Background: Quality of life (QoL) may change in female patients with breast cancer over time due to its dynamic construct. Recent studies have provided statistics on the diverse predictors of QoL in breast cancer patients. Still, the literature findings on the prognostication of QoL are scarce. Objectives: With this meta-analysis, we aim to describe the dynamics of overall QoL and its dimensions: mental status, physical functioning, and social interactions. Methods: To reach this aim, we will systematically evaluate the peer-reviewed literature on QoL of women who have undergone either mastectomy or breast-conserving surgery. The proposed study will focus on, but not be limited to, the analysis of BIRS, FACT-B, and EORTC QLQ-C30 questionnaires, which are most commonly used in the assessment of the quality of life of cancer patients. Then, we will extract the following outcome measures: The participants’ age, time since surgery, type of tumor removal procedure (mastectomy or breast-conserving surgery), breast reconstruction technique, mean value, and standard deviation in a QoL score. A random-effects model will then compute the pooled QoL and construct the trend lines for scores received from each diagnostic tool. The findings will be adjusted according to the reconstruction techniques and tumor removal surgery. Finally, we will model the QoL dynamics with a set of predictors identified in the extracted studies. Conclusions: The study findings may serve as a tool for stratifying female patients with breast cancer by the risk of significant reduction in QoL. Full article

On 21 January 2024, asteroid 2024BX1, discovered the three hours before, fell to Earth south of Ribbeck in the Havelland region of Germany. In this study, fragments of the Ribbeck meteorite, characterized by white and gray colors lithology, were examined for their chemical and phase compositions. The white lithology fragment exhibited a homogeneous chemical and phase structure typical of orthopyroxene, which crystallizes in the orthorhombic system. The gray lithology fragment showed a greater diversity in chemical and phase compositions. Raman spectra analysis revealed that, in addition to the pyroxenes found in the white lithology fragment, minerals from the olivine group (fayalite and forsterite) were also present, along with plagioclase and sulfur in pure crystalline form. The serotonin 5-HT6 receptor (5-HT6R), expressed almost exclusively in the brain, affects the Cdk5 signaling as well as the mTOR pathway. Due to the association of 5-HT6R signaling with pathways involved in cancer progression, we decided to check the usefulness of 5-HT6R ligands in the treatment of CNS tumors. For this purpose, a new group of low-base 5-HT6R ligands was developed, belonging to arylsulfonamide derivatives of cyclic arylguanidines. The selected group of molecules was also tested for their antiproliferative activity on astrocytoma (1321N1) and glioblastoma (U87MG, LN-229, U-251) cell lines. Some of the molecules were subjected to ADMET tests in vitro, including lipophilicity, drug binding to plasma proteins, affinity for phospholipids, drug–drug interaction (DDI), the penetration of the membrane (PAMPA), metabolic stability, and hepatotoxicity as well as in vivo cardiotoxicity in the Danio rerio model. Two antagonists with an affinity constant Ki < 50 nM (PR 68 Ki = 37 nM) were selected. These compounds were characterized by very high selectivity. An analysis of pharmacokinetic parameters for the lead compound PR 68 confirmed favorable properties for administration, including passive diffusion and acceptable metabolic stability (metabolized in 49%, MLMs). The compound did not exhibit the potential for drug–drug interactions. Full article

The aim of the article was to compare the landscape and cultural value of cemeteries located around wooden churches on the Polish–Czech border in the Lower Silesian Voivodeship. Research regarding the history of the villages was undertaken, describing their development and the construction of the churches and the cemeteries so as to compare changes in cemetery spatial layout, architecture, and landscape. The villages involved were Grzmiąca, Kamieńczyk, Międzygórze, Nowa Bystrzyca, Rybnica Leśna, and Zalesie. The following analyses were conducted: assessment of the visual aspects of the landscape, evaluation of the architectural value, and assessment of land use. In the landscape of Lower Silesia, churchyard cemeteries, which bear witness to the rich past of this region, are gradually but remorselessly deteriorating. Adverse changes are occurring to their spatial layout, to church buildings, as well as in architecture and greenery. The article presents a new approach to the subject of sacred sites through multi-faceted research aimed at protecting the fragile and changing religious landscape. Full article

GSK-3β, IKK-β, and ROCK-1 kinases are implicated in the pathomechanism of Alzheimer’s disease due to their involvement in the misfolding and accumulation of amyloid β (Aβ) and tau proteins, as well as inflammatory processes. Among these kinases, GSK-3β plays the most crucial role. In this study, we present compound 62, a novel, remarkably potent, competitive GSK-3β inhibitor (IC50 = 8 nM, Ki = 2 nM) that also exhibits additional ROCK-1 inhibitory activity (IC50 = 2.3 µM) and demonstrates anti-inflammatory and neuroprotective properties. Compound 62 effectively suppresses the production of nitric oxide (NO) and pro-inflammatory cytokines in the lipopolysaccharide-induced model of inflammation in the microglial BV-2 cell line. Furthermore, it shows neuroprotective effects in an okadaic-acid-induced tau hyperphosphorylation cell model of neurodegeneration. The compound also demonstrates the potential for further development, characterized by its chemical and metabolic stability in mouse microsomes and fair solubility. Full article

We explore the crystal structure and luminescent properties of a new 1D organic–inorganic hybrid, MHy2SbI5, based on methylhydrazine. The compound reveals the red photoluminescence (PL) originating from the 5s2 electron pairs of Sb(III) as well as complex structural behavior. MHy2SbI5 crystalizes in two polymorphic forms (I and II) with distinct thermal properties and structural characteristics. Polymorph I adopts the acentric P212121 chiral space group confirmed by SHG, and, despite a thermally activated disorder of MHy, does not show any phase transitions, while polymorph II undergoes reversible low-temperature phase transition and high-temperature reconstructive transformation to polymorph I. The crystal structures of both forms consist of 1D perovskite zig-zag chains of corner-sharing SbI6 octahedra. The intriguing phase transition behavior of II is associated with the unstable arrangement of the [SbI5]2− chains in the structure. The energy band gap (Eg) values, estimated based on the UV-Vis absorption spectra, indicate that both polymorphs have band gaps, with Eg values of 2.01 eV for polymorph I and 2.12 eV for polymorph II. The research presented in this article concerns the issue of rural tourism, which is associated with significant economic and landscape changes in the countryside. It is important that tourist development in these areas be subject to the principles of sustainable development, which are important for the preservation of the environment in which tourism operates. The research topic is related to two of the most salient features of rural tourism today, environmental sustainability and pro-environmental behavior, which are considered the right ways to achieve sustainability goals. The main research aim presented is to indicate which of the postulates of sustainable development are being implemented and how concerning the functioning and development of rural tourism is on the Polish tourism market. Another goal is to indicate what types of resources in the rural environment are the basis for the preparation of a tourist product; for whom and how these resources are to be used; as well as the participation of the local community in the management of the resources that constitute its own environment, one of the basic principles of sustainable development. The primary method used was a document analysis and an analysis of selected websites presenting rural environmental resources used in this form of tourism. The research identified that sustainable approaches to tourism development in rural areas in Poland are appropriate with respect to the resources available there and to both natural and cultural values. For resources used in a rural environment, emphasis is placed on the protection of both elements. It has been identified that one idea for their protection is sustainable sharing, which assumes rational use to an extent that would not affect the nature of the assets but at the same time could benefit the local community. Both values are used in different ways but, in addition to their purely recreational value, their educational value is equally important each time. Full article

This article presents the tourism potential of the Polish–Czech borderlands (Klodzko Land, Lower Silesia, Poland), in which historical sacred sites, namely cemeteries and churches, play a significant role. An analysis of the most important features of sacred sites was carried out to present the diversity of the architectural richness of the region. At the same time, attention was paid to additional elements, such as their visual aspects and their impact on shaping space. The research is based on the sacred sites located on the Polish–Czech borderland. As part of the work, an analysis of historical, natural, and landscape conditions was performed along with the spatial layout of 106 cemeteries. During the field research, inventories of landscape and natural values were carried out. The assessment of landscape value was carried out using an original concept to enable an indication of site’s tourism potential on the basis of visual aspects of its space. The article and the research methods contained within concern interdisciplinary research combining architecture, landscape architecture, socio-economic geography, and tourism. Full article

Alzheimer’s disease (AD) is a neurodegenerative disorder, for which there is no effective cure. Current drugs only slow down the course of the disease, and, therefore, there is an urgent need to find effective therapies that not only treat, but also prevent it. Acetylcholinesterase inhibitors (AChEIs), among others, have been used for years to treat AD. Histamine H3 receptors (H3Rs) antagonists/inverse agonists are indicated for CNS diseases. Combining AChEIs with H3R antagonism in one structure could bring a beneficial therapeutic effect. The aim of this study was to find new multitargetting ligands. Thus, continuing our previous research, acetyl- and propionyl-phenoxy-pentyl(-hexyl) derivatives were designed. These compounds were tested for their affinity to human H3Rs, as well as their ability to inhibit cholinesterases (acetyl- and butyrylcholinesterases) and, additionally, human monoamine oxidase B (MAO B). Furthermore, for the selected active compounds, their toxicity towards HepG2 or SH-SY5Y cells was evaluated. The results showed that compounds 16 (1-(4-((5-(azepan-1-yl)pentyl)oxy)phenyl)propan-1-one) and 17 (1-(4-((6-(azepan-1-yl)hexyl)oxy)phenyl)propan-1-one) are the most promising, with a high affinity for human H3Rs (Ki: 30 nM and 42 nM, respectively), a good ability to inhibit cholinesterases (16: AChE IC50 = 3.60 µM, BuChE IC50 = 0.55 µM; 17: AChE IC50 = 1.06 µM, BuChE IC50 = 2.86 µM), and lack of cell toxicity up to 50 µM. Paenibacillus larvae is a deadly pathogen for bee brood, which can lead to the death of entire colonies. The presence of specific volatile organic compounds (VOCs) in the hive may be related to the occurrence of this bacterium in brood. Compositions of those volatile fractions present in healthy brood from control colonies and the brood without symptoms of infection collected from the colonies infected by P. larvae were compared using gas chromatography coupled with mass spectrometry (GC-MS) and solid phase microextraction (SPME). Among the seven compounds detected and quantified, the relative concentrations of 3-carene and limonene significantly differentiated the brood from healthy and infected colonies. Based on the ratio analysis, the samples were differentiated in terms of the number of emitted VOCs. Full article

The multitarget-directed ligands demonstrating affinity to histamine H3 receptor and additional cholinesterase inhibitory potency represent a promising strategy for research into the effective treatment of Alzheimer’s disease. In this study, a novel series of benzophenone derivatives was designed and synthesized. Among these derivatives, we identified compound 6 with a high affinity for H3R (Ki = 8 nM) and significant inhibitory activity toward BuChE (IC50 = 172 nM and 1.16 µM for eqBuChE and hBuChE, respectively). Further in vitro studies revealed that compound 6 (4-fluorophenyl) (4-((5-(piperidin-1-yl)pentyl)oxy)phenyl)methanone) displays moderate metabolic stability in mouse liver microsomes, good permeability with a permeability coefficient value (Pe) of 6.3 × 10−6 cm/s, and its safety was confirmed in terms of hepatotoxicity in the HepG2 cell line. Therefore, we investigated the in vivo activity of compound 6 in the Passive Avoidance Test and the Formalin Test. While compound 6 did not show a statistically significant influence on memory and learning, it showed analgesic properties in both acute (ED50 = 20.9 mg/kg) and inflammatory (ED50 = 17.5 mg/kg) pain. The article presented methods of urban development in terms of the application of the ‘sponge city’ concept, as well as the possibility of introducing different hydro-engineering solutions into the urban fabric that allow infiltration and retention at various scales of spatial planning. The aim of the paper was to indicate which specific solutions can be used in the city in multi-dimensional and multi-functional systems. As a result of the research, the concept of a ‘5-scales’ diffusion of blue-green infrastructure elements was presented. Elements of this system are based on multi-scale blue-green infrastructure, creating a patchwork of ‘blue connections’ that fit into the city ‘green’ natural system and have a connection with urban rainwater drainage. These five elements together allow for the infiltration and retention of rainwater, and can be used in the design of ecologically sustainable water-oriented cities in the future. Full article

Previously identified N-hexyl trazodone derivatives, 2-(6-(4-(3-chlorophenyl)piperazin-1-yl)hexyl)-[1,2,4]triazolo[4,3-a]pyridin-3(2H)-one hydrochloride (7a·HCl), with high affinity for 5-HT1AR and 2-(6-(4-([1,1′-biphenyl]-2-yl)piperazin-1-yl)hexyl)-[1,2,4]triazolo[4,3-a]pyridin-3(2H)-one hydrochloride (7b·HCl), a dual-acting 5-HT1A/5-HT7 receptor ligand, were prepared with a new microwave-assisted method. The protocol for the synthesis of 7a and 7b involved reductive alkylation under a mild reducing agent. We produced the final compounds with yield of 56–63% using ethanol or 51–56% in solvent-free conditions in 4 min. We then determined the 5-HT7R binding mode for compounds 7a and 7b using in silico methods and assessed the preliminary ADME and safety properties (hepatotoxicity and CYP3A4 inhibition) using in vitro methods for 7a·HCl and 7b·HCl. Furthermore, we evaluated antidepressant-like activity of the dual antagonist of 5-HT1A/5-HT7 receptors (7b·HCl) in the forced swim test (FST) in mice. The 5-HT1AR ligand (7a·HCl) with a much lower affinity for 5-HT7R compared to that of 7b·HCl was tested comparatively. Both compounds showed antidepressant activity, while 5-HT1A/5-HT7 double antagonist 7b·HCl showed a stronger and more specific response. Full article

In this study, we examined the fate and future of sewage irrigation fields; historic urban wetlands that served as sewer drainage before modern sewage treatment plants were built. Our aim in this study was to reappraise sewage irrigation fields in the urban fabric of modern cities and to analyse the possibility of re-integrating them into the ecosystem services system, as well as into green and blue infrastructure, providing leisure and recreational opportunities, stabilising the city’s biodiversity and microclimate, and increasing water retention in these areas. The research was based on the identification of the location of sewage irrigation fields in green and blue infrastructure systems, determination of the scale and extent of their connections to the urban fabric and an analysis of their multi-functionality including: ecological, climatic, hydrological, landscape, spatial, environmental, cultural and social, educational, and tourist and recreational functions. Probiotic microorganisms that are potentially beneficial to the health of the host are commercially available in a great variety of products. Not all microorganism strains present in products have proven beneficial to the health properties. These products include not only foodstuffs but also dietary supplements, food for special medical purposes, medicinal products, as well as cosmetics and medical devices. These products contain from one to a dozen bacterial strains of the same or different species and sometimes also fungal strains. Since the pro-health effects of probiotics depend on a specific strain, the number of its cells in a dose, and the lack of pathogenic microorganisms, it is extremely important to control the quality of probiotics. Depending on the classification of a given product, its form, and its content of microorganisms, the correct determination of the number of microorganisms and their identification is crucial. This article describes the culture-dependent and culture-independent methods for testing the contents of probiotic microorganisms, in addition to biochemical and genetic methods of identification. The microbiological purity requirements for various product categories are also presented. Due to numerous reports on the low quality of probiotic products available on the market, it is important to standardise research methods for this group of products and to increase the frequency of inspections of these products.
