechanism that fits well to equip beneficiary autonomy in terms of knowledge and capacity of production in the recipient country. The local governments need to set policy environments and institutional frameworks that encourage and support the agricultural technology transfer to benefit the rural farmers. Full article

The aim of the present study was to assess the effects of a 10 week shoulder home based exercise program (SHEP) on shoulder pain (SP) and range of motion (ROM) in a group of elite wheelchair basketball (WB) players. A convenience sample of elite WB players (n = 36, 15 males and 21 females), mean age of 26 years (SD 7.6, range 15–45)) were assigned to either an exercise or a control group, according to the use of the wheelchair during daily activities. The shoulder pain index for wheelchair basketball players (SPI-WB), functional tests and ROM were measured at baseline and after a 10 week intervention. In the analysis of the SPI-WB scores, for the exercise and control groups separately, there were no significant reductions of SPI-WB scores after intervention. Related to the analysis between groups after 10 weeks of intervention, there were no significant differences in changes between the exercise and control groups (Z = 0.840, p > 0.05, r = 0.743). In this regard, there was a significant change after the intervention for shoulder extension ROM (Z = 2.81, p ≤ 0.05, r = 0.249). Shoulder Pain did not increase along the 10 weeks of the SHEP development in WB players who reported SP before the intervention program. However, in those players who started the intervention without SP, as no increase in SP was observed and players were free of injury. An exercise program could be a tool to maintain shoulder health and prevent injuries in elite WB players. Full article

An outstanding question in developmental neurobiology is how RNA processing events contribute to the regulation of neurogenesis. RNA processing events are increasingly recognized as playing fundamental roles in regulating multiple developmental events during neurogenesis, from the asymmetric divisions of neural stem cells, to the generation of complex and diverse neurite morphologies. Indeed, both asymmetric cell division and neurite morphogenesis are often achieved by mechanisms that generate asymmetric protein distributions, including post-transcriptional gene regulatory mechanisms such as the transport of translationally silent messenger RNAs (mRNAs) and local translation of mRNAs within neurites. Additionally, defects in RNA splicing have emerged as a common theme in many neurodegenerative disorders, highlighting the importance of RNA processing in maintaining neuronal circuitry. RNA-binding proteins (RBPs) play an integral role in splicing and post-transcriptional gene regulation, and mutations in RBPs have been linked with multiple neurological disorders including autism, dementia, amyotrophic lateral sclerosis (ALS), spinal muscular atrophy (SMA), Fragile X syndrome (FXS), and X-linked intellectual disability disorder. Despite their widespread nature and roles in neurological disease, the molecular mechanisms and networks of regulated target RNAs have been defined for only a small number of specific RBPs. This review aims to highlight recent studies in Drosophila that have advanced our knowledge of how RBP dysfunction contributes to neurological disease. Full article
