Morphometric and Biochemical Analysis with Seed Protein Profiling of Passiflora Species Found in the Northeastern Himalayan Region of India

Passion fruit is an underutilised fruit in Northeastern India, known for its unique flavour and health benefits. This study analysed 15 genotypes (P1 to P15) to explore their morphological and biochemical traits related to fruit quality and yield. P. quadrangularis L. (P15) exhibited maximum flower length, fruit size, weight, juice content, shelf-life, and yield. P. edulis f. flavicarpa (P3, P5, and P2) had the highest seed count per fruit and antioxidant activity, along with greater chlorophyll and anthocyanin levels. Passiflora edulis Sims (P8 and P11) showed superior total soluble solids, carotenoids, and vitamin C. The study found that fruit shelf life positively correlated with seed weight, while the number of fruits per vine negatively correlated with seed traits and peel weight. Additionally, certain traits, such as total carotenoids, had strong positive correlations with reducing sugar and flavonoids. Principal component analysis revealed distinct trait relationships, particularly for genotypes P7 and P10. SDS-PAGE protein profiling indicated a significant distance between P3 and P14, emphasising genetic diversity. In conclusion, this research highlights the diverse morphological and biochemical characteristics of passion fruit genotypes, paving the way for the region’s improved fruit quality, yield, and breeding strategies. Some species of the genus Passiflora have leaf morphological adaptations that grow to influence the development of the plant in producing areas. Hence, the objective of this work is to quantify and characterize the leaf anatomy of passion fruit species distributed in the South American region, which can become an important strategy in the selection of species more adapted to the environment where they will be grown. This work evaluates the abaxial and adaxial cuticle thickness (ABCT and ADCT), abaxial and adaxial epidermis thickness (ABET and ADET), xylem diameter (XD), phloem diameter (PD), and thickness of the palisade parenchyma (TPP), of the species Passiflora quadrangularis L., Passiflora foetida L., Passiflora edulis Sims, Passiflora gibertii N.E Brown, Passiflora coccinea Aubl, Passiflora alata Curtis, Passiflora tenuifila Killip, Passiflora caerulea L., and Passiflora cincinnata Mast. Passion fruit species present differences in leaf anatomy, which may influence the plant’s development. The species Passiflora quadrangularis L. showed a greater thickness of cuticles, epidermis, conducting vessels, and palisade parenchyma. The species Passiflora edulis has higher density and stomatal functionality. All Passiflora species formed druses on their leaves. Full article

Nanoemulsions Containing Passiflora quadrangularis L. Fruit Extracts for Cosmetic Application and Skin Efficacy Study

This study aimed to determine the anti-collagenase, anti-elastase, and anti-hyaluronidase activities of Passiflora quadrangularis fruit extracts (epicarp, mesocarp, endocarp, and seed), develop stable nanoemulsions, and evaluate the efficacy of the nanoemulsions containing extracts in human volunteers. The results indicated that the epicarp and seed extracts exhibited collagenase, elastase, and hyaluronidase inhibition effects. Gallic acid was identified in the extracts, with the highest concentration found in the endocarp extract (1449.35 mg gallic acid/g extract) followed by the seed extract (839.63 mg gallic acid/g extract). The endocarp and seed extracts demonstrated good stability at different temperatures. Consequently, these extracts were selected for incorporation into nanoemulsions due to their high yield, excellent anti-tyrosinase and anti-aging activity, and good stability, making them suitable for cosmetic products. The nanoemulsions were prepared using the ultrasonication method, resulting in a particle size of 133.1 ± 0.8 nm, a polydispersity index of 0.33 ± 0.03, and a zeta potential of −61.8 ± 0.6 mV, indicating good stability. The nano-serum containing extracts was non-irritating and safe for the skin. Skin evaluation among human volunteers after 60 days of application revealed improvements in skin hydration and reduction in wrinkles. In conclusion, nanoemulsions containing P. quadrangularis fruit extracts have the potential to be effective cosmetic products. Full article

This research was designed to provide the first protocol to establish an efficient solution for direct organogenesis regeneration in Passiflora quadrangularis using nodal explants from young shoots. Passifloraceae tissue culture has been associated with problems such as recalcitrance, sensitivity to ethylene accumulation and browning of explants due to the presence of phenols in the tissues. Due to the high rate of endogenous contamination of the explants, a preliminary experiment was performed. The best results of surface sterilization were obtained using the pretreatment with 70% EtOH, 1 min and 50% NaOCl, 10 min along with the treatment of Rifampicin 15 µg/mL and Benomyl 2 g/L. The effects of plant growth regulators on the induction of direct organogenesis, multiplication of shoots in subcultures and in vitro rooting were evaluated. Additional compounds such as AgNO3 and Pluronic F-68 were added to the culture media in order to reduce the effects of phenols and the sudden browning of the explants. Shoot proliferation increased to the sixth subculture after which it decreased. A maximum of 7.17 shoots were obtained from one shoot on Murashige and Skoog (MS) medium supplemented with 2 mg/L 6-benzylaminopurine and 1 mg/L thidiazuron. Supplementation of ½ MS medium with 1 mg/L 1-naphthaleneacetic acid was conducing to root formation in 61.11% of shoots. After acclimatization, the plants showed vigorous growth, green leaves, and well-developed roots. Although this species has previously shown difficulty in in vitro propagation, this protocol established based on the results proved to be efficient and reproducible. Full article

Chemical Composition and Antioxidant Activity of the Main Fruits Consumed in the Western Coastal Region of Ecuador as a Source of Health-Promoting Compounds

We studied 19 different tropical fruits traditionally consumed in the coastal lowlands of Ecuador to determine their chemical composition and antioxidant activity. Carambola (Averrhoa carambola L.) had the highest total phenolic, flavonoid, and total antioxidant capacity values, whereas guava fruits (Psidium guajava L.) had the highest vitamin C values. The main organic acids identified were lactic, citric, and acetic acids, and the highest amount of lactic acid was found in soursop fruits (Annona muricata L.), whereas Ecuadorian ivory palm (Phytelephas aequatorialis Spruce) and guava fruits had the highest acetic acid content. Guava also had the highest citric acid content; the highest concentration of oxalic acid was found in carambola. In terms of sugar content, giant granadilla (Passiflora quadrangularis L.) had the highest values of glucose, and red mombin (Spondias mombin L.) had the largest values for fructose and guava for sucrose. Chili pepper (Capsicum chinense Jacq) proved to be the main source of carotenoids, lutein, and β-carotene, anthocyanins, and vitamin C. The results here increase our knowledge regarding the composition of the main fruits consumed on the west coast of Ecuador to facilitate recommendations as potential sources of health-promoting compounds.
