This study assessed the effectiveness of a trastuzumab-targeted 177Lu-labeled mesoporous Carbon@Silica nanostructure (DOTA@TRA/MC@Si) for HER2-positive breast cancer treatment, focusing on its uptake, internalization, and efflux in breast cancer cells. The synthesized PEI-MC@Si nanocomposite was reacted with DOTA-NHS-ester, confirmed by the Arsenazo(III) assay. Following this, TRA was conjugated to the DOTA@PEI-MC@Si for targeting. DOTA@PEI-MC@Si and DOTA@TRA/MC@Si nanocomposites were labeled with 177Lu, and their efficacy was evaluated through in vitro radiolabeling experiments. According to the results, the DOTA@TRA/MC@Si nanocomposite was successfully labeled with 177Lu, yielding a radiochemical yield of 93.0 ± 2.4%. In vitro studies revealed a higher uptake of the [177Lu]Lu-DOTA@TRA/MC@Si nanocomposite in HER2-positive SK-BR-3 cells (44.0 ± 4.6% after 24 h) compared to MDA-MB-231 cells (21.0 ± 2.3%). The IC50 values for TRA-dependent uptake in the SK-BR-3 and BT-474 cells were 0.9 µM and 1.3 µM, respectively, indicating affinity toward HER-2 receptor-expressing cells. The lipophilic distribution coefficients of the radiolabeled nanocomposites were determined to be 1.7 ± 0.3 for [177Lu]Lu-DOTA@TRA/MC@Si and 1.5 ± 0.2 for [177Lu]Lu-DOTA@PEI-MC@Si, suggesting sufficient passive transport through the cell membrane and increased accumulation in target tissues. The [177Lu]Lu-DOTA@TRA/MC@Si nanocomposite showed an uptake into HER2-positive cell lines, marking a valuable step toward the development of a nanoparticle-based therapeutic agent for an improved treatment strategy for HER2-positive breast cancer. The aim of this study is to introduce a new filament and novel 3D printing technique to adjust the density of a printing job in order to mimic the radiological properties of different tissues. We used a special filament, Light Weight PLA (LW-PLA), which utilizes foaming technology triggered by temperature. Cylindrical samples were printed at various temperatures, flow rates, print speeds, and diameters. A computed tomography (CT) scan was performed to identify their radiological properties in terms of the mean Hounsfield Unit (HU). The densities of the samples ranged from 0.36 g/cm3 to 1.21 g/cm3, corresponding to mean HU values between −702.7 ± 13.9 HU and +141.4 ± 7.1 HU. Strong linear correlations were observed between the flow rate and density as well as the flow rate and mean HU. The axial homogeneity of the samples was reported as being comparable to that of distilled water. A reduction in the mean HU was observed at a lower print speed and it changed slightly with respect to the sample size. Reproducibility assessments confirmed consistent results for identical printing jobs. Comparisons with regular PLA samples revealed a superior homogeneity in the LW-PLA samples. The findings of this study suggest a practical and accessible solution for mimicking all of the soft tissues, including the lungs, by using a single filament. Full article

Conducted by the nonprofit organization Breathe Mongolia—Clean Air Coalition, this study investigated ambient air pollution in Ulaanbaatar, focusing on the significant role of coal briquette combustion within ger areas. This combustion not only contributes to outdoor air pollution but also significantly degrades indoor air quality within these traditional dwellings, leading to substantial health concerns. To address this challenge, the study assessed indoor air pollution in gers—traditional Mongolian yurts—that had implemented Cooking, Heating, and Insulation Packages (CHIP), a program offering subsidized electrical heating, cooking, and insulation materials. The study encompassed 28 gers, among which 25 were equipped with CHIP while 3 were not, enabling a comparative analysis. Employing cost-effective technology, carbon monoxide levels were monitored across all 28 gers using Binary System monitors. Fine particulate matter concentrations were measured using AirVisual monitors in 14 of these gers. Data collection occurred during the winter of 2022–2023. To comprehensively assess indoor air quality within gers and ascertain the efficacy of interventions like CHIP in diminishing indoor air pollution, Breathe Mongolia intends to continue monitoring efforts within ger areas. These initiatives strive to address data gaps and inform strategies aimed at enhancing indoor air quality. Full article

Objective: Obstructive sleep apnea syndrome (OSAS) is a common sleep disorder that is caused by the reduction or cessation of airflow in the upper airway. Irisin, retinol-binding protein-4 (RBP-4), and adiponectin are the three significant factors in the metabolic process of the human body. The objective of this study was to investigate whether plasma irisin, RBP-4, and adiponectin levels are associated with the severity of OSAS. Methods: According to inclusion and exclusion criteria, 125 patients with OSAS and 46 healthy, gender-matched controls were included in this study. The patients were classified according to the apnea hypopnea index (AHI) as 14 mild cases (5 < AHI < 15), 23 moderate OSAS cases (15 < AHI < 30), and 88 severe OSAS cases (AHI > 30). The plasma irisin, RBP-4, and adiponectin levels were measured and compared between groups. Results: RBP-4 levels were higher in severe OSAS compared to other groups, and irisin levels were significantly lower in severe OSAS compared to other groups. There was a negative correlation between irisin and RBP-4 (r = −0.421; p < 0.001), and irisin and AHI (r = −0.834; p < 0.001), and a positive correlation between irisin and adiponectin (r = 0.240; p = 0.002). There was a negative correlation between RBP-4 and adiponectin (r = −0.507; p < 0.001) and a positive correlation between RBP-4 and AHI (r = 0.473; p < 0.001). As a predictor of OSAS, adiponectin showed the highest specificity (84.8%) and RBP-4 the highest sensitivity (92.0%). Conclusion: Circulating adiponectin, irisin, and RBP-4 may be new biomarkers in OSAS patients in addition to risk factors such as diabetes, obesity, and hypertension. When polysomnography is not available, these parameters and clinical data can be used to diagnose the disease. As a result, patients with an AHI score greater than thirty should be closely monitored for metabolic abnormalities. Full article

Air quality has important climate and health effects. There is a need, therefore, to monitor air quality both indoors and outdoors. Methods of measuring air quality should be cost-effective if they are to be used widely, and one such method is low-cost sensors (LCS). This study reports on the use of LCSs in Ulaanbataar, Mongolia to measure PM2.5 concentrations inside yurts or “gers”. Some of these gers were part of a non-government agency (NGO) initiative to improve insulating properties of these housing structures. The goal of the NGO was to decrease particulate emissions inside the gers; a secondary result was to lower the use of coal and other biomass material. LCSs were installed in gers heated primarily by coal, and interior air quality was measured. Gers that were modified by increasing their insulating capacities showed a 17.5% reduction in PM2.5 concentrations, but this is still higher than recommended by health organizations. Gers that were insulated and used a combination of both coal and electricity showed a 19.1% reduction in PM2.5 concentrations. Insulated gers that used electricity for both heating and cooking showed a 48% reduction in PM2.5 but still had higher concentrations of PM2.5 that were 6.4 times higher than recommended by the World Health Organization (WHO). Nighttime and daytime trends followed similar patterns and trends in PM2.5 concentrations with slight variations. It was found that at nighttime the outside PM2.5 concentrations were generally higher than the inside concentrations of the gers in this study, meaning that PM2.5 would flow into the ger whenever the doors were opened, causing spikes in PM2.5 concentrations. Research on the psychological and physiological well-being of captive animals has focused on investigating different types of social and structural enrichment. Consequently, cognitive enrichment has been understudied, despite the promising external validity, comparability, and applicability. As we aim to fill this gap, we developed an interactive, multiple-choice interface for cage-mounted touchscreen devices that rhesus monkeys (Macaca mulatta) can freely interact with, from within their home enclosure at the Cognitive Neuroscience Laboratory of the German Primate Center. The multiple-choice interface offers interchangeable activities that animals can choose and switch between. We found that all 16 captive rhesus macaques tested consistently engaged with the multiple-choice interface across 6 weekly sessions, with 11 of them exhibiting clear task preferences, and displaying proficiency in performing the selected tasks. Our approach does not require social separation or dietary restriction and is intended to increase animals’ sense of competence and agency by providing them with more control over their environment. Thanks to the high level of automation, our multiple-choice interface can be easily incorporated as a standard cognitive enrichment practice across different facilities and institutes working with captive animals, particularly non-human primates. We believe that the multiple-choice interface is a sustainable, scalable, and pragmatic protocol for enhancing cognitive well-being and animal welfare in captivity. Full article

Machining hard-to-cut materials, such as cobalt (Co)-based superalloys, is a common problem in manufacturing industries. Background: wire electrical discharge machining (W-EDM) is one of the widely used cutting processes that causes layer (white layer—WL and heat-affected zone—HAZ) formation, and microcracks on the material’s surface. Purpose: this study investigates the effects of WL and HAZ on the electrochemical grooving (EC grooving) performance of Co-based superalloys. Two different surface types (W-EDMed and VFed) were used in the experiments. Result: the experiments showed that material removal rate (MRR) values increased up to 212.49% and 122.23% for vibratory finished (VFed) and wire-electrical-discharge-machined (W-EDMed) surfaces, respectively. Conclusion: This result indicates the presence of HAZ and WL that prevent current transition between two electrodes. However, increased voltage causes an increase in surface roughness, with increment rates at 71.13% and 36.08% for VFed and W-EDMed surfaces, respectively. Moreover, for the VFed surfaces, the groove lost its flatness at the bottom after an approximately 100 µm depth due to the different electrochemical machineabilities of HAZ and real surface texture. This result can be attributed to the different microstructures (HAZ and surface texture) showing different electrochemical dissolution rates. Therefore, high-depth distance HAZ and WL must be removed from the workpiece. Full article

Glomerular disease due to podocyte malfunction is a major factor in the pathogenesis of chronic kidney disease. Identification of podocyte-specific signaling pathways is therefore a prerequisite to characterizing relevant disease pathways and developing novel treatment approaches. Here, we employed loss of function studies for EPB41L5 (Yurt) as a central podocyte gene to generate a cell type-specific disease model. Loss of Yurt in fly nephrocytes caused protein uptake and slit diaphragm defects. Transcriptomic and proteomic analysis of human EPB41L5 knockout podocytes demonstrated impaired mechanotransduction via the YAP/TAZ signaling pathway. Further analysis of specific inhibition of the YAP/TAZ-TEAD transcription factor complex by TEADi led to the identification of ARGHAP29 as an EPB41L5 and YAP/TAZ-dependently expressed podocyte RhoGAP. Knockdown of ARHGAP29 caused increased RhoA activation, defective lamellipodia formation, and increased maturation of integrin adhesion complexes, explaining similar phenotypes caused by loss of EPB41L5 and TEADi expression in podocytes. Detection of increased levels of ARHGAP29 in early disease stages of human glomerular disease implies a novel negative feedback loop for mechanotransductive RhoA—YAP/TAZ signaling in podocyte physiology and disease. The residential Mongolian yurt is representative of nomadic culture and its mobile residences. Behavior forms the closest connection the environment and people. There is almost no research about the residence and lifestyle of Mongolian yurt dwellers from the perspective of housing behavior. This study applied grounded theoretical methods to analyze the theoretical model of the influence mechanism of Mongolian yurt dwelling on behavior. Interviews were conducted according to the principle of purposive sampling, and were summarized in five categories: production lifestyle, natural environment, residential characteristics, cultural beliefs, and emotional experience. Production lifestyle is the core category and the critical factor in dwelling behavior, which interacts with the other main categories. Pastoral policy is the factor that has worked most quickly and directly to influence housing in yurts. Mongolians living in yurts on the prairie show higher satisfaction compared to those in urban housing, which is related to the Mongolian advocacy for nature and freedom. Full article

Farming seabass (Dicentrarchus labrax) is an essential activity in the Mediterranean basin including the Aegean Sea. The main seabass producer is Turkey accounting for 155,151 tons of production in 2021. In this study, skin swabs of seabass farmed in the Aegean Sea were analysed with regard to the isolation and identification of Pseudomonas. Bacterial microbiota of skin samples (n = 96) from 12 fish farms were investigated using next-generation sequencing (NGS) and metabarcoding analysis. The results demonstrated that Proteobacteria was the dominant bacterial phylum in all samples. At the species level, Pseudomonas lundensis was identified in all samples. Pseudomonas, Shewanella, and Flavobacterium were identified using conventional methods and a total of 46 viable (48% of all NGS+) Pseudomonas were isolated in seabass swab samples. Additionally, antibiotic susceptibility was determined according to standards of the European Committee on Antimicrobial Susceptibility Testing (EUCAST) and Clinical and Laboratory Standards Institute (CLSI) in psychrotrophic Pseudomonas. Pseudomonas strains were tested for susceptibility to 11 antibiotics (piperacillin-tazobactam, gentamicin, tobramycin, amikacin, doripenem, meropenem, imipenem, levofloxacin, ciprofloxacin, norfloxacin, and tetracycline) from five different groups of antibiotics (penicillins, aminoglycosides, carbapenems, fluoroquinolones, and tetracyclines). The antibiotics chosen were not specifically linked to usage by the aquaculture industry. According to the EUCAST and CLSI, three and two Pseudomonas strains were found to be resistant to doripenem and imipenem (E-test), respectively. All strains were susceptible to piperacillin-tazobactam, amikacin, levofloxacin, and tetracycline. Our data provide insight into different bacteria that are prevalent in the skin microbiota of seabass sampled from the Aegean Sea in Turkey, and into the antibiotic resistance of psychrotrophic Pseudomonas spp. Full article

Cardiac magnetic resonance (CMR) is an essential clinical tool for the assessment of cardiovascular disease. Deep learning (DL) has recently revolutionized the field through image reconstruction techniques that allow unprecedented data undersampling rates. These fast acquisitions have the potential to considerably impact the diagnosis and treatment of cardiovascular disease. Herein, we provide a comprehensive review of DL-based reconstruction methods for CMR. We place special emphasis on state-of-the-art unrolled networks, which are heavily based on a conventional image reconstruction framework. We review the main DL-based methods and connect them to the relevant conventional reconstruction theory. Next, we review several methods developed to tackle specific challenges that arise from the characteristics of CMR data. Then, we focus on DL-based methods developed for specific CMR applications, including flow imaging, late gadolinium enhancement, and quantitative tissue characterization. Finally, we discuss the pitfalls and future outlook of DL-based reconstructions in CMR, focusing on the robustness, interpretability, clinical deployment, and potential for new methods. Full article

Coenzyme Q10 (CoQ10) is a vital substance found throughout body. It helps convert food into energy and is eaten small amounts in foods. CoQ10 has gained great interest in recent years as a potential candidate for the treatment of various diseases. The content of CoQ10 in food samples is a crucial quality index for foods. Therefore, the development of sensitive separation and quantification method for determining the amount of CoQ10 in various samples, especially in foods, is an important issue, especially for food nutrition. In this study, a new, miniaturized monolithic column was developed and applied for the determination of CoQ10 in pistachio samples by nano-liquid chromatography (nano-LC). The monolithic column with a 50 µm i.d. was prepared by in situ polymerization using laurylmethacrylate (LMA) as the main monomer and ethylene dimethacrylate (EDMA) as the crosslinker. Methanol (MeOH) and polyethyleneglycol (PEG) were used as porogenic solvents. The final monolithic column was characterized by using scanning electron microscopy (SEM) and chromatographic analyses. The monolithic column with a 50 µm i.d. was applied to the analysis of CoQ10 in pistachio samples in nano-LC. This analytical method was validated by means of sensitivity, linearity, precision, recovery, and repeatability. The LOD and LOQ values were 0.05 and 0.48 µg/kg, respectively. The developed method using the monolithic column was optimized to achieve very sensitive analyses of CoQ10 content in the food samples. The applicability of the method was successfully demonstrated by the analysis of CoQ10 in pistachio samples. Full article

The new coronavirus disease (COVID-19), which was detected in the Wuhan region of China in 2019 and spread rapidly all over the world, was declared a pandemic by the WHO in 2020. Since then, despite widespread recommendations to prevent the spread of the disease and provide treatment for sick people, 6,573,968 people died all over the world, 101,203 of which in Turkey. According to the international adult vaccination guidelines, pregnant women have been recommended to get vaccinated against the new coronavirus disease, as well as influenza and tetanus, during pregnancy. Before this study, not enough information was available about the vaccination awareness and vaccination hesitancy rates of pregnant women living in Turkey. For this reason, we believe that our study will contribute to filling this gap in the literature. The main objective of this study was to investigate the vaccination rates of pregnant women in a local hospital in Turkey and the reasons for vaccine hesitancy in this patient group. The minimum sample size of the study was found to be 241, with 80% power, 0.2 effect size, and 95% confidence interval, at p < 0.05 significance level. We included 247 consecutive pregnant women who applied to the Samsun Training and Research Hospital Gynecology and Obstetrics Outpatient Clinics between January 2022 and April 2022. The researchers prepared a questionnaire by taking into account the characteristics of the local community. A preliminary survey with these questions was also conducted before starting the main study. The mean age of the pregnant women participating in the study was 28.7 ± 5.3 years, and the mean gestational age was 28.2 ± 7.9 weeks. Among the participants, 26.3% were university graduates or had a higher degree, and 17% were actively working; in addition, 93 (37.7%) of the 247 pregnant women had received the COVID-19 vaccine, 203 (82.2%) had received at least one dose of the tetanus vaccine, and only 1 (0.4%) person had been administered the influenza vaccine during pregnancy. The most common reason for COVID-19 vaccine refusal and hesitancy was safety concerns, while the low rates of tetanus and influenza vaccination were due to a lack of knowledge. These results show that it is important to inform and educate the pregnant population on this subject to improve their vaccination behavior. Full article

Evaluation of the Prevalence of Barotrauma and Affecting Factors in Patients with COVID-19 during Follow-Up in the Intermediate Care Unit

It is known that pneumothorax (PX) and pneumomediastinum (PM) develop due to COVID-19 disease. The objective of our study was to determine the prevalence of PX/PM due to COVID-19 in the intermediate intensive care unit (IMCU) and to evaluate the factors causing barotrauma and also the clinical outcomes of these patients. A total of 283 non-intubated patients with COVID-19 pneumonia followed up in the IMCU in a 1-year period were included in the study. The patients were classified as group 1 (having barotrauma) and group 2 (without barotrauma). The rate of barotrauma was 8.1% (n = 23, group 1). PX developed on the right hemithorax in 12 (70.6%) patients. Group 1 had statistically significantly higher 28-day mortality rates compared with group 2 (p = 0.014). The eosinophil and d-dimer levels of the patients in group 1 were higher, while C-reactive protein (CRP), fibrinogen, and albumin levels were lower than Group 2 (p < 0.001, p = 0.017, p = 0.001, p < 0.001), and p < 0.001, respectively). The similar rates of NIMV administration in our study groups support that barotrauma is not the only mechanism in the development of PX/PM. The findings of high blood eosinophil count and low blood levels of CRP, albumin, and fibrinogen in the barotrauma group of our study might be a pathfinder for future studies. Full article

Community-based tourism represents an opportunity for sustainable socioeconomic development, helping local populations to emerge out of lower living conditions. This paper investigates the perceptions of tourists and stakeholders engaged in Community-Based Tourism (CBT) in Central Asian countries (including Kazakhstan, Uzbekistan, Kyrgyzstan, Tajikistan, and Turkmenistan) as an opportunity for sustainable development. This study’s purpose is to point out the usefulness of a combination approach of stakeholders’ and tourists’ perceptions to address the opportunities of CBT that can improve the quality of life of the tourism community in Central Asia by identifying the pitfalls of practices and determining challenges for tourism policy. Using a mixed-method approach, two complementary methodologies are simultaneously conducted. (1) An in-depth interview approach with sixteen selected experts in the region was processed with semantic network analysis for the definition of the main challenges and opportunities facing CBT as an enabler of sustainable development, considering the perceptions of sustainability from the point of view of stakeholders. (2) An online survey involving one hundred twenty-five tourists to the region was carried out to focus on tourists’ perceptions of sustainability. A structural equation modelling technique was used to identify the influence of tourists’ sustainability preferences and their involvement with local communities on sustainability perceptions. The results highlight the benefits the community receives, as well as future opportunities to obtain more advantages from tourism practices within the scope of sustainable tourism planning.
