Background: There is an urgent need for novel treatment options for Neisseria gonorrhoeae. Methenamine is an interesting urinary antiseptic with a very low propensity to induce antimicrobial resistance. Methods: We assessed the MICs of methenamine-hippurate for 18 N. gonorrhoeae isolates. We then assessed the in vivo efficacy of methenamine-hippurate against N. gonorrhoeae using the Galleria mellonella infection model. Results: We found that all the gonococcal isolates had a methenamine-hippurate MIC of 300 mg/L. This MIC was not higher in isolates with higher ceftriaxone MICs. No toxicity of methenamine at the doses tested was found, and doses as low as 200 mg/kg were effective in the G. mellonella model. Conclusions: Further studies in mice and humans are required to assess if methenamine-hippurate could be used to treat gonococcal urethritis alone or in combination with other agents such as ceftriaxone. Landslide susceptibility assessment is critical for hazard mitigation and land-use planning. This study evaluates the impact of two different non-landslide sampling methods—random sampling and sampling constrained by the Global Landslide Hazard Map (GLHM)—on the performance of various machine learning and deep learning models, including Naïve Bayes (NB), Support Vector Machine (SVM), SVM-Random Forest hybrid (SVM-RF), and XGBoost. The study area is a 2 km buffer zone along the Duku Highway in Xinjiang, China, with 102 landslide and 102 non-landslide points extracted by aforementioned sampling methods. Models were tested using ROC curves and non-parametric significance tests based on 20 repetitions of 5-fold spatial cross-validation data. GLHM sampling consistently improved AUROC and accuracy across all models (e.g., AUROC gains: NB +8.44, SVM +7.11, SVM–RF +3.45, XGBoost +3.04; accuracy gains: NB +11.30%, SVM +8.33%, SVM–RF +7.40%, XGBoost +8.31%). XGBoost delivered the best performance under both sampling strategies, reaching 94.61% AUROC and 84.30% accuracy with GLHM sampling. SHAP analysis showed that GLHM sampling stabilized feature importance rankings, highlighting STI, TWI, and NDVI as the main controlling factors for landslides in the study area. These results highlight the importance of hazard-informed sampling to enhance landslide susceptibility modeling accuracy and interpretability. Full article

HIV disproportionately affects key populations (MSM, transgender people, sex workers and psychoactive substance users), who face greater social vulnerability and limited healthcare access. This study aimed at analyzing sexual orientation and gender identity and their association with sexual practices, sexually transmitted infections and psychoactive substance use among PrEP users. Method: A cross-sectional study was conducted between January 2018 and June 2021 with 736 Brazilian PrEP users from a health service in São Paulo. Sociodemographic data, sexual behaviors, STI history (past 3 months) and psychoactive substances use (past 3 months) were extracted from clinical records. The associations were analyzed using binomial logistic regression (p < 0.05). Results: Most of the participants were cisgender men (93.4%) and homosexual (84.8%), with a mean age of 34.9 years old. Condomless sex was reported by 98.5%, and 18.4% had some recent sexually transmitted infection, mainly syphilis. Psychoactive substance use was reported by 55.4%, especially marijuana, club drugs, erectile stimulants and poppers. Transgender and cisgender women were more likely to report sex work and crack use. Homosexual and bisexual participants had higher odds of using erectile stimulants. Conclusions: The study reveals key links between gender, sexual orientation and risk behaviors, highlighting the need for inclusive, targeted prevention. Full article

Adult entertainment work may be associated with increased vulnerability to sexually transmitted infections, particularly HIV. In Brazil, pre-exposure prophylaxis (PrEP) for HIV infection has been available through the Brazilian Unified Health System (SUS) since November 2017, representing a significant advancement in public sexual health policy. The objective of this study was to understand the individual and social determinants that promote PrEP use among adult entertainment workers. This was a cross-sectional, analytical, and quantitative study. A multivariate modeling approach was employed to identify factors independently associated with PrEP use. The study included 254 adult entertainment workers using oral PrEP through the SUS, predominantly young adults (141; 55.5%), SUS users (248; 97.6%), single (213; 83.9%), non-white (142; 55.9%), cisgender (148; 58.3%), and heterosexual (152; 59.8%). Factors independently associated with greater PrEP use included having adult entertainment as the main source of income (aPR: 2.69; 95% CI: 1.86–3.95), prior use of PEP (aPR: 2.49; 95% CI: 1.63–3.81), undergoing any type of health treatment (aPR: 1.56; 95% CI: 1.15–2.12), and having a history of STIs (aPR: 1.51; 95% CI: 1.10–2.08). Conclusion: PrEP use in this population was strongly influenced by structural and contextual factors, indicating that the availability of the technology alone does not ensure its effectiveness. Background: The objective of this study is to investigate the prevalence and epidemiological changes of major sexually transmitted infections (STIs) in Korea over the past decade. Methods: From 2010 to 2021, patients diagnosed with STIs based on ICD-10 codes were analyzed using Korean Health insurance data. The analysis included the number of patients, prevalence, and age-specific prevalence (in 5-year intervals) over this period. We examined changes in disease patterns over time by analyzing the annual trends and age-specific prevalence of bacterial STIs such as chlamydia, mycoplasma, gonorrhea, and syphilis; viral STIs such as genital herpes, human papillomavirus (HPV), and human immunodeficiency virus (HIV); and other infections including scabies, pubic lice, and trichomoniasis. Results: In 2010, the STI with the highest prevalence due to an infectious pathogen was trichomoniasis (256.65/100,000), while latent syphilis had the lowest prevalence (5.29/100,000). In 2021, the STI with the highest prevalence was genital herpes (254.54 per 100,000 persons), and latent syphilis continued to have the lowest prevalence. Bacterial STIs showed a decreasing trend. Viral STIs showed a continuous increase throughout the study period, with anogenital warts (AGW) having the highest rate of increase. Other infections showed a decreasing trend. HIV and AGW in men showed a rapid increase. Gender differences varied depending on the disease. Conclusions: While bacterial STIs have gradually declined, viral STIs have continued to increase during last decade. The characteristics of each pathogen vary according to age and gender, necessitating the establishment of risk groups for each pathogen and the development of prevention policies accordingly. Background/Objectives: Mycoplasma and Ureaplasma species are pathogens commonly associated with urogenital infections in sexually active individuals. Despite their clinical relevance, these organisms are less frequently studied than other sexually transmitted infections (STIs), leading to limited data on their antimicrobial susceptibility and resistance profiles. This study aimed to characterize the antimicrobial susceptibility and resistance patterns of Mycoplasma hominis and Ureaplasma spp. among individuals in Salvador, Bahia, Brazil, and to identify the potential associated risk factors. Methods: We conducted a retrospective descriptive study during 2022–2024 using secondary data obtained from the SMARTLab® diagnostic system. Sociodemographic and epidemiological data, along with results from IST2 and IST3 diagnostic tests, were analyzed. Absolute and relative frequencies were calculated by sex, age group, and antimicrobial susceptibility profile. Results: Our results revealed a predominance of M. hominis and Ureaplasma spp. infection among women (98.5%), and in individuals aged 38 to 47 years. Ureaplasma spp. accounted for the majority of positive cases. High rates of resistance were observed in the IST2 test, with 75.0% of M. hominis and 84.1% of Ureaplasma urealyticum resistant to ciprofloxacin. In the IST3 test, Ureaplasma spp. demonstrated a 7.3% resistance rate to levofloxacin, which increased to 22.2% in cases of co-infection. Conclusions: These findings underscore the growing threat of antimicrobial resistance in Mycoplasma and Ureaplasma species and highlight the need for targeted public health strategies and diagnostic tools to manage infections caused by these organisms, particularly in high-risk populations. Background/Objectives: Sexual health research involving young adolescents remains scarce despite rising rates of early sexual debut, pregnancies, and sexually transmitted infections (STIs) in this population. We explored community stakeholders’ perspectives on engaging young adolescents in sexual health research in Western New York to inform strategies for engaging young adolescents in sexual health research. Methods: This qualitative descriptive study was conducted from April 2022 to June 2023. Seventeen community stakeholders, including health education teachers, youth counselors, and adolescent health providers, participated in semi-structured in-depth interviews. Data were analyzed using conventional content analysis, managed by MAXQDA 2020. The rigor and trustworthiness of the data were ensured through triangulation with observations, peer debriefing, team analysis, and respondent validation. Results: Participants were predominantly female (94.1%), 52.9% Black/African American, 41.2% White, and 5.9% Caucasian–Indian American, and aged 23–59 years. Four themes emerged: perspectives on conducting sexual health research with young adolescents, recruitment strategies, sexual health questions appropriate for young adolescents, and building readiness for participation in sexual health research. Participants reported the need for sexual health research with young adolescents and recommended building a trusting relationship and involving schools, parents, and trusted community organizations in the research process. Suggested research questions included those related to awareness of sex, STIs, available resources, experiences with sexual education, and desired support. The findings also revealed the need to initiate sexual health conversations early when children start asking questions, as a foundation for meaningful participation in sexual health research. Conclusions: The findings suggest that sexual health research with young adolescents is feasible and necessary, with implications for the design of developmentally appropriate sexual health research and interventions grounded in trust and community collaboration. Future research should explore the perspectives of caregivers and young adolescents to inform studies and programs that are attuned to young adolescents’ developmental needs. Full article

Global climate change has increased the frequency of compound drought–heatwave events (CDHEs), seriously threatening tropical forest ecosystems. However, due to the complex structure of natural tropical forests, related research remains limited. To address this, we focused on rubber plantations on Hainan Island, which have simpler structures, to explore the impacts of CDHEs on their primary productivity. We used Pearson and Spearman correlation analyses to select the optimal combination of drought and heatwave indices. Then, we constructed a Compound Drought–Heatwave Index (CDHI) using Copula functions to describe the temporal patterns of CDHEs. Finally, we applied a Bayes–Copula conditional probability model to estimate the probability of GPP loss under CDHE conditions. The main findings are as follows: (1) The Standardized Precipitation Evapotranspiration Index (SPEI-3) and Standardized Temperature Index (STI-1) formed the best index combination. (2) The CDHI successfully identified typical CDHEs in 2001, 2003–2005, 2010, 2015–2016, and 2020. (3) Temporally, CDHEs significantly increased the probability of GPP loss in April and May (0.58 and 0.64, respectively), while the rainy season showed a reverse trend due to water buffering (lowest in October, at 0.19). (4) Spatially, the northwest region showed higher GPP loss probabilities, likely due to topographic uplift. This study reveals how tropical plantations respond to compound climate extremes and provides theoretical support for the monitoring and management of tropical ecosystems. Full article

Trichomonas vaginalis infection (TVI) is the most common curable sexually transmitted infection (STI). In this study, we aimed to assess the performances of different tests for TVI diagnosis in symptomatic Greek women, evaluating the TVI prevalence rate (PR) in Greece and comparing the latter with TVI-PR estimates from Europe. A laboratory-based cross-sectional analysis and a meta-analysis were conducted. Of 399 symptomatic Greek women, 17 had TVI, corresponding to a TVI-PR of 4.3%. The commercial nucleic acid amplification test (NAAT) achieved a sensitivity of 94.1%, which was 6% higher than the sensitivity of the culture method, 35% higher than that of the wet mount test, and 59% higher than that of the Giemsa stain test. The wet mount test achieved the lowest positive predictive value of 76.9%. All the tests had high specificity levels and negative predictive values. Data from 34 European TVI-PR studies in symptomatic women were pooled. The TVI-PR established in our study was similar to the TVI-PR estimates of 4.8% in Europe and 4.5% in Greece, with the second being higher than those of 2.1% in Northwestern Europe and 1.5% in Southern Europe but closer to that of 6.7% in Türkiye. In Greece, a European country with a relatively high TVI-PR among symptomatic women, the highly sensitive and specific, automated, point-of-care NAAT would facilitate rapid, accurate TVI diagnosis and the treatment of this target population to meet the WHO’s goal of ending STI epidemics by 2030. The architecture of contemporary museums often emphasizes visual aesthetics, such as large volumes, open-plan layouts, and highly reflective finishes, resulting in acoustic challenges, such as excessive reverberation, poor speech intelligibility, elevated background noise, and reduced privacy. This study quantified the impact of surface—specific absorption treatments on acoustic metrics across eight gallery spaces. Room impulse responses calibrated virtual models, which simulated nine absorption scenarios (low, medium, and high on ceilings, floors, and walls) and evaluated reverberation time (T20), speech transmission index (STI), clarity (C50), distraction distance (rD), Spatial Decay Rate of Speech (D2,S), and Speech Level at 4 m (Lp,A,S,4m). The results indicate that going from concrete to a wooden floor yields the most rapid T20 reductions (up to −1.75 s), ceiling treatments deliver the greatest STI and C50 gains (e.g., STI increases of +0.16), and high-absorption walls maximize privacy metrics (D2,S and Lp,A,S,4m). A linear regression model further predicted the STI from T20, total absorption (Sabins), and room volume, with an 84.9% conditional R2, enabling ±0.03 accuracy without specialized testing. These findings provide empirically derived, surface-specific “first-move” guidelines for architects and acousticians, underscoring the necessity of integrating acoustics early in museum design to balance auditory and visual objectives and enhance the visitor experience. The concept of static and dynamic reservoirs is presented, and their performances are evaluated. The static reservoir is a simple reservoir with constant volume, and the dynamic one has a volume which varies as a function of the position of an internal piston coupled to a spring. The spring is compressed when the pressure in the chamber rises and exerts a proportional force on it. The two reservoirs are components to be used in compressed air energy storage systems. The study comprises a model of the compression machine as well as models of the two reservoirs. The filling processes are simulated, and the different variables are represented as a function of time. A reduced scale experimentation set-up is presented, and its behavior is first simulated. Then. the results are compared to the experimental records. Full article

In healthy conditions and cardiovascular diseases, the most abundant microparticles (MPs) in the bloodstream are those of platelet origin, but the direct effect of these microparticles on endothelial activation is poorly understood. The objective of this paper is to measure endothelial cell activation, as evaluated by the expression of the adhesion molecules E-selectin, VCAM-1, ICAM-1, and PECAM-1 in endothelial cell line HMEC-1 when stimulated with MPs produced by platelets stimulated in vitro with thrombin (TH), adenosine diphosphate (ADP), calcium ionophore (ICa), N-acetylglucosamine (NAcGlc), and without any stimulus. Platelets from healthy individuals induced the formation of MPs with different agonists. The results from the determination of the phenotype of the MPs showed that the expression of GPIIb/IIIa was significant, with median fold changes of TH = 2.2, ADP = 5.2, Ica = 7.0, and NAcGlc = 10.0. However, in HMEC-1 cells, the expression of adhesion molecules stimulated with MPs had a median change slightly higher for E-Sel expression (ranging from 1.4 to 4.2) and ICAM-1 expression (range 2.2 to 3.0), especially VCAM-1 expression (ranging from 15 to 18.8), all of which were significant. For PECAM-1, only stimulation with ICa (1.5) was significant, demonstrating that MPs elicit stimulus-dependent responses in endothelial cells. Platelet-derived MPs may have a potential role in modulating inflammation and other endothelial functions. Full article

Engineering Safety-Oriented Blasting-Induced Seismic Wave Signal Processing: An EMD Endpoint Suppression Method Based on Multi-Scale Feature

Blasting-induced seismic waves are typically nonlinear and non-stationary signals. The EMD-Hilbert transform is commonly used for time–frequency analysis of such signals. However, during the empirical mode decomposition (EMD) processing of blasting-induced seismic waves, endpoint effects occur, resulting in varying degrees of divergence in the obtained intrinsic mode function (IMF) components at both ends. The further application of the Hilbert transform to these endpoint-divergent IMFs yield artificial time–frequency analysis results, adversely impacting the assessment of blasting-induced seismic wave hazards. This paper proposes an improved EMD endpoint effect suppression algorithm that considers local endpoint development trends, global time distribution, energy matching, and waveform matching. The method first analyzes global temporal characteristics and endpoint amplitude variations to obtain left and right endpoint extension signal fragment S(t)L and S(t)R. Using these as references, the original signal is divided into “b” equal segments S(t)1, S(t)2 … S(t)b. Energy matching and waveform matching functions are then established to identify signal fragments S(t)i and S(t)j that match both the energy and waveform characteristics of S(t)L and S(t)R. Replacing S(t)L and S(t)R with S(t)i and S(t)j effectively suppresses the EMD endpoint effects. To verify the algorithm’s effectiveness in suppressing EMD endpoint effects, comparative studies were conducted using simulated signals to compare the proposed method with mirror extension, polynomial fitting, and extreme value extension methods. Three evaluation metrics were utilized: error standard deviation, correlation coefficient, and computation time. The results demonstrate that the proposed algorithm effectively reduces the divergence at the endpoints of the IMFs and yields physically meaningful IMF components. Finally, the method was applied to the analysis of actual blasting seismic signals. It successfully suppressed the endpoint effects of EMD and improved the extraction of time–frequency characteristics from blasting-induced seismic waves. This has significant practical implications for safety assessments of existing structures in areas affected by blasting. Full article

Multidrug-resistant (MDR) Mycoplasma genitalium (M. genitalium) presents a significant risk of treatment failure in many sexually transmitted infections (STIs) and can result in persistent and recurrent urethritis or cervicitis. This case report describes a recurrent M. genitalium urethritis resistant to sulfamethoxazole-trimethoprim (TMP-SMX), doxycycline, and moxifloxacin. The infection was ultimately cured after both the removal of a nidus of infection and through the use of Lefamulin. Lefamulin is a novel agent approved for use in community-acquired bacterial pneumonia and bacterial skin infections that may be useful in difficult sexually transmitted infections. Background/Objectives: Deciding whether or not to treat M. genitalium can be challenging as it can be a colonizer, or present with a symptomatic pathogen, and even if it is causing symptoms, it can be drug-resistant. Our objective here is to highlight important considerations on whether or not to treat and, if so, what options exist. Conclusions: In a world of increasing drug-resistant STIs, this case highlights the challenges of managing MDR M. genitalium and how foreign bodies can allow reoccurrence. Also highlighted in this case, Lefamulin appears to be a viable alternative line of treatment of MDR M. genitalium that defies other first-line antibiotics. Full article

Disrupted Vaginal Microbiota and Increased HPV Infection Risk Among Non-Vaccinated Women: Findings from a Prospective Cohort Study in Kazakhstan

Introduction: Vaginal microbiota has emerged as an important factor influencing human papillomavirus (HPV) persistence and host immunity. While HPV infection is often transient, persistent infections with high-risk HPV genotypes significantly increase the risk of cervical carcinogenesis. Thus, this study aims to investigate the association between microflora/sexually transmitted infections (STIs) and HPV infection, with a focus on the prevalence of coinfection and the potential role of genital tract microecological disorders. Methods: A prospective cohort study was conducted at a tertiary care center in Astana, Kazakhstan, between November 2024 and March 2025. A total of 396 non-pregnant women aged 18–45 years were enrolled during routine gynecological screening. Cervical samples were collected for high-risk HPV genotyping and the detection of 11 other vaginal microorganisms using real-time PCR. Results: HPV-positive women were significantly younger and more likely to be single compared to HPV-negative participants. They also had fewer pregnancies and deliveries and were more likely to use barrier contraception. Among STIs, Mycoplasma hominis demonstrated a significant association with HPV infection (adjusted OR = 2.16, 95% CI: 1.15–4.05, p = 0.017). Overall STI presence (adjusted OR = 2.16, p = 0.017) and STI multiplicity (adjusted OR = 1.36 per additional STI, p = 0.017) were also significantly associated with HPV positivity. Correlation analysis revealed a moderate association between Chlamydia trachomatis and Trichomonas vaginalis (ϕ = 0.39, p < 0.001), suggesting shared ecological or transmission pathways. Conclusion: The findings highlight the relevance of specific vaginal pathogens, particularly Mycoplasma hominis, and co-infection patterns in increasing the risk of HPV infection. These results underscore the importance of comprehensive STI screening and microbial profiling in cervical cancer prevention strategies, especially in populations with limited access to HPV vaccination. Further longitudinal and mechanistic studies are warranted to elucidate causal pathways and progression to cervical neoplasia. Full article
