Extended-spectrum cephalosporin-resistant Escherichia coli (ESC-EC) poses a significant public health concern, with its presence increasingly detected in healthy humans and various animal species. This study explores the transmission dynamic of ESC-EC within the Danish population as well as the transmission impact of a range of food and animal sources. We developed a compartmental model encompassing farmers, pet owners, and the general population. Additionally, we applied an established source attribution model to estimate the contributions to the transmission of different sources using Danish surveillance data on the distribution of resistance genes in E. coli. Our findings highlight the central role of human-to-human transmission while also showing the significant contributions of food and animal sources to the spread of ESC-EC in sporadic human infections. Imported food, pets, and livestock were estimated to contribute importantly to human infections. The results emphasize the complexity of ESC-EC transmission dynamics and the critical value of employing a One Health approach in modeling disease transmission and in the development of targeted intervention strategies. Jaguars play a crucial role in population control across multiple biomes. They are endangered and protected by in situ and ex situ conservation mechanisms to ensure their conservation. Cardiovascular diseases in wild mammals, including jaguars, often have unclear etiopathogenies, underscoring the need for research into novel hemodynamic parameters. This study evaluates the cardiovascular health of fifteen clinically healthy jaguars using conventional and Holter electrocardiography, non-invasive systemic blood pressure measurement, and echocardiography. Chemical restraint was achieved with medetomidine (0.08–0.1 mg/kg) and ketamine (5 mg/kg), with anesthesia reversed using atipamezole (0.25 mg/kg). The average heart rate was 72 ± 18 bpm, with sinus rhythm in ten animals and sinus arrhythmia in five. Six animals exhibited first and second-degree atrioventricular blocks, one had supraventricular complexes, and another had premature ventricular complexes. Non-invasive systolic blood pressure remained stable at 163 ± 29 mmHg during anesthesia. Echocardiographic examination revealed mitral, tricuspid, pulmonary, and aortic valve insufficiencies via color Doppler. The transmitral flow showed a normal E/A ratio and E` < A`, suggesting a pseudonormal ventricular filling pattern. No significant anesthetic complications were observed, affirming the protocol’s safety. This study provides valuable data, validating the anesthetic protocol and establishing reference cardiovascular values for jaguars, thus paving the way for future research in other veterinary species. Background: Transcranial Magnetic Stimulation–Electroencephalography (TMS-EEG) is a non-operative technique that allows for magnetic cortical stimulation (TMS) and analysis of the electrical currents generated in the brain (EEG). Despite the regular utilization of both techniques independently, little is known about the potential impact of their combination in neurosurgical practice. Methods: This scoping review, conducted following PRISMA guidelines, focused on TMS-EEG in epilepsy, neuro-oncology, and general neurosurgery. A literature search in Embase and Ovid MEDLINE returned 3596 records, which were screened based on predefined inclusion and exclusion criteria. After full-text review, three studies met the inclusion criteria. Two independent investigators conducted study selection and data extraction, with mediators resolving disagreements. The NHLBI tool was used to assess risk of bias in the included studies. Results: A total of 3596 articles were screened following the above-mentioned criteria: two articles and one abstract met the inclusion criteria. TMS-EEG is mentioned as a promising tool to evaluate tumor–brain interaction, improve preoperative speech mapping, and for lateralization epileptic focus in patients undergoing epilepsy surgery. Lack of detailed patient and outcome information preclude further considerations about TMS-EEG use beyond the potential applications of this technique. Conclusions: TMS-EEG research in neurosurgery is required to establish the role of this non-invasive brain stimulation-recording technique. Tumor–brain interaction, preoperative mapping, and seizure lateralization are in the front row for its future applications. Full article

While the wine industry has already adopted some sustainable and environmentally friendly practices, special attention should be paid to the greenness of the analytical methodologies used. In this study, a new tool called “Green Wine Analytical Procedure Evaluation” (GWAPE) was developed to fulfill these “green” requirements. This framework offers a structured approach to evaluating the environmental impact of wine analysis processes, covering all the analytical steps from sample collection to the final results. GWAPE provides quantitative information, uses schematic representations, assigns varying levels of importance to green chemistry principles, and conducts detailed evaluations of hazard structures. Since wine analytical methods typically involve fewer highly hazardous reagents, certain criteria previously applied in green analytical chemistry should be integrated or omitted. In summary, GWAPE offers a customized and precise solution to help the laboratories of enology and the wineries assess their analytical methodologies’ environmental impact. As an example of application, the proposed metric was used to evaluate the greenness of three different standard analytical methodologies to determine sugars in wine, showing good discrimination ability. This paper examines the influence of emotions on political polarization, looking at online propagation of conspiracy thinking by extreme right movements in Southern Europe. Integrating insights from psychology, political science, media studies, and system theory, we propose the ‘polarization loop’, a causal mechanism explaining the cyclical relationship between extreme messages, emotional engagement, media amplification, and societal polarization. We illustrate the utility of the polarization loop observing the use of the Great Replacement Theory by extreme right movements in Italy, Portugal, and Spain. We suggest possible options to mitigate the negative effects of online polarization in democracy, including public oversight of algorithmic decission-making, involving social science and humanities in algorithmic design, and strengthening resilience of citizenship to prevent emotional overflow. We encourage interdisciplinary research where historical analysis can guide computational methods such as Natural Language Processing (NLP), using Large Language Models fine-tunned consistently with political science research. Provided the intimate nature of emotions, the focus of connected research should remain on structural patterns rather than individual behavior, making it explicit that results derived from this research cannot be applied as the base for decisions, automated or not, that may affect individuals. The combination of cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) with endocrine therapy (ET) is the standard-of-care for estrogen receptor (ER)-positive, HER2-negative (ER+/HER2− advanced/metastatic breast cancer (mBC). However, the impact of CDK4/6i on circulating immune cells and circulating tumor cells (CTCs) in patients receiving CDK4/6i and ET (CDK4/6i+ET) remains poorly understood. This was a prospective cohort study including 44 patients with ER+/HER2− mBC treated with CDK4/6i+ET in either first or second line. Peripheral blood samples were collected before (baseline) and 3 months (t2) after therapy. Immune cell’s subsets were quantified by flow cytometry, and microfluidic-captured CTCs were counted and classified according to the expression of cytokeratin and/or vimentin. Patients were categorized according to response as responders (progression-free survival [PFS] ≥ 6.0 months; 79.1%) and non-responders (PFS < 6.0 months; 20.9%). CDK4/6i+ET resulted in significant changes in the hematological parameters, including decreased hemoglobin levels and increased mean corpuscular volume, as well as reductions in neutrophil, eosinophil, and basophil counts. Specific immune cell subsets, such as early-stage myeloid-derived suppressor cells, central memory CD4+ T cells, and Vδ2+ T cells expressing NKG2D, decreased 3 months after CDK4/6i+ET. Additionally, correlations between the presence of CTCs and immune cell populations were observed, highlighting the interplay between immune dysfunction and tumor dissemination. This study provides insights into the immunomodulatory effects of CDK4/6i+ET, underscoring the importance of considering immune dynamics in the management of ER+/HER2− mBC. Full article

The growing activity in the textile industry has been demanding the search for new and innovative technologies to meet consumers’ needs regarding more sustainable and ecological processes, with functionality receiving more attention. Bee products are known for their wide spectra of properties, including antioxidant and antibacterial activities. Propolis and honey are the most popular and used since ancient times for the most diverse applications due to their health benefits. With the increasing need for safer and more sustainable practices, the use of natural products for the functional finishing process can be a suitable alternative due to their safety and eco-friendly nature. For that, a biosolution, composed of a mixture of propolis and honey in water, was used to perform the functional finishing of cotton knits, both in the presence and in the absence of potassium alum as a chemical mordant. The fastness strength was also evaluated after three washing cycles. The antioxidant potential of the biosolution, assessed with the in vitro ABTS scavenging assay, provided textiles with the capacity to reduce more than 90% of the ABTS radical, regardless of the mordant presence and even after three washing cycles. Furthermore, biofunctional textiles decreased the growth of Bacillus subtilis, Propionibacterium acnes, Escherichia coli, and, particularly, Staphylococcus aureus cultures after 24 h of incubation with an increase in antibacterial activity when potassium alum was used. These findings show that bee products are promising and effective alternatives to be used in the textile industry to confer antioxidant and antibacterial properties to cotton textiles, thereby enhancing human health. Cadherins are cell–cell adhesion proteins which have been strongly implicated in cancer invasion, dissemination and metastasis capacity; thus, they are key players in the epithelial-to-mesenchymal transition (EMT) program. However, their role in glioblastoma (GBM), a primary central nervous system aggressive tumor, remains to be clarified. N-, E- and P-cadherin expression was analyzed on a large series of GBMs, characterized with clinical, imaging and neuropathological parameters, as well as with patients’ survival data. In addition, cadherins’ expression was studied in match-recurrent cases. Using TCGA data, cadherin expression profiles were also evaluated according to GBM transcription subtypes. N-cadherin expression was observed in 81.5% of GBM, followed by E-cadherin in 31% and P-cadherin in 20.8%. Upon tumor recurrence, P-cadherin was the only significantly upregulated cadherin compared with the primary tumor, being positive in 65.8% of the cases. Actually, P-cadherin gain was observed in 51.4% of matched primary-recurrent cases. Cadherins’ co-expression was also explored. Interestingly, E- and N-cadherin co-expression identified a GBM subgroup with frequent epithelial differentiation and a significant survival benefit. On the other hand, subgroups with P-cadherin expression carried the worse prognosis. P- and N-cadherin co-expression correlated with the presence of a mesenchymal phenotype. Expressions of isolated P-cadherin or E- and P-cadherin co-expression were associated with imaging characteristics of aggressiveness, to highly heterogeneous tumors, an d to worse patient survival. Classical cadherins co-expression subgroups present consistent clinical, imaging, neuropathological and survival differences, which probably reflect different states of an EMT-like program in GBM. Despite decades of literature on (de)hydration in healthy individuals, many unanswered questions remain. To outline research and policy priorities, it is fundamental to recognize the literature trends on (de)hydration and identify current research gaps, which herein we aimed to pinpoint. From a representative sample of 180 (de)hydration studies with 4350 individuals, we found that research is mainly limited to small-scale laboratory-based sample sizes, with high variability in demographics (sex, age, and level of competition); to non-ecological (highly simulated and controlled) conditions; and with a focus on recreationally active male adults (e.g., Tier 1, non-athletes). The laboratory-simulated environments are limiting factors underpinning the need to better translate scientific research into field studies. Although, consistently, dehydration is defined as the loss of 2% of body weight, the hydration status is estimated using a very heterogeneous range of parameters. Water is the most researched hydration fluid, followed by alcoholic beverages with added carbohydrates (CHO). The current research still overlooks beverages supplemented with proteins, amino acids (AA), and glycerol. Future research should invest more effort in “real-world” studies with larger and more heterogeneous cohorts, exploring the entire available spectrum of fluids while addressing hydration outcomes more harmoniously. Among the many lysosomal storage disorders (LSDs) that would benefit from the establishment of novel cell models, either patient-derived or genetically engineered, is mucopolysaccharidosis type II (MPS II). Here, we present our results on the establishment and characterization of two MPS II patient-derived stem cell line(s) from deciduous baby teeth. To the best of our knowledge, this is the first time a stem cell population has been isolated from LSD patient samples obtained from the dental pulp. Taking into account our results on the molecular and biochemical characterization of those cells and the fact that they exhibit visible and measurable disease phenotypes, we consider these cells may qualify as a valuable disease model, which may be useful for both pathophysiological assessments and in vitro screenings. Ultimately, we believe that patient-derived dental pulp stem cells (DPSCs), particularly those isolated from human exfoliated deciduous teeth (SHEDs), may represent a feasible alternative to induced pluripotent stem cells (iPSCs) in many labs with standard cell culture conditions and limited (human and economic) resources. Source Attribution of Antibiotic Resistance Genes in Estuarine Aquaculture: A Machine Learning Approach

Aquaculture located in urban river estuaries, where other anthropogenic activities may occur, has an impact on and may be affected by the environment where they are inserted, namely by the exchange of antimicrobial resistance genes. The latter may ultimately, through the food chain, represent a source of resistance genes to the human resistome. In an exploratory study of the presence of resistance genes in aquaculture sediments located in urban river estuaries, two machine learning models were applied to predict the source of 34 resistome observations in the aquaculture sediments of oysters and gilt-head sea bream, located in the estuaries of the Sado and Lima Rivers and in the Aveiro Lagoon, as well as in the sediments of the Tejo River estuary, where Japanese clams and mussels are collected. The first model included all 34 resistomes, amounting to 53 different antimicrobial resistance genes used as source predictors. The most important antimicrobial genes for source attribution were tetracycline resistance genes tet(51) and tet(L); aminoglycoside resistance gene aadA6; beta-lactam resistance gene blaBRO-2; and amphenicol resistance gene cmx_1. The second model included only oyster sediment resistomes, amounting to 30 antimicrobial resistance genes as predictors. The most important antimicrobial genes for source attribution were the aminoglycoside resistance gene aadA6, followed by the tetracycline genes tet(L) and tet(33). This exploratory study provides the first information about antimicrobial resistance genes in intensive and semi-intensive aquaculture in Portugal, helping to recognize the importance of environmental control to maintain the integrity and the sustainability of aquaculture farms. The NEET phenomenon in southern Europe is particularly alarming. Most studies have focused on analysing the socio-demographic characteristics of NEET and policies directed towards them, but more research is needed regarding the psychological aspects that underlie this condition. This paper, grounded in the bioecological model, aims to analyse the psychological dimensions regarding the functioning of Public Employment Services (PESs) in three countries that rely on the South Mediterranean welfare model. Between November 2022 and September 2023, research was conducted across six rural PES locations in Italy, Spain, and Portugal, based on interviews and focus groups with PES operators and young NEET users. Thematic analysis identified two central axes: Enhancing Relationships and Building Strategic Action, which were crucial for improving NEET-PES interactions and fostering connections between PES and other community entities. The results highlighted the importance of psychological elements in the effective involvement of NEETs in policy initiatives. Full article

Background: There has been a growing interest in environmental education programs to improve environmental awareness and behavior change among school-aged youth. Yet, assessment of the approaches aligned with citizen science principles emphasizing community participation and empowerment is scarce. The present study aimed to explore the acceptability of the “ECOCIDADANIA” environmental education program targeted at youth in rural areas. Methods: The program was applied for two years in the municipality of Gouveia, and it involved nature walks methodologies combined with citizen science. A total of 107 students participated in the program. Eighteen participants (13 youth and 5 teachers) were selected as key informants, and semistructured interviews were conducted to assess the appropriateness of the intervention and the perceived benefits. Results: The program was demonstrated to be acceptable to youth and teachers, and its benefits included increasing students’ knowledge, skills and self-efficacy, discovering the territory, promoting environmental awareness, strengthening social relationships between peers and educators, and contributing to the promotion of individual and collective well-being. All the participants were satisfied with the program’s activities and recommend its continuation in the school context. Conclusions: Programs of this nature could foster environmental activism and enhance environmental sustainability at the local level, which reinforces the entrepreneurship profile of the youth. Pancreas Rejection in the Artificial Intelligence Era: New Tool for Signal Patients at Risk

Introduction: Pancreas transplantation is currently the only treatment that can re-establish normal endocrine pancreatic function. Despite all efforts, pancreas allograft survival and rejection remain major clinical problems. The purpose of this study was to identify features that could signal patients at risk of pancreas allograft rejection. Methods: We collected 74 features from 79 patients who underwent simultaneous pancreas–kidney transplantation (SPK) and used two widely-applicable classification methods, the Naive Bayesian Classifier and Support Vector Machine, to build predictive models. We used the area under the receiver operating characteristic curve and classification accuracy to evaluate the predictive performance via leave-one-out cross-validation. Results: Rejection events were identified in 13 SPK patients (17.8%). In feature selection approach, it was possible to identify 10 features, namely: previous treatment for diabetes mellitus with long-term Insulin (U/I/day), type of dialysis (peritoneal dialysis, hemodialysis, or pre-emptive), de novo DSA, vPRA_Pre-Transplant (%), donor blood glucose, pancreas donor risk index (pDRI), recipient height, dialysis time (days), warm ischemia (minutes), recipient of intensive care (days). The results showed that the Naive Bayes and Support Vector Machine classifiers prediction performed very well, with an AUROC and classification accuracy of 0.97 and 0.87, respectively, in the first model and 0.96 and 0.94 in the second model. Conclusion: Our results indicated that it is feasible to develop successful classifiers for the prediction of graft rejection. The Naive Bayesian generated nomogram can be used for rejection probability prediction, thus supporting clinical decision making. Full article

Artificial intelligence and machine learning have been increasingly used in the medical imaging field in the past few years. The evaluation of medical images is very subjective and complex, and therefore the application of artificial intelligence and deep learning methods to automatize the analysis process would be very beneficial. A lot of researchers have been applying these methods to image analysis diagnosis, developing software capable of assisting veterinary doctors or radiologists in their daily practice. This article details the main methodologies used to develop software applications on machine learning and how veterinarians with an interest in this field can benefit from such methodologies. The main goal of this study is to offer veterinary professionals a simple guide to enable them to understand the basics of artificial intelligence and machine learning and the concepts such as deep learning, convolutional neural networks, transfer learning, and the performance evaluation method. The language is adapted for medical technicians, and the work already published in this field is reviewed for application in the imaging diagnosis of different animal body systems: musculoskeletal, thoracic, nervous, and abdominal.
