Accelerometer-based wearables offer a cost-effective solution for managing match and training loads in invasion team sports. However, a multitude of acceleration-derived metrics, each employing different algorithms, has led to inconsistent and incomparable outcomes across studies and devices. This article reviews the mathematical procedures underlying whole-body mechanical load metrics, clarifies their conceptual differences, and proposes refinements to enhance standardization. Synthetic data were employed to investigate conceptual differences, while experimental accelerometric data (463 time series) from a set of elite handball training sessions (involving 16 players) were used to implement the corrected equations and analyze statistical relationships. Analysis of synthetic data revealed that derivative-based metrics, such as Jerk Modulus (typically referred to as Player Load) and corrected Accel’Rate (cAccel’Rate), tend to amplify noise compared to acceleration-based metrics, such as universal Dynamic Stress Load (uDSL) and Body Load. Experimental results indicated that when metrics were summed, their values were nearly identical. In time-series comparisons, Jerk Modulus and cAccel’Rate were predictably found to be nearly identical, while Body Load was the most distinct. Acceleration-based metrics are preferable to derivative-based ones. Sports scientists should lead the design and validation of such metrics, ensuring methodological rigor, transparency, and innovation while preventing commercial interests from introducing rebranded variables with undisclosed scaling factors and unclear calculations. Corneal endothelial cells (CE) are critical for the cornea’s transparency. For severe corneal damage, corneal tissue transplantation is the most promising option for restoring vision. However, CE apoptotic cell death occurs during the storage of donor corneas for transplantation. This study used small interfering (si)RNA-mediated silencing of pro-apoptotic proteins as a novel strategy to protect CE against apoptosis. Therefore, the pro-apoptotic proteins Bax and Bak were silenced in the human corneal endothelial cell line (HCEC-12) by transfection with Accell™siRNA without any adverse effects on cell viability. When apoptosis was induced, e.g., etoposide, the caspase-3 activity and Annexin V-FITC/PI assay indicated a significantly reduced apoptosis rate in Bax+Bak-siRNA transfected HCECs compared to control (w/o siRNA). TUNEL assay in HCECs exposed also significantly lower cell death in Bax+Bak-siRNA (7.5%) compared to control (w/o siRNA: 32.8%). In ex vivo donor corneas, a significant reduction of TUNEL-positive CEs in Bax+Bak-siRNA corneas (8.1%) was detectable compared to control-treated corneas (w/o siRNA: 27.9%). In this study, we demonstrated that suppressing pro-apoptotic siRNA leads to inhibiting CE apoptosis. Gene therapy with siRNA may open a new translational approach for corneal tissue treatment in the eye bank before transplantation, leading to graft protection and prolonged graft survival. Sport coaches increasingly rely on external load metrics for designing effective training programs. However, their accuracy in estimating internal load is inconsistent, and their ability to predict autonomic nervous system (ANS) deterioration is unknown. This study aimed to evaluate the relationships between internal and external training load metrics and ANS recovery and function in college football players. Football athletes were recruited from a D1 college in the southeastern US and prospectively followed for 27 weeks. Internal load was estimated via exercise cardiac load (ECL; average training heartrate (HR) × session duration) and measured with an armband monitor equipped with electrocardiographic capabilities (Warfighter MonitorTM (WFM), Tiger Tech Solutions, Miami, FL, USA). External load was estimated via the summation and rate of acceleration and decelerations as measured by a triaxial accelerometer using the WFM and an accelerometer-based (ACCEL) device (Catapult Player Load, Catapult Sports, Melbourne, Australia) worn on the mid-upper back. Baseline HR, HR variability (HRV) and HR recovery served as the indicators for ANS recovery and function, respectively. For HRV, two, time-domain metrics were measured: the standard deviation of the NN interval (SDNN) and root mean square of the standard deviation of the NN interval (rMSSD). Linear regression models evaluated the associations between ECL, ACCEL, and the indicators of ANS recovery and function acutely (24 h) and cumulatively (one- and two-week). Athletes (n = 71) were male and, on average, 21.3 ± 1.4 years of age. Acute ECL elicited stronger associations for 24 h baseline HR (R2 0.19 vs. 0.03), HR recovery (R2 0.38 vs. 0.07), SDNN (R2 0.19 vs. 0.02) and rMSSD (R2 0.19 vs. 0.02) compared to ACCEL. Similar results were found for one-week: 24 h baseline HR (R2 0.48 vs. 0.05), HR recovery (R2 0.55 vs. 0.05), SDNN (R2 0.47 vs. 0.05) and rMSSD (R2 0.47 vs. 0.05) and two-week cumulative exposures: 24 h baseline HR (R2 0.52 vs. 0.003), HR recovery (R2 0.57 vs. 0.05), SDNN (R2 0.52 vs. 0.003) and rMSSD (R2 0.52 vs. 0.002). Lastly, the ACCEL devices weakly correlated with ECL (rho = 0.47 and 0.43, p < 0.005). Our findings demonstrate that ACCEL poorly predicted ANS deterioration and underestimated internal training load. ACCEL devices may “miss” the finite window for preventing ANS deterioration by potentially misestimating training loads acutely and cumulatively. Full article

It has been shown that the loss of function of the BRCA1, BRCA2, and PALB2 genes due to a number of hereditary mutations or chromosomal aberrations can affect the effectiveness of chemotherapy treatment and disease prognosis in patients with various types of cancer, and in particular in breast cancer. Thus, the aim of the work was to evaluate the predictive and prognostic potential of DNA copy number aberrations and mutations in the BRCA1, BRCA2, and PALB2 genes in breast tumors. Materials and Methods: The study included 66 patients with breast cancer. DNA copy number aberrations (CNA) were assessed by high-density CytoScanHD™ Array micro matrix analysis. Gene mutations were assessed by sequencing on the MiSeq™ Sequencing System using the Accel-Amplicon BRCA1, BRCA2, and PALB2 Panel. Results: It has been established that the presence of a normal copy number of PALB2 is associated with a lack of response to chemotherapy in Taxotere-containing treatment regimens (p = 0.05). In addition, the presence of a PALB2 deletion is associated with 100% metastatic survival rates (log-rank test p = 0.04). As a result of sequencing, 25 mutations were found in the BRCA1 gene, 42 mutations in BRCA2, and 27 mutations in the PALB2 gene. The effect of mutations on the effectiveness of treatment is controversial, but an effect on the survival of patients with breast cancer has been shown. So, in the presence of pathogenic mutations in the BRCA2 gene, 100% metastatic survival is observed (log-rank test p = 0.05), as well as in the elimination of PALB2 mutations during treatment (log-rank test p = 0.07). Conclusion: Currently, there is little data on the effect of chromosomal aberrations and mutations in the BRCA1/2 and PALB2 genes on the effectiveness of treatment and prognosis of the disease. At the same time, the study of these genes has great potential for testing focused on a personalized approach to the treatment of patients with breast cancer. Full article

We investigated the use of multi-sensor physical activity monitors, body mass index (BMI), and heart rate (HR) to measure energy expenditure (EE) of various physical activity levels among Chinese collegiate students, compared with portable indirect calorimetry. Methods: In a laboratory experiment, 100 college students, 18–25 years old, wore the SenseWear Pro3 Armband™ (SWA; BodyMedia, Inc., Pittsburg, PA, USA) and performed 7 different physical activities. EE was measured by indirect calorimetry, while body motion and accelerations were measured with an SWA accelerometer. Special attention was paid to the analysis of unidirectional and three-directional accelerometer output. Results: Seven physical activities were recorded and distinguished by SWA, and different physical activities demonstrated different data features. The mean values of acceleration ACz (longitudinal accel point, axis Z) and VM (vector magnitude) were significantly different (p = 0.000, p < 0.05) for different physical activities, whereas no significant difference was found in one single physical activity with varied speeds (p = 0.9486, p > 0.05). When all physical activities were included in a correlation regression analysis, a strong linear correlation between the EE and accelerometer reporting value was found. According to the correlation analysis, sex, BMI, HR, ACz, and VM were independent variables, and the EE algorithm model demonstrated a high correlation coefficient R2 value of 0.7. Conclusions: The predictive energy consumption model of physical activity based on multi-sensor physical activity monitors, BMI, and HR demonstrated high accuracy and can be applied to daily physical activity monitoring among Chinese collegiate students. The Jiaju ancient landslide is a giant landslide located upstream of the Dadu River, eastern Tibetan Plateau, with a volume of approx. 7.04 × 108 m3. The Jiaju ancient landslide is complex and comprises five secondary sliding bodies, e.g., the Jiaju landslide (H01), Niexiaping landslide (H02), Xiaobawang landslide (H03), Niela landslide (H04), and Mt.-peak landslide (H05). Affected by regional neotectonic movement, heavy rainfall, river erosion, and lithology, the secondary sliding bodies of the Jiaju ancient landslide are undergoing significantly different creep-sliding deformation, which will cause great damage to villages, roads, and rivers around the sliding bodies. Combined with the SBAS-InSAR method, Sentinel-1A data from June 2018 to August 2021, remote sensing and field surveys, this study obtained the Jiaju ancient landslide deformation characteristics and deformation rate in the line-of-sight direction (VLOS), slope (VSlope), and vertical (VVertical). It is concluded that the maximum deformation rate of the Jiaju ancient landslide is significant. The maximum of VLOS, VSlope, and VVertical are −179 mm/a, −211 mm/a, and −67 mm/a, respectively. The Niela landslide (H04), Jiaju landslide (H01), and Mt.-peak landslide (H05) are very large and suffer strong deformation. Among these, the Niela landslide (H04) is in the accelerative deformation stage and at the Warn warning level, and the Jiaju landslide (H01) is in the creep deformation and attention warning level, especially heavy rainfall, which will accelerate landslide deformation and trigger reactivation. Because the geological structure is very complex for the Jiaju ancient landslide and strong neotectonic movement, under heavy rainfall, the secondary landslide creep-sliding rate of the Jiaju ancient landslide is easily accelerated and finally slides in part or as a whole, resulting in river blocking. It is suggested to strengthen the landslide deformation monitoring of the Niela landslide and Jiaju landslide and provide disaster mitigation and prevention support to the government and residents along the Dadu River watershed. While the Player Load is a widely-used parameter for physical demand quantification using wearable accelerometers, its calculation is subjected to potential errors related to rotational changes of the reference frame. The aims of this study were (i) to assess the concurrent validity of accelerometry-based Player Load against force plates; (ii) to validate a novel metric, the Accel’Rate overcoming this theoretical issue. Twenty-one recreational athlete males instrumented with two triaxial accelerometers positioned at the upper and lower back performed running-based locomotor movements at low and high intensity over six in-series force plates. We examined the validity of the Player Load and the Accel’Rate by using force plates. Standard error of the estimate was small to moderate for all tested conditions (Player Load: 0.45 to 0.87; Accel’Rate: 0.25 to 0.95). Accel’Rate displayed trivial to small mean biases (−1.0 to 6.1 a.u.) while the Player Load displayed systematic very large to extremely large mean biases (17.1 to 226.0 a.u.). These findings demonstrate a better concurrent validity of the Accel’Rate compared to the Player Load. This metric could be used to improve the estimation of whole-body mechanical load, easily accessible in sport training and competition settings. Full article

Skilled birth attendance and institutional delivery have been advocated for reducing maternal, neonatal mortality and infant mortality (NMR and IMR). This paper examines the role of place of delivery with respect to neo-natal and infant mortality in India using four rounds of the Indian National Family Health Survey conducted in 2015–2016. The place of birth has been categorized as “at home” or “public and private institution.” The role of place of delivery on neo-natal and infant mortality was examined by using multivariate hazard regression models adjusted for clus-tering and relevant maternal, socio-economic, pregnancy and new-born characteristics. There were 141,028 deliveries recorded in public institutions and 54,338 in private institutions. The esti-mated neonatal mortality rate in public and private institutions during this period was 27 and 26 per 1000 live births respectively. The study shows that when the mother delivers child at home, the chances of neonatal mortality risks are higher than the mortality among children born at the health facility centers. Regression analysis also indicates that a professionally qualified provider′s antenatal treatment and assistance greatly decreases the risks of neonatal mortality. The results of the study illustrate the importance of the provision of institutional facilities and proper pregnancy in the prevention of neonatal and infant deaths. To improve the quality of care during and imme-diately after delivery in health facilities, particularly in public hospitals and in rural areas, accel-erated strengthening is required. Full article

The gene ABK52392 from the thermophilic bacterium Acidothermus cellulolyticus 11B was predicted to be endoglucanase and classified into glycoside hydrolase family 12. ABK52392 encodes a protein containing a catalytic domain and a carbohydrate binding module. ABK52392 was cloned and functionally expressed in Escherichia coli. After purification by Ni-NTA agarose affinity chromatography and Q-Sepharose® Fast Flow chromatography, the properties of the recombinant protein (AcCel12B) were characterized. AcCel12B exhibited optimal activity at pH 4.5 and 75 °C. The half-lives of AcCel12B at 60 and 70 °C were about 90 and 2 h, respectively, under acidic conditions. The specific hydrolytic activities of AcCel12B at 70 °C and pH 4.5 for sodium carboxymethylcellulose (CMC) and regenerated amorphous cellulose (RAC) were 118.3 and 104.0 U·mg−1, respectively. The Km and Vmax of AcCel12B for CMC were 25.47 mg·mL−1 and 131.75 U·mg−1, respectively. The time course of hydrolysis for RAC was investigated by measuring reducing ends in the soluble and insoluble phases. The total hydrolysis rate rapidly decreased after the early stage of incubation and the generation of insoluble reducing ends decreased earlier than that of soluble reducing ends. High thermostability of the cellulase indicates its potential commercial significance and it could be exploited for industrial application in the future. Full article
