Objectives: Anisakidosis is an emerging, cosmopolitan, and underdiagnosed parasitic disease caused by the accidental ingestion of third-stage anisakid larvae when consuming raw or improperly prepared seafood. Within hours to days of consuming infected raw seafood, patients may develop acute gastrointestinal symptoms including pain, nausea, vomiting, diarrhea and/or constipation, as live anisakid larvae attach to the gastric, or more rarely, the intestinal mucosa. Cases have been reported in which the nematodes succeed at migrating from the stomach upwards to the esophagus and then the oral cavity. Therefore, the purpose of the present literature review is to collect, analyze, summarize and present the relevant epidemiological, clinical, diagnostic, parasitological, therapeutic, and prognostic data concerning anisakidosis localized inside the oral cavity. Methods: An electronic search of the PubMed, Scopus, and Ovid databases was performed with them being accessed for the last time on 29 March 2025. Results: The present literature review identified 13 individual case reports of oral mucosa anisakidosis, which were published in the period 1971–2022. Conclusions: Our review aims to summarize the relevant epidemiological, clinical, diagnostic, parasitological, therapeutic, and prognostic data regarding the oral localization of anisakidosis, a helminthic infection caused by the accidental ingestion of live anisakid larvae and which manifests mainly with gastrointestinal symptoms. Its localization in the oral mucosa appears to be exceptionally rare and, in most cases, occurs with a characteristic clinical picture, defined by the onset of acute mouth or throat pain immediately after the consumption of raw seafood and by the observation of one or more larvae, either lying on or penetrating the oral mucosa. Despite its rarity, dental health professionals and other clinicians should be aware of this disease and the possibility of its intraoral localization, since environmental factors on the one hand, and the adoption of foreign dietary habits on the other, will likely make anisakidosis a much more common disease worldwide in the near future. The Atlantic chub mackerel, Scomber colias, is sold in Spain, as well as another species of the genus, like the Atlantic mackerel, S. scombrus, often mistaken to be the same fish. This study aims to analyse the risk of human anisakiosis through the consumption of S. colias, and to clarify if these two species differ regarding this risk. A total of 250 S. colias (125 originating from the Atlantic and 125 from the Mediterranean) were helminthologically analysed using conventional parasitological techniques. Concerning Ascaridoid nematodes, the prevalence of Anisakis type I was higher in the Atlantic than in the Mediterranean. The presence of three other helminth parasites stands out, two other Ascaridoid nematodes, the larvae of Contracaecum spp. and Hysterothylacium spp., the latter a food-spoiling and non-pathogenic parasite. The other helminth found was the intestinal adult of the acanthocephalan Rhadinorhynchus pristis, also non-pathogenic. The comparison of the current results with previously published ones by our research group on S. scombrus show that Anisakis type I prevalence was higher in S. scombrus than in S. colias, making the correct differentiation between them necessary, given their varying risk of human anisakiosis. Furthermore, fish label information is important to prevent the risk of anisakiosis for consumers. Full article

Ecdysterone controls moulting and reproduction in insects, crustaceans, and helminths. It is also produced by many plants, probably as an insect deterrent. The steroid is not made by vertebrates but has anabolic effects in mammals and could be useful for treating sarcopenia in aged horses. However, ecdysterone is banned in horseracing and equestrian sports, and with no limit of reporting, the risk of unintended exposure to this naturally occurring prohibited substance is a concern. To explore this risk, pasture plants and hay samples were analysed for ecdysterone content, as well as samples of blood, faeces, and intestinal mucosa from horses (euthanized for non-research purposes) with varying degrees of endo-parasite infestation. The variability in serum ecdysterone concentrations between different horses after administering a fixed dose was also examined. Ecdysterone was detected in 24 hay samples (0.09 to 3.74 µg/g) and several weeds, with particularly high concentrations in Chenopodium album (244 µg/g) and Solanum nigrum (233 µg/g). There was a positive correlation between faecal ecdysterone and faecal egg counts, but no effect of anthelmintic treatment and no relation to the number of encysted cyathostome larvae in the large intestine mucosa. Certain horses maintained an unusually high serum ecdysterone concentration over several weeks and/or displayed an abnormally large response to oral ecdysterone administration. Thus, the risk of environmental exposure to ecdysterone is apparent, and several factors must be considered when determining an appropriate dosage for clinical studies or setting a reporting threshold for equine sports. Estimating the human impact on parasites in wildlife is essential to create efficient strategies to control diseases and improve public health. This study assessed the human influence on the parasite burden of the two commonest canine mesocarnivores, the red fox (N = 194) and the golden jackal (N = 151), in Europe. We compared the infracommunity crowding index, parasite diversity, prevalence, and abundance of different parasite taxa, and the presence of Echinococcus multilocularis between groups of hosts, such as jackals and foxes hunted at different distances from human settlements. The analysis of hosts’ distribution ascertained that 61.2% of the investigated wild canids were hunted in a 500–1750 m ring around the human settlements. Ancylostomatidae proved the most prevalent taxon (>60%), while Echinococcus multilocularis and Dipylidium caninum occurred least frequently. The two taxa that mostly contributed to the crowding of infracommunities were Ancylostomatidae and Toxocaridae, with a negative correlation between their abundances (Spearman’s ρ = −0.445, p < 0.001). Parasite infracommunities were more crowded in Echinococcus multilocularis infected foxes (p = 0.02) and jackals (p = 0.001). Our study highlighted that human modification of landscapes worsened the parasite burden of wild mesocarnivores, which could increase the public health risk of zoonotic parasites. Full article

Feline parasitism affects animals’ health and welfare. Faeces from 472 client-owned cats from Greece were examined to provide updated data on the epizootiology of metazoan endo- and ectoparasites (namely, Toxocara cati, Ancylostomatidae, Dipylidium caninum, lungworms, Toxascaris leonina, Otodectes cynotis, fleas, ticks and Notoedres cati). All positive animals received a topical formulation containing esafoxolaner, eprinomectin and praziquantel (NexGard® Combo, Boehringer Ingelheim), and its efficacy was evaluated. The overall prevalence of parasitism was 22.9%, while that of multiparasitism was 16.3%. Toxocara cati (18.4%) was the most prevalent endoparasite, followed by Ancylostomatidae (10.8%), D. caninum (4.7%), lungworms (2.5%) and T. leonina (0.4%). Regarding ectoparasites, O. cynotis (3.2%), fleas (2.3%), ticks (0.6%) and N. cati (0.4%) were found. To estimate the efficacy of treatment, the geometric means of the number of parasitic elements before the first treatment and post-treatment, (i) 14 days for intestinal helminths, (ii) 28 and 56 days for lungworms and (iii) 28 days for O. cynotis and fleas, were estimated and compared. Following statistical analyses (paired t-test and McNemar’s test), an efficacy of 100% was recorded against the most commonly detected parasites (gastrointestinal helminths and mites) and a notable statistically significant effect against fleas and lungworms after one dose, while 100% efficacy against lungworms was achieved after two doses of the product. No adverse effects were reported. The prevalence of parasitism in owned cats in Greece remains high, highlighting the demand for targeted preventive antiparasitic schemes. This study demonstrated high-level efficacy and tolerance of NexGard® Combo against common endoparasites and ectoparasites of household cats in Greece. Full article

Elasmobranchs (Chondrichthyes, Elasmobranchii) are exposed to a variety of gastrointestinal parasites acquired through the ingestion of infected prey. An increasing amount of evidence suggests the usefulness of parasitological information to elucidate aspects of the biology and ecology of sharks and rays, to inform the correct management and conservation of their stocks and the appropriate husbandry of captive specimens. This study aims to identify at the morphological and molecular level the helminth parasites found in the stomachs and intestines of various elasmobranchs accidentally caught by Mediterranean fisheries, with the aim of updating and providing new information on the parasitic fauna of these species. Specimens of smooth-hound Mustelus mustelus, blackspotted smooth-hound Mustelus punctulatus, blue shark Prionace glauca, spiny dogfish Squalus acanthias, lesser-spotted dogfish Scyliorhinus canicula, pelagic stingray Pteroplatytrygon violacea and Mediterranean starry ray Raja asterias were examined. The parasitological examination allowed us to identify the nematode Acanthocheilus rotundatus in the two species of smooth-hounds analyzed, the tapeworm species Scyphophyllidium exiguum, S. prionacis, Anthobothrium caseyi and Nybelinia indica in P. glauca, the nematodes Hysterothylacium aduncum and Proleptus obtusus in S. acanthias and S. canicula, respectively, and finally the nematode Pseudanisakis rajae and the tapeworm Nybelinia sp. in Raja asterias. Some observations represent new reports at a geographical level, in particular, those on A. caseyi in P. glauca and H. aduncum in S. acanthias from the Adriatic Sea, or first host records, such as S. exiguum and N. indica in P. glauca or P. rajae. in R. asterias. The results of this survey represent a contribution to broadening the knowledge of the parasitic fauna of these elasmobranchs in the Mediterranean Sea. From more in-depth future studies, it will be possible to reach more solid evidence and general conclusions on aspects relating to the biology, ecology, and health of the investigated species, offering useful information for their conservation and management. The prevalence of human intestinal helminth parasitic infections is extensive, with over half of the global population estimated to suffer from these infections. Traditionally, various plant species, including Ricinus communis L. (Euphorbiaceae), are used to treat helminth infections. In this study, ricinoleic acid was isolated from the base hydrolysate of the petroleum ether seed extract of R. communis using column chromatography and transformed into ricinoleic acid methyl ester through esterification. The extract, ricinoleic acid and its methyl ester were evaluated for their anthelmintic activities against the model organism Caenorhabditis elegans. The results revealed that at a concentration of 1 mg/mL, ricinoleic acid and its methyl ester killed 97.40% and 97.83% of C. elegans worms, respectively. Molecular docking studies of ricinoleic acid on succinate dehydrogenase (SDH), glucose-6-phosphate 1-dehydrogenase (G6PD), and tubulin beta-2 chain (TBB2C) revealed that ricinoleic acid has a more favorable interaction with succinate dehydrogenase (−5.408 kcal/mol) compared to glucose-6-phosphate 1-dehydrogenase (−3.758 kcal/mol) and tubulin beta-2 chain (−1.444 kcal/mol). Furthermore, Absorption, Distribution, Metabolism, and Excretion (ADME) analyses unveiled that ricinoleic acid adheres to Lipinski’s rule of five, positioning it as a potential compound to treat helminths. The current study demonstrated that R. communis seed oil possesses genuine anthelmintic activity against C. elegans, which is likely due to ricinoleic acid. Full article

Interactions between parasites and hosts are not fully understood, though the dynamic pattern of infection and reinfection in humans varies with different demographic variables and behavioral changes. A community-based non-equivalent control group post-test-only design, an aspect of quasi-experimental design (QED), was carried out between March 2019 and February 2020. For the extraction of data from respondents, structural questionnaires were filled. Their CD4 count and viral load from the database of the National Health Laboratory Services, Mthatha were recorded. The method applied for the identification of intestinal parasites was a direct examination of the stool and the use of concentration methods. The post-test analysis showed that the intervention sites that received THEdS (Treatment, Health education, and Sanitation) bundle had a cure proportion of 60% and a re-infection proportion of 40%. The post-test results on control sites (treatment-only group) showed that the cure proportion was 51.4% and the re-infection proportion was 48.6%. The viral load significantly reduced from 377 to 44 copies/mL with a significant increment in CD4 count from 244 to 573 (cells µL) and (p-value) = 0.002. The combination of THEdS is an effective measure to reduce infection and reinfection of intestinal parasites. The THEdS bundle is a sustainable control and prevention method for the control of helminthes and protozoan associated with unsanitary environment and poor personal hygiene among immune-compromised individuals like HIV/AIDS patients. Poor sanitation, improper food handling, limited access to safe drinking water sources, and limited access to healthcare services contribute to a high prevalence of infections caused by soil-transmitted helminths (STHs) among the BaAka Pygmies, an indigenous community living in Central Africa. The aim of this study was to determine the rates of STH infections in the BaAka people inhabiting the rain forests of the south-western parts of the Central African Republic (CAR) as well as to examine the validity of biannual deworming with a single dose of albendazole 400 mg in high-risk communities exposed to extreme environmental conditions. The study was conducted in August 2021 and involved a sample of 49 BaAka Pygmies inhabiting the rain forest of the Sangha-Mbaéré Prefecture, CAR. The study consisted of collecting single stool samples from each participant and examining the samples for intestinal parasites by light microscopy methods. The collected samples were fixed in SAF fixative and next transported from Africa to Europe, where they were analyzed by light microscopy using three different diagnostic methods (direct smear in Lugol’s solution, the Fülleborn’s flotation, the Kato–Katz thick smear) at the Department of Epidemiology and Tropical Medicine in Gdynia, Poland. Microscopic examination found that 61.2% of the study group were infected with at least one helminthic species. The parasitological screening found invasions with four different species of nematodes, of which hookworm invasions were the most prevalent. The study results demonstrated that although the WHO-recommended mass deworming, which is provided to the BaAka Pygmies in healthcare centers set up on the premises of catholic missions, can effectively reduce the number of infections with soil-transmitted helminths, the prevalence of STH infections remains high in the region. The study findings suggest that in order to contain the spread of STHs in the local community, it will be necessary to implement additional preventive measures, apart from only conducting mass deworming programs. Infecting humans with controlled doses of small intestinal helminths, such as human hookworm, is proposed as a therapy for the colonic inflammatory disease ulcerative colitis. Strengthening the colonic mucus barrier is a potential mechanism by which small intestinal helminths could treat ulcerative colitis. In this study, we compare C57BL/6 mice infected with the small intestinal helminth Heligmosomoides polygyrus and uninfected controls to investigate changes in colonic mucus. Histology, gene expression, and immunofluorescent analysis demonstrate that this helminth induces goblet cell hyperplasia, and an upregulation of mucin sialylation, and goblet-cell-derived functional proteins resistin-like molecule-beta (RELM-β) and trefoil factors (TFFs), in the colon. Using IL-13 knockout mice, we reveal that these changes are predominantly IL-13-dependent. The assessment of the colonic mucus microbiome demonstrates that H. polygyrus infection increases the abundance of Ruminococcus gnavus, a commensal bacterium capable of utilising sialic acid as an energy source. This study also investigates a human cohort experimentally challenged with human hookworm. It demonstrates that TFF blood levels increase in individuals chronically infected with small intestinal helminths, highlighting a conserved mucus response between humans and mice. Overall, small intestinal helminths modify colonic mucus, highlighting this as a plausible mechanism by which human hookworm therapy could treat ulcerative colitis. Full article

Madagascar is one of the poorest countries in the world. The country’s extreme weather conditions, poor sanitation, and weak economy facilitate the spread of parasitic diseases. Infections with intestinal parasites are particularly dangerous for children because they can cause malnutrition and anemia, which, in turn, have a negative effect on children’s cognitive functions and physical development. The aim of the present study was to analyze the prevalence of intestinal parasites and to assess hematological parameters in a group of children living in northern Madagascar. The screening was conducted in May 2024 in the Clinique Medicale Beyzym in Manerinerina, the Ambatoboeny district. It involved a sample of 208 children aged 0–17 years. Single stool samples were collected from all study participants. The samples were fixed in SAF fixative and then transported from Africa to Europe for further diagnostics at the Department of Epidemiology and Tropical Medicine, the Military Institute of Medicine–the National Research Institute in Poland. First, the samples were analyzed by light microscopy methods using three different diagnostic techniques (direct smear, decantation with distilled water, and the Fülleborn method). Next, they were tested by molecular biology methods (real-time PCR). Blood samples for the assessment of hematological parameters were collected at the healthcare center in Madagascar. The prevalence of intestinal parasites in the study sample was 61.5%. Helminths were found in 15.2% of the investigated children, and Giardia intestinalis (20.5%) was found to be the most prevalent parasite in the study population. Most infections were caused by potentially pathogenic stramenopila Blastocystis spp. (32.0%). Mean Hb, HCT, MCV, MCH, and MCHC levels in the study participants were below normal values. However, no correlation was found between the presence of a parasitic infection and low hematological parameters, which are a clinical sign of anemia. High rates of infections with intestinal parasites in children living in northern Madagascar support the necessity to introduce long-term preventive measures, which would limit the spread of parasitic diseases in the Malagasy population. Low hematological parameters in non-infected children may be indicative of persistent malnutrition or infection with other parasites, e.g., malaria or schistosomiasis. Full article

The human gut hosts a diverse and active community of bacteria that symbiotically support the physiology, metabolism, and immunity of the intestinal lining. Nevertheless, a dynamic community of parasites (helminths and protozoa) may share a habitat with gut-dwelling microbiota. Both microbiota and parasites can significantly change the physical and immunological environment of the gut, thus generating several mechanisms of interaction. Studying this field is crucial for understanding the pathogenesis of parasitic diseases. Additionally, intestinal microbiota and gut-dwelling parasites may interact with each other and with the host immunity to alleviate or exacerbate the disease. These interactions can alter the pathogenicity of both parasites and microbiota, thereby changing the infection outcomes and the overall disease profile. Parasites and microbiota interactions occur via several mechanisms, including physical alteration in both the gastrointestinal microenvironment and the adaptive and innate immune responses. By modulating the microbiota, treating parasitic infections and microbiota dysbiosis may be improved through knowing the mechanisms and consequences of the interactions between intestinal parasites and the microbiota. Thus, new biological tools of treatment including probiotics can be introduced, particularly with the emergence of drug resistance and adverse effects. Full article

Anthelmintic resistance has been documented worldwide, but few cases have been reported in dogs. Shelter dogs are a great opportunity to study intestinal helminths and assess their potential resistance to anthelmintic compounds. With these two goals in mind, 361 faecal samples were collected from dogs in 18 municipalities in Portugal, from 19 shelters and 11 private tutors. These samples were analysed using the Mini-FLOTAC before and 13 days after deworming. The percentage of faecal egg count reduction was calculated. Parasitic forms were identified in 22.4% of the samples collected: Trichuris vulpis (10.5%), Toxocara canis (8.0%), members of the family Ancylostomatidae (6.4%), Toxascaris leonina (0.8%), Dipylidium caninum (0.6%) and members of the family Taeniidae (0.3%). The first three nematode parasites showed the highest intensity of infection (2900, 1325, and 1265 eggs per gram, respectively). In the second faecal collection, parasites were present in 20.8% of the samples. The anthelmintics febendazole + pyrantel embonate + praziquantel and praziquantel + fenbendazole were ineffective for T. vulpis and members of the family Ancylostomatidae in 16.2% and 6.9% of the samples, respectively. The parasites identified are all potentially zoonotic. This study is the first to suggest a potential resistance of T. vulpis to anthelmintics. Full article

Nematophagous, or helminthophagous fungi of the genera Duddingtonia, Arthrobotrys, Monacrosporium, Pochonia, Paecilomyces, and Mucor, have been used over the years in in vitro and in vivo experiments to control helminth parasites that are potentially zoonotic. These fungi have shown efficacy against the following helminth genera: Ancylostoma, Toxocara, Enterobius, Strongyloides, Angiostrongylus, Taenia, Fasciola, and Schistosoma. The results obtained from these experiments, together with studies on soil contamination, suggest the viability of their use as a sustainable and effective strategy to reduce environmental contamination by these zoonotic parasites. Therefore, the aim of this review was to address the role of helminthophagous fungi in the biological control of potentially zoonotic helminths. To this end, we describe (1) a brief history of helminthophagous fungi; (2) a discussion of some potentially zoonotic intestinal parasites; (3) the importance of helminthophagous fungi in the control of nematodes, cestodes, and trematodes; and (4) the potential of helminthophagous fungi as a practical and sustainable strategy. Although previous studies have characterized the helminth fauna of wild boars kept in captivity in Brazil, records on these helminths in free-ranging animals are still scarce. In view of this, we aimed in our work to investigate the occurrence and morphological and morphometric characteristics of gastrointestinal helminths in wild Sus scrofa from the northwest region of the State of São Paulo, Brazil. The digestive systems of 10 animals (5 males and 5 females of different ages) were used in this study. Each anatomical segment was washed and sieved under running water, and the helminths were separated and identified using light and scanning electron microscopy, according to their morphological characteristics. A total of 2750 (1152 males and 1598 females) nematode specimens were collected from the small intestine of these wild boars, and all of them presented the morphological characteristics of Globocephalus urosubulatus. However, one characteristic is of particular interest because it has not yet been reported in the literature: a marked asymmetry between the lobes and their respective rays of the copulatory bursa, with the left one being larger than the right one. In this research, we identified the presence of G. urosubulatus in all the examined free-ranging wild boars and reported for the first time in the literature the asymmetry in the copulatory bursa.
