Metabolic dysfunction-associated steatotic liver disease (MASLD) is one of the most prevalent chronic diseases in the world, lacking specific pharmacological interventions or well-established treatments. MASLD involves intricate pathological mechanisms characterized by oxidative stress and robust inflammatory responses. Selenium, an essential trace element, plays a critical role in antioxidation, regulation of inflammation, anticancer activity, and so on. Recent studies have reported that supplementation with selenium could alleviate MASLD and associated hepatic disorders, while excessive consumption may result in insulin resistance or even selenosis. Therefore, supranutritional selenium supplementation can be more suitable for the therapy and prevention of MASLD. This paper comprehensively reviews research about selenium and MASLD to highlight the potential applications and risks of supranutritional selenium supplementation in MASLD, following three steps: conducting a search, reviewing research articles and reviews, and discussing results. The keywords for the search include but are not limited to selenium, MASLD, supranutritional, hepatic diseases, selenoproteions, and selenium nanoparticles (SeNPs). We have reached the following conclusions: supranutritional selenium supplementation exhibits promising potential as a strategy to treat MASLD, but there are still some risks, depending on the dose and form of selenium; evaluating MASLD severity and selenium nutritional status accurately, as well as supplementing with superior forms of selenium (e.g., organic selenium and SeNPs), can further ensure the safety and efficacy of selenium supplementation. However, relationships between selenium homeostasis disorders and the occurrence and development of MASLD have not been fully elucidated. Methods for comprehensively assessing selenium status and mechanisms of selenosis require further investigation and research. Full article

(1) Background: The role of selenium in cancer biology remains poorly understood. Our aim was to study the course of selenium serum levels and the use of selenium supplements during breast cancer therapy. (2) Methods: Serum selenium levels, clinical–pathological data, selenium supplementation, and lifestyle factors were monitored quarterly over one year. (3) Results: A total of 110 non-metastatic breast cancer patients were enrolled in the prospective observational “BEGYN-1” study. At baseline, 2.9% of patients were selenium-deficient (<50 ng/mL), 1.9% were overdosed (>120 ng/mL), and 6.4% received substitution. The median selenium level was 81.5 ng/mL and ranged between 78.7 and 84.5 ng/mL within the year. A total of 25.3% of the patients received supplementation, resulting in significantly higher selenium levels (p < 0.05). A total of 8.7–28.6% of the patients using supplements were overdosed. Selenium levels strongly correlated with mushroom consumption (p = 0.003), but no association was found with therapy or clinical characteristics. (4) Conclusions: Although selenium deficiency is rare, serum selenium levels should be assessed in breast cancer patients. Mushrooms and nuts should be preferred over supplements to correct selenium deficiency. Ruling out selenium deficiency helps prevent the risk of selenosis and avoid unnecessary, costly supplementation in patients who are often financially burdened due to their disease. Full article

Selenium (Se) is an essential trace element for humans and animals, and an excess of or deficiency in Se is harmful to health. Research on the selenium enrichment zone began in the late 1970s in Shuang’an, Ziyang, southern Shaanxi Province. Naore village is only one selenosis area in Shuang’an, Ziyang, China. Different scholars have conducted systematic studies on the occurrence of selenium, its organic geochemistry and biomarkers, and its content and enrichment patterns in this area. This study applied the TIMA (TESCAN integrated mineral analyzer) for the first time to conduct detailed mineralogical work. The minerals included quartz, carbonate minerals (calcite and dolomite), feldspar (plagioclase, albite, and orthoclase), biotite and muscovite, clay minerals (chlorite and kaolinite), hematite, pyrite, and accessory minerals (almandine, olivine, zircon, and apatite) in Naore village, Ziyang, Shaanxi Province. The ATi index (100 × apatite/(apatite + tourmaline)) is used to determine the source of heavy minerals and the degree of heavy minerals’ weathering. The content POS (100 × (pyroxene + olivine + spinel)/transparent heavy mineral) of olivine, pyroxene, and spinel in heavy minerals can reflect the contribution of basic and ultrabasic rocks in the source area. The ATi and POS indexes for the heavy minerals in the research area were 91.83~99.96 and 0.01~18.75, respectively, reflecting the abundance of volcanic rock material in their source. In addition, the migration, transformation, bioavailability, and toxicity of selenium in the environment are closely related to its species. The species of selenium in various selenium-rich areas (Naore, Wamiao, and Guanquan) mainly include unusable residues and organic forms, followed by humic-acid-bound selenium. The proportions of water-soluble, exchangeable, and carbonate-bound selenium are relatively small, and the proportion of Fe-Mn oxide-bound selenium is the lowest. Full article

Selenium (Se)-enriched habitats have led to chronic selenosis, seriously affecting the health and survival of Procapra przewalskii (P. przewalskii). Our targets were to explore the molecular mechanisms of chronic selenosis and to look for a new way to protect endangered species. The mineral contents of soils, grass, blood, and muscle were analyzed. The biochemical indices, antioxidant capability, and immune function were also investigated. The analyses of proteomics and metabolomics were also carried out. The results showed that the Se contents in the muscle and blood of P. przewalskii, and the soil and grass in the Se-enriched habitats were significantly higher than those in healthy pastures. The P. przewalskii in the Se-enriched habitats showed symptoms of anemia, decreased antioxidant capability, and low immune function. A total of 44 differential proteins and 36 differential metabolites were screened by analyzing their proteomics and metabolomics. These differential proteins and metabolites were involved in glycolysis pathway, amino acid biosynthesis, carbon metabolism, phenylalanine metabolism, and energy metabolism. In particular, phenylalanine metabolism was the common pathway of proteomics and metabolomics, which was an important finding in studying the mechanism of chronic selenosis in animals. This study will help us to further understand the mechanism of chronic selenosis in P. przewalskii, and it provides a scientific basis for the protection of endangered species in Se-enriched habitats. The agricultural soils of West Limerick, Ireland, contain very localised, extremely high natural Se concentrations that reach levels that are very toxic to grazing livestock. The Carboniferous shales that formed in anoxic deep-water marine environments are the source of the selenium, which, along with the other redox-sensitive elements of molybdenum, uranium, arsenic and vanadium, were mobilised and reprecipitated in post-glacial anoxic marshes. The result has been a history of selenosis and molybdenosis in livestock in this important dairy province. Soils collected at 10–20 cm from five different agricultural sites were analysed, and all yielded concentrations greatly in excess of the safe Se limits of 3–10 mg kg−1; the highest value recorded was 1265.8 mg kg−1 Se. The highest recorded value for Mo in these soils was 1627.5 mg kg−1, and for U, 658.8 mg kg−1. There was a positive correlation between Se, Mo U and organic matter in the soils. Analysis of non-accumulator pasture grasses (Lolium perenne (perennial ryegrass), Festuca arundinacea (tall fescue), Dactylis glomerata (cocksfoot) and Phleum pretense (timothy grass)) revealed the shoot/leaf to contain up to 78.05 mg kg−1 Se while Trifolium repens (white clover) leaves contained 296.15 mg kg−1 Se. An in situ growing experiment using the Se accumulator species Brassica oleracea revealed 971.2 mg kg−1 Se in the leaves of premier kale, which also contained 1000.4 mg kg−1 Mo. Translocation factors (TFs) were generally higher for Mo than Se across all plant species. Combined X-ray absorption near edge spectroscopy (XANES) with micro-X-ray fluorescence (μ-XRF) showed the Se was present in the soil predominantly as the reduced immobile phase, elemental Se (Se0), but also as bioavailable organoselenium species, mainly selenomethionine (SeMet). SeMet was also the main species identified within both the Se non-accumulator and Se accumulator plants. The Se soil–plant system in West Limerick is dominated by SeMet, and uptake into the cattle pasture results in selenosis in the grazing dairy herds. The hyperaccumulating Brassica oleracea species could be used to extract both the Se and Mo to reduce the toxicity of the blighted fields. Full article

Selenium (Se) enrichments or deficiency in maize (Zea mays L.), one of the world’s most important staple foods and livestock feeds, can significantly affect many people’s diets, as Se is essential though harmful in excess. In particular, Se-rich maize seems to have been one of the factors that led to an outbreak of selenosis in the 1980s in Naore Valley in Ziyang County, China. Thus, this region’s geological and pedological enrichment offers some insight into the behavior of Se in naturally Se-rich crops. This study examined total Se and Se species in the grains, leaves, stalks, and roots of 11 maize plant samples, Se fractions of soils around the rhizosphere, and representative parent rock materials from Naore Valley. The results showed that total Se concentrations in the collected samples were observed in descending order of soil > leaf > root > grain > stalk. The predominant Se species detected in maize plants was SeMet. Inorganic Se forms, mainly Se(VI), decreased from root to grain, and were possibly assimilated into organic forms. Se(IV) was barely present. The natural increases of Se concentration in soils mainly affected leaf and root dry-weight biomasses of maize. In addition, Se distribution in soils markedly correlated with the weathered Se-rich bedrocks. The analyzed soils had lower Se bioavailability than rocks, with Se accumulated predominantly as recalcitrant residual Se. Thus, the maize plants grown in these natural Se-rich soils may uptake Se mainly from the oxidation and leaching of the remaining organic-sulfide-bound Se fractions. A viewpoint shift from natural Se-rich soils as menaces to possibilities for growing Se-rich agricultural products is also discussed in this study. Full article

This short review deals with the properties and significance of the determination of selenium, which is in trace amounts an essential element for animals and humans, but toxic at high concentrations. It may cause oxidative stress in cells, which leads to the chronic disease called selenosis. Several analytical techniques have been developed for its detection, but electroanalytical methods are advantageous due to simple sample preparation, speed of analysis and high sensitivity of measurements, especially in the case of stripping voltammetry very low detection limits even in picomoles per liter can be reached. A variety of working electrodes based on mercury, carbon, silver, platinum and gold materials were applied to the analysis of selenium in various samples. Only selenium in oxidation state + IV is electroactive therefore the most of voltammetric determinations are devoted to it. However, it is possible to detect also other forms of selenium by indirect electrochemistry approach. Full article

Next year (2017), the micronutrient Selenium (Se) is celebrating its birthday—i.e., 200 years after first being identified by the Swedish chemist Jöns Jakob Berzelius. Despite its impressive age, research into the functions of this essential trace element is very alive and reaching out for new horizons. This special issue presents some recent fascinating, exciting, and promising developments in Se research in the form of eight original contributions and seven review articles. Collectively, aspects of Se supply, biochemical, physiological, and chemotherapeutic effects, and geobiological interactions are covered by leading scientists in the areas of nutritional, basic, and clinical research. It is obvious from the contributions that the bicentennial anniversary will celebrate a micronutrient still in its infancy with respect to being understood in terms of its biomedical importance. Use of dietary supplements in the U.S. has increased steadily over the last 25 years. While misformulation is uncommon, the consequences can be serious. A March 2008 voluntary market recall removed supplement products responsible for the most serious selenium toxicity outbreak that has occurred in the U.S. We quantified selenium concentrations in the misformulated supplement products, measured the temporal response in the nail biologic monitor, and associated exposure to self-reported selenosis symptoms. Subjects recruited through state health departments and referrals provided samples of the misformulated supplement products, exposure information, monthly toenail and or fingernail clippings or onycholysitic nail fragments, and listed their newly onset adverse health effects attributed to selenium toxicity. Ninety-seven subjects enrolled and submitted at least one test sample. Peak selenium concentrations (up to 18.3 and 44.1 μg/g for toenails and fingernails, respectively) were measured. Multiple samples (52 total) of all six recalled supplement lots were analyzed ranging from 22,300 to 32,200 μg selenium per daily dose. Average consumption was 30.9 ± 13.9 doses; 73 subjects provided follow-up data on selenosis symptoms at 2.50 ± 0.14 years. Nail samples accurately reflect exposure in this selenium toxicity outbreak, which resulted in long-term/permanent adverse health effects. Full article

Enshi is a high selenium (Se) region in Hubei, China, where human selenosis was observed between 1958 and 1963. This study investigated the daily dietary Se intake of residents in Shadi, a town located 72 km northeast of Enshi City, to assess the risk of human selenosis in the high Se area. Foods consumed typically by the local residents and their hair samples were analyzed for total Se concentration. Concentrations of Se in different diet categories were as follows: cereals: 0.96 ± 0.90 mg kg−1 DW in rice and 0.43 ± 0.55 mg kg−1 DW in corn; tuber: 0.28 ± 0.56 mg kg−1 in potato and 0.36 ± 0.12 mg kg−1 in sweet potato; vegetables: ranging from 0.23 ± 1.00 mg kg−1 in carrot to 1.57 ± 1.06 mg kg−1 in kidney bean; animal proteins: 1.99 ± 1.11 mg kg−1 in chicken and egg. Based on the food Se concentrations and the daily per-capita consumption, the estimated daily Se intake in Shadi was 550 ± 307 µg per capita. Moreover, the Se concentrations in the hairs of local adult residents were 3.13 ± 1.91 mg kg−1 (n = 122) and 2.21 ± 1.14 mg kg−1 (n = 122) for females and males, respectively, suggesting that females might be exposed to higher levels of Se from daily cooking. Although there was no human selenosis occurrence in recent years, the high level of the daily Se intake suggested that the potential risk of selenosis for local residents, especially females, might be a matter of concern.
