Severe kinds of uterine inflammation in animals cause reproductive and economic problems. Although there are changes in prostaglandin (PG) production and noradrenergic uterine innervation during endometritis, the role of noradrenaline (NA) and adrenoreceptors (ARs) in PGF2α formation is not yet fully understood. To recognize noradrenergic control of the PGF2α generation on the cellular level during endometritis, the action of NA as well as α1-, α2- and β-ARs on protein abundances of PGF synthase (PGFS) and PG 9-ketoreductase/carbonyl reductase (CBR1) in the Escherichia coli lipopolysaccharide (LPS)-influenced pig endometrial epithelial cells and PGF2α release from these cells were studied. The epithelial cells were exposed to LPS and NA alone; LPS with NA; LPS with agonists of α1-, α2- and β-Ars; LPS with antagonists of β1-, β2- and β3-ARs with NA; and LPS with antagonists of β1-, β2- and β3-ARs in combinations with agonists of β1-, β2-, and β3-ARs for 24 h. PGFS and CBR1 protein abundances in cells were determined by Western blotting and PGF2α medium content by ELISA. LPS alone increased CBR1 protein abundance and PGF2α release by epithelial cells in reference to the control value. NA alone exerted a stimulatory effect on PGFS and CBR1 protein abundances and PGF2α secretion. After the exposure of cells to LPS with NA together, CBR1 protein abundance, as well as PGF2α release, was higher than in response to LPS and NA alone. PGFS protein abundance was increased by LPS with NA together compared to LPS action alone. In LPS-exposed endometrial epithelial cells, NA acting by β2- and β3-ARs leads to a rise in CBR1 protein abundance and PGF2α secretion. β2-ARs also participate in the NA excitatory effect on PGFS protein abundance. The NA effect on all the parameters tested is not mediated by α1- and α2-ARs. β2- and β3-ARs mediate the stimulatory effect of NA on PGF2α generation and secretion by the LPS-exposed pig endometrial epithelial cells. The results suggest that these cells may be a significant source of PGF2α under noradrenergic stimulation in the inflamed endometrium, and NA affects processes controlled by PGF2α during endometritis in an indirect manner. The gut microbiota plays a vital role in the physiological functions and health of animals. Domestic pigs (Sus scrofa domesticus) and wild boars (Sus scrofa scrofa), which share a common ancestor, present a unique model for exploring the evolutionary changes in gut microbiota due to domestication. Comparing the gut microbiota of these two groups provides a unique opportunity to identify microorganisms that may have been lost or altered during domestication. This study employed 16S rRNA sequencing to compare the gut microbiota compositions of domestic pigs and wild boars, analyzing 69 fecal samples from both groups. Our results indicate significant differences in the taxonomic profiles of the gut microbiota between the two groups. Specifically, the genera Bifidobacterium and Lactobacillus, were found in higher abundance in wild boars. In contrast, domestic pigs exhibited a decreased variety of these potentially health-enhancing bacteria, likely due to factors such as simplified diets and antibiotic use. Functional gene analysis revealed that pathways related to carbohydrate metabolism, lipid metabolism, immune response, and environmental adaptability were more enriched in wild boars. These findings demonstrate significant differences in the gut microbiota composition between domestic pigs and wild boars, underscoring the impact of domestication on microbial diversity. Further research is necessary to explore the potential of transferring beneficial microbes from wild to domestic populations. Full article

African Swine Fever (ASF) is an infectious disease affecting pigs and wild boars, causing significant economic losses. Epidemiological surveillance plays an important role in minimizing its impact. The aim of this study was to evaluate the usefulness of β-actin DNA and mitochondrial DNA (mtDNA) levels in determining the age of animal remains from the Suidae family, which could be helpful in epidemiological investigations. The study included selected tissues and internal organs of Sus scrofa domesticus, exposed to natural environmental conditions or kept in stable 4 °C conditions, to assess the levels of β-actin and mtDNA over a period of 18 months. The levels of both tested parameters exhibited the expected decreasing trend over time. However, in most tissues, some discrepancies from this general pattern were observed. The results obtained for bone marrow showed a consistent downward trend and a strong correlation between mtDNA and β-actin, with long-term detectability (up to the 13th month of the study). Therefore, bone marrow could be considered a matrix of choice for age assessment. However, due to various influencing factors, further studies are required. Full article

Domesticated animals are artificially selected to exhibit desirable traits, however not all traits of domesticated animals are the result of deliberate selection. Loss of olfactory capacity in the domesticated pig (Sus scrofa domesticus) is one example. We used whole transcriptome analysis (RNA-Seq) to compare patterns of gene expression in the olfactory mucosa of the pig and two subspecies of wild boar (Sus scrofa), and investigate candidate genes that could be responsible for the loss of olfactory capacity. We identified hundreds of genes with reductions in transcript abundance in pig relative to wild boar as well as differences between the two subspecies of wild boar. These differences were detected mainly in genes involved in the formation and motility of villi, cilia and microtubules, functions associated with olfaction. In addition, differences were found in the abundances of transcripts of genes related to immune defenses, with the highest levels in continental wild boar subspecies. Overall, the loss of olfactory capacity in pigs appears to have been accompanied by reductions in the expression of candidate genes for olfaction. These changes could have resulted from unintentional selection for reduced olfactory capacity, relaxed selection for maintaining olfactory capacity, pleiotropic effects of genes under selection, or other non-selective processes. Our findings could be a cornerstone for future researches on wild boars, pigs, feral populations, and their evolutionary trajectories, aimed to provide tools to better calibrate species management as well as guidelines for breeders. Domesticated animals have undergone significant changes in their behavior, morphology, and physiological functions during domestication. To identify the changes in gene expression associated with domestication, we collected the RNA-seq data of pigs, chickens, wild boars, and red junglefowl from public databases and performed a meta-analysis. Gene expression was quantified, and the expression ratio between domesticated animals and their wild ancestors (DW-ratio) was calculated. Genes were classified as “upregulated”, “downregulated”, or “unchanged” based on their DW-ratio, and the DW-score was calculated for each gene. Gene set enrichment analysis revealed that genes upregulated in pigs were related to defense from viral infection, whereas those upregulated in chickens were associated with aminoglycan and carbohydrate derivative catabolic processes. Genes commonly upregulated in pigs and chickens are involved in the immune response, olfactory learning, epigenetic regulation, cell division, and extracellular matrix. In contrast, genes upregulated in wild boar and red junglefowl are related to stress response, cell proliferation, cardiovascular function, neural regulation, and energy metabolism. These findings provide valuable insights into the genetic basis of the domestication process and highlight potential candidate genes for breeding applications. Full article

Agonic aspiration of blood (AAB) may result from an inadequate exsanguination with accidental trachea severing, that can be favoured by ineffective stunning of pigs (Sus scrofa domesticus). This study aimed to evaluate AAB as an animal-based indicator of electrical stunning ineffectiveness, which could be used by official veterinarians during the post-mortem inspection of pigs. Information on 3584 finishing pigs was collected at a Portuguese abattoir that performs head-to-body electrical stunning with horizontal bleeding. Of them, 15.5% of the pigs presented signs of ineffective stunning. AAB lung lesions were found in 27.8% of lungs. Despite what was predicted, a strong correlation was found between well stunned animals and the presence of blood lesions in lungs (p = 0.006). Statistical significances were found between pigs’ lateral recumbency at the conveyor and the presence of blood affecting one lung. Under the conditions of this study, the authors cannot point to AAB lung lesions as an animal-based indicator of electrical stunning ineffectiveness. Further studies should be conducted to establish a better understanding of the causes of aspiration of blood, in particular how horizontal bleeding may affect the occurrence of similar lesions. Post-weaning diarrhoea and enterotoxaemia caused by Escherichia coli are serious threats in the pig (Sus scrofa domesticus) livestock industry and are responsible for economic losses related to mortality, morbidity and stunted growth. The aim of this study was to evaluate the effect of an engineered tobacco seeds-based edible vaccine in O138 Escherichia coli-challenged piglets throughout a multidisciplinary approach. Thirty-six weaned piglets were enrolled and randomly divided into two experimental groups, a control (C; n = 18) group and a tobacco edible vaccination group (T, n = 18), for 29 days of trial. At days 0, 1, 2, 5 and 14, piglets of the T group were fed with 10 g of the engineered tobacco seeds line expressing F18 and VT2eB antigens, while the C group received wild-type tobacco seeds. After 20 days, 6 piglets/group were orally challenged with the Escherichia coli O138 strain (creating four subgroups: UC = unchallenged control, CC = challenged control, UT = unchallenged tobacco, CT = challenged tobacco) and fed with a high protein diet for 3 consecutive days. Zootechnical, clinical, microbiological, histological and immunological parameters were assayed and registered during the 9 days of post-challenge follow up. At 29 days post-challenge, the CT group displayed a lower average of the sum of clinical scores compared to the CC group (p < 0.05), while the CC group showed a higher average sum of the faecal score (diarrhoea) (p < 0.05) than the CT group. A decreased number of days of shedding of the pathogenic strain was observed in the CT compared to the CC group (p < 0.05). Specific anti-F18 IgA molecules were significantly higher in the CT group compared to the CC group’s faecal samples during the post-challenge period (p < 0.01). In conclusion, edible vaccination with engineered tobacco seeds showed a protective effect on clinical symptoms and diarrhoea incidence during the post-challenge period, characterized by a limited time of pathogenic strain shedding in faeces. It is well understood that intrinsic factors of bone contribute to bone diagenesis, including bone porosity, crystallinity, and the ratio of organic to mineral components. However, histological analyses have largely been limited to adult bones, although with some exceptions. Considering that many of these properties are different between juvenile and adult bone, the purpose of this study is to investigate if these differences may result in increased degradation observed histologically in fetal and juvenile bone. Thirty-two fetal (n = 16) and juvenile (n = 16) Sus scrofa domesticus femora subject to different depositions over a period of two years were sectioned for histological observation. Degradation was scored using an adapted tunneling index. Results showed degradation related to microbial activity in both fetal and juvenile remains across depositions as early as three months. Buried juvenile remains consistently showed the greatest degradation over time, while the blanket fetal remains showed more minimal degradation. This is likely related to the buried remains’ greater contact with surrounding soil and groundwater during deposition. Further, most of the degradation was seen in the subendosteal region, followed by the subperiosteal region, which may suggest the initial microbial attack is from endogenous sources. Full article

Rift valley fever (RVF), caused by the RVF virus (RVFV), is a vector-borne zoonotic disease that primarily affects domestic ruminants. Abortion storms and neonatal deaths characterise the disease in animals. Humans develop flu-like symptoms, which can progress to severe disease. The susceptibility of domestic pigs (Sus scrofa domesticus) to RVFV remains unresolved due to conflicting experimental infection results. To address this, we infected two groups of pregnant sows, neonates and weaners, each with a different RVFV isolate, and a third group of weaners with a mixture of the two viruses. Serum, blood and oral, nasal and rectal swabs were collected periodically, and two neonates and a weaner from group 1 and 2 euthanised from 2 days post infection (DPI), with necropsy and histopathology specimens collected. Sera and organ pools, blood and oronasorectal swabs were tested for RVFV antibodies and RNA. Results confirmed that pigs can be experimentally infected with RVFV, although subclinically, and that pregnant sows can abort following infection. Presence of viral RNA in oronasorectal swab pools on 28 DPI suggest that pigs may shed RVFV for at least one month. It is concluded that precautions should be applied when handling pig body fluids and carcasses during RVF outbreaks. Full article

For more than a decade, next-generation sequencing (NGS) has been emerging as the mainstay of agrigenomics research. High-throughput technologies have made it feasible to facilitate research at the scale and cost required for using this data in livestock research. Scale frameworks of sequencing for agricultural and livestock improvement, management, and conservation are partly attributable to innovative informatics methodologies and advancements in sequencing practices. Genome-wide sequence-based investigations are often conducted worldwide, and several databases have been created to discover the connections between worldwide scientific accomplishments. Such studies are beginning to provide revolutionary insights into a new era of genomic prediction and selection capabilities of various domesticated livestock species. In this concise review, we provide selected examples of the current state of sequencing methods, many of which are already being used in animal genomic studies, and summarize the state of the positive attributes of genome-based research for cattle (Bos taurus), sheep (Ovis aries), pigs (Sus scrofa domesticus), horses (Equus caballus), chickens (Gallus gallus domesticus), and ducks (Anas platyrhyncos). This review also emphasizes the advantageous features of sequencing technologies in monitoring and detecting infectious zoonotic diseases. In the coming years, the continued advancement of sequencing technologies in livestock agrigenomics will significantly influence the sustained momentum toward regulatory approaches that encourage innovation to ensure continued access to a safe, abundant, and affordable food supplies for future generations. Over 80% proteins consumed by the local people in Nujiang Prefecture of Southwest China, a remote and mountainous area in the Eastern Himalayas, are from pork, or Gaoligongshan pig (a landrace of Sus scrofa domestica Brisson). Previous research on the Gaoligongshan pig has focused on nutritional composition, production performance, and genetic resource characteristics, but neglected the reasons behind the local people’s practice. From 2019 to 2022, we have used ethnobiological research methods to comprehensively document the traditional rearing and management patterns and the traditional culture associated with Gaoligongshan pigs. The results show that Gaoligongshan pigs graze in mixed herds with cattle and sheep during the day and prefer to eat 23 wild plant species, in which 17 species have medicinal values. At night, the pigs are artificially fed and rest in the pigsty. The local Bai and Lisu people have developed a creative food culture, rituals, and festivals culture associated with Gaoligongshan pigs. Overall, the biocultural diversity of Gaoligongshan pig contributes to the in situ conservation of genetic diversity of this important hog landrace, and supports rural development in this remote area. Early Neolithic communities penetrated in southwestern Transylvania and were established in the Mureș Valley where they found suitable territories for domestic animal herds and probably plant cultivation. The present study tries to answer questions related to the beginnings of the neolithisation in this area, from an interdisciplinary perspective, valuing archeozoologically and archeobotanically the Starčevo-Criș site of Soimuș-Teleghi (Hunedoara County), dating to the 7th–6th millennium BC. Animal skeletal remains and phytoliths are the proxies analysed in this paper, offering data about the palaeoeconomy and palaeoenvironment of the Early Neolithic settlement. In the archaeozoological samples, the remains coming from domestic mammals are the most frequent, being identified as cattle (Bos taurus), sheep/goat (Ovis aries/Capra hircus), pig (Sus domesticus), and dog (Canis familiaris). The fact that pig remains are almost absent in the samples is significant, suggesting a mobility of the evaluated communities. The skeletal remains of wild mammal species are rare, belonging to red deer (Cervus elaphus), roe deer (Capreolus capreolus), aurochs (Bos primigenius), wild boar (Sus scrofa), and polecat (Mustela putorius). The strong dominance of grasses is attested through the phytoliths’ assemblages. ELONGATE DENDRITIC phytoliths are well represented. This morphotype, which originates from the inflorescence or the husk of Poaceae, is most likely derived from cultivated plants (cereals such as wheat, barley, etc.). Archaeozoological and archaeobotanical data resulting from this study suggest an open environment around the Early Neolithic settlement, where communities of the Starčevo-Criș culture mainly raised herds of cattle and sheep/goat. Sporadically, hunting and gathering molluscs were practised by the inhabitants, as indicated by archaeozoological results. Full article

Eight genotypes of the hepatitis E virus (Orthohepevirus A; HEV) designated HEV-1 to HEV-8 have been reported from various mammalian hosts. Notably, domestic pigs and wild boars are the natural reservoirs of HEV-3 and HEV-4 genotypes with zoonotic propensity. Since HEV infection in domestic pigs is usually subclinical, it may remain undetected, facilitating zoonotic spillover of HEV to the exposed human populations. A previous study from our group in 2021, using deep sequencing of a pooled saliva sample, generated various swine enteric virus genomes, including a near full-length swine HEV genome (7040 nt; 97.7% genome coverage) from five-month-old grower pigs at a backyard pig farm in the uMgungundlovu District, KwaZulu-Natal, South Africa. In the present study, we describe the further characterization, including genotyping and subtyping of the swine HEV isolate using phylogenetics and ‘HEVnet Typing Tool’. Our analyses confirmed that the South African swine HEV genome characterized in this study belonged to HEV genotype 3 subtype 3c (HEV-3c). While HEV-3c infections in domestic pigs have been previously reported from Brazil, Germany, Italy, and the Netherlands, they only generated partial genome sequences of open reading frame 1 (ORF1) and/or ORF2. To our knowledge, this is the first near full-length swine HEV-3c genome generated from naturally infected domestic pigs (Sus scrofa domesticus) in South Africa. However, due to the gap in the information on the HEV-3c genome sequences in various geographical locations worldwide, including South Africa, the epidemiology of the South African swine HEV genome characterized in this study remains inconclusive. Molecular and genomic surveillance of HEV in domestic pig populations in South Africa would be useful to determine their prevalence, circulating subtypes, and zoonosis risk. Full article

The demand for the development of novel medicines with few side effects and no proarrhythmic properties is increasing. Extensive research on herbal extracts has been conducted with the expectation that the compounds will exert precise effects without harmful side effects. Elsholtzia ciliata (Thunb.) Hyl. essential oil (EO) possesses antiarrhythmic properties similar to those of class 1B antiarrhythmics, such as prolonging myocardial activation of the QRS complex and shortening the QT interval. In this study, we determined the kinetic profile of EO phytocompounds and the effects of EO on heart electrical activity and arterial blood pressure. For this study, we chose to use local breed pigs that were anaesthetized. The effects of an intravenous bolus of EO on ECG parameters, arterial blood pressure, heart rate variability, and blood levels of haematological and biochemical parameters were registered and evaluated. Following an intravenous injection of a bolus, EO exerted a vasodilatory effect, resulting in significant reductions in arterial blood pressure. EO also increased the heart rate and altered ECG parameters. The bolus of EO prolonged the QRS complex, shortened the QT interval, and nonmonotonically altered the PQ interval. After the administration of a bolus of EO, the activity of the autonomic nervous system was altered. This study confirms that EO possesses similar properties to class 1B antiarrhythmics and exerts a hypotensive effect; it reduces arterial blood pressure possibly by modulating peripheral vascular resistance. Fluoride exposure from natural, agricultural, and industrial sources has harmed people living in fluoride-affected areas. Fluoride accumulates in the human body after being exposed to it through the food chain. The population consisted of 371 community health volunteers who were surveyed and chosen based on personal fluoride information. Only 39 residents were chosen to be interviewed and take part in the trial, which involved drinking fluoride-containing groundwater (>1.5 part per million: ppm) and urine testing that revealed urine fluoride level (>0.7 ppm). In addition, 47 biological samples and eight commercially bottled water specimens were examined. The information was gathered in four ways: (1) a questionnaire-based survey of fluoride knowledge, (2) food consumption behavior with locally grown vegetables, fruits, poultry, and meat, and commercially bottled water produced by groundwater in fluoride-affected areas, (3) a semi-food frequency questionnaire, and (4) fluoride content measurements using an ion-selective electrode. According to the analyses, the participants ranged in age from 51 to 60 years, with approximately 60.38% of them female and born and raised in polluted areas. The majority of subjects had a low level of fluoride knowledge (65.23%). The respondents’ primary source of drinking water (100.00%) was commercially bottled water; they chewed camellia sinensis 11.56% of the time (1 to 5 years) and they drank tea 9.16% of the time (during 1 to 5 years). Sus scrofa domesticus was responsible for the intake of vegetables and fruits, whereas Brassica chinensis, Jusl var para-chinensis (Bailey), and Tsen and Lee were responsible for the intake of poultry and animal flesh. They were all purchased at a local farm. The hazard quotient was greater than one, and the fluoride concentration (ppm) ranged between 75.00% (0.29–5.20), 57.14% (0.01–0.46), 88.89% (0.07–0.91), 100.00% (0.43–3.07), 100.00% (0.58–0.77), 42.86% (0.12–0.62 ppm.), 60.00% (0.11–1.44), and 33.33% (0.10–0.80) in drinking water, fruit, young and mature plants. Fluoride ingestion may pose a health concern. Under the 95th percentile condition, 74.47% consumed water with a high fluoride level, vegetables and fruits, and poultry and meats. Full article
