The so-called “counterfeit monks and nuns” 僧尼偽濫 is regarded as an important reason for the “Huichang Persecution of Buddhism” 會昌滅佛, but it reflects the central views of the Tang Dynasty. When we delve into the local society of the Mid-to-Late Tang period, we find that they developed their own narrative logic. From the perspective of the imperial court, Li Deyu 李德裕 criticized Wang Zhixing 王智興 for establishing an ordination altar in Sizhou 泗州 for personal gain. However, in the biographical inscription of monk Mingyuan 明遠 in Sizhou, Wang Zhixing is portrayed as a key figure who collaborated with Mingyuan to ensure the survival of the Kaiyuan Monastery 開元寺, with the inauguration of the ordination altar 戒壇 serving as a necessary means to obtain financial resources. In fact, Mingyuan had previously undertaken a similar operation at the Lingju Monastery 靈居寺 in Liuhe County 六合縣, Yangzhou 揚州. The inscription of the Lingju Monastery Stele 大唐揚州六合縣靈居寺碑 reflects the cooperation between local monks and secular people at that time. During the process of rebuilding the monasteries, Mingyuan cleverly exploited the cult of the divine monk Sengqie 僧伽 within the Society of Jianghuai 江淮. The cult of Sengqie had become a national belief during the Mid-to-Late Tang period, and the existence of the Sengqie pagoda 僧伽塔 made the Kaiyuan Monastery in Sizhou uniquely significant. Later on, Youxuan 幽玄 also carried out similar initiatives by establishing an ordination altar for the restoration at the Baoli Monastery 寶曆寺 in Hongzhou 洪州. If we set aside the shadow of the overarching theme of the Huichang Persecution of Buddhism on the history of Buddhism during the Mid-to-Late Tang period, we may uncover a more vibrant picture of local Buddhism. The aging communities within traditional historical and cultural districts in the post-epidemic era face numerous challenges, including declining vitality, deteriorating environments, sluggish economies, and heightened public health risks. This paper aims to explore strategies for promoting the internal self-development of aging communities and enhancing their resilience. It analyzes the resilience of these communities through four dimensions: social factors, built environments, economic factors, and ecological environments. Using the Sizhou’an community in China as a case study, this paper examines the underlying causes of vulnerability and proposes renewal strategies. These strategies include stimulating the vitality of elderly groups, improving the community’s spatial environment, leveraging regional cultural advantages, and establishing urban community agricultural gardens. Furthermore, it presents a community renewal model characterized by “government guidance + community leadership + capital introduction + resident participation” to bolster community resilience and vitality, thereby ensuring sustainable development. The incidence rate of invasive mucormycosis (IM) in patients with hematological malignancies (HMs) is increasing year by year, ranging from 0.07% to 4.29%, and the mortality rate is mostly higher than 50%. With the ongoing pandemic of COVID-19, COVID-19-associated mucormycosis (CAM) also became a global health threat. Patients with high risk factors such as active HMs, relapsed/refractory leukemia, prolonged neutropenia may still develop breakthrough mucormycosis (BT-MCR) even under the prophylaxis of Mucorales-active antifungals, and such patients often have higher mortality. Rhizopus spp. is the most common genus associated with IM, followed by Mucor spp. and Lichtheimia spp. Pulmonary mucormycosis (PM) is the most common form of IM in patients with HMs, followed by rhino-orbital-cerebral mucormycosis (ROCM) and disseminated mucormycosis. The prognosis of IM patients with neutrophil recovery, localized IM and receiving early combined medical–surgical therapy is usually better. As for management of the disease, risk factors should be eliminated firstly. Liposome amphotericin B (L-AmB) combined with surgery is the initial treatment scheme of IM. Those who are intolerant to L-AmB can choose intravenous formulations or tablets of isavuconazole or posaconazole. Patients who are refractory to monotherapy can turn to combined antifungals therapy. Studies have shown that Sargassum fusiforme and its extracts are effective herbal treatments for leukemia. We previously found that a polysaccharide from Sargassum fusiforme, SFP 2205, stimulated apoptosis in human erythroleukemia (HEL) cells. However, the structural characterization and antitumoral mechanisms of SFP 2205 remain uncertain. Here, we studied the structural characteristics and anticancer mechanisms of SFP 2205 in HEL cells and a xenograft mouse model. The results demonstrated that SFP 2205, with a molecular weight of 41.85 kDa, consists of mannose, rhamnose, galactose, xylose, glucose, and fucose with monosaccharides composition of 14.2%, 9.4%, 11.8%, 13.7%, 11.0%, and 38.3%, respectively. On animal assays, SFP 2205 significantly inhibited growth of HEL tumor xenografts with no discernible toxicity to normal tissues. Western blotting showed that SFP 2205 therapy improved Bad, Caspase-9, and Caspase-3 protein expression, and ultimately induced HEL tumor apoptosis, indicating mitochondrial pathway involvement. Furthermore, SFP 2205 blocked the PI3K/AKT signaling pathway and 740 Y-P, an activator of the PI3K/AKT pathway, rescued the effects of SFP 2205 on HEL cell proliferation and apoptosis. Overall, SFP 2205 may be a potential functional food additive or adjuvant for preventing or treating leukemia. Full article

Minichromosome maintenance protein 5 (MCM5) is a critical cell cycle regulator; its role in DNA replication is well known, but whether it is involved in the regulation of organogenesis in a cell cycle-independent way, is far from clear. In this study, we found that a loss of mcm5 function resulted in a mildly smaller liver, but that mcm5 overexpression led to liver bifida. Further, the data showed that mcm5 overexpression delayed endodermal migration in the ventral–dorsal axis and induced the liver bifida. Cell cycle analysis showed that a loss of mcm5 function, but not overexpression, resulted in cell cycle delay and increased cell apoptosis during gastrulation, implying that liver bifida was not the result of a cell cycle defect. In terms of its mechanism, our data proves that mcm5 represses the expression of cxcr4a, which sequentially causes a decrease in the expression of itgb1b during gastrulation. The downregulation of the cxcr4a-itgb1b cascade leads to an endodermal migration delay during gastrulation, as well as to the subsequent liver bifida during liver morphogenesis. In conclusion, our results suggest that in a cell cycle-independent way, mcm5 works as a gene expression regulator, either partially and directly, or indirectly repressing the expression of cxcr4a and the downstream gene itgb1b, to coordinate endodermal migration during gastrulation and liver location during liver organogenesis. This article explores regional Buddhist monasteries in Tang Dynasty (618–907 CE) China, including their arrangement, functions, and sources for their study. Specifically, as a case study, it considers the reconstruction of the Kaiyuan monastery 開元寺 in Sizhou 泗州 (present-day Jiangsu Province) with reference to the works of three prominent state officials and scholars: Bai Juyi 白居易 (772–846), Li Ao 李翱 (772–841), and Han Yu 韓愈 (768–824). The writings of these literati allow us to trace the various phases of the monastery’s reconstruction, fundraising activities, and the network of individuals who participated in the project. We learn that the rebuilt multi-compound complex not only provided living areas for masses of pilgrims, traders, and workers but also functioned as a barrier that protected the populations of Sizhou and neighboring prefectures from flooding. Moreover, when viewed from a broader perspective, the renovation of the Kaiyuan monastery demonstrates that Buddhist construction projects played a pivotal role in the social and economic development of Tang China’s major metropolises as well as its regions. Given the large-scale exploitation and utilization of wind power, the problems caused by the high stochastic and random characteristics of wind speed make researchers develop more reliable and precise wind power forecasting (WPF) models. To obtain better predicting accuracy, this study proposes a novel compound WPF strategy by optimal integration of four base forecasting engines. In the forecasting process, density-based spatial clustering of applications with noise (DBSCAN) is firstly employed to identify meaningful information and discard the abnormal wind power data. To eliminate the adverse influence of the missing data on the forecasting accuracy, Lagrange interpolation method is developed to get the corrected values of the missing points. Then, the two-stage decomposition (TSD) method including ensemble empirical mode decomposition (EEMD) and wavelet transform (WT) is utilized to preprocess the wind power data. In the decomposition process, the empirical wind power data are disassembled into different intrinsic mode functions (IMFs) and one residual (Res) by EEMD, and the highest frequent time series IMF1 is further broken into different components by WT. After determination of the input matrix by a partial autocorrelation function (PACF) and normalization into [0, 1], these decomposed components are used as the input variables of all the base forecasting engines, including least square support vector machine (LSSVM), wavelet neural networks (WNN), extreme learning machine (ELM) and autoregressive integrated moving average (ARIMA), to make the multistep WPF. To avoid local optima and improve the forecasting performance, the parameters in LSSVM, ELM, and WNN are tuned by backtracking search algorithm (BSA). On this basis, BSA algorithm is also employed to optimize the weighted coefficients of the individual forecasting results that produced by the four base forecasting engines to generate an ensemble of the forecasts. In the end, case studies for a certain wind farm in China are carried out to assess the proposed forecasting strategy. Full article

Accurate wind speed prediction plays a crucial role on the routine operational management of wind farms. However, the irregular characteristics of wind speed time series makes it hard to predict accurately. This study develops a novel forecasting strategy for multi-step wind speed forecasting (WSF) and illustrates its effectiveness. During the WSF process, a two-stage signal decomposition method combining ensemble empirical mode decomposition (EEMD) and variational mode decomposition (VMD) is exploited to decompose the empirical wind speed data. The EEMD algorithm is firstly employed to disassemble wind speed data into several intrinsic mode function (IMFs) and one residual (Res). The highest frequency component, IMF1, obtained by EEMD is further disassembled into different modes by the VMD algorithm. Then, feature selection is applied to eliminate the illusive components in the input-matrix predetermined by partial autocorrelation function (PACF) and the parameters in the proposed wavelet neural network (WNN) model are optimized for improving the forecasting performance, which are realized by hybrid backtracking search optimization algorithm (HBSA) integrating binary-valued BSA (BBSA) with real-valued BSA (RBSA), simultaneously. Combinations of Morlet function and Mexican hat function by weighted coefficient are constructed as activation functions for WNN, namely DAWNN, to enhance its regression performance. In the end, the final WSF values are obtained by assembling the prediction results of each decomposed components. Two sets of actual wind speed data are applied to evaluate and analyze the proposed forecasting strategy. Forecasting results, comparisons, and analysis illustrate that the proposed EEMD/VMD-HSBA-DAWNN is an effective model when employed in multi-step WSF. Full article
