‘Fengtang’ plum is increasingly cultivated in southern China under mountain cultivation conditions, which involve lower temperatures, higher irradiance, and thinner soils. These conditions may uniquely shape the rhizosphere microbiome and influence tree health, yet their effects remain poorly understood. This study aimed to characterize the structural and taxonomic characteristics of the rhizosphere microbial community of ‘Fengtang’ plum grown under mountain cultivation conditions, and to identify key microbial groups associated with plant growth. We performed 16S rRNA and ITS high-throughput sequencing on rhizosphere and non-rhizosphere soil samples from ‘Fengtang’ plum orchards. Significant differences in α-diversity and β-diversity were observed between rhizosphere and non-rhizosphere communities. Dominant bacterial phyla included Pseudomonadota, Actinobacteriota, Acidobacteriota, and Chloroflexi; Ascomycota and Basidiomycota dominated in fungal communities. Dominant microbial groups were consistent across phylum, class, and order levels. Beneficial genera such as Streptomyces, Bacillus, and Rhizobium were enriched in the rhizosphere, and are considered putative core functional genera based on their known plant-growth-promoting traits. The microbial community in the rhizosphere shows distinct compositional patterns that may be linked to microecological balance. This study seeks to provide a comprehensive perspective for understanding the microbial communities associated with the plum tree rhizosphere. Full article

‘Fengtang’ plum, a specialty fruit variety from Guizhou Province, is characterized by its high total soluble solids (TSS) and well-balanced sugar–acid ratio. In this study, widely targeted metabolomics (UPLC-MS/MS) combined with multivariate statistical analysis was employed to systematically monitor dynamic changes in sugar–acid metabolism across three critical developmental stages of the fruit: pre-veraison stage (PR), veraison stage (VR), and full veraison stage (FR). The aim was to elucidate the intrinsic mechanisms underlying high sugar accumulation. The results indicated that TSS content increased significantly during fruit maturation, reaching up to 17%, while titratable acid (TA) content continuously decreased. Consequently, the sugar–acid ratio rose substantially from 21.5 at the early stage to 43.8. A comprehensive metabolomic analysis led to the identification of 2003 metabolites, with notable emphasis on phenolic acids, amino acid derivatives, sugars, and other major classes. Key metabolic pathways from the KEGG database were also examined. Enrichment analysis of these pathways revealed that central metabolic routes—including fructose and mannose metabolism, galactose metabolism, and the tricarboxylic acid (TCA) cycle—played crucial roles in the dynamic accumulation of sugar–acid components. Notably, oligosaccharides such as cottonseed sugar and pine trisaccharide were observed to accumulate significantly during fruit ripening. These findings clarify the metabolic foundation and mechanisms contributing to the high sugar quality of ‘Fengtang’ plum, thereby providing a theoretical basis for precise fruit quality regulation and the optimization of post-harvest preservation strategies. Fengtang plum, a novel cultivar recently developed in China, has gained huge popularity due to its large fruit size, crisp sweetness, distinctive aroma, and notable resistance to brown rot caused by Monilinia spp. To investigate microbial community dynamics during fruit development, we analyzed samples from three phenological stages: fruit-setting (BSP1), veraison (BSP2), and maturity (BSP3). Our results demonstrated stage-specific microbial succession patterns: alpha diversity indices (observed species, ACE, PD_whole_tree) significantly increased at BSP2/BSP3 versus BSP1, accompanied by diverging Shannon index trends between bacteria (progressive enhancement) and fungi (stage-dependent reduction). Bacterial communities maintained Proteobacteria and Firmicutes dominance while accumulating low-abundance species (18.06–61.84%), whereas Ascomycota constituted the persistent fungal phylum with Trichoderma, reaching 95.91% dominance at BSP3. Community differentiation primarily arose from stage-specific bacteria Ralstonia, Brevundimonas, and Limnobacter, and dominant fungi Trichoderma and Cladosporium. Bacterial metabolic shifts were predicted to transition from basic energy production to complex organic/aromatic compound utilization, contrasting with fungal transitions from pathogen–saprophyte competition to saprophytic dominance. While the enrichment of Lactobacillus and Trichoderma during mid-to-late stages may suggest potential associations with aromatic compound production and fungal pathogen resistance, these hypotheses require validation through targeted metabolomics and pathogen challenge experiments. This study elucidates microbial community succession patterns during Fengtang plum development; notably, functional predictions were inferred from 16S/ITS sequencing data rather than direct metagenomic or metatranscriptomic analyses, thus limiting mechanistic interpretations, though future work integrating multi-omics approaches would strengthen functional insights. To investigate the differences in the volatile compounds of plum fruit samples from different cultivars, the volatile compounds of the ‘Fengtang’ plum, ‘Kongxin’ plum, and ‘Cuihong’ plum fruits were analyzed using headspace solid-phase microextraction–gas chromatography–mass spectrometry (HS-SPME-GC-MS). The results demonstrated that a total of 938 volatile compounds were identified in three plum fruits, including 200 terpenoids, 171 esters, 116 heterocyclic compounds, 89 hydrocarbons, 82 ketones and alcohols, 63 aldehydes, 54 aromatic hydrocarbons, 21 amines, 18 acids, 17 phenols, 10 nitrogenous compounds, 7 sulfur compounds, and other compounds, 470 of which were common to all the cultivars. Moreover, 704, 691, and 704 volatile substances were detected, respectively, in the ‘Fengtang’ plum, ‘Kongxin’ plum, and ‘Cuihong’ plum, with terpenoids, esters, and heterocycles as the main compounds, accounting for 62.12~72.03% of the volatile compounds. The results of principal component analysis (PCA) and cluster analysis (CA) illustrated that the ‘Fengtang’ plum and ‘Cuihong’ plum were similar in terms of volatile compounds; the ‘Kongxin’ plum compounds were different from those in the other cultivars. Orthogonal partial least squares discriminant analysis was performed, revealing the typical volatile compounds that differed among the plum fruits of the different varieties; thus, the three plum fruits could be better distinguished. These results can provide a theoretical basis for the studies of plum fruit flavor, quality, and geographical origin identification. Full article

Herein, an accurate and sensitive method was developed for detecting four stereoisomers of propiconazole in “Fengtang” plum by LC-MS/MS. The mean recovery of four propiconazole stereoisomers ranged from 79.42 to 104.10% at three adding levels with reasonable RSD of 1.54–11.68%, and the LOD and LOQ of the four stereoisomers was 0.0005 mg/kg and 0.004 mg/kg, respectively. In addition, the residue and selective degradation of propiconazole stereoisomers in plums were investigated by storage at 20 °C and 4 °C. The half-lives of propiconazole stereoisomeric during storage were 9.49–15.40 d at 20 °C, and 21.00–28.88 d at 4 °C. The degradation of (2R,4R)-propiconazole and (2R,4S)-propiconazole in stored plums was slightly slower than that of the corresponding enantiomers (2S,4S)-propiconazole and (2S,4R)-propiconazole. The total residues of propiconazole were 0.026–0.487 mg/kg in the plum storage period, and the water washing could remove 49.35% to 54.65% of the propiconazole residue in plum. The hardness of plums treated with propiconazole was generally higher than that of control in the middle and late stages of storage. The effects of propiconazole on the total soluble solid content of plums were different at 20 °C and 4 °C. This study provides a scientific reference for the food safety evaluation of the “Fengtang” plum after the application of propiconazole during the storage period. Full article

In the present study, the cuticular wax morphology, composition and the relationship with storage quality in three plum cultivars of Prunus salicina ‘Kongxin’ (KXL), Prunus salicina ‘Fengtang’ (FTL) and Prunus salicina ‘Cuihong’ (CHL) were investigated during storage at room temperature of 25 ± 1 °C. The results illustrated that the highest cuticular wax concentration was discovered in KXL, followed by FTL and the lowest in CHL. The fruit wax composition of the three plum cultivars was similar and principally composed of alkanes, alcohols, fatty acids, ketones, aldehydes, esters, triterpenes and olefins. Alcohols, alkanes and triterpenes were the dominant fruit wax compounds of the three plum cultivars. After storage for 20 d at room temperature, the variation of cuticular wax crystal structure and composition showed significant cultivar-associated differences. The total wax content decreased for FTL and CHL and increased for KXL, and the wax crystal degraded and melted together over time. The higher contents of the main components in the three plum cultivars were nonacosane, 1-triacontanol, 1-heneicosanol, nonacosan-10-one, octacosanal, ursolic aldehyde and oleic acid. Alcohols, triterpenes, fatty acids and aldehydes were most dramatically correlated with the softening of fruit and storage quality, and alkanes, esters and olefins were most significantly correlated with the water loss. Nonacosane and ursolic aldehyde can enhance the water retention of fruit. Overall, this study will provide a theoretical reference for the further precise development of edible plum fruit wax. Plums are one of the most popular stone fruits worldwide owing to their high nutritional value. After harvest, plum fruit quality and flavor change during storage; however, little is known about the changes in metabolites during this period. A comprehensive comparison of primary metabolites in ‘Fengtang’ plum fruits during storage is performed using widely targeted primary metabolomics. A total of 272 primary metabolites were identified by means of ultra-performance liquid chromatography and tandem mass spectrometry (UPLC-MS/MS) in the plums at different storage periods. There was a significant increase in the relative amounts of twenty-eight lipids, twenty amino acids and their derivatives, thirteen organic acids, ten saccharides and alcohols, six nucleotides and their derivatives, and two vitamins. A Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis of differential metabolites revealed that glucosinolate biosynthesis, starch and sucrose metabolism, ascorbate and aldarate metabolism, lysine degradation, and other metabolic pathways were significantly enriched; therefore, changes in these metabolic pathways may be key to the quality and flavor change in ‘Fengtang’ plum fruits during storage. Our results provide a theoretical foundation and technical support to evaluate ‘Fengtang’ plum fruit quality. Full article
